We have investigated if changes in hepatic lipid metabolism produced by old age are related to changes in liver peroxisome proliferator-activated receptor alpha (PPARalpha). Our results indicate that 18-month-old rats showed a marked decrease in the expression and activity of liver PPARalpha, as shown by significant reductions in PPARalpha mRNA, protein and binding activity, resulting in a reduction in the relative mRNA levels of PPARalpha target genes, such as liver-carnitine-palmitoyl transferase-I (CPT-I) and mitochondrial medium-chain acyl-CoA dehydrogenase (MCAD). Further, in accordance with a liver PPARalpha deficiency in old rats, treatment of old animals with a therapeutic dose of gemfibrozil (GFB) (3mg/kg per day, 21 days) was ineffective in reducing plasma triglyceride concentrations (TG), despite attaining a 50% reduction in TG when GFB was administered to young animals at the same dose and length of treatment. We hypothesize that the decrease in hepatic PPARalpha can be related to a state of leptin resistance present in old animals.

Abstract : Post-translational modifications such as proteolytic processing, phosphorylation, and glycosylation, add extra layers of complexity to proteomes and allow a finely tuned regulation of the activity of many proteins. The evolutionarily conserved cell-cycle and transcriptional regulator HCP-] is regulated by proteolytic maturation via which a stable heterodirneric complex of two cleaved subunits is formed from a single precursor protein. The human HCF-1 precursor is cleaved at six nearly identical 26 amino acid sequence repeats, called HCF-1pro repeats, which represent uncommon protease recognition sites dedicated to human HCF-1 proteolysis. This proteolytic maturation process is conserved in vertebrate HCF-1 homologues and is essential for the functions of the human protein in cell-cycle regulation; the mechanisms that execute and control HCF-1 proteolysis, however, remain poorly understood. In this dissertation I investigate the mechanisms of proteolytic maturation of HCF-1 proteins in different species. I show that the Drosophila homolog of human HCF-1, called dHCP, is proteolytically cleaved via a different mechanism than human HCF-1. dHCP is processed by the same protease, called Taspase], which cleaves one of the key developmental regulators in flies, the Trithorax protein. Maturation of HCP proteins via Taspase] cleavage is probably not particular to dHCP as many invertebrate HCP proteins, particularly insects and flatworms, possess Taspase] recognition sites. In contrast, the vertebrate HCF-1 proteins lack Taspase] recognition sites and the HCF-1pro repeats are not Taspase1 substrates, suggesting that multiple mechanisms for HCF-1 proteolytic maturation have appeared during evolution. I also show that the proteolytic activity responsible for the cleavage of the HCP- 1pro repeats is very difficult to characterize, being resistant to most protease inhibitors and very sensitive to biochemical fractionation. Moreover, the HCF-1pro repeats represent complex protease recognition sites and I demonstrate that, in addition to be the HCF-1 cleavage sites, these repeated sequences, also recruit the OG1cNAc transferase OGT. The OGT protein and the OG1cNAc modification of HCF-1 are both important for HCF-1pro repeat proteolysis. Interestingly, a human recombinant OGT purified from insect cells is able to induce cleavage of a HCF-1pro-repeat precursor in vitro, indicating that OGT either (i) induces HCF-1 autoproteolysis,(ii) is the HCF-1pro- repeat proteolytic activity itself, or (iii) physically associates with a proteolytic activity that is conserved in insect cells. In any case, OGT plays an important role in HCF-1 proteolytic maturation and perhaps a broader role in HCF-1 biological function. Résumé : Les modifications post-traductionelles pomme le clivage protéolytique, la phosphorylation, et la glycosylation, augmentent significativement la complexité des protéomes et permettent une régulation fine de l'activité de beaucoup de protéines. La protéine HCF-1, qui est un régulateur du cycle cellulaire et de la transcription, est elle- même régulée par clivage protéolytique. La protéine HCF-1 est en effet coupée en deux sous-unités qui s'associent l'une a l'autre pour former la protéine mature. Le précurseur de la protéine HCF-1 humaine est clivé à six sites correspondant à six séquences répétées nommées les HCF-1pro repeats, chacune composée de 26 acide aminés. Les HCF-1pro- repeats ne ressemblent ai aucune séquence de clivage protéolytique connue et sont présentes seulement dans les protéines HCF-1 chez les vertébrés. Bien que la maturation protéolytique d'HCF-1 soit essentielle pour les activités de cette protéine pendant le cycle cellulaire, les mécanismes qui la contrôlent restent inconnus. Au cours de mon travail de thèse, j'ai analysé les mécanismes de clivage protéolytique des protéines HCF dans différentes espèces. J'ai montré que la protéine de Drosophile homologue d'HCF-1 humaine nommée dHCF est clivée par une protéase nommée Taspase1. Ainsi, dHCF est clivé par la même protéase que celle qui induit la maturation protéolytique d'un des principaux facteurs du développement chez la mouche, la protéine Trithorax. La maturation de dHCF via le clivage par la Taspase1 n'est pas spécifique à la mouche, mais est probablement étendu à plusieurs protéines HCF chez les invertébrés, surtout dans les familles des insectes et des plathehninthes, car ces protéines HCF présentent des sites de reconnaissance pour la Taspasel. Par contre, les protéines HCF-1 chez les vertébrés n'ont pas de sites de reconnaissance pour la Taspasel et cela suggère que différents mécanismes de maturation des protéines HCF- ls ont apparu au cours de l'évolution. J'ai montré aussi que les HCF-1pro-repeats sont clivés par une activité protéolytique très difficile a identifier, car elle est résistante à la plupart des inhibiteurs de protéases, mais elle est très sensible au fractionnement biochimique. En plus, les HCF-1pro-repeats sont un site de protéolyse complexe qui ne sert pas seulement au clivage des protéines HCF- chez les vertébrés mais aussi à recruter l'enzyme responsable de la O- GlcNAcylation nommée OGT. La protéine OGT et la O-GlcNAcylatio d'HCF-1 sont toutes les deux importantes pour le clivage protéolytique des HCF1pro-repeats. Curieusement, la protéine OGT humaine produite dans des cellules d'insectes est capable de cliver les HCF-1pro repeats in vitro et cela suggère que OGT soit (i) induit le clivage autocatalytique cl'HCF-1, soit (ii) est elle-même l'activité protéolytique qui clive HCF4, soit (iii) est associée à une activité protéolytique conservée dans les cellules d'insectes qui a été co-purifiée avec OGT. En conclusion, OGT joue un rôle important dans la maturation protéolytique d'HCF-1 et peut-être aussi un rôle plus large dans les fonctions biologiques de la protéine HCF-1.

