Nach Homogenisation ejakulierter Eberspermien und Zentrifugation des Homogenates blieben mehr als 60% der Aktivität des glykolytischen Enzyms Pyruvatkinase (PK) an Zellfragmenten im Sediment gebunden. Diese strukturgebundene PK wurde als PK-S bezeichnet. Das Detergenz Triton X-100 führte nicht zur Ablösung der PK-S; mit Trypsin konnten jedoch rund 80% der PK-S ohne Verlust an Aktivität von den Strukturen gelöst und durch kombinierte Kationenaustausch- und Hydrophobizitätschromatographie gereinigt werden (spezifische Aktivität: 116,7 U/mg Protein). Die lösliche PK aus Eberspermien konnte ebenfalls durch ein ähnliches Verfahren angereichert werden. Im Gel (SDS-PAGE) zeigten die Untereinheiten der PK-S mit 64.400 eine geringfügig größere relative Molekülmasse als die der PK-M1 aus Kaninchenmuskel (62.000). Die kinetischen Eigenschaften der abgelösten PK-S als auch der noch an Spermienstrukturen gebundenen PK-S und der löslichen PK aus Eberspermien waren sehr ähnlich und entsprachen der M1-Isoform der PK. Antikörper gegen Kaninchenmuskel-PK (Anti-PK-M1) reagierten auch mit der löslichen PK und der PK-S aus Eberspermien. Edman-Abbau der ersten 19 Aminosäuren zeigte, dass die tryptisch abgelöste PK-S am N-Terminus um 5 Aminosäuren gegenüber nativer PK-M1 verlängert ist, während der C-Terminus der erhaltenen PK-S-Sequenz mit einem meist nahe dem N-Terminus gelegenen Sequenzabschnitt der PK-M1 und -M2 übereinstimmt. Die N-terminale Verlängerung der nativen PK-S enthält sicherlich mehr als die nach tryptischer Lyse nachgewiesenen 5 Aminosäuren. Vergleiche der Aminosäure- und übersetzten Nukleotidsequenzen sowie die kinetischen Eigenschaften lassen vermuten, dass die PK-S, wie die PK-M1 und PK-M2, vom PKM-Gen codiert wird. Gegen die gereinigte PK-S wurden Antikörper in Kaninchen produziert. Da das Antiserum nicht ausreichend spezifisch für PK-S war, wurden aus ihm affinitätschromatographisch Antikörper (Anti-PK-S) isoliert, die hohe Affinität zu einem synthetisierten PK-S-Peptid (13 N-terminale Aminosäuren der tryptisch abgelösten PK-S) hatten. Dieses Anti-PK-S-Präparat war spezifisch für PK-S; es reagierte weder mit Kaninchenmuskel-PK noch mit löslicher PK oder anderen Proteinen aus Eberspermien. Anti-PK-S und Anti-PK-M1 wurden zur Lokalisierung von PK-S und löslicher PK in Spermien von Eber, Bulle und Mensch sowie in Schnitten von Eberhoden eingesetzt. Mit Anti-PK-S wurden der Bereich des Akrosoms und das lange flagellare Hauptstück sowie der Übergangsbereich zwischen Kopf und Mittelstück von Eberspermien fluoreszenzmarkiert, wogegen das kurze, die Mitochondrien enthaltende Mittelstück des Flagellums und der postakrosomale Kopfbereich nur mit Anti-PK-M1 markiert wurden. Immunogoldmarkierung in elektronenmikroskopischen Bildern bestätigte die Lokalisierung von PK-S im Akrosombereich. Im Hauptstück banden Anti-PK-M1 und Anti-PK-S an die fibröse Scheide. Glyzerinaldehyd-3-phosphat Dehydrogenase (GAPDH) konnte von mir ebenfalls im Akrosombereich, im Übergangsbereich zwischen Kopf und Mittelstück und an der fibrösen Scheide detektiert werden. Auch an Bullen- und Humanspermien konnte über Immunogoldmarkierung PK und vermutlich GAPDH an der fibrösen Scheide gezeigt werden. Im Akrosombereich dieser Spermien waren die Nachweise von PK und GAPDH jedoch nicht sicher. In Eberhodenschnitten war die PK-S erstmals, oder zumindest vermehrt, in den elongierenden Spermatiden über Fluoreszenzmarkierung nachweisbar, während andere, vermutlich somatische PK vermehrt in den früheren Stadien (Spermatogonien, aber auch in den Spermatozyten und runden Spermatiden) auftrat. Für die GAPDH zeigte sich ein ähnlicher Entwicklungsverlauf. Die Ergebnisse zeigen, dass in Eberspermien zwei Isoformen der PK auftreten: eine N-terminal verlängerte, strukturgebundene Form, die PK-S, und eine lösliche Form, die beide der PK-M1 ähneln. Der ungewöhnliche N-Terminus der PK-S dient vermutlich der spezifischen räumlichen Anordnung der PK-S im Akrosombereich und an der fibrösen Scheide, nicht aber der Modulation kinetischer Eigenschaften. Meine Untersuchungen stützen die Hypothese, dass in bestimmten Kompartimenten von Säugerspermien die Glykolyse durch Verankerung einiger ihrer Enzyme strukturell hochgeordnet ist. Dadurch wird vermutlich die Versorgung der Mitochondrien-freien Regionen mit ATP sichergestellt. Man kann diese Organisation als Anpassung des Stoffwechsels von Spermien deuten, bei denen die Mitochondrien in einem kleinen Bereich (Mittelstück) hinter dem Spermienkopf kompartimentiert sind. Im Hauptstück des Flagellums könnte die Glykolyse ATP für die Spermienmotilität liefern, im Akrosombereich für die Verhinderung einer vorzeitigen Akrosomreaktion. Somit käme der strukturierten Glykolyse eine essentielle Bedeutung für die Befruchtungsfähigkeit von Säugerspermien zu.

