Foi conduzido um ensaio numa plantação comercial de café de variedade Mundo Novo de 9 anos de idade, com uma população de 1904 covas/ha, destinada a avaliar a quantidade de biomassa e de nutrientes removidas por diferentes tipos de poda: recepa a 0,40m; decote a 1,00, 1,50 e 2,00 m; decote a 1,50m com esqueletamento. A análise do material e dos dados permitiu tirar-se as seguintes conclusões: (1) a biomassa removida pela poda foi maior na recepa (24,3 t de matéria fresca e 11,9 de matéria seca) e no decote a 1,00 m (20,6 e 10,1 t, respectivamente); seguia-se o decote a 1,50 m com esqueletamento que deu 19,4 e 8,3 t de matéria fresca e seca por hectare; os pesos da matéria fresca e seca correspondentes aos decotes a 1,50 m e 2,00 m foram: 12,1 e 5,4; 5,6 e 2,5 t/ha; (2) a relação existente entre a altura de poda e quantidade de fitomassa removida é descrita por equações de regressão simples; (3) as quantidades de nutrientes removidas são proporcionais as quantidades de material podado sendo as seguintes de acordo com a ordem dos tratamentos dado, em kg/ha: N - 320, 294, 162, 80 e 261; P - 18, 15, 10, 44 e 16; K - 286, 266, 168, 78 e 273; Ca - 149, 139, 63, 33 e 101: Mg - 30, 33, 16, 8 e 26; S - 10, 7,6, 3 e 10; as quantidades de micronutrientes removidas foram, em g/ha: B - 306, 337, 163, 83 e 268; Cu - 229, 219, 121, 51 e 191; Fe - 2783, 2328, 1367, 544 e 2,088; Mn - 437, 779, 264, 142 e 412; Zn - 174, 152, 74, 28 e 121; (3) foram derivadas equações de regressão simples que relacionam quantidade extraídas e altura da poda; (4) a reciclagem de fitomassa contribui com economia substancial de fertilizantes para a nova vegetação. Cerca de dois terços e três quartos de nutrientes, entretanto, estão contidos no material lenhoso de caules e ramos o que deve fazer que a sua disponibilidade seja mais lenta.

Metamysidopsis atlantica elongata (Bascescu, 1968) is a common mysid in the surf zone of sandy beaches from the state of Rio Grande do Sul, Brazil, where it is frequently recorded forming dense aggregations. Trough laboratory trials, behavioral responses to salinity (10, 20, 25, 28, 30, 40 e 45), temperature (10, 15, 20, 30±1ºC) and light (yes/no) were tested using adult males, adult females and juveniles. Although there was no response to temperature, the species showed clear response to salinity and light. In the presence of light, organisms remained in the bottom of the aquaria, but moved to surface when bottom salinities were increased. In the absence of light, adults moved to the surface. However, juveniles moved down to or remained on the bottom, maybe as a response to avoid adult predation.

BACKGROUND: Immunotherapy offers a promising novel approach for the treatment of cancer and both adoptive T-cell transfer and immune modulation lead to regression of advanced melanoma. However, the potential synergy between these two strategies remains unclear. METHODS: We investigated in 12 patients with advanced stage IV melanoma the effect of multiple MART-1 analog peptide vaccinations with (n̴1;=̴1;6) or without (n̴1;=̴1;6) IMP321 (LAG-3Ig fusion protein) as an adjuvant in combination with lymphodepleting chemotherapy and adoptive transfer of autologous PBMCs at day (D) 0 (Trial registration No: NCT00324623). All patients were selected on the basis of ex vivo detectable MART-1-specific CD8 T-cell responses and immunized at D0, 8, 15, 22, 28, 52, and 74 post-reinfusion. RESULTS: After immunization, a significant expansion of MART-1-specific CD8 T cells was measured in 83% (n̴1;=̴1;5/6) and 17% (n̴1;=̴1;1/6) of patients from the IMP321 and control groups, respectively (P̴1;<̴1;0.02). Compared to the control group, the mean fold increase of MART-1-specific CD8 T cells in the IMP321 group was respectively >2-, >4- and >6-fold higher at D15, D30 and D60 (P̴1;<̴1;0.02). Long-lasting MART-1-specific CD8 T-cell responses were significantly associated with IMP321 (P̴1;<̴1;0.02). At the peak of the response, MART-1-specific CD8 T cells contained higher proportions of effector (CCR7S15; CD45RAS14;/S15;) cells in the IMP321 group (P̴1;<̴1;0.02) and showed no sign of exhaustion (i.e. were mostly PD1S15;CD160S15;TIM3S15;LAG3S15;2B4S14;/S15;). Moreover, IMP321 was associated with a significantly reduced expansion of regulatory T cells (P̴1;<̴1;0.04); consistently, we observed a negative correlation between the relative expansion of MART-1-specific CD8 T cells and of regulatory T cells. Finally, although there were no confirmed responses as per RECIST criteria, a transient, 30-day partial response was observed in a patient from the IMP321 group. CONCLUSIONS: Vaccination with IMP321 as an adjuvant in combination with lymphodepleting chemotherapy and adoptive transfer of autologous PBMCs induced more robust and durable cellular antitumor immune responses, supporting further development of IMP321 as an adjuvant for future immunotherapeutic strategies.