Sphingomonas paucimobilis B90A contains two variants, LinA1 and LinA2, of a dehydrochlorinase that catalyzes the first and second steps in the metabolism of hexachlorocyclohexanes (R. Kumari, S. Subudhi, M. Suar, G. Dhingra, V. Raina, C. Dogra, S. Lal, J. R. van der Meer, C. Holliger, and R. Lal, Appl. Environ. Microbiol. 68:6021-6028, 2002). On the amino acid level, LinA1 and LinA2 were 88% identical to each other, and LinA2 was 100% identical to LinA of S. paucimobilis UT26. Incubation of chiral alpha-hexachlorocyclohexane (alpha-HCH) with Escherichia coli BL21 expressing functional LinA1 and LinA2 S-glutathione transferase fusion proteins showed that LinA1 preferentially converted the (+) enantiomer, whereas LinA2 preferred the (-) enantiomer. Concurrent formation and subsequent dissipation of beta-pentachlorocyclohexene enantiomers was also observed in these experiments, indicating that there was enantioselective formation and/or dissipation of these enantiomers. LinA1 preferentially formed (3S,4S,5R,6R)-1,3,4,5,6-pentachlorocyclohexene, and LinA2 preferentially formed (3R,4R,5S,6S)-1,3,4,5,6-pentachlorocyclohexene. Because enantioselectivity was not observed in incubations with whole cells of S. paucimobilis B90A, we concluded that LinA1 and LinA2 are equally active in this organism. The enantioselective transformation of chiral alpha-HCH by LinA1 and LinA2 provides the first evidence of the molecular basis for the changed enantiomer composition of alpha-HCH in many natural environments. Enantioselective degradation may be one of the key processes determining enantiomer composition, especially when strains that contain only one of the linA genes, such as S. paucimobilis UT26, prevail.

