NiAl intermetallic compound was synthesized by mechanical alloying technique in planetary and attritor mills. The starting powders consisted of elemental mixtures of Ni and Al at Ni50Al50(at%) composition. In the planetary mill, compound formation occurred gradually during mechanical alloying, while the occurrence of a mechanically induced self-propagating reaction (MSR) can be suggested in the attritor mill. The NiAl obtained in both mill types was partially disordered with long-range order parameter not inferior to 0.66. Quantitative phase analysis using the Rietveld method was performed in as-milled samples, and this method was also employed to estimate changes in crystallite size and lattice strain of the NiAl produced during mechanical alloying. The objective of this study was to determine the size and composition of atmospheric aerosols in the downtown area of the city of So Paulo, Brazil, for a polluted and an unpolluted period. Aerosols were sampled with a portable air sampler (PAS), Micro-Orifice Uniform Deposit Impactor (MOUDI), and Scanning Mobility Particle Sizer. At the study site, air quality is poor, especially during the winter, high concentrations of pollutants being emitted primarily by the light- and heavy-duty vehicle fleet. We analyzed mass, black carbon (BC), Al, Si, P, S, Cl, K, Ca, Ti, V, Cr, Mn, Fe, Ni, Cu, Zn, Br, Rb, Sn, Zr, and Pb. During the polluted period, diurnal PM(10) was higher than nocturnal PM(10), whereas the inverse was true during the unpolluted period. The FPM was rich in BC, S, and Pb, whereas CPM was rich in Al, Si, Ca, Ti, and Fe. Mass balance was performed by category: ammonium sulfate, sodium chloride, crustal material, BC, and other. The PAS-determined FPM was mainly BC. The MOUDI-determined FPM crustal material explained more mass than did ammonium sulfate and BC during the polluted period, whereas ammonium sulfate had the largest mass during the unpolluted period. Crustal material was the major CPM component, followed by ammonium sulfate and BC. During the unpolluted period, FPM concentrations were lower, whereas those of ammonium sulfate were relatively higher, especially at night, and particle number was inversely proportional to particle size. Aerosol growth was more intense during the polluted period.

Clinical evidence has identified the pulmonary circulation as an important target of air pollution. It was previously demonstrated that in vitro exposure to fine particulate matter (aerodynamic diameter <= 2.5 mu m, PM2.5) induces endothelial dysfunction in isolated pulmonary arteries. We aimed to investigate the effects of in vivo exposure to urban concentrated PM2.5 on rat pulmonary artery reactivity and the mechanisms involved. For this, adult Wistar rats were exposed to 2 weeks of concentrated Sao Paulo city air PM2.5 at an accumulated daily dose of approximately 600 mu g/m(3). Pulmonary arteries isolated from PM2.5-exposed animals exhibited impaired endothelium-dependent relaxation to acetylcholine without significant changes in nitric oxide donor response compared to control rats. PM2.5 caused vascular oxidative stress and enhanced protein expression of Cu/Zn- and Mn-superoxide dismutase in the pulmonary artery. Protein expression of endothelial nitric oxide synthase (eNOS) was reduced, while tumor necrosis factor (TNF)-alpha was enhanced by PM2.5 inhalation in pulmonary artery. There was a significant positive correlation between eNOS expression and maximal relaxation response (E-max) to acetylcholine. A negative correlation was found between vascular TNF-alpha expression and E-max to acetylcholine. Plasma cytokine levels, blood cells count and coagulation parameters were similar between control and PM2.5-exposed rats. The present findings showed that in vivo daily exposure to concentrated urban PM2.5 could decrease endothelium-dependent relaxation and eNOS expression on pulmonary arteries associated with local high TNF-alpha level but not systemic pro-inflammatory factors. Taken together, the present results elucidate the mechanisms underlying the trigger of cardiopulmonary diseases induced by urban ambient levels of PM2.5. (C) 2012 Elsevier Ireland Ltd. We present a detailed study of carbon-enhanced metal-poor (CEMP) stars, based on high-resolution spectroscopic observations of a sample of 18 stars. The stellar spectra for this sample were obtained at the 4.2 m William Herschel Telescope in 2001 and 2002, using the Utrecht Echelle Spectrograph, at a resolving power R similar to 52 000 and S/N similar to 40, covering the wavelength range lambda lambda 3700-5700 angstrom. The atmospheric parameters determined for this sample indicate temperatures ranging from 4750 K to 7100 K, log g from 1.5 to 4.3, and metallicities -3.0 <= [Fe/H]<=-1.7. Elemental abundances for C, Na, Mg, Sc, Ti, Cr, Cu, Zn, Sr, Y, Zr, Ba, La, Ce, Nd, Sm, Eu, Gd, Dy are determined. Abundances for an additional 109 stars were taken from the literature and combined with the data of our sample. The literature sample reveals a lack of reliable abundance estimates for species that might be associated with the r-process elements for about 67% of CEMP stars, preventing a complete understanding of this class of stars, since [Ba/Eu] ratios are used to classify them. Although eight stars in our observed sample are also found in the literature sample, Eu abundances or limits are determined for four of these stars for the first time. From the observed correlations between C, Ba, and Eu, we argue that the CEMP-r/s class has the same astronomical origin as CEMP-s stars, highlighting the need for a more complete understanding of Eu production.

