Epigenetic silencing of tumor suppressor genes by DNA hypermethylation at promoter regions is a common event in carcinogenesis and tumor progression. Abrogation of methylation and reversal of epigenetic silencing is a very potent way in cancer treatment. However, the reactivation mechanisms are poorly understood. In this study, we first developed a cell line model system named YB5, derived from SW48 cancer cell line, which bears one copy of stably integrated EGFP gene on Chromosome 1p31.1 region. The GFP gene expression is transcriptionally silenced due to the hypermethylated promoter CMV. However, the GFP expression can be restored using demethylating agent 5-aza-2' deoxycytidine (DAC), and detected by FACS and fluorescent microscopy. Using this system, we observed the heterogeneous reactivation induced by DAC treatment. After flow sorting, GFP negative cells exhibited similar level of incomplete demethylation compared to GFP positive cells on repetitive LINE1 element, tumor suppressor genes such as P16, CDH13, and RASSF1a, and CMV promoter as well. However, the local chromatin of CMV-GFP locus altered to an open structure marked by high H3 lysine 9 acetylation and low H3 lysine 27 tri-methylation in GFP positive cells, while the GFP negative cells retained mostly the original repressive marks. Thus, we concluded that DAC induced DNA hypomethylation alone does not directly determine the level of re-expression, and the resetting of the local chromatin structure under hypomethylation environment is required for gene reactivation. Besides, a lentivirus vector-based shRNA screening was performed using the YB5 system. Although it is the rare chance that vector lands in the neighboring region of GFP, we found that the exogenous vector DNA inserted into the upstream region of GFP gene locus led to the promoter demethylation and reactivated the silenced GFP gene. Thus, epigenetic state can be affected by changing of the adjacent nucleic acid sequences. Further, this hypermethylation silenced system was utilized for epigenetic drug screening. We have found that DAC combined with carboplatin would enhance the GFP% yield and increase expression of other tumor suppressor genes than DAC alone, and this synergistic effect may be related to DNA repair process. In summary, these studies reveal that reversing of methylation silencing requires coordinated alterations of DNA methylation, chromatin structure, and local microenvironment. ^

Transglutaminases are a family of enzymes that catalyze the covalent cross-linking of proteins through the formation of $\varepsilon$-($\gamma$-glutaminyl)-lysyl isopeptide bonds. Tissue transglutaminase (Tgase) is an intracellular enzyme which is expressed in terminally differentiated and senescent cells and also in cells undergoing apoptotic cell death. To characterize this enzyme and examine its relationship with other members of the transglutaminase family, cDNAs, the first two exons of the gene and 2 kb of the 5$\sp\prime$ flanking region, including the promoter, were isolated. The full length Tgase transcript consists of 66 bp of 5$\sp\prime$-UTR (untranslated) sequence, an open reading frame which encodes 686 amino acids and 1400 bp of 3$\sp\prime$-UTR sequence. Alignment of the deduced Tgase protein sequence with that of other transglutaminases revealed regions of strong homology, particularly in the active site region.^ The Tgase cDNA was used to isolate and characterize a genomic clone encompassing the 5$\sp\prime$ end of the mouse Tgase gene. The transcription start site was defined using genomic and cDNA clones coupled with S1 protection analysis and anchored PCR. This clone includes 2.3 kb upstream of the transcription start site and two exons that contain the first 256 nucleotides of the mouse Tgase cDNA sequence. The exon intron boundaries have been mapped and compared with the exon intron boundaries of three members of the transglutaminase family: human factor XIIIa, the human keratinocyte transglutaminase and human erythrocyte band 4.1. Tissue Tgase exon II is similar to comparable exons of these genes. However, exon I bears no resemblance with any of the other transglutaminase amino terminus exons.^ Previous work in our laboratory has shown that the transcription of the Tgase gene is directly controlled by retinoic acid and retinoic acid receptors. To identify the region of the Tgase gene responsible for regulating its expression, fragments of the Tgase promoter and 5$\sp\prime$-flanking region were cloned into the chloramphenicol actetyl transferase (CAT) reporter constructs. Transient transfection experiments with these constructs demonstrated that the upstream region of Tgase is a functional promoter which contains a retinoid response element within a 1573 nucleotide region spanning nucleotides $-$252 to $-$1825. ^

El artículo presenta al autor de Ariel, Rodó, como integrante de la promoción literaria uruguaya, como uno de los exponentes del modernismo y como pensador que pertenece a la generación de los Fundadores. Parte de la problemática Ariel- Calibán como símbolo latinoamericano y de la postulación de Ariel como discurso antiimperialista para situar la pregunta respecto de la función de la educación en la constitución del joven latinoamericano como sujeto social portador de sentido del proyecto nacional. Se tratan luego las figuras que se desprenden de los Prósperos, los Calibanes y los Arieles concebidos como la pluralidad de perspectivas desde las que transcurren las vidas de los sujetos y de los espacios desde los cuales las naciones se paran para mirarse unas a otras.