Over the past decade a series of trials of the EORTC Brain Tumor Group (BTG) has substantially influenced and shaped the standard-of-care of primary brain tumors. All these trials were coupled with biological research that has allowed for better understanding of the biology of these tumors. In glioblastoma, EORTC trial 26981/22981 conducted jointly with the National Cancer Institute of Canada Clinical Trials Group showed superiority of concomitant radiochemotherapy with temozolomide over radiotherapy alone. It also identified the first predictive marker for benefit from alkylating agent chemotherapy in glioblastoma, the methylation of the O6-methyl-guanyl-methly-transferase (MGMT) gene promoter. In another large randomized trial, EORTC 26951, adjuvant chemotherapy in anaplastic oligodendroglial tumors was investigated. Despite an improvement in progression-free survival this did not translate into a survival benefit. The third example of a landmark trial is the EORTC 22845 trial. This trial led by the EORTC Radiation Oncology Group forms the basis for an expectative approach to patients with low-grade glioma, as early radiotherapy indeed prolongs time to tumor progression but with no benefit in overall survival. This trial is the key reference in deciding at what time in their disease adult patients with low-grade glioma should be irradiated. Future initiatives will continue to focus on the conduct of controlled trials, rational academic drug development as well as systematic evaluation of tumor tissue including biomarker development for personalized therapy. Important lessons learned in neurooncology are to dare to ask real questions rather than merely rapidly testing new compounds, and the value of well designed trials, including the presence of controls, central pathology review, strict radiology protocols and biobanking. Structurally, the EORTC BTG has evolved into a multidisciplinary group with strong transatlantic alliances. It has contributed to the maturation of neurooncology within the oncological sciences.

In order to identify useful parameters for maize genetic breeding programs aiming at a more efficient use of N, two maize varieties of contrasting N efficiency, Sol da Manhã NF (efficient) and Catetão (inefficient) were compared. Experiments were carried out under field and greenhouse conditions, at low and high N levels. The parameters analysed included total and relative plant and grain N content, biomass and the activities of nitrate reductase and glutamine synthetase in different parts of the plant. It was found that the translocation efficiency of N and photoassimilates to the developing seeds and the source-sink relations were significantly different for the two varieties. N content of the whole plant and grain, cob weight and the relative ear dry weight were useful parameters for characterizing the variety Sol da Manhã NF as to its efficient use of N. Enzymes activity of glutamine synthetase (transferase reaction) and nitrate reductase did not differ among the varieties.

The management of gliomas in daily clinical practice is challenging. It requires a multidisciplinary and coordinated approach involving neurosurgery, radiotherapy and, finally, chemotherapy. Important progress has been made during the last years with the introduction of a combined treatment associating standard radiotherapy with concomitant chemotherapy using temozolomide, a novel alkylating agent. For the first time in many years a new treatment strategy translated into a significant prolongation of survival. In parallel, molecular markers (e.g. loss of heterozygosity on chromosomes 1p and 19q or methylation of the methyl-guanine methyl transferase [MGMT] gene promoter) allowed for identification of distinct subtypes of glioma or prediction of treatment response. In this "Practical Guide", we describe the daily practice and aim at answering some common questions in the management of patients suffering from glioblastoma, astrocytoma, oligodendroglioma and low grade glioma. The therapeutic options presented here are based on evidences from the literature. In the absence of documented evidence, the empirical choices from our local practice are explained and justified.