Background: The majority of studies have investigated the effect of exercise training (TR) on vascular responses in diabetic animals (DB), but none evaluated nitric oxide (NO) and advanced glycation end products (AGEs) formation associated with oxidant and antioxidant activities in femoral and coronary arteries from trained diabetic rats. Our hypothesis was that 8-week TR would alter AGEs levels in type 1 diabetic rats ameliorating vascular responsiveness. Methodology/Principal Findings: Male Wistar rats were divided into control sedentary (C/SD), sedentary diabetic (SD/DB), and trained diabetic (TR/DB). DB was induced by streptozotocin (i.p.: 60 mg/kg). TR was performed for 60 min per day, 5 days/week, during 8 weeks. Concentration-response curves to acetylcholine (ACh), sodium nitroprusside (SNP), phenylephrine (PHE) and tromboxane analog (U46619) were obtained. The protein expressions of eNOS, receptor for AGEs (RAGE), Cu/Zn-SOD and Mn-SOD were analyzed. Tissues NO production and reactive oxygen species (ROS) generation were evaluated. Plasma nitrate/nitrite (NOx-), superoxide dismutase (SOD), catalase (CAT), thiobarbituric acid reactive substances (TBARS) and N-epsilon-(carboxymethyl) lysine (CML, AGE biomarker). A rightward shift in the concentration-response curves to ACh was observed in femoral and coronary arteries from SD/DB that was accompanied by an increase in TBARS and CML levels. Decreased in the eNOS expression, tissues NO production and NOx- levels were associated with increased ROS generation. A positive interaction between the beneficial effect of TR on the relaxing responses to ACh and the reduction in TBARS and CML levels were observed without changing in antioxidant activities. The eNOS protein expression, tissues NO production and ROS generation were fully re-established in TR/DB, but plasma NOx- levels were partially restored. Conclusion: Shear stress induced by TR fully restores the eNOS/NO pathway in both preparations from non-treated diabetic rats, however, a massive production of AGEs still affecting relaxing responses possibly involving other endothelium-dependent vasodilator agents, mainly in coronary artery.

