The grazing trial at Kidston Gold Mine, North Queensland, was aimed specifically to assess the uptake of metals from the tailing and the potential for unacceptable contamination of saleable meat. Further aims included estimating metal dose rates and identifying potential exposure pathways including plant uptake of heavy metals, mine tailings adhered to plants and direct ingestion of mine tailing. It was found that of the 11 metals analysed (As, Zn, Co, Cd, Cr, Sn, Pb, Sb, Hg, Se and Ni) in the animal's liver, muscle and blood during the 8-month trial period, only accumulation of arsenic and zinc occurred. A risk assessment including these two metals was conducted to determine the potential for chronic metal toxicity and long-term contamination, using the estimates of metal dose rate. It was concluded that no toxicity or long-term contamination in cattle was likely at this site. Management procedures were therefore not required at this site; however, the results highlight percent ground cover and standing dry matter (DM) as important factors in decreasing metal exposure from direct ingestion of tailings and dust adhered to plants. A detailed analysis procedure is described for evaluating rates of volumetric change in brain structures based on structural magnetic resonance (MR) images. In this procedure, a series of image processing tools have been employed to address the problems encountered in measuring rates of change based on structural MR images. These tools include an algorithm for intensity non-uniforniity correction, a robust algorithm for three-dimensional image registration with sub-voxel precision and an algorithm for brain tissue segmentation. However, a unique feature in the procedure is the use of a fractional volume model that has been developed to provide a quantitative measure for the partial volume effect. With this model, the fractional constituent tissue volumes are evaluated for voxels at the tissue boundary that manifest partial volume effect, thus allowing tissue boundaries be defined at a sub-voxel level and in an automated fashion. Validation studies are presented on key algorithms including segmentation and registration. An overall assessment of the method is provided through the evaluation of the rates of brain atrophy in a group of normal elderly subjects for which the rate of brain atrophy due to normal aging is predictably small. An application of the method is given in Part 11 where the rates of brain atrophy in various brain regions are studied in relation to normal aging and Alzheimer's disease. We present global and regional rates of brain atrophy measured on serially acquired T1-weighted brain MR images for a group of Alzheimer's disease (AD) patients and age-matched normal control (NC) subjects using the analysis procedure described in Part I. Three rates of brain atrophy: the rate of atrophy in the cerebrum, the rate of lateral ventricular enlargement and the rate of atrophy in the region of temporal lobes, were evaluated for 14 AD patients and 14 age-matched NC subjects. All three rates showed significant differences between the two groups, However, the greatest separation of the two groups was obtained when the regional rates were combined. This application has demonstrated that rates of brain atrophy, especially in specific regions of the brain, based on MR images can provide sensitive measures for evaluating the progression of AD. These measures will be useful for the evaluation of therapeutic effects of novel therapies for AD. (C) 2002 Elsevier Science Inc. All rights reserved.

Human R183H-GH causes autosomal dominant GH deficiency type II. Because we show here that the mutant hormone is fully bioactive, we have sought to locate an impairment in its progress through the secretory pathway as assessed by pulse chase experiments. Newly synthesized wild-type and R183H-GH were stable when expressed transiently in AtT20 cells, and both formed equivalent amounts of Lubrol-insoluble aggregates within 40 min after synthesis. There was no evidence for intermolecular disulfide bond formation in aggregates of wild-type hormone or the R183H mutant. Both wildtype and R183H-GH were packaged into secretory granules, assessed by the ability of 1 mm BaCl2 to stimulate release and by immunocytochemistry. The mutant differed from wildtype hormone in its retention in the cells after packaging into secretory granules; 50% more R183H-GH than wild-type aggregates were retained in AtT20 cells 120 min after synthesis, and stimulated release of R183H-GH or a mixture of R183H-GH and wild-type that had been retained in the cell was reduced. The longer retention of R183H-GH aggregates indicates that a single point mutation in a protein contained in secretory granules affects the rate of secretory granule release.

Telemedicine promises to revolutionize medical care delivery in rural and remote areas. The ability to accurately evaluate physical impairment via the Internet is important to the possible future provision of Internet-based physiotherapy. This study evaluated the reliability and validity of assessing knee range of motion via the Internet. Two therapists evaluated knee angle on a single subject via two methods of assessment: the Internet and the traditional method (face-to-face). Nine random positions of the knee were chosen with the principal examiner performing 20 face-to-face and two sets of 20 Internet measures in each position (n=540). The secondary therapist performed Internet assessments only. The Internet connection was established at a readily available speed of 17 kbit/s. The Internet-based goniometer was found to be a valid tool for measuring both knee flexion and extension angles. It was shown to possess both high intra and inter-rater reliability. Difference average plots of the scores verified the consistency of measurement between both modes of assessment. The successful evaluation of the physical outcome measure of knee range of motion via the Internet assists the further development of Internet-based physiotherapy applications. Purpose. The flux of a topically applied drug depends on the activity in the skin and the interaction between the vehicle and skin. Permeation of vehicle into the skin can alter the activity of drug and the properties of the skin barrier. The aim of this in vitro study was to separate and quantify these effects. Methods. The flux of four radiolabeled permeants (water, phenol, diflunisal, and diazepam) with log K-oct/water values from 1.4 to 4.3 was measured over 4 h through heat-separated human epidermis pretreated for 30 min with vehicles having Hildebrand solubility parameters from 7.9 to 23.4 (cal/cm(3))(1/2). Results. Enhancement was greatest after pretreatment with the more lipophilic vehicles. A synergistic enhancement was observed using binary mixtures. The flux of diazepam was not enhanced to the same extent as the other permeants, possibly because its partitioning into the epidermis is close to optimal (log K-oct 2.96). Conclusion. An analysis of the permeant remaining in the epidermis revealed that the enhancement can be the result of either increased partitioning of permeant into the epidermis or an increasing diffusivity of permeants through the epidermis.