The low-energy β− emitter 161Tb is very similar to 177Lu with respect to half-life, beta energy and chemical properties. However, 161Tb also emits a significant amount of conversion and Auger electrons. Greater therapeutic effect can therefore be expected in comparison to 177Lu. It also emits low-energy photons that are useful for gamma camera imaging. The 160Gd(n,γ)161Gd→161Tb production route was used to produce 161Tb by neutron irradiation of massive 160Gd targets (up to 40 mg) in nuclear reactors. A semiautomated procedure based on cation exchange chromatography was developed and applied to isolate no carrier added (n.c.a.) 161Tb from the bulk of the 160Gd target and from its stable decay product 161Dy. 161Tb was used for radiolabeling DOTA-Tyr3-octreotate; the radiolabeling profile was compared to the commercially available n.c.a. 177Lu. A 161Tb Derenzo phantom was imaged using a small-animal single-photon emission computed tomography camera. Up to 15 GBq of 161Tb was produced by long-term irradiation of Gd targets. Using a cation exchange resin, we obtained 80%–90% of the available 161Tb with high specific activity, radionuclide and chemical purity and in quantities sufficient for therapeutic applications. The 161Tb obtained was of the quality required to prepare 161Tb–DOTA-Tyr3-octreotate. We were able to produce 161Tb in n.c.a. form by irradiating highly enriched 160Gd targets; it can be obtained in the quantity and quality required for the preparation of 161Tb-labeled therapeutic agents.

An improved chemical strategy for processing of the generator produced 68Ga was developed based on processing of the original 68Ge/68Ga generator eluate on a micro-column. Direct pre-concentration and purification of the eluted 68Ga is performed on a cation-exchange resin in hydrochloric acid/acetone media. A supplementary step based on a second micro-column filled with a second resin allows direct re-adsorption of 68Ga eluted from the cation exchanger. 68Ga is finally striped from the second resin with a small volume of pure water. For this purpose a strong anion exchanger and a novel extraction chromatographic resin based on tetraalkyldiglycolamides are characterized. The strategy allows online pre-concentration and purification of 68Ga from the original generator eluate. The supplementary column allows transferring 68Ga with high radionuclide and chemical quality in the aqueous solution with small volume and low acidity useful for direct radiolabeling reactions.