Abstract Background Obesity has been associated with a variety of disease such as type II diabetes mellitus, arterial hypertension and atherosclerosis. Evidences have shown that exercise training promotes beneficial effects on these disorders, but the underlying mechanisms are not fully understood. The aim of this study was to investigate whether physical preconditioning prevents the deleterious effect of high caloric diet in vascular reactivity of rat aortic and mesenteric rings. Methods Male Wistar rats were divided into sedentary (SD); trained (TR); sedentary diet (SDD) and trained diet (TRD) groups. Run training (RT) was performed in sessions of 60 min, 5 days/week for 12 weeks (70–80% VO2max). Triglycerides, glucose, insulin and nitrite/nitrate concentrations (NOx-) were measured. Concentration-response curves to acetylcholine (ACh) and sodium nitroprusside (SNP) were obtained. Expression of Cu/Zn superoxide dismutase (SOD-1) was assessed by Western blotting. Results High caloric diet increased triglycerides concentration (SDD: 216 ± 25 mg/dl) and exercise training restored to the baseline value (TRD: 89 ± 9 mg/dl). Physical preconditioning significantly reduced insulin levels in both groups (TR: 0.54 ± 0.1 and TRD: 1.24 ± 0.3 ng/ml) as compared to sedentary animals (SD: 0.87 ± 0.1 and SDD: 2.57 ± 0.3 ng/ml). On the other hand, glucose concentration was slightly increased by high caloric diet, and RT did not modify this parameter (SD: 126 ± 6; TR: 140 ± 8; SDD: 156 ± 8 and TRD 153 ± 9 mg/dl). Neither high caloric diet nor RT modified NOx- levels (SD: 27 ± 4; TR: 28 ± 6; SDD: 27 ± 3 and TRD: 30 ± 2 μM). Functional assays showed that high caloric diet impaired the relaxing response to ACh in mesenteric (about 13%), but not in aortic rings. RT improved the relaxing responses to ACh either in aortic (28%, for TR and 16%, to TRD groups) or mesenteric rings (10%, for TR and 17%, to TRD groups) that was accompanied by up-regulation of SOD-1 expression and reduction in triglycerides levels. Conclusion The improvement in endothelial function by physical preconditioning in mesenteric and aortic arteries from high caloric fed-rats was directly related to an increase in NO bioavailability to the smooth muscle mostly due to SOD-1 up regulation.

Background: Reactive oxygen species (ROS) are formed under natural physiological conditions and are thought to play an important role in many human diseases. A wide range of antioxidants are involved in cellular defense mechanisms against ROS, which can be generated in excess during stressful conditions, these include enzymes and non-enzymatic antioxidants. The aim of this study was to evaluate the antioxidant responses of mice to two diets control, commercial and the purified AIN 93 diet, commonly used in experiments with rodents. Results: Malondialdehyde (MDA) and hydrogen peroxide (H2O2) concentrations and superoxide dismutase (SOD) and glutathione reductase (GR) activities determined in the liver were lower in the group of mice fed with the AIN 93 diet, while catalase (CAT) activity was higher in the same group, when compared to the group fed on the commercial diet. Liver glutathione peroxidase (GSH-Px) activity was similar in the groups fed on either AIN 93 or the commercial diets. Two SOD isoforms, Mn-SODII and a Cu/Zn-SODV, were specifically reduced in the liver of the AIN 93 diet fed animals. Conclusions: The clear differences in antioxidant responses observed in the livers of mice fed on the two diets suggest that the macro- and micro-nutrient components with antioxidant properties, including vitamin E, can promote changes in the activity of enzymes involved in the removal of the ROS generated by cell metabolism.

Waste products from the forest industry are to be spread in forests in Sweden to counteract nutrient depletion due to whole tree harvesting. This may increase the bioavailability of calcium (Ca) and heavy metals, such as cadmium (Cd), copper (Cu) and zinc (Zn) in forest soils. Heavy metals, like Cd, have already been enriched in forest soils in Sweden, due to deposition of air pollutions, and acidification of forest soils has increased the bioavailability of toxic metals for plant uptake. Changes in the bioavailability of metals may be reflected in altered accumulation of Ca and heavy metals in forest trees, changes in tree growth, including wood formation, and altered tree species composition. This thesis aims at examining: A) if inter- or intra- specific differences in sensitivity to Cd occur in the most common tree species of Sweden, and if so, to study if these can be explained by the uptake and distribution of Cd within the plant: B) how elevated levels of