Este trabajo se enfoca en la doctrina que Tomás de Aquino establece con respecto al sentido del gusto, estructurándose en tres partes. En primer lugar, se discute la ubicación que el autor otorga al gusto dentro del esquema de los cinco sentidos externos a partir de algunos aportes contemporáneos. Luego, se expone la relación que establece el Aquinate entre el gusto y sus objeto propio, el sabor, con todas las implicancias de orden biológico y antropológico que conlleva. Finalmente, se trata la valencia moral del sentido del gusto a partir de uno de los vicios capitales que se engendra en él, la gula, y la originalidad que Tomás aporta a esta discusión con respecto a la doctrina cristiana anterior.

Escrito en el apogeo de la Revolución Cubana, “Tengo," que comparte el nombre del libro donde aparece, es una celebración eufórica del triunfo del hombre común y corriente. También documenta el punto de cambio de la doxa, al mostrar cómo “Juan sin nada" se convirtió en “Juan con todo". Esta investigación utiliza la teoría bourdieusiana para documentar cómo los cambios sociales deben aparecer en el nivel de habitus antes de manifestarse en el nivel físico. Este fallo tiene consecuencias muy importante para el poeta como “maestro de memes".

Serpentinite seamounts in the Mariana forearc have been explained as diapirs rising from the Benioff zone. This hypothesis predicts that the serpentinites should have low strengths as well as low densities relative to the surrounding rocks. Drilling during Leg 125 showed that the materials forming Conical Seamount in the Mariana forearc and Torishima Forearc Seamount in the Izu-Bonin forearc are water-charged serpentinite muds of density <2 g/cm**3. Wykeham-Farrance torsion-vane tests showed that they are plastic solids with a rheology that bears many similarities to the idealized Cam clay soil model and is well described by critical-state soil mechanics. The serpentinite muds have ultimate strengths of 1.3 to 273.7 kPa and yield strengths of approximately 1.0 to 50 kPa. These muds thus are orders of magnitude weaker than salt and are, in fact, comparable in density and strength to common deep-sea clay muds. Such weak and low-density materials easily become diapiric. Serpentinite muds from the summit of Conical Seamount are weaker and more ductile than those on its flanks or on Torishima Forearc Seamount. Moreover, the summit muds do not contain the pronounced pure- and simple-shear fabrics that characterize those on the seamount flanks. The seamounts are morphologically similar to shield volcanoes, and anastomosing serpentinite debris flows descending from their summits are similar in map view to pahoehoe flows. These morphologic features, together with the physical properties of the muds and their similarities to other oceanic muds and the geochemistry of the entrained waters, suggest that many forearc serpentinite seamounts are gigantic (10-20 km wide, 1.5-2.0 km high) mud volcanoes that formed by the eruption of highly liquid serpentinite muds. Torishima Forearc Seamount, which is blanketed by more ìnormalî pelagic/volcaniclastic sediment, has probably been inactive since the Miocene. Conical Seamount, which seems to consist entirely of serpentinite mud and is venting fresh water of unusual chemistry from its summit, is presently active. Muds from the flanks of Conical Seamount are stronger and more brittle than those from the summit site, and muds from Torishima Forearc Seamount are stronger yet; this suggests that the serpentinite debris flows are compacted and dewatered as they mature. The shear fabrics probably result from downslope creep and flow, but may also be inherited.

We have compiled the first stratigraphically continuous high-resolution benthic foraminiferal stable isotope record for the Paleocene from a single site utilizing cores recovered at Pacific ODP Site 1209. The long-term trend in the benthic isotope record suggests a close coupling of volcanic CO2 input and deep sea warming. Over the short-term the record is characterized by slow excursions with a pronounced periodic beat related to the short (100-kyr) and long (405-kyr) eccentricity cycle. The phase relationship between the benthic isotope record and eccentricity is similar to patterns documented for the Oligocene and Miocene confirming the role of orbital forcing as the pace maker for paleoclimatic variability on Milankovitch time scales. In addition, the record documents an unusual transient warming of 2°C coeval with a 0.6 per mil carbon isotope excursion and a decrease in carbonate content at 61.75 Ma. This event, which bears some resemblance to Eocene hyperthermals, marks the onset of a long-term decline in d13C. The timing indicates it might be related to the initiation of volcanism along Greenland margin.

