The aim of this study was to evaluate the hygienic and sanitary conditions of pettreats from an export industry. The occurrence of Salmonella, enumerated sulfite-reducing Clostridium (SRC), coagulase-positive Staphylococcus (CPS) and total(TC) and thermotolerant coliforms (TTC) was studied. Of the 108 samples ana-lyzed, 22 were contaminated by some microorganism. TC were detected at twosamples (3.5 × 101and 4.5 × 101colony-forming units [cfu]/g), SRC in 19 samples(1.0 × 101to 1.3 × 105cfu/g) and Salmonella spp. at only one sample; CPS andTTC were not detected. These findings indicate the microbiological safety of mostitems, except for a mix of dried bovine offal, with 1.3 × 105cfu/g of SRC, and onemix of dehydrated bovine viscera contaminated with Salmonella. The current pro-duction processes and the food safety management system are adequate to guar-antee the production of microbiologically safe foods, but that some improvementscan still be made with regard to cleaning and disinfection and hygiene training.

Forty eight piglets from a commercial strain weaned at an average age of 21 days were used in order to evaluate effects of carbohydrates sources (lactose or maltodextrin) and the weights of piglets at weaning on the stomach pH, colon and rectum contents, and the amount of total coliform, Escherichia coli and Lactobacillus spp in digesta of ileum and cecum. The experiment was conducted in a randomized block design and analyzed in split plot, considering class of weight and carbohydrates as main plots and the slaughter age of animal as sub-plots. The stomach pH and intestinal contents were not influenced by the diets provided as well as the amount of total coliform. It was found that the presence of lactose in the feed decreased counts of Escherichia coli and Lactobacillus ssp in the ileum. Regarding the class of weight, the lighter piglets had lower counts of Escherichia coli and Lactobacillus. The maltodextrin can be used as an alternative to replace the lactose in the diet, regardless of the piglets weight at weaning, as it does not adversely affect the pH and the Lactobacillus population in the gastrointestinal tract.

Sensorial and microbiological characteristics of a Brazilian fresh cheese samples with Bifidobacterium animalis subps. lactis as well as samples with this probiotic and polydextrose, a prebiotic ingredient, were evaluated. The addition of this microorganism was studied as: (1) lyophilized probiotic added to cheese curd and (2) by using milk previously fermented by this probiotic to produce the cheese. Cheese samples were microbiologically characterized after 0, 7, 14, 21 and 28 days of storage at a temperature of 4 °C. The microbiological analyses conducted were quantification of total lactic acid bacteria, mesophilic microorganisms, Bif. animalis subps. lactis, coliforms at 30 °C and 45 °C. Affective sensory test was conducted for two different cheese samples (with probiotic and with probiotic and prebiotic) as well as for control one week after manufacturing date. Cheese samples provided acceptable results for coliform counts at 30 °C and 45 °C in compliance with legislation. The cheese samples produced using milk fermented by probiotic showed counts of 107 -108 CFU/g after 28 days of storage, which assures functional property for this product to be claimed.

Milk that is adequate for consumption must be of hygienic quality, nutritional value, and should maintain its organoleptic properties. Isolation of fecal and/or total coliforms from bovine milk is considered an indicator of hygiene and good management practices, and can be used as a quality indicator. This study aimed to isolate, identify, and assess the resistance profile of coliforms isolated from collective bulk tanks and individual milk tanks. A total of 89 milk samples were collected from collective bulk tanks and, from these, 21 Klebsiella spp., one E. coli, and 29 Enterobacter spp. were isolated, whereas 102 milk samples from individual tanks showed isolation of one Klebsiella spp. and seven Enterobacter spp. In collective bulk tanks, at least 47% of Klebsiella spp. and Enterobacter spp. were resistant to cephalexin and 30% to ampicillin. From these, at least 24% showed multidrug resistance. Among the microorganisms isolated from the individual tanks, 85% or more were resistant to ampicillin. The ESBL phenotype and the blaTEM gene were detected in strains of Klebsiella spp. isolated from both tanks. It was concluded that contamination of milk with resistant total coliforms, and especially the storage of raw milk from several small producers in the collective bulk tank increase the risk of contamination.