Ca, Cd, Cu and Zn affect the accumulation and attachment of metals in bark and wood, and growth of young Norway spruce (Picea abies): C) how waste products from the forest industry, such as wood ash, influence the contents of Ca, Cd, Cu and Zn in wood and bark of young Norway spruce. Sensitivity to Cd, and its uptake and distribution, in seedlings of Picea abies, Pinus sylvestris and Betula pendula from three regions (southern, central and northern parts) of Sweden, treated with varying concentrations of Cd, were compared. Differences in root sensitivity to Cd both among and within woody species were found and the differences could to some extent be explained by differences in uptake and translocation of Cd. The root sensitivity assays revealed that birch was the least, and spruce the most, sensitive species, both to the external and to tissue levels of Cd. The central ecotype of the species tested tended to be most Cd resistant. The radial distribution, accumulation and attachment of, and interactions between Ca and heavy metals in stems of two-year-old Norway spruce trees treated with elevated levels of Cd, Cu, Zn and/or Ca, were investigated. Further, the influence of these metals on growth, and on root metal content, was examined. Accumulation of the metals was enhanced in wood, bark and/or roots at elevated levels of the metal in question. Even at low levels of the metals, similar to after application of wood ash, an enhanced accumulation was apparent in wood and/or bark, except for Cd. The increased accumulation of Zn and Cu in the stem did not affect the growth. However, Cu decreased the accumulation of Ca in wood. Higher levels of Cu and Cd reduced the stem diameter and the toxic effect was associated with a reduced Ca content in wood. Copper and Cd also decreased the accumulation of Zn in the stem. On the other hand, elevated levels of Ca increased the stem diameter and reduced the accumulation of Cd, Cu, Zn and Mn in wood and/or bark. When metals interacted with each other the firmly bound fraction of the metal reduced was in almost all cases not affected. As an exception, Cd decreased the firmly bound fraction of Zn in the stem. The influence of pellets of wood ash (ash) or a mixture of wood ash and green liquor dregs (ash+GLD), in the amount of 3000 kg ha-1, on the contents of Ca, Cd, Cu and Zn in wood and bark of young Norway spruce in the field was examined. The effect of the treatments on the metal content of bark and wood was larger after 3 years than after 6 years. Treatment with ash+GLD had less effect on the heavy metal content of bark and wood than treatment with ash alone. The ash treatment increased the Cu and Zn content in bark and wood, respectively, after 3 years, and decreased the Ca content of the wood after 6 years. The ash+GLD treatment increased the Ca content of the bark and decreased the Zn content of bark and wood after 3 years. Both treatments reduced, or tended to decrease, the Cd content in wood and bark at both times. To conclude, small changes in the bioavailability of Ca, Cu, Cd and Zn in forest soils, such as after spreading pellets of wood ash or a mixture of wood ash and green liquor dregs from the forest industry, will be reflected in an altered accumulation of metals in wood and bark of Norway spruce. It will not only be reflected in changed accumulation of those metals in which bioavailability in the soil has been enhanced, but also of other metals, probably partly due to interactions between metals. When metals interact the exchangeable bound fraction of the metal reduced is suggested to be the main fraction affected. The small alterations in accumulation of metals should not affect the growth of Norway spruce, especially since the changes in accumulation of metals are low, and further since these decrease over time. However, as an exception, one positive and maybe persistent effect of the waste products is that these may decrease the accumulation of Cd in Norway spruce, which partly may be explained by competition with Ca for uptake, translocation and binding. A decreased accumulation of Cd in Norway spruce will probably affect the trees positively, since Norway spruce is one of the most sensitive species to Cd of the forest trees in Sweden. Thus, spreading of waste products from the forest industry may be a solution to decrease the accumulation of Cd in Norway spruce. In a longer perspective, this will decrease the risk of Cd altering the tree species composition of the forest ecosystem. An elevated bioavailability of Ca in forest soils will, in addition to Cd, probably also decrease the accumulation of other less competitive heavy metals, like Zn and Mn, in the stem.