Heavy (magnetic & non-magnetic) minerals are found concentrated by natural processes in many fluvial, estuarine, coastal and shelf environments with a potential to form economic placer deposits. Understanding the processes of heavy mineral transport and enrichment is prerequisite to interpret sediment magnetic properties in terms of hydro- and sediment dynamics. In this study, we combine rock magnetic and sedimentological laboratory measurements with numerical 3D discrete element models to investigate differential grain entrainment and transport rates of magnetic minerals in a range of coastal environments (riverbed, mouth, estuary, beach and near-shore). We analyzed grain-size distributions of representative bulk samples and their magnetic mineral fractions to relate grain-size modes to respective transport modes (traction, saltation, suspension). Rock magnetic measurements showed that distribution shapes, population sizes and grain-size offsets of bulk and magnetic mineral fractions hold information on the transport conditions and enrichment process in each depositional environment. A downstream decrease in magnetite grain size and an increase in magnetite concentration was observed from riverine source to marine sink environments. Lower flow velocities permit differential settling of light and heavy mineral grains creating heavy mineral enriched zones in estuary settings, while lighter minerals are washed out further into the sea. Numerical model results showed that higher heavy mineral concentrations in the bed increased the erosion rate and enhancing heavy mineral enrichment. In beach environments where sediments contained light and heavy mineral grains of equivalent grain sizes, the bed was found to be more stable with negligible amount of erosion compared to other bed compositions. Heavy mineral transport rates calculated for four different bed compositions showed that increasing heavy mineral content in the bed decreased the transport rate. There is always a lag in transport between light and heavy minerals which increases with higher heavy mineral concentration in all tested bed compositions. The results of laboratory experiments were validated by numerical models and showed good agreement. We demonstrate that the presented approach bears the potential to investigate heavy mineral enrichment processes in a wide range of sedimentary settings.

Sediment dynamics in limnic, fluvial and marine environments can be assessed by granulometric and rock-magnetic methodologies. While classical grain-size analysis by sieving or settling mainly bears information on composition and transport, the magnetic mineral assemblages reflect to a larger extent the petrology and weathering conditions in the sediment source areas. Here, we combine both methods to investigate Late Quaternary marine sediments from five cores along a transect across the continental slope off Senegal. This region near the modern summer Intertropical Convergence Zone is particularly sensitive to climate change and receives sediments from several aeolian, fluvial and marine sources. From each of the investigated five GeoB sediment cores (494-2956 m water depth) two time slices were processed which represent contrasting climatic conditions: the arid Heinrich Stadial 1 (~ 15 kyr BP) and the humid Mid Holocene (~ 6 kyr BP). Each sediment sample was split into 16 grain-size fractions ranging from 1.6 to 500 µm. Concentration and grain-size indicative magnetic parameters (susceptibility, SIRM, HIRM, ARM and ARM/IRM) were determined at room temperature for each of these fractions. The joint consideration of whole sediment and magnetic mineral grain-size distributions allows to address several important issues: (i) distinction of two aeolian sediment fractions, one carried by the north-easterly trade winds (40-63 µm) and the other by the overlying easterly Harmattan wind (10-20 µm) as well as a fluvial fraction assigned to the Senegal River (< 10 µm); (ii) identification of three terrigenous sediment source areas: southern Sahara and Sahel dust (low fine-grained magnetite amounts and a comparatively high haematite content), dust from Senegalese coastal dunes (intermediate fine-grained magnetite and haematite contents) and soils from the upper reaches of the Senegal River (high fine-grained magnetite content); (iii) detection of partial diagenetic dissolution of fine magnetite particles as a function of organic input and shore distance; (iv) analysis of magnetic properties of marine carbonates dominating the grain-size fractions 63-500 µm.