ButsDans la littérature actuelle, peu d'études existent sur la relation entre la consommation d'alcool et le syndrome métabolique. Les quelques données disponibles sont contradictoires et très limitées chez les buveurs à haut risque. Quant au diabète, une association est connue entre la consommation à bas risque d'alcool et une prévalence diminuée de la maladie. Là encore, les données sur la consommation à haut risque sont très limitées. Par conséquent, notre but était d'étudier la relation entre la consommation d'alcool, le syndrome métabolique et le diabète dans la cohorte lausannoise (CoLaus), où la consommation moyenne d'alcool est nettement plus élevée que dans la plupart des études disponibles, notamment celles des États-Unis.MéthodesNous avons analysé les données de 6172 hommes et femmes, âgés de 35 à 75 ans. La consommation d'alcool a été catégorisée en 0,1-6, 7-13, 14-20, 21-27, 28-34 et >35 boissons par semaine ou comme non-buveurs (0), buveurs à bas risque (1-13), à risque moyen à élevé (14-34) et à très haut risque (>35). Nous avons confirmé la consommation d'alcool par la y- glutamyl transferase et la transferrine déficiente en hydrates de carbone (CDT). Après l'analyse des caractéristiques des groupes de consommateurs, nous avons utilisé des régressions multivariées pour évaluer la relation entre la consommation d'alcool, la prévalence du syndrome métabolique et du diabète ainsi que la résistance à l'insuline, déterminée par le modèle d'homéostasie de la résistance à l'insuline (HOMA-IR). Dans le modèle d'ajustement, nous avons inclus l'âge, le genre, le status tabagique, l'activité physique et le niveau de formation. Nous avons aussi comparé la relation du type d'alcool (vin, bière et spiritueux) avec le syndrome métabolique, le diabète et le HOMA-IR en testant l'hypothèse d'égalité de leurs coefficients de régression, après ajustement.RésultatsParmi les participants, 73% buvaient de l'alcool, 16% étant buveurs à risque moyen à élevé et 2% à risque très élevé. En analyse multivariée, la prévalence du syndrome métabolique et du diabète ainsi que le HOMA-IR moyen diminuaient avec la consommation d'alcool à bas risque et augmentaient avec la consommation à très haut risque, montrant une relation en U. La prévalence ajustée du syndrome métabolique était de 24% chez les non-buveurs, 19% chez les buveurs à bas risque (p<0.001 vs. non-buveurs), 20% chez ceux à risque moyen à élevé et 29% chez ceux à très haut risque (p=0.005 vs. bas risque). La prévalence ajustée du diabète était de 6.0% chez les non-buveurs, 3.6% chez les buveurs à bas risque (p<0.001 vs. non-buveurs), 3.8% chez ceux à risque moyen à élevé et 6.7% chez ceux à très haut risque (p=0.046 vs. bas risque). Le HOMA-IR moyen ajusté était de 2.47 chez les non-buveurs, 2.14 chez ceux à bas risque (pcO.OOl vs. non-buveurs), 2.27 chez ceux à risque moyen à élevé et 2.53 chez ceux à très haut risque (p=0.04 vs. bas risque). Ces relations ne différaient pas selon les types de boissons.ConclusionsLa prévalence du syndrome métabolique, du diabète et le HOMA-IR baissent pour les faibles consommations d'alcool, mais augmentent à nouveau avec les plus fortes consommations, sans différence entre les types de boissons.

O objetivo deste trabalho foi avaliar as concentrações séricas de minerais e as funções hepática e renal de frangos de corte intoxicados, experimentalmente, com 3 ppm de aflatoxina, e submetidos a diferentes concentrações de montmorilonita sódica na dieta. Foram utilizados 720 frangos, machos, da linhagem Cobb, divididos em seis tratamentos: T1, dieta normal; T2, dieta com aflatoxina (3 ppm); T3, dieta com montmorilonita sódica (0,25%); T4, dieta com aflatoxina (3 ppm) + montmorilonita sódica (0,25%); T5, dieta com montmorilonita sódica (0,5%); T6, dieta com aflatoxina (3 ppm) + montmorilonita sódica (0,5%), com seis repetições por tratamento. A dieta com 3 ppm de aflatoxina (T2) resultou em uma diminuição significativa na concentração sérica de proteínas totais, albumina, globulinas e aspartato amino transferase; a concentração sérica de ácido úrico diminuiu, significativamente, na dieta com aflatoxina (3 ppm) + momtmorilonita sódica a 0,25%; ocorreu diminuição significativa nas concentrações séricas de fósforo na dieta com aflatoxina + montmorilonita sódica a 0,5%. A aflatoxina na concentração de 3 ppm altera a função hepática de frangos de corte; o uso da montmorilonita sódica, na concentração de 0,5%, é eficaz na prevenção dos efeitos tóxicos da aflatoxina, mas diminui os níveis séricos de fósforo.