L’idrogeno è un elemento di elevato interesse economico, con una produzione industriale che supera i 55 x 1010 m3/anno e notevoli prospettive di sviluppo delle sue applicazioni. Attualmente l’idrogeno è prodotto principalmente in impianti di larga scala (circa 1000 m3/h) da combustibili fossili attraverso processi di steam reforming ed ossidazione parziale catalitica. Per aumentare la produzione di idrogeno un ruolo fondamentale è svolto dalla reazione di water gas shift (WGS) che abbatte il contenuto di CO, massimizzando la produzione di idrogeno. La reazione è condotta industrialmente in due stadi, operanti ad alta temperatura (HTS, circa 350 °C) e bassa temperatura (LTS, circa 250 °C), utilizzando rispettivamente catalizzatori a base di ferro o rame. Tuttavia, è evidente l’interesse per nuove formulazioni in grado di operare in un unico stadio a temperatura intermedia (MTS), mantenendo le caratteristiche ottimali di attività e stabilità. In questo lavoro di tesi, condotto in collaborazione con AIR LIQUIDE (F), è stato affrontato uno studio della reazione di WGS finalizzato allo sviluppo di nuove formulazioni attive e stabili nell’MTS. In particolare, sono stati sintetizzati precursori idrotalcitici Cu/Zn/Al (contenenti carbonati o silicati), con bassi contenuti di rame (diversamente da quanto riportato in letteratura), modulandone le proprietà chimico-fisiche, l’attività catalitica e la stabilità con il tempo di reazione. Si è osservato come i catalizzatori con minori contenuti di rame ed ottenuti da precursori contenenti carbonati mostrassero un’elevata attività e selettività nell’MTS, raggiungendo valori di conversione del CO analoghi a quelli all’equilibrio termodinamico già a 300 °C, indipendentemente dai valori del rapporto S/DG e del tempo di contatto. Tutti i catalizzatori mostrano un’elevata stabilità con il tempo di reazione, con incrementi del quantitativo del CO in uscita dopo 100h di circa lo 0,7 % v/v. I catalizzatori scaricati dopo le prove catalitiche evidenziano gli effetti dei processi di sinterizzazione (diminuzione dell’area superficiale ed incremento delle dimensioni dei cristalliti), la cui entità diminuisce al diminuire del contenuto di rame. Infine, confrontando l’attività dei migliori catalizzatori preparati in questo lavoro di tesi con quella di uno dei più utilizzati catalizzatori commerciali per la reazione di WGS a bassa temperatura, si sono osservati valori di attività analoghi, raggiungendo quelli di equilibrio per temperature 300°C, ma con una attività significativamente superiore nelle condizioni LTS, soprattutto considerando il valore del tempo di contatto inferiore a quelli comunemente utilizzati negli impianti industriali.

La contaminazione da fonti diffuse e locali può determinare perdite di funzionalità del suolo e ripercuotesi a cascata nelle catene alimentari, attraverso la contaminazione di falde e corpi d'acqua, l'assorbimento da parte dei vegetali e degli animali fino all'uomo. Industrializzazione, urbanizzazione e pratiche agricole sono le tre principali sorgenti di metalli nei suoli. In questo lavoro di tesi, che ha avuto come area oggetto di studio parte del territorio del Comune di Ravenna, sono stati analizzati gli arricchimenti di Cr, Ni, Cu, Zn e Pb applicando una metodica impostata sul confronto tra le concentrazioni totali rinvenute in superficie e in profondità mediante il calcolo dell'Indice di Geoaccumulo. La sola determinazione delle aliquote dei metalli negli orizzonti superficiali del suolo, non è, di per sé, in grado di fornire indicazioni esaustive circa lo stato di contaminazione dei suoli. Infatti, tale informazione, non permette la distinzione tra origine naturale o arricchimento determinato da attività antropica. In più lo studio ha valutato la possibile relazione con le caratteristiche del substrato (origine del sedimento) e l'uso del suolo. Alte concentrazioni di Cr e Ni, in corrispondenza della Pineta San Vitale (area naturale) non sono attribuibili a contaminazioni di natura antropica, bensì dipendono dal parent material. La distribuzione delle anomalie per Cu, Zn e Pb, invece, sembra dipendere invece dall'uso dei suoli presenti nell'area oggetto di studio. Il contenuto naturale di questi metalli può variare notevolmente a seconda del materiale su cui si è sviluppato il suolo e questo riflette i tenori di fondo per le diverse unità. I tenori di fondo del Cr calcolati per il deposito di cordone litorale (DCL), dove oggi è impostata la Pineta San Vitale (area naturale), sono maggiori rispetto ai limiti di concentrazione previsti dal Decreto Legislativo n°152 del 2006 per questa tipologia di aree. Cu, Zn e Pb, invece, presentano tenori di fondo bassi in corrispondenza del deposito del cordone litorale (DCL), costituito da materiale a granulometria grossolana. Per questi metalli è il fattore granulometria ad avere un ruolo fondamentale nello stabilire i valori di background negli altri depositi. In ultima analisi, per valutare gli effetti e i rischi associati in presenza di alte concentrazioni di Cr, Ni, Cu, Zn e Pb, è stata quantificata la frazione che può essere resa biodisponibile con DTPA. Le anomalie riscontrate sono state confrontate in primo luogo con alcuni campioni di controllo, coerenti con i valori di background per i cinque metalli. La bassa biodisponibilità osservata per Cr e Ni riflette il forte legame di questi metalli con la fase mineralogica, non rappresentando un pericolo per le piante e di conseguenza per gli animali superiori. Cu, Zn e Pb presentano, in alcuni casi, concentrazioni biodisponibili sia proporzionali all'entità dell'anomalia evidenziata con il calcolo dell'Indice di Geoaccumulo, sia maggiori rispetto a quelle dei campioni selezionati come controlli.