Clay minerals have a fundamental importance in many processes in soils and sediments such as the bioavailability of nutrients, water retention, the adsorption of common pollutants, and the formation of an impermeable barrier upon swelling. Many of the properties of clay minerals are due to the unique environment present at the clay mineral/water interface. Traditional techniques such as X-ray diffraction (XRD) and absorption isotherms have provided a wealth of information about this interface but have suffered from limitations. The methods and results presented herein are designed to yield new experimental information about the clay mineral/water interface.A new method of studying the swelling dynamics of clay minerals was developed using in situ atomic force microscopy (AFM). The preliminary results presented here demonstrate that this technique allows one to study individual clay mineral unit layers, explore the natural heterogeneities of samples, and monitor swelling dynamics of clay minerals in real time. Cation exchange experiments were conducted monitoring the swelling change of individual nontronite quasi-crystals as the chemical composition of the surrounding environment was manipulated several times. A proof of concept study has shown that the changes in swelling are from the exchange of interlayer cations and not from the mechanical force of replacing the solution in the fluid cell. A series of attenuated total internal reflection Fourier transform infrared spectroscopy (ATR-FTIR) experiments were performed to gain a better understanding of the organization of water within the interlayer region of two Fe-bearing clay minerals. These experiments made use of the Subtractive Kramers-Kronig (SKK) Transform and the calculation of difference spectra to obtain information about interfacial water hidden within the absorption bands of bulk water. The results indicate that the reduction of structural iron disrupts the organization of water around a strongly hydrated cation such as sodium as the cation transitions from an outer-sphere complex with the mineral surface to an inner-sphere complex. In the case of a less strongly hydrated cation such as potassium, reduction of structural iron actually increases the ordering of water molecules at the mineral surface. These effects were only noticed with the reduction of iron in the tetrahedral sheet close to the basal surface where the increased charge density is localized closer to the cations in the interlayer.

Carnitine is an amino acid derivative that plays a key role in energy metabolism. Endogenous carnitine is found in its free form or esterified with acyl groups of several chain lengths. Quantification of carnitine and acylcarnitines is of particular interest for screening for research and metabolic disorders. We developed a method with online solid-phase extraction coupled to high-performance liquid chromatography and tandem mass spectrometry to quantify carnitine and three acylcarnitines with different polarity (acetylcarnitine, octanoylcarnitine, and palmitoylcarnitine). Plasma samples were deproteinized with methanol, loaded on a cation exchange trapping column and separated on a reversed-phase C8 column using heptafluorobutyric acid as an ion-pairing reagent. Considering the endogenous nature of the analytes, we quantified with the standard addition method and with external deuterated standards. Solid-phase extraction and separation were achieved within 8 min. Recoveries of carnitine and acylcarnitines were between 98 and 105 %. Both quantification methods were equally accurate (all values within 84 to 116 % of target concentrations) and precise (day-to-day variation of less than 18 %) for all carnitine species and concentrations analyzed. The method was used successfully for determination of carnitine and acylcarnitines in different human samples. In conclusion, we present a method for simultaneous quantification of carnitine and acylcarnitines with a rapid sample work-up. This approach requires small sample volumes and a short analysis time, and it can be applied for the determination of other acylcarnitines than the acylcarnitines tested. The method is useful for applications in research and clinical routine.