Background: Pharmacogenetic studies are essential in understanding the interindividual variability of drug responses. DNA sample collection for genotyping is a critical step in genetic studies. A method using dried blood samples from finger-puncture, collected on DNA-cards, has been described as an alternative to the usual venepuncture technique. The purpose of this study is to evaluate the implementation of the DNA cards method in a multicentre clinical trial, and to assess the degree of investigators' satisfaction and the acceptance of the patients perceived by the investigators.Methods: Blood samples were collected on DNA-cards. The quality and quantity of DNA recovered were analyzed. Investigators were questioned regarding their general interest, previous experience, safety issues, preferences and perceived patient satisfaction. Results: 151 patients' blood samples were collected. Genotyping of GST polymorphisms was achieved in all samples (100%). 28 investigators completed the survey. Investigators perceived patient satisfaction as very good (60.7%) or good (39.3%), without reluctance to finger puncture. Investigators preferred this method, which was considered safer and better than the usual methods. All investigators would recommend using it in future genetic studies. Conclusion: Within the clinical trial setting, the DNA-cards method was very well accepted by investigators and patients (in perception of investigators), and was preferred to conventional methods due to its ease of use and safety.

O objetivo deste trabalho foi avaliar a assimilação de enxofre elementar (S0), aplicado nas folhas de soja, e sua eficiência comparada à adubação feita ao solo, de acordo com a dose e a natureza da fonte do nutriente. O S0 aplicado às folhas, independentemente da dose e fonte, foi assimilado pela planta, o que acarretou em aumento no teor de proteína total na folha. Todas as fontes de S aplicadas às folhas aumentaram a produção de grãos, semelhantemente à aplicação ao solo. Observou-se uma mesma produtividade com o uso de 20 kg ha-1 de S0 no solo ou de 6 kg ha-1 via foliar. A eficiência da aplicação de S via foliar, com base no conteúdo de proteína solúvel total, foi superior à da aplicação ao solo.

O objetivo deste trabalho foi avaliar os minerais séricos e as funções hepática e renal de frangos de corte, alimentados com dietas com aflatoxinas e dietas com aflatoxinas e argila clinoptilolita natural. Foram utilizados 528 frangos de corte machos, da linhagem Ross, distribuídos em seis tratamentos com quatro repetições cada: T1 - testemunha: ração sem aflatoxina ou clinoptilolita; T2 - ração com 5 ppm de aflatoxinas; T3 - ração com 0,25% de clinoptilolita; T4 - ração com 5 ppm de aflatoxinas e 0,25% de clinoptilolita; T5 - ração com 0,5% de clinoptilolita; e T6 - ração com 5 ppm de aflatoxinas e 0,5% de clinoptilolita. As aves foram submetidas aos tratamentos do 1º ao 42º dia de idade. Foram abatidos 72 animais, e foram analisadas as concentrações séricas de cálcio, fósforo, uréia, creatinina, ácido úrico, colesterol, triglicerídeos e gamaglutamil-transferase (GGT). As aflatoxinas diminuem o nível sérico de colesterol. A clinoptilolita diminui o nível sérico do ácido úrico, quando na concentração de 0,25%. As aflatoxinas com 0,25% de clinoptilolita diminuem os níveis séricos da creatinina, do ácido úrico e do colesterol. As aflatoxinas acrescidas por 0,5% de clinoptilolita diminuem os níveis séricos da creatinina e do colesterol e elevam os de gamaglutamil-transferase.

The objective of this work was to develop a genetic transformation system for tropical maize genotypes via particle bombardment of immature zygotic embryos. Particle bombardment was carried out using a genetic construct with bar and uidA genes under control of CaMV35S promoter. The best conditions to transform maize tropical inbred lines L3 and L1345 were obtained when immature embryos were cultivated, prior to the bombardment, in higher osmolarity during 4 hours and bombarded at an acceleration helium gas pressure of 1,100 psi, two shots per plate, and a microcarrier flying distance of 6.6 cm. Transformation frequencies obtained using these conditions ranged from 0.9 to 2.31%. Integration of foreign genes into the genome of maize plants was confirmed by Southern blot analysis as well as bar and uidA gene expressions. The maize genetic transformation protocol developed in this work will possibly improve the efficiency to produce new transgenic tropical maize lines expressing desirable agronomic characteristics.

