Metarhizium is a soil-inhabiting fungus currently used as a biological control agent against various insect species, and research efforts are typically focused on its ability to kill insects. In section 1, we tested the hypothesis that species of Metarhizium are not randomly distributed in soils but show plant rhizosphere-specific associations. Results indicated an association of three Metarhizium species (Metarhizium robertsii, M. brunneum and M. guizhouense) with the rhizosphere of certain types of plant species. M. robertsii was the only species that was found associated with grass roots, suggesting a possible exclusion of M. brunneum and M. guizhouense, which was supported by in vitro experiments with grass root exudate. M. guizhouense and M. brunneum only associated with wildflower rhizosphere when co-occurring with M. robertsii. With the exception of these co-occurrences, M. guizhouense was found to associate exclusively with the rhizosphere of tree species, while M. brunneum was found to associate exclusively with the rhizosphere of shrubs and trees. These associations demonstrate that different species of Metarhizium associate with specific plant types. In section 2, we explored the variation in the insect adhesin, Madl, and the plant adhesin, Mad2, in fourteen isolates of Metarhizium representing seven different species. Analysis of the transcriptional elements within the Mad2 promoter region revealed variable STRE, PDS, degenerative TATA box, and TATA box-like regions. Phylogenetic analysis of 5' EF-Ia, which is used for species identification, as well as Madl and Mad2 sequences demonstrated that the Mad2 phylogeny is more congruent with 5' EF-1a than Madl. This suggests Mad2 has diverged among Metarhizium lineages, contributing to clade- and species-specific variation. While other abiotic and biotic factors cannot be excluded in contributing to divergence, it appears that plant associations have been the driving factor causing divergence among Metarhizium species.

Here we wanted to explore whether there was more variation in genes devoted to plant association (Mad2) or to insect association (Mad1) overall in the genus Metarhizium. Greater divergence within the genus Metarhizium in one of these genes may provide evidence for whether host insect or plant is a driving force in adaptation and evolution in the genus Metarhizium. We compared differences in variation in the insect adhesin gene, Mad1, which enables attachment to insect cuticle, and the plant adhesin gene, Mad2, which enables attachment to plants. Overall variation for the Mad1 promoter region (7.1%), Mad1 open reading frame (6.7%), and Mad2 open reading frame (7.4%) were similar, while it was higher in the Mad2 promoter region (9.9%). Analysis of the transcriptional elements within the Mad2 promoter region revealed variable STRE, PDS, degenerative TATA box, and TATA box-like regions, while this level of variation was not found for Mad1. Sequences were also phylogenetically compared to EF-1a, which is used for species identification, in 14 isolates representing 7 different species in the genus Metarhizium. Introduction: L’arthrose est une maladie fréquente chez le chien. Même si des traitements palliatifs ont prouvé leur efficacité pour soulager les signes cliniques associés, aucun n’a permis de restaurer la structure des cartilages ou la fonction des patients. De nouvelles modalités de traitement sont proposées mais très peu d’évidences existent quant à leur efficacité. Un problème récurrent pour apporter plus d’évidence est le manque d’outils d’évaluation dûment validés. Objectif: Evaluer l’efficacité d’une diète contenant de la moule verte, ainsi que la validité de différentes méthodes pour évaluer les effets de ce traitement sur la condition arthrosique de chiens de propriétaire. Méthodes: Une étude prospective croisée contrôlée en aveugle a été conduite sur 30 chiens de propriétaires. Leur condition était stable et non traitée. Ils ont reçu une diète contrôle pendant 30 jours puis la diète thérapeutique pendant 60 jours. A J0 (inclusion), J30 (fin de la période contrôle) et J90 (fin du traitement), les concentrations plasmatiques en acides gras ω–3 (EPA et DHA), le pic de force verticale (PVF – critère de référence), un questionnaire multifactoriel standardisé (MFQ), et la conductance électrodermique (EDA) ont été mesurés. Deux fois par semaine, les propriétaires remplissaient un questionnaire sur la fonction spécifique à leur animal (CODI). L’activité locomotrice (actimétrie) a été enregistrée en continu sur 7 chiens. Résultats: Aucun effet secondaire n’a été documenté excepté une augmentation du poids des chiens durant la période traitement. La localisation -thoracique ou pelvienne- du membre étudié et le poids ont interagi avec PVF. Les taux d’EPA et DHA ont augmenté durant la période traitement (p=0.026 et p<0.001, respectivement), tout comme PVF corrigé pour l’effet du poids et de la localisation du membre (p=0.003). La validité technique de PVF a été vérifiée. La validité technique et selon-critère du CODI ont été vérifiées. MFQ n’a pas montré d’homogénéité suffisante entre ses différents composants, bien que les résultats du composite convergent partiellement avec PVF. L’EDA n’a pas montré de validité technique suffisante. L’actimétrie a montré une validité technique satisfaisante ainsi que des résultats convergeant avec ceux de PVF. Conclusions: La méthode choisie comme référence a montré un effet bénéfique de la diète testée et sa validité technique a été vérifiée. À la fois, CODI et l’actimétrie ont été validés. La validité d’EDA et de MFQ a été remise en question. L’actimétrie a donné des résultats prometteurs, probablement complémentaires de PVF.