Natural zeolites are crystalline aluminosilicates with unique adsorption, cation-exchange, and catalytic properties that have multiple uses in industry and agriculture. TMAZ, a natural zeolite clinoptilolite with enhanced physicochemical properties, is the basis of the dietary supplements Megamin and Lycopenomin, which have demonstrated antioxidant activity in humans. The aim of this prospective, open, and controlled parallel-group study was to investigate the effects of supplementation with TMAZ on the cellular immune system in patients undergoing treatment for immunodeficiency disorder. A total of 61 patients were administered daily TMAZ doses of 1.2 g (Lycopenomin) and 3.6 g (Megamin) for 6 to 8 weeks, during which the patients' primary medical therapy was continued unchanged. Blood and lymphocyte counts were performed at baseline and at the end of the study. Blood count parameters were not relevantly affected in either of the two treatment groups. Megamin administration resulted in significantly increased CD4+, CD19+, and HLA-DR+ lymphocyte counts and a significantly decreased CD56+ cell count. Lycopenomin was associated with an increased CD3+ cell count and a decreased CD56+ lymphocyte count. No adverse reactions to the treatments were observed.

The specific gravity of urine (SG) indicates the number and weight of solute particles in urine; its measurement is helpful in interpreting proteinuria detected by dipstick tests and in monitoring adequate hydration in patients with nephrolithiasis. Four methods for measuring SG or osmolality of urine are currently available (depression of the freezing-point, urometry, refractometry, cation exchange on a reagent strip). Using a recently developed reagent strip, we have measured SG in morning urines of 340 non-selected outpatients and compared the results with SG measurements by refractometry of the same urines. In 86.2% of all urines, a good positive correlation between SG measured by reagent strip and refractometry was noted (r = 0.913, p = 0.0001). In 13.8% of the urines, however, the SG measured by reagent strip deviated by more than +/- 5 from the value obtained by refractometry; in 90% of these urines, glucosuria (reagent strip values too low or too high), proteinuria (values too high), or bacteriuria/leukocyturia (values too low or too high) could be found. In alkaline urine (pH > 7.0), SG values obtained by reagent strip have to be corrected by +5.

In recent years, implementation of 68Ga-radiometalated peptides for PET imaging of cancer has attracted the attention of clinicians. Herein, we propose the use of 44Sc (half-life = 3.97 h, average β+ energy [Eβ+av] = 632 keV) as a valuable alternative to 68Ga (half-life = 68 min, Eβ+av = 830 keV) for imaging and dosimetry before 177Lu-based radionuclide therapy. The aim of the study was the preclinical evaluation of a folate conjugate labeled with cyclotron-produced 44Sc and its in vitro and in vivo comparison with the 177Lu-labeled pendant. Methods: 44Sc was produced via the 44Ca(p,n)44Sc nuclear reaction at a cyclotron (17.6 ± 1.8 MeV, 50 μA, 30 min) using an enriched 44Ca target (10 mg 44CaCO3, 97.00%). Separation from the target material was performed by a semiautomated process using extraction chromatography and cation exchange chromatography. Radiolabeling of a DOTA-folate conjugate (cm09) was performed at 95°C within 10 min. The stability of 44Sc-cm09 was tested in human plasma. 44Sc-cm09 was investigated in vitro using folate receptor–positive KB tumor cells and in vivo by PET/CT imaging of tumor-bearing mice Results: Under the given irradiation conditions, 44Sc was obtained in a maximum yield of 350 MBq at high radionuclide purity (>99%). Semiautomated isolation of 44Sc from 44Ca targets allowed formulation of up to 300 MBq of 44Sc in a volume of 200–400 μL of ammonium acetate/HCl solution (1 M, pH 3.5–4.0) within 10 min. Radiolabeling of cm09 was achieved with a radiochemical yield of greater than 96% at a specific activity of 5.2 MBq/nmol. In vitro, 44Sc-cm09 was stable in human plasma over the whole time of investigation and showed folate receptor–specific binding to KB tumor cells. PET/CT images of mice injected with 44Sc-cm09 allowed excellent visualization of tumor xenografts. Comparison of cm09 labeled with 44Sc and 177Lu revealed almost identical pharmacokinetics. Conclusion: This study presents a high-yield production and efficient separation method of 44Sc at a quality suitable for radiolabeling of DOTA-functionalized biomolecules. An in vivo proof-of-concept study using a DOTA-folate conjugate demonstrated the excellent features of 44Sc for PET imaging. Thus, 44Sc is a valid alternative to 68Ga for imaging and dosimetry before 177Lu-radionuclide tumor therapy.