Este estudo teve como objetivo investigar o impacto das atividades antropogênicas da cidade de Belém pela comparação da qualidade da água e das comunidades de larvas de peixes em dois igarapés que desembocam no rio Guamá. Um dos igarapés atravessa um subúrbio pobre e populoso de Belém, enquanto o outro é localizado em uma ilha de Belém, declarada Área de Proteção Ambiental desde 1997. Dois pontos de coletas foram definidos em cada igarapé e monitorados durante oito horas, a cada três meses e durante um ano. O igarapé em região urbana apresentou fortes alterações na qualidade da água, durante o ano todo e em todas as marés, e isto deve se essencialmente a presença de um elevado número de coliformes termotolerantes. Poucas larvas foram encontradas. A água foi considerada imprópria para uso e atividades humanas, assim como para a vida aquática. O igarapé da ilha apresentou primeiros sinais de contaminação por nutrientes e bactéria durante o período chuvoso, parcialmente decorrente de fontes de poluição difusa. Em ambos os córregos, as comunidades larvais foram quase exclusivamente compostas de clupeiformes. Todos os estágios de desenvolvimento larval foram encontrados. Densidades e proporções mais elevadas de larvas recém eclodidas foram registradas durante a estação seca e associadas à presença de nitrato. Resultados apontam a necessidade de desenvolver um sistema de drenagem urbano para esgoto e água pluvial na maior brevidade, e recomenda um estudo de monitoramento integrado do igarapé na Área de Proteção Ambiental.

Os dejetos suínos contribuem de forma significativa em prol da degradação dos recursos naturais e para a diminuição da qualidade de vida na região sul do estado de Santa Catarina, Brasil. O excesso de dejetos gerados pelo grande número dos suínos por unidade de área dificulta a solução do problema. Este trabalho teve como objetivo avaliar o comportamento de um sistema de tratamento de dejetos suínos, em série e em escala real, composto de unidades anaeróbias, unidades aeróbias e pós-tratamento. Os parâmetros analisados foram: DBO, DQO, PT, P-PO4, N-NH3, NTK, pH, clorofila a, biomassa algal, coliformes totais e coliformes fecais (Escherichia coli). O sistema mostrou que após um ano de funcionamento as eficiências de remoção foram satisfatórias para matéria orgânica e nutrientes, alcançando valores de 97% para DBO, 95% para DQO, 88% para N-NH3 e 74% para PT/P-PO4, mesmo com as variações de vazão e de carga orgânica aplicada. Entretanto, a remoção de coliformes fecais nas unidades aeróbias foi baixa, reduzindo somente 0,86 unidades log na lagoa facultativa aerada e 0,80 unidades log na lagoa de maturação.

In this study, a total of 172 samples of minimally processed vegetables (MPV) were collected from supermarkets in the city of Campinas, Brazil. The MPV were analyzed using traditional and/or alternative methods for total aerobic mesophilic bacteria, total coliforms, Escherichia coil, coagulase positive staphylococci, Salmonella and Listeria monocytogenes. All the MPV analyzed presented populations of aerobic mesophilic microorganisms and total coliforms were >4 log(10) CFU/g and 1.0-3.4 log(10) CFU/g, respectively. E. coil was enumerated in only 10 samples out of 172 collected, while none of the 172 samples of MPV presented contamination by coagulase positive Staphylococcus (<10(1) CFU/g). Among the four methods used for detection of Salmonella in MPV (Vidas, 1,2 Test, Reveal, and Traditional), when Reveal was used a total of 29 positive samples were reported. For L monocytogenes, the four methods tested (Vidas, Vip, Reveal, and traditional) performed similarly. The presence of Salmonella and L monocytogenes in MPV was confirmed in one (watercress) and two samples (watercress and escarole), respectively. In conclusion, it has been observed that the microbiological quality of MPV commercialized in Campinas is generally satisfactory. Besides, the choice of microbiological method should be based not only on resource and time issues, but also on parameters such as sensitivity and specificity for the specific foods under ahalysis. PHYSICO-CHEMICAL EVALUATION OF AN EFFLUENT TREATED IN ANAEROBIC BIODIGESTER REGARDING ITS EFFICIENCE AND APPLICATION AS FERTILIZER. The use of biodigester for basic and environmental sanitation has large demand in Brazil. A biodigester was built to treat conjunctly the human and pig feces and urine, regarding to its future application in rural small towns. The results show that the biodigester can reduce 90% of COD and BOD and, up to 99.99% of thermotolerant coliforms. The treated effluent has variable quantities of macro- and micro-nutrients; and organic matter. However, the concentration variability of the nutrients makes difficult a dosed application into soil. The soluble salts (mainly as Na+ form) make necessary a controlled use to avoid environmental degradation.