Bei der amyotrophen Lateralsklerose 1 (ALS1) handelt es sich um eine altersabhängige Motoneuronenerkrankung, die durch Mutationen im Gen der Cu/Zn-Superoxid Dismutase (hSOD1mut) ausgelöst wird. Die toxischen Eigen¬schaften von hSOD1mut (z. B. Aggregations- oder oxidative Stress-Hypothese) und der Einfluss wildtypischer hSOD1 (hSOD1WT) auf den Krankheitsverlauf sind weithin ungeklärt. Das Ziel dieser Arbeit war es, die Auswirkungen von hSOD1mut-hSOD1WT-Heterodimeren im Vergleich zu mutanten Homodimeren auf die Pathogenese der ALS1 zu untersuchen. Nachdem gezeigt werden konnte, dass es in humanen Zellen in der Tat zu einer Bil¬dung hetero- und homodimerer mutanter hSOD1-Spezies kommt, wurden Dimerfusionsproteine aus zwei hSOD1-Monomeren generiert, die durch einen flexiblen Peptidlinker verbunden und C-terminal mit eGFP markiert waren. Neben hSOD1WT-WT wurden hSOD1mut-mut- und hSOD1mut-WT-Dimere mit vier verschiedenen hSOD1-Mu¬tanten untersucht. Die biochemische Charakterisierung zeigte, dass alle Dimere, die wildtyp-ähnliche hSOD1mut enthielten, eine Dismutaseaktivität aufwiesen. Im Gegensatz dazu war das Homodimer aus zwei metalldefizienten hSOD1G85R inaktiv, wobei interessanterweise hSOD1G85R mit hSOD1WT ein Dismutase-aktives Dimer bilden konnte. Sowohl in Zellkultursystemen als auch in einem C. elegans-Modell bildeten alle mutanten Homodimere vermehrt Aggregate im Vergleich zu den dazugehörigen Heterodimeren. Dieses Aggregationsverhalten korrelierte aber nicht mit der Toxizität der Dimerproteine in Überlebensassays und einer C. elegans Bewe¬gungs¬analyse. In diesen funktionellen Studien assoziierte die Toxizität der dimeren Fusionsproteine mit der enzy¬matischen Aktivität. In Übereinstimmung mit diesen Ergebnissen konnte gezeigt werden, dass hSOD1WT nicht in hSOD1mut-abhängigen Aggregaten vorkommt. Die Ergebnisse dieser Studie sprechen gegen die Aggregation als primäre toxische Eigen¬schaft der hSOD1mut und unterstützen die oxidative Stress-Hypothese. Dis¬mutase-inaktive hSOD1mut können eine untypische Enzymaktivität durch die Heterodimerisierung mit hSODWT erlangen, die auf diese Weise maßgeblich an der Pathogenese der ALS1 beteiligt sein könnte.

Negli ultimi anni l’interesse nei confronti dell’H2 è cresciuto notevolmente per l’aumento della richiesta energetica mondiale. Uno dei processi più importanti per la produzione di H2 utilizza la reazione di Water-Gas Shift (WGS) per il trattamento delle correnti in uscita dai processi di steam reforming o di ossidazione parziale catalitica. CO + H2O CO2 + H2 ∆H0298 = -41,2 KJ/mol Sono quindi stati sviluppati sistemi catalitici attivi nella reazione di WGS a media temperatura (circa 300 °C). Partendo da sistemi catalitici a base di Cu/Zn/Al, ottenuti da precursori idrotalcitici e sviluppati in lavori di tesi precedenti, sono state effettuate modifiche nella composizione al fine di aumentarne l’attività e la stabilità. L’aggiunta di piccole quantità di Mg ha un effetto positivo sull’attività dei sistemi catalitici, con effetti più evidenti a 250 °C. Tuttavia, l’aumento del contenuto di Mg, sebbene migliori le proprietà fisiche del catalizzatore (area superficiale e dispersione del Cu) sia del campione calcinato che di quello scaricato dopo reazione, peggiora drasticamente l’attività catalitica. L’aggiunta di piccole quantità di Mg sembra portare alla stabilizzazione della specie attiva Cu+ e promuovere un meccanismo redox superficiale (Cu0 e Cu+). E’ possibile correlare la conversione del CO con il rapporto ZnO/Cu, confermando il ruolo nella reazione di WGS dell’interazione Cu0/ZnO libero. La sostituzione di Mg con Ba comporta un miglioramento delle prestazioni catalitiche, in particolare nelle condizioni MTS (300 °C), suggerendo una più facile dissociazione dell’acqua legata alla stabilizzazione degli ossidrili da parte dei siti basici. È però accompagnato da una diminuzione della stabilità nelle condizioni di reazione. L’aggiunta di piccole quantità di La, Ce o Zr (con un rapporto Al/R = 50 mol/mol) incrementa la stabilità termica, sia in termini di proprietà fisiche che di attività catalitica. A seguito dei cicli termici di invecchiamento accelerato, infatti, non si riscontrano importanti diminuzioni di attività catalitica, evidenziando un’elevata stabilità della fase attiva.