We assessed the suitability of the radiolanthanide 155 Tb (t1/2 = 5.32 days, Eγ = 87 keV (32%), 105 keV (25%)) in combination with variable tumor targeted biomolecules using preclinical SPECT imaging. Methods 155Tb was produced at ISOLDE (CERN, Geneva, Switzerland) by high-energy (~ 1.4 GeV) proton irradiation of a tantalum target followed by ionization and on-line mass separation. 155 Tb was separated from isobar and pseudo-isobar impurities by cation exchange chromatography. Four tumor targeting molecules – a somatostatin analog (DOTATATE), a minigastrin analog (MD), a folate derivative (cm09) and an anti-L1-CAM antibody (chCE7) – were radiolabeled with 155 Tb. Imaging studies were performed in nude mice bearing AR42J, cholecystokinin-2 receptor expressing A431, KB, IGROV-1 and SKOV-3ip tumor xenografts using a dedicated small-animal SPECT/CT scanner. Results The total yield of the two-step separation process of 155 Tb was 86%. 155 Tb was obtained in a physiological l-lactate solution suitable for direct labeling processes. The 155 Tb-labeled tumor targeted biomolecules were obtained at a reasonable specific activity and high purity (> 95%). 155 Tb gave high quality, high resolution tomographic images. SPECT/CT experiments allowed excellent visualization of AR42J and CCK-2 receptor-expressing A431 tumors xenografts in mice after injection of 155 Tb-DOTATATE and 155 Tb-MD, respectively. The relatively long physical half-life of 155 Tb matched in particular the biological half-lives of 155 Tb-cm09 and 155 Tb-DTPA-chCE7 allowing SPECT imaging of KB tumors, IGROV-1 and SKOV-3ip tumors even several days after administration. Conclusions The radiolanthanide 155 Tb may be of particular interest for low-dose SPECT prior to therapy with a therapeutic match such as the β--emitting radiolanthanides 177Lu, 161 Tb, 166Ho, and the pseudo-radiolanthanide 90Y.

An accurate and efficient determination of the highly toxic Cr(VI) in solid materials is important to determine the total Cr(VI) inventory of contaminated sites and the Cr(VI) release potential from such sites into the environment. Most commonly, total Cr(VI) is extracted from solid materials following a hot alkaline extraction procedure (US EPA method 3060A) where a complete release of water-extractable and sparingly soluble Cr(VI) phase is achieved. This work presents an evaluation of matrix effects that may occur during the hot alkaline extraction and in the determination of the total Cr(VI) inventory of variably composed contaminated soils and industrial materials (cement, fly ash) and is compared to water-extractable Cr(VI) results. Method validation including multiple extractions and matrix spiking along with chemical and mineralogical characterization showed satisfying results for total Cr(VI) contents for most of the tested materials. However, unreliable results were obtained by applying method 3060A to anoxic soils due to the degradation of organic material and/or reactions with Fe2+-bearing mineral phases. In addition, in certain samples discrepant spike recoveries have to be also attributed to sample heterogeneity. Separation of possible extracted Cr(III) by applying cation-exchange cartridges prior to solution analysis further shows that under the hot alkaline extraction conditions only Cr(VI) is present in solution in measurable amounts, whereas Cr(III) gets precipitated as amorphous Cr(OH)3(am). It is concluded that prior to routine application of method 3060A to a new material type, spiking tests are recommended for the identification of matrix effects. In addition, the mass of extracted solid material should to be well adjusted to the heterogeneity of the Cr(VI) distribution in the material in question.