Purpose. As reductions in dermal clearance increase the residence time of solutes in the skin and underlying tissues we compared the topical penetration of potentially useful vasoconstrictors (VCs) through human epidermis as both free bases and ion-pairs with salicylic acid (SA). Methods. We determined the in vitro epidermal flux of ephedrine, naphazoline, oxymetazoline, phenylephrine, and xylometazoline applied as saturated solutions in propylene glycol: water (1: 1) and of ephedrine, naphazoline and tetrahydrozoline as 10% solutions of 1: 1 molar ratio ion-pairs with SA in liquid paraffin. Results. As free bases, ephedrine had the highest maximal flux, Jmax = 77.4 +/- 11.7 mug/cm(2)/h, being 4-fold higher than tetrahydrozoline and xylometazoline, 6-fold higher than phenylephrine, 10-fold higher than naphazoline and 100-fold higher than oxymetazoline. Stepwise regression of solute physicochemical properties identified melting point as the most significant predictor of flux. As ion-pairs with SA, ephedrine and naphazoline had similar fluxes (11.5 +/- 2.3 and 12.0 +/- 1.6 mug/cm(2)/h respectively), whereas tetrahydrozoline was approximately 3-fold slower. Corresponding fluxes of SA from the ion-pairs were 18.6 +/- 0.6, 7.8 +/- 0.8 and 1.1 +/- 0.1 respectively. Transdermal transport of VC's is discussed. Conclusions. Epidermal retention of VCs and SA did not correspond to their molar ratio on application and confirmed that following partitioning into the stratum corneum, ion-pairs separate and further penetration is governed by individual solute characteristics.

Nitrate leaching below the crop root-zone in variable charge soils may be adsorbed at anion exchange sites, thereby temporarily reducing the risk of contamination of water bodies. The objectives of this study were (i) to investigate whether nitrate adsorption, accumulation, and retention in the Johnstone River Catchment of Far North Queensland wet tropics is widespread; (ii) to assess the capacity of soil in the Johnstone River Catchment to retain nitrate; and (iii) to deduce the consequences of nitrate adsorption/desorption on contamination of water bodies. Soil cores ranging from 8 to 12.5 m depth were taken from 28 sites across the catchment, representing 9 Ferrosol soil types under sugarcane (Saccharum officinarum-S) cultivation for at least 50 years and from rainforest. The cores were segmented at 0.5-m depth increments and subsamples were analysed for nitrate-N, cation and anion exchange capacities, pH, exchangeable cations (Ca, Mg, K, Na), soil organic C, electrical conductivity, sulfate-S, and chloride. Nitrate-N concentration under sugarcane ranged from 0 to 72.5 mg/kg, compared with 0 to 0.31 mg/kg under rainforest, both Pin Gin soils. The average N load in 1-12 m depth across 19 highly oxidic profiles of the Pin Gin soil series was 1550 kg/ha, compared with 185 kg/ha under 8 non-Pin Gin soils and 11 kg/ha in rainforest on a Pin Gin soil. Most of the nitrate retention was observed at depth of 2-12 m, particularly at 4-10 m, indicating that the accumulation was well below the crop root-zone. The average maximum potential nitrate retention capacity was 10.8 t/ha for the Pin Gin and 4.7 t/ha for the non-Pin Gin soil. Compared with the current N load, the soils still possess a large capacity to adsorb and retain nitrate in profiles. Retention of large quantities of the leached nitrate deep in most of the profiles has reduced the risk of contamination of water bodies. However, computations show that substantial quantities of the nitrate leached below the root-zone were not adsorbed and remain unaccounted for. This unaccounted nitrate might have entered both on- and off-site water bodies and/or have been denitrified.