Die vorliegende Dissertation beinhaltet Untersuchungen zur Expression und Funktion der respiratorischen Proteine Neuroglobin (Nbg) und Cytoglobin (Cygb) in Vertebraten. rnrnUm die Expression der Globine während der Entwicklung des Säugerhirns zu untersuchen, wurden die Hirne von Maus-Embryonen ab dem Fötalstadium MF10 bis zum Tag eins nach der Geburt (T1) mit Adulttieren verglichen. Quantifiziert wurde sowohl die mRNA- als auch die Protein-Expression. Beide Globine zeigten im Verlauf der Entwicklung einen stetigen Anstieg der mRNA-Expression, wobei Ngb zu Beginn in zehnfach höherer Konzentration vorlag und im zeitlichen Verlauf einen 130-fachen Anstieg zeigte. Cygb zeigte lediglich einen 16-fachen Anstieg bis zum Adultstadium. Auf Proteinebene konnte die Expressionszunahme beider Globine im Laufe der Entwicklung bestätigt werden. Weder in den hypoxieresistenten Frühembryonalstadien noch während der mit Sauerstoff-Stress verbundenen Geburt zeigte sich ein Expressionsmaximum. Dies spricht gegen eine Globin-Funktion in der Oxidanz-Abwehr. Eher ist zumindest Ngb mit der Reifung der Neurone und dem damit einhergehenden, gesteigerten oxidativen Stoffwechsel assoziiert.rnrnDes Weiteren sollte die zelluläre und intrazelluläre Lokalisation beider Globine anhand einer primären Zellkultur aus dem Hippocampus pränataler Ratten und in immortalen Zelllinien untersucht werden. Neuroglobin wurde dabei nur in Neuronen, nicht jedoch in Gliazellen nachgewiesen. Das Färbemuster war in allen Ngb-exprimierenden Zellen zytoplasmatisch. Cytoglobin wurde in der Primärkultur in den Neuronen jedoch ebenso in den mit anti-GFAP markierten Gliazellen beobachtet. In beiden Zellpopulationen war auch der Kern durch das CyGB-Antiserum markiert. rnEine genauere Untersuchung der intrazellulären Lokalisation sollte durch die Transfektion von Globin-pEGFP-Fusionsproteinen erfolgen. Nach Transfektion der Fusionskonstrukte wurde die GFP-Färbung bei beiden Globinen sowohl im Zytoplasma als auch im Kern beobachtet. Eine rein nukleäre Lokalisation, die insbesondere für Cygb von anderen Autoren postuliert wurde, konnte somit ausgeschlossen werden. rnrnIn primären Zellkulturen aus Cerebellum und Kortex, die mit Hilfe von Paraquat oxidativem Stress ausgesetzt wurden, wurde der Verlauf der Globin-mRNA-Expression mit dem unregulierten 18s rRNA-Referenzgen und mit den Antioxidanz-Enzymen Cu-Zn-SOD und Gpx verglichen. Neuroglobin zeigte einen Expressionsverlauf ähnlich dem der beiden Antioxidanz-Enzyme, jedoch liegt seine mRNA im Hirngewebe in hundertfach niedrigerer Menge als Cu-Zn-SOD und Gpx vor. Cytoglobin zeigte keine Veränderung der Expression. Eine Funktion der Globine im Sinne einer ROS-Abwehr kann aus den Befunden nicht abgeleitet werden. rnrnUntersuchungen von Tumor und Normalgewebe mittels eines cDNA-Cancer-Arrays zeigten, dass NGB in Tumoren verschiedenen Ursprungs nicht exprimiert wird, CyGB dagegen keine Änderung seiner Expression in Tumor versus Normalgewebe erfährt. Eine Induktion der beiden Globine z.B. durch Hypoxie in soliden Tumoren kann daher ausgeschlossen werden.rn