This study reports the implementation of GMPs in a mozzarella cheese processing plant. The mozzarella cheese manufacturing unit is located in the Southwestern region of the state of Parana, Brazil, and processes 20,000 L of milk daily. The implementation of GMP took place with the creation of a multi-disciplinary team and it was carried out in four steps: diagnosis, report of the diagnosis and road map, corrective measures and follow-up of GMP implementation. The effectiveness of actions taken and GMP implementation was compared by the total percentages of non-conformities and conformities before and after implementation of GMR Microbiological indicators were also used to assess the implementation of GMP in the mozzarella cheese processing facility. Results showed that the average percentage of conformity after the implementation of GMP was significant increased to 66%, while before it was 32% (p < 0.05). The populations of aerobic microorganisms and total coliforms in equipment were significantly reduced (p < 0.05) after the implementation of GMP, as well as the populations of total coliforms in the hands of food handlers (p < 0.05). In conclusion, GMP implementation changed the overall organization of the cheese processing unity, as well as managers and food handlers' behavior and knowledge on the quality and safety of products manufactured. Florianopolis, a city located in the Santa Catarina State in southern Brazil, is the national leading producer of bivalve mollusks. The quality of bivalve mollusks is closely related to the sanitary conditions of surrounding waters where they are cultivated. Presently, cultivation areas receive large amounts of effluents derived mainly from treated and non-treated domestic, rural, and urban sewage. This contributes to the contamination of mollusks with trace metals, pesticides, other organic compounds, and human pathogens such as viruses, bacteria, and protozoan. The aim of this study was to perform a thorough diagnosis of the shellfish growing areas in Florianopolis, on the coast of Santa Catarina. The contamination levels of seawater, sediments, and oysters were evaluated for their microbiological, biochemical, and chemical parameters at five sea sites in Florianopolis, namely three regular oyster cultivation areas (Sites 1, 2, and oyster supplier), a polluted site (Site 3), and a heavily polluted site (Site 4). Samples were evaluated at day zero and after 14 days. Seawater and sediment samples were collected just once, at the end of the experiment. Antioxidant defenses, which may occur in contaminated environments in response to the increased production of reactive oxygen species (ROS) by organisms, were analyzed in oysters, as well as organic compounds (in oysters and sediment samples) and microbiological contamination (in oysters and seawater samples). The results showed the presence of the following contaminants: fecal coliforms in seawater samples (four sites), human adenovirus (all sites), human noroviruses GI and GII (two sites), Hepatitis A viruses (one site), JC Polyomavirus in an oyster sample from the oyster supplier, Giardia duodenalis cysts, and Cryptosporidium sp oocysts (one site). Among organochlorine pesticides, only DDT (dichlorodiphenyltrichloroethane) and HCH (hexachlorocyclohexane) were detected in some sediment and oysters samples in very low levels; site 4 had the highest concentrations of total aliphatic hydrocarbons. PAHs, and linear alkylbenzenes (LABs) found either in oysters or in sediment samples. The major concentration of fecal sterol coprostanol was found at site 4, followed by site 3. After 14 days of allocation in the four selected sites, there was a significant difference in the enzymes analyzed at the monitored spots. The detection of different contaminants in oysters, seawater, and sediment samples in the present study shows the impact untreated or inadequately treated effluents have on coastal areas. These results highlight the need for public investment in adequate wastewater treatment and adequate treatment of oysters, ensuring safe areas for shellfish production as well as healthier bivalve mollusks for consumption.

Analyses of living foraminiferal and environmental parameters near an outfall at Mar Grosso Beach (Laguna, SC, Brazil) demonstrate its usefulness as indicators of domestic sewage pollution. The low species diversity may be due to sand accumulation in the central part. Higher diversity was noted closer to the mouth of Laguna estuarine system where reduced salinity and higher temperatures indicate freshwater influence, suggesting a relationship between increased diversity and greater availability of terrestrial food. On the basis of foraminiferal diversity and average coliform count the higher values are closer to the mouth of the estuarine system and under the influence of the outfall. Due to the effect of local hydrodynamics, the particulate organic waste derived from the outfall does not settle down locally, and thus, do not accumulate nearby. Our hypothesis is that the fine material derived from the outfall is accumulating on the southwestern and northwestern parts of the beach.

This study aimed to assess the sanitary quality of water, and wet and dry sand from three beaches located in the South Coast region of Sao Paulo State, Brazil, selected taking into account the frequency of tourists and the water quality (good, fair and poor). Thirty-six water samples each of wet and dry sand and seawater were collected monthly over a period of one year and analyzed for fecal indicator bacteria (FIB: thermotolerant coliforms, Escherichia coli, and enterococci), presumptive Staphylococcus aureus, Pseudomonas aeruginosa, Candida albicans and dermatophytes. The results revealed FIB concentrations more elevated in dry sand followed by wet sand and water. P. aeruginosa and presumptive S. aureus were detected with a similar frequency in water and sand samples, but maximum concentrations and geometric means were higher in dry sand. C. albicans was detected only in water samples whereas the dermatophyte Microsporum sp. was isolated exclusively from dry and wet sand samples. This evaluation showed also that the environment had a significant influence on P. aeruginosa but not on presumptive S. aureus concentrations. According to threshold values proposed in the literature for E. coli and enterococci dry sand densities, none of the beaches would be considered of sufficient quality for recreational activities.