O consumo de suco de frutas vem aumentando no Brasil. Entre 2002 e 2009 o consumo de sucos, sejam eles concentrados, em pó, sucos ou néctares, aumentou em 21%. Devido ao seu sabor agradável e doce, e ao seu valor nutricional, o suco de laranja é o suco mais comum fabricado pela indústria de processamento de bebidas. Diversos fatores podem afetar a qualidade do suco de laranja. A microbiota típica presente no suco de laranja pode ser proveniente de várias etapas de sua produção. Em relação às enzimas, a pectinametilesterase (PME) é a principal causadora de alterações em suco laranja. A pasteurização e a esterilização comercial são os métodos de conservação mais comuns utilizados para inativar enzimas e micro-organismos, porém podem causar efeitos adversos em relação às características sensoriais (cor, sabor, aroma, e outros) dos produtos. A tecnologia de ultrassom vem sendo estudada recentemente como uma forma de conservar os alimentos sem causar efeitos indesejáveis como os provocados pelos tratamentos térmicos. O objetivo deste trabalho foi avaliar a utilização da tecnologia de ultrassom e de ultrassom aliado a temperaturas brandas, como forma de conservar suco de laranja. Para isto, foram analisadas a contagem de mesófilos totais e bolores e leveduras, a atividade da pectinametilesterase, o teor de vitamina C, a cor, o pH, o teor de sólidos solúveis e a estabilidade em relação à turbidez. Ainda, avaliou-se a aceitação sensorial de suco de laranja submetido à termossonicação. Os resultados foram comparados com os obtidos para o suco natural e o suco pasteurizado. Utilizou-se um ultrassom de 40 kHz, associado às temperaturas de 25 ºC, 30 ºC, 40 ºC, 50 ºC e 60 ºC durante 10 minutos. Os tratamentos utilizando ultrassom a 50 ºC e 60 ºC foram capazes de reduzir a contagem de bolores e leveduras e de mesófilos totais, apresentando uma redução de 3 ciclos logarítmicos. Resultado similar foi encontrado quando realizado o tratamento térmico a 90 ºC por 30 segundos. Observou-se que a aplicação da termossonicação permitiu uma redução significativa na atividade de PME e uma menor perda de vitamina C. O tratamento que apresentou melhor redução na atividade de PME foi utilizando ultrassom 40 kHz com temperatura de 60 ºC. Em relação ao ácido ascórbico, quanto menor a temperatura utilizada em conjunto com a sonicação, menor foi a perda deste composto. O teor de sólidos solúveis, o pH e a cor do suco não foram alterados ao longo do processamento. Avaliando a aceitabilidade do suco, verificou-se que a cor não foi influenciada por nenhum tratamento. Em relação ao aroma, sabor e aceitação global o suco submetido a termossonicação obteve aceitação sensorial superior à encontrada para o suco pasteurizado. Concluiu-se então que a utilização da termossonicação como uma forma de conservação para suco de laranja é viável.

A região da Caatinga é caracterizada pelas altas temperaturas durante o ano e má distribuição das chuvas. Em virtude desses fatores de clima regional, tem-se a necessidade de adoção por práticas que elevem à eficiência e sustentabilidade agrícola local. Assim, objetivou-se avaliar a aptidão de leguminosas herbáceas perenes como cobertura permanente de solo no cultivo de bananeira. Foram conduzidos três experimentos, para avaliação das leguminosas, utilizou-se um delineamento experimental em blocos casualizados, com quatro repetições, em esquema de parcelas subdivididas no espaço e para algumas variáveis, sub-subdividida no tempo, sendo: Fator “A” os dois diferentes ambientes de plantio: municípios de Itaobim/MG e Virgem da Lapa/MG; Fator “B”, nas subparcelas, dois manejos de cobertura do solo e para algumas variáveis, três manejos, constituídos pelas leguminosas: cudzu tropical (Pueraria phaseoloides) calopogônio (Calopogonium mucunoides) e solo descoberto (solo capinado); Fator “C” épocas de coleta de dados. Para avaliação das bananeiras, foram dois experimentos em blocos casualizados, com quatro repetições, em esquema de parcelas sudivididas no espaço, sendo: nas parcelas, fator “A” constituído por três manejos de cobertura do solo, pelas leguminosas: cudzu tropical e o calopogônio, e solo descoberto (solo capinado); fator “B”, nas subparcelas, plantas de bananeiras em três idades morfofisiológicas (diferentes ciclos e tamanhos); Para algumas variáveis que foram submetidas a coletas periódicas, utilizou-se o esquema de parcelas subsubdivididas no tempo, acrescentando-se o fator “C”, datas das coletas nas sub-subparcelas, tendo como referência os dias após semeadura (DAS) das leguminosas. Foram avaliadas as seguintes variáveis: taxa de cobertura do solo; potencial de deposição de folhas e a ciclagem de nutrientes; capacidade de inibição da vegetação espontânea; conservação da temperatura e promoção da retenção de umidade do solo. Também foi avaliado o crescimento vegetativo e produtividade das bananeiras. Como resultados principais, notou-se que as leguminosas proporcionaram eficiente cobertura do solo, o calopogônio apresentou o maior acúmulo de N, P, K, e Ca, via deposição de material senescente, tal como maior inibição das plantas espontâneas nos pomares de bananeiras. Essa cobertura também promoveu uma eficiente redução da temperatura do solo, possibilitando menor variação térmica nas camadas de maior concentração radicular da bananeira, e consequentemente, obtendo maior acúmulo de umidade no solo. As bananeiras cultivadas sobre coberturas vivas de solo apresentaram aumento gradativo no crescimento e peso de cacho. Os resultados reforçam o potencial uso dessas espécies na fruticultura, principalmente em regiões de severas restrições hídricas, como forma de adubação e otimização de diversos processos biológicos em seu ambiente de cultivo.