Per massimizzare la quantità di H2 nel gas di sintesi ottenuto dal processo di Steam Reforming (SR) si utilizza la reazione di Water Gas Shift (WGS): CO + H2O ⇆ CO2 + H2 ∆H0298 = - 41,2 KJ/mol Sulla base di sistemi catalitici Cu/Zn/Al si è cercato di modificarne la composizione per ottenere catalizzatori attivi e stabili in un intervallo di temperatura tra 350-450 °C, (High Temperature Shift o HTS), al fine di sostituire i tradizionali sistemi a base di Fe/Cr, in relazione alle limitazioni ambientali all’utilizzo del Cr e per poter operare con valori inferiori del rapporto S/DG. Si sono inoltre studiate le caratteristiche dei catalizzatori e le condizioni di reazione che favoriscono la produzione di metilammine ed alcoli nel processo a temperature intermedie, tra 300-350 °C (Middle Temperature Shift o MTS), in relazione alla disattivazione che questi composti comportano nel processo di SR a seguito del riciclo della fase acquosa dal reattore di WGS.

The blood-brain barrier (BBB) and the blood-spinal cord barrier (BSCB) separate the brain and the spinal cord from the circulating blood and are important for the maintenance of the CNS homeostasis. They build a physical barrier thereby protecting the CNS from pathogens and toxic agents, and their disruption plays a crucial role in the pathogenesis of several CNS disorders. In this thesis, the blood-CNS-barriers were studied via in vitro models in two case studies for neurodegenerative disorders, in particular Alzheimer’s disease (AD) and amyotrophic lateral sclerosis (ALS). The first model evaluates treatment possibilities of AD using nanotechnology-based strategies. Since the toxic amyloid-β42 (Aβ42) peptide plays a crucial role in the pathogenesis of AD, reduced generation or enhanced clearance of Aβ42 peptides are expected to modify the disease course in AD. Therefore, several Aβ42-lowering drugs like flurbiprofen had been tested in clinical trials, but most of them failed due to their low brain penetration. Here, flurbiprofen was embedded in polylactide (PLA) nanoparticles and its transport was examined in an in vitro BBB model. The embedding of flurbiprofen into the nanoparticles disguised its cytotoxic potential and enabled the administration of higher drug concentrations which resulted in a sufficient transport of the drug across an endothelial cell monolayer. These results demonstrate that non-permeable drugs can be transported efficiently via nanoparticles and that these nanotechnology-based strategies are a promising tool to generate novel therapeutic options for AD and other CNS diseases. rnThe focus of the second project was to investigate the impaired integrity of the BSCB in a mouse model for ALS. About 20% of all familial ALS cases are associated with missense mutations or small deletions in the gene that encodes Cu/Zn-superoxide dismutase 1 (SOD1). To date, the molecular mechanisms resulting in ALS are still unknown, but there is evidence that the disruption of the BSCB is one of the primary pathological events. In both familial and sporadic ALS patients, loss of endothelial integrity and endothelial cell damage was observed, and studies with SOD1 transgenic mice demonstrated that the BSCB disruption was found prior to motor neuron degeneration and neurovascular inflammation. Thus, an in vitro model for ALS endothelial cells was generated which exhibited comparable integrity characteristics and tight junction (TJ) protein expression profiles as isolated primary endothelial cells of the BSCB of SOD1 transgenic mice. In this, an alteration of the βcat/AKT/FoxO1 pathway, which regulates the expression of the TJ protein claudin-5, could be observed. These data furthermore indicate that ALS is a neurovascular disease, and understanding of the primary events in ALS pathogenesis will hopefully provide ideas for the development of new therapeutic strategies. rn