Este trabajo analiza el fenómeno intertextual de la ?cita? en Petronio, Sat. 118, donde el poeta de la obra, Eumolpo, expone su particular ?ars poetica?. En este discurso, Eumolpo cita los nombres de Homero, Virgilio y Horacio, al tiempo que cita el primer verso de la Oda 3.1 del Venusino, lo que evidencia que el poetastro tiene muy en cuenta la figura horaciana. El análisis de las citas en este pasaje se relaciona con que Petronio es un autor que alude constantemente, pero que cita muy rara vez, por lo que, cuando lo hace, se debe prestar mucha atención

Los rojos de Ultramar (2004) del mexicano Jordi Soler es una novela que sondea la memoria de la guerra civil española y la dictadura franquista, indagando en sus consecuencias a nivel individual y colectivo a partir de la experiencia de los exiliados. El narrador de la novela, trasunto del propio autor, es nieto de un republicano exiliado que se propone investigar a fondo la accidentada biografía de su abuelo. La obra comparte muchos rasgos formales y temáticos con varias novelas de la memoria recientes de autores españoles, tales como Soldados de Salamina de Javier Cercas y Mala gente que camina de Benjamín Prado, en las que la investigación histórica de un narrador-personaje funciona como motor narrativo. Sin embargo, la obra aporta al debate español en torno a la memoria histórica una novedosa visión desde fuera de las fronteras nacionales. Por medio de la voz del narrador, portador de dos culturas (la mexicana y la catalana) y dos lenguas (el castellano y el catalán), la novela de Soler proporciona una perspectiva transnacional y multicultural, que se aproxima a lo que Michael Rothberg ha denominado memoria multidireccional. El objetivo de este artículo es, por un lado, identificar y discutir las características narrativas que Los rojos de ultramar comparte con las novelas-investigación de autores españoles y, por otro lado, profundizar en la aportación particular de esta obra, que reivindica la hibridez cultural y utiliza la memoria de una comunidad cultural específica (la de los republicanos exiliados) de modo ejemplar para crear solidaridad entre diferentes grupos culturales.

The reconstruction of the stable carbon isotope evolution in atmospheric CO2 (d13Catm ), as archived in Antarctic ice cores, bears the potential to disentangle the contributions of the different carbon cycle fluxes causing past CO2 variations. Here we present a new record of d13Catm before, during and after the Marine Isotope Stage 5.5 (155 000 to 105 000 years BP). The record was derived with a well established sublimation method using ice from the EPICA Dome C (EDC) and the Talos Dome ice cores in East Antarctica. We find a 0.4 permil shift to heavier values between the mean d13Catm level in the Penultimate (~ 140 000 years BP) and Last Glacial Maximum (~ 22 000 years BP), which can be explained by either (i) changes in the isotopic composition or (ii) intensity of the carbon input fluxes to the combined ocean/atmosphere carbon reservoir or (iii) by long-term peat buildup. Our isotopic data suggest that the carbon cycle evolution along Termination II and the subsequent interglacial was controlled by essentially the same processes as during the last 24 000 years, but with different phasing and magnitudes. Furthermore, a 5000 years lag in the CO2 decline relative to EDC temperatures is confirmed during the glacial inception at the end of MIS 5.5 (120 000 years BP). Based on our isotopic data this lag can be explained by terrestrial carbon release and carbonate compensation.

La literatura en la enseñanza de una lengua permite el desarrollo integral del niño a través de la exploración de contenidos con valor cultural relevantes. En el caso de una lengua extranjera permite implementar secuencias didácticas que ayudan a andamiar la lengua en su enseñanza al mismo tiempo que desarrollan una competencia intercultural. Se abordará la enseñanza del inglés a partir de una variedad de géneros y tipos textuales permitiendo de esta manera el desarrollo de una perspectiva intercultural en los niños. Con este fin trabajaremos desde una didáctica centrada en el enfoque de resolución de tareas, debido a que estas actividades poseen objetivos lingüísticos específicos desde una perspectiva discursiva de la lengua y ocurren en un contexto determinado con una meta y destinatarios particulares. Estas les permitirán a los niños moverse, pensar, utilizar la lengua y planear estrategias con un claro propósito en mente. Goldilocks and the Three Little Bears será el puntapié inicial para ilustrar estos conceptos

En este artículo se postula que la geografía posee una cierta capacidad para establecer marcos analíticos que sirven a la prospectiva en ciencias sociales. Para ello se acude a las posibilidades del análisis multiescalar como instrumento que, partiendo de los procesos territoriales estudiados a diferentes escalas, da cuenta también de los diferentes tempos que los afectan. Esas distintas temporalidades, al conjugarse en determinados espacios, ponen condiciones a los desarrollos futuros que son de interés para el análisis sociológico. Las actuales condiciones en que se desarrolla la sociedad urbana argentina son analizadas desde esta perspectiva.