IL-24 is an unusual member of the IL-10 family, which is considered a Th1 cytokine that exhibits tumor cell cytotoxicity. I describe the purification of this novel cytokine from the supernatant of IL-24 gene transfected human embryonic kidney cells and define the biochemical and functional properties of the soluble, human IL-24 protein. ^ I showed IL-24 non-covalently associates with bovine albumin. Immunoaffinity purification followed by cation exchange chromatography resulted in the significant enrichment of N-glycosylated IL-24. This protein elicited dose-dependent secretion of TNF-α and IL-6 from purified human monocytes and TNF-α secretion from PMA differentiated U937 cells. I showed this same protein was cytotoxic to melanoma tumor cells via the induction of IFN-α. ^ I reported IL-24 associates as at least two disulfide linked, N-glycosylated dimers. Enzymatic removal of N-linked-glycosylation from purified IL-24 partially diminished its cytokine and cytotoxic functions. Disruption of IL-24 dimers via reduction and alkylation of intermolecular disulfide bonds nearly abolished IL-24s cytokine function. ^ I elucidated IL-24 induced TNF-α secretion was pSTAT1, pSTAT3 as well as the class II heterodimeric receptors IL-20R1/IL-22R2 independent. I identified a requirement for the heterodimer of Toll-like Receptors 1 and 2 for IL-24s cytokine function and show a physical interaction between IL-24 and the extracellular domain of TLR-1. ^ Thus, I demonstrated that purified N-glycosylated, soluble, dimeric, human IL-24 exhibits both immunomodulatory and anti-cancer activities and these functions remain associated during purification. IL-24 induced TNF-α secretion required an interaction with the heterodimeric receptor TLR-1/2 and IL-24s cytotoxic affect to melanoma tumor cells was in part due to its induction of IFN-β. ^

Los residuos del sector avícola, principalmente guano (aves ponedoras) y cama de parrilleros (aves de engorde), pueden generar un impacto negativo en el ambiente contribuyendo a la contaminación de suelo, agua y aire. La estabilización aeróbica a través del compostaje es una alternativa de tratamiento para reducir la contaminación. El objetivo de este trabajo fue evaluar el proceso de compostaje en dos mezclas con diferentes porcentajes de residuos avícolas (guano de aves ponedoras y cama de pollos parrilleros). Se compostaron dos mezclas que contenían 81% y 70% de residuos avícolas durante 16 semanas. Las variables analizadas fueron: temperatura (T°), pH, conductividad eléctrica (CE), humedad (H), capacidad de intercambio catiónico (CIC), carbono orgánico total (COT), amonio (NH4+), nitrato (NO3 - ), nitrógeno total (NT ) y carbono soluble (CS). Las características finales de los compost A y B fueron: pH 7,1 - 6,8, CE 3,3 - 2,9 (mS. cm- 1), COT 14,8 - 17,9 %, NT 0,97 - 0,88 %, NH4 + 501 - 144,9 mg kg-1, NO3-552,3 - 543,0 mg kg-1 respectivamente. El proceso de compostaje podría ser una herramienta para estabilizar los residuos avícolas minimizando su impacto en el ambiente.

El objetivo de este trabajo fue comparar el poder de neutralización de enmiendas calcáreo- magnésicas y evaluar su efecto sobre el complejo de intercambio catiónico en suelos de la pampa llana santafesina. Para ello, se aplicaron tres enmiendas (caliza, dolomita y enmienda mezcla) a dos dosis (neutralización del 100% y 150% del hidrógeno de intercambio). Se evaluó: pH actual, pH potencial, hidrógeno, calcio y magnesio intercambiable luego de 60 días de incubación. El pH actual no mostró diferencias entre dosis, mientras que el incremento de pH de las enmiendas fue caliza > dolomita > mezcla. El pH potencial fue más sensible al tipo de enmienda y dosis. Todos los tratamientos redujeron el nivel de insaturación, aunque ninguno neutralizó la totalidad del hidrógeno intercambiable. En relación con el calcio intercambiable, todos los tratamientos produjeron aumentos y la mayor saturación cálcica se obtuvo con la aplicación de caliza o enmienda mezcla con la dosis necesaria para neutralizar 1,5 veces el hidrógeno intercambiable. En cuanto al magnesio, las enmiendas dolomita y mezcla aumentaron significativamente el contenido independientemente de la dosis. La información aquí presentada es de utilidad para la prescripción de enmiendas calcáreo- magnésica en los suelos de la Pampa llana santafesina.