O objetivo deste trabalho foi avaliar os efeitos da suplementação da dieta de tambacus com concentrações de vitamina E sobre variáveis de desempenho e hematológicas, bem como determinar a relação da atividade da glutationa peroxidase com a inclusão de vitamina E e selênio na dieta. Foram utilizados 250 juvenis de tambacu, divididos em 25 aquários. A dieta basal constituiu-se de ração peletizada com 32% de proteína bruta e 3.300 kcal kg-1 de energia digestível, e inclusão de 0,40 mg kg-1 de Se. As dietas-teste - isoprotéicas e isoenergéticas - foram compostas da dieta basal com diferentes concentrações de vitamina E (0, 100, 200, 300, 400 mg kg-1), em delineamento inteiramente ao acaso, com cinco tratamentos e cinco repetições. Não foi observado efeito significativo da suplementação com vitamina E sobre o comprimento total, volume corpuscular médio, concentração de hemoglobina corpuscular média e número de eritrócitos, nem sobre a atividade da glutationa peroxidase. Peso final e conversão alimentar aparente, no entanto, sofreram efeito dos tratamentos suplementados. O teor de 400 mg kg-1 de vitamina E melhorou o comprimento padrão e ganho de peso. A suplementação da dieta com vitamina E resulta em melhor desempenho produtivo de tambacus.

Using the yeast two-hybrid system, we identified the mu 2 subunit of the clathrin adaptor complex 2 as a protein interacting with the C-tail of the alpha 1b-adrenergic receptor (AR). Direct association between the alpha 1b-AR and mu 2 was demonstrated using a solid phase overlay assay. The alpha 1b-AR/mu 2 interaction occurred inside the cells, as shown by the finding that the transfected alpha 1b-AR and the endogenous mu 2 could be coimmunoprecipitated from HEK-293 cell extracts. Mutational analysis of the alpha 1b-AR revealed that the binding site for mu 2 does not involve canonical YXX Phi or dileucine motifs but a stretch of eight arginines on the receptor C-tail. The binding domain of mu 2 for the receptor C-tail involves both its N terminus and the subdomain B of its C-terminal portion. The alpha 1b-AR specifically interacted with mu 2, but not with the mu 1, mu 3, or mu 4 subunits belonging to other AP complexes. The deletion of the mu 2 binding site in the C-tail markedly decreased agonist-induced receptor internalization as demonstrated by confocal microscopy as well as by the results of a surface receptor biotinylation assay. The direct association of the adaptor complex 2 with a G protein-coupled receptor has not been reported so far and might represent a common mechanism underlying clathrin-mediated receptor endocytosis.

The biosynthetic genes pchDCBA and pchEF, which are known to be required for the formation of the siderophore pyochelin and its precursors salicylate and dihydroaeruginoate (Dha), are clustered with the pchR regulatory gene on the chromosome of Pseudomonas aeruginosa. The 4.6-kb region located downstream of the pchEF genes was found to contain three additional, contiguous genes, pchG, pchH, and pchI, probably forming a pchEFGHI operon. The deduced amino acid sequences of PchH and PchI are similar to those of ATP binding cassette transport proteins with an export function. PchG is a homolog of the Yersinia pestis and Y. enterocolitica proteins YbtU and Irp3, which are involved in the biosynthesis of yersiniabactin. A null mutation in pchG abolished pyochelin formation, whereas mutations in pchH and pchI did not affect the amounts of salicylate, Dha, and pyochelin produced. The pyochelin biosynthetic genes were expressed from a vector promoter, uncoupling them from Fur-mediated repression by iron and PchR-dependent induction by pyochelin. In a P. aeruginosa mutant lacking the entire pyochelin biosynthetic gene cluster, the expressed pchDCBA and pchEFG genes were sufficient for salicylate, Dha, and pyochelin production. Pyochelin formation was also obtained in the heterologous host Escherichia coli expressing pchDCBA and pchEFG together with the E. coli entD gene, which provides a phosphopantetheinyl transferase necessary for PchE and PchF activation. The PchG protein was purified and used in combination with PchD and phosphopantetheinylated PchE and PchF in vitro to produce pyochelin from salicylate, L-cysteine, ATP, NADPH, and S-adenosylmethionine. Based on this assay, a reductase function was attributed to PchG. In summary, this study completes the identification of the biosynthetic genes required for pyochelin formation from chorismate in P. aeruginosa.