The thesis deals with different aspects connected with the distribution and toxicity of PHC in crude oils in the aquatic environment and marine molluscs. Information has been gathered and presented on the present status of research in the field based on a thorough review of recent literature. It is seen from the literature that the resident time of PHC becomes lesser and lesser when crude oil is discharged in large quantities into the coastal waters where excess oxygenation light penetration, temperature availability and strong tidal currents persist. This results in the reduction of the onslaught of oil pollution. This probably shows that taxic insult of crude oil in tropical shores will be relatively lesser than that occurring in temperate and boreal waters. However, there is no record of major episodal oil pollution comparable in quantum and extent to that of Torrey Canyon or Amoco Cadiz which prevents a more reliable analysis on the issue. Considerable controversy exists on the method of estimation of PHC in seawater. However, Ultra-violet fluorescence spectroscopy has been widely recommended as an analytical tool for the determination of total hydrocarbons. Similarly representation of values as chrysene equivalents has been recognised as the most reliable method of Quantum representation.

In the attempt to find out catalytic potency and properties of the endoglucanase of green mussel, it could be highlighted that the enzyme is efficient in degrading carboxymethylcellulose to reducing sugars. The immobilized enzyme will find applications in the food industry, paper and pulp industry, wood preservation, alcohol and pharmaceutical industry.The purification method employed i.e. Sephadex G100 chromatography employing affinity and exclusion principles simplify the purification procedure.Addition of Mg2+ and Co2+ at 10mM concentrations enhances endoglucanase activity of green mussel.The immobilized endoglucanase can be used for deinking mixed office waste paper. The endoglucanase if supplemented with exoglucanase and B-glucosidase under appropriate conditions would help in the recycling of paper.

The present scientific investigation of the effects of copper, mercury and cadmium has focussed on their effects on two commercially important marine bivalve species, Perna indica (brown mussel) and Donax incarnatus (wedge clam), conspicuous representatives of the tropical intertidal areas. The investigation centred around delineating the cause and effects of heavy metal stress, individually and in combination on these species under laboratory conditions. A clear understanding of the cause and effect can be had only if laboratory experiments are conducted employing sub-lethal concentrations of the above toxicants. Therefore, during the course of the investigation, sub-lethal concentrations of copper, mercury and cadmium were employed to assess the concentration dependent effects on survival, ventilation rate, O:N ratio and tissues. The results obtained are compared with the already available information and partitioned in sections to make a meaningful presentation.The thesis is presented in five chapters comprising INTRODUCTION, ACUTE TOXICITY, VENTILATION RATE, OXYGEN : NITROGEN RATIO and HISTOPATHOLOGY. In this thesis certain important aspects of heavy metal toxicity have been worked out. Recent studies have clearly shown that when experimental media contained more than one heavy metals, such metals could conspicuously influence the toxic reaction of the animals both in terms of quantity and nature. The experimental results available on individual metal toxicity show that, in majority of such results, unrealistically high concentrations of dissolved metals are involved. A remarkable number of factors have been shown to influence metal toxicity such as various environmental factors particularly temperature and salinity, the condition of the organism and the ability of some of the marine organisms to adapt to metallic contamination. Further, some of the more sensitive functions like embryonic and larval development, growth and fecundity, oxygen utilization and the function of various enzymes are found to be demonstrably sensitive in the presence of heavy metals. However, some of the above functions could be compensated for by adaptive process. If it is assumed that the presence of a single metal in higher concentrations could affect the life function of marine animals, more than one metal in the experimental media should manifest such effects in a greater scale. Commonly known as synergism or more than additivity, majority of heavy metals bring about synergistic reaction .The work presented in this thesis comprises lethal and sublethal toxicities of different salt forms of copper and silver on the brown mussel Perna indica. during the present investigation sublethal concentrations of copper and silver in their dent effects on survival, oxygen consumption, filtration, accumulation and depuration on Perna indica. The results are presented under different sections to make the presentation meaningful .

Lobsters are renowned the world over as tasty delicacies and are in great demand for Epicurian gourests. As a result these animals found in different parts of the world from countries like India they are mostly frozen and exported thus earning considerable foreign exchange for the country. Importance of this crustacean is thus well known and whatever harvested from nature are utilized to the maximum extent. The demand being much more than what is being produced the need for artificial culture becomes evident. In this context a complete knowledge of the lifecycle ,larval biology and their culture is very essential. Thus with the object of studying the life history of the commercially important lobsters fished from Indian waters the larval biology of panulirus homarus has been taken up as the project for the present study