The distribution of Li isotopes in pore waters to a depth of 1157 m below seafloor is presented for ODP Sites 918 and 919 in the Irminger Basin, offshore Greenland. Lithium isotope data are accompanied by strontium isotope ratios to decipher diagenetic reactions in the sediments which are characterized by the pervasive presence of volcanic material, as well as by very high accumulation rates in the upper section. The lowering of the 87Sr/86Sr ratio below contemporaneous seawater values indicates several zones of volcanic material alteration. The Li isotope profiles are complex suggesting a variety of exchange reactions with the solid phases. These include cation exchange with NH4+ and mobilization from sediments at depth, in addition to the alteration of volcanic matter. Lithium isotopes are, therefore, a sensitive indicator of sediment-water interaction. d6Li values of pore waters at these two sites vary between -42 and -25?. At shallow depths (<100 mbsf), rapid decreases in the Li concentration, accompanied by a shift to heavier isotopic compositions, indicate uptake of Li into alteration products. A positive anomaly of d6Li observed at both sites is coincident with the NH4+ maximum produced by organic matter decomposition and may be related to ion exchange of Li from the sediments by NH4+. In the lower sediment column at Site 918, dissolved Li increases with depth and is characterized by enrichment of 6Li. The Li isotopic compositions of both the waters and the solid phase suggest that the enrichment of Li in deep interstitial waters is a result of release from pelagic sediments. The significance of sediment diagenesis and adsorption as sinks of oceanic Li is evaluated. The maximum diffusive flux into the sediment due to volcanic matter alteration can be no more than 5% of the combined inputs from rivers and submarine hydrothermal solutions. Adsorption on to sediments can only account for 5-10% of the total inputs from rivers and submarine hot springs.

Leg 119 of the Ocean Drilling Program (ODP) provided the first opportunity to study the interstitial-water chemistry of the eastern Antarctic continental margin. Five sites were cored in a northwest-southeast transect of Prydz Bay that extended from the top of the continental slope to within 30 km of the coastline. Geological studies of the cores reveal a continental margin that has evolved through terrestrial, glacial, and glacial-marine environments. Chemical and stable isotopic analyses of the interstitial-waters were performed to determine the types of depositional environments and the diagenetic and hydrologic processes that are operating in this unusual marine environment. Highly compacted glacial sediments provide an effective barrier to the vertical diffusion of interstitial-water solutes. Meteoric water from the Antarctic continent appears to be flowing into Prydz Bay sediments through the sequence of terrestrial sediments that lie underneath the glacial sediments. The large amounts of erosion associated with glacial advances appear to have had the effect of limiting the amount of marine organic matter that is incorporated into the sediments on the continental shelf. Although all of the sites cored in Prydz Bay exhibit depletions in dissolved sulfate with increasing depth, the greatest bacterial activity is associated with a thin layer of diatom ooze that coats the seafloor of the inner bay. Results of alkalinity modeling, thermodynamic calculations, and strontium analyses indicate that (1) ocean bottom waters seaward of Site 740 are undersaturated with respect to both calcite and aragonite, (2) interstitial waters at each site become saturated or supersaturated with respect to calcite and aragonite with increasing depth, (3) precipitation of calcium carbonate reduces the alkalinity of the pore waters with increasing depth, and (4) recrystallization of aragonite to calcite accounts for 24% of the pore-water strontium. Weathering of unstable terrestrial debris and cation exchange between clay minerals and pore fluids are the most probable chemical processes affecting interstitial water cation gradients.