Inflammatory bowel disease (IBD) is a systemic disease associated with a large number of extraintestinal manifestations (EIM). EIM are present in 15-20% of patients with ulcerative colitis and in 20-40% of patients with Crohn's disease. The management of EIM is best provided by a multidisciplinary team, which includes specialists in the affected organ systems with training in the treatment of IBD. Therapeutic strategy is often empirical. This is explained by the paucity of randomized-controlled studies for the specific treatment of EIM in IBD and by the fact that treatment models are based on extrapolation from patients with similar conditions but without IBD. For most EIM, the mainstay of therapy is the treatment of the underlying active IBD. However, some EIM such as axial arthritis, pyoderma gangrenosum, uveitis and primary sclerosing cholangitis run a clinical course independent of IBD activity and need specific therapy (e.g. TNF antagonists in ankylosing spondylitis and skin manifestations). This review summarizes the conventional and novel (e.g. anti-TNF) treatment modalities, and the therapeutic implications for the management of extraintestinal symptoms in IBD, in order to assist clinicians in optimizing treatment strategies for IBD patients with EIM.

In Brazil, until 2004, the immunization policy against diphtheria involved childhood vaccination with no official routine booster dose administered after 15 years of age. This study assessed functional antibody levels against diphtheria among blood donors. A total of 140 blood samples were collected, and diphtheria antitoxin levels were evaluated by Vero cell neutralization test. The mean age of the population was 34 years old (range: 18-61 years); 37.8% females and 62.2% males. Overall, 30.7% (95%, CI: 23.4-38.7) individuals presented neutralizing antitoxin antibody titers < 0.01 IU/ml; 42.1% (95%, CI: 34.1-50.4) showed values between 0.01-0.09 IU/ml and, 27.1% (95%, CI: 20.2-34.9) had ³ 0.1 IU/ml. In the subgroup of individuals with history of diphtheria immunization during childhood (85%), a number of 28.5% showed unprotective levels of circulating neutralizing antibody (< 0.01 IU/ml). Despite the continuous progress of immunization programs directed to Brazilian population, currently healthy adults remain susceptible to diphtheria.

PURPOSE. To evaluate potential risk factors for the development of multiple sclerosis in Brazilian patients. METHOD. A case control study was carried out in 81 patients enrolled at the Department of Neurology of the Hospital da Lagoa in Rio de Janeiro, and 81 paired controls. A standardized questionnaire on demographic, social and cultural variables, and medical and family history was used. Statistical analysis was performed using descriptive statistics and conditional logistic regression models with the SPSS for Windows software program. RESULTS. Having standard vaccinations (vaccinations specified by the Brazilian government) (OR=16.2; 95% CI=2.3-115.2), smoking (OR=7.6; 95% CI=2.1-28.2), being single (OR=4.7; 95% CI=1.4-15.6) and eating animal brain (OR=3.4; 95% CI=1.2-9.8) increased the risk of developing MS. CONCLUSIONS. RESULTS of this study may contribute towards better awareness of the epidemiological characteristics of Brazilian patients with multiple sclerosis.

PURPOSE: We have investigated the expression and regulation of 15-hydroxyprostaglandin dehydrogenase (15-PGDH) in gastric cancer. EXPERIMENTAL DESIGN: Clinical gastric adenocarcinoma samples were analyzed by immunohistochemistry and quantitative real-time PCR for protein and mRNA expression of 15-PGDH and for methylation status of 15-PGDH promoter. The effects of interleukin-1beta (IL-1beta) and epigenetic mechanisms on 15-PGDH regulation were assessed in gastric cancer cell lines. RESULTS: In a gastric cancer cell line with a very low 15-PGDH expression (TMK-1), the 15-PGDH promoter was methylated and treatment with a demethylating agent 5-aza-2'-deoxycytidine restored 15-PGDH expression. In a cell line with a relatively high basal level of 15-PGDH (MKN-28), IL-1beta repressed expression of 15-PGDH mRNA and protein. This effect of IL-1beta was at least in part attributed to inhibition of 15-PGDH promoter activity. SiRNA-mediated knockdown of 15-PGDH resulted in strong increase of prostaglandin E(2) production in MKN-28 cells and increased cell growth of these cells by 31% in anchorage-independent conditions. In clinical gastric adenocarcinoma specimens, 15-PGDH mRNA levels were 5-fold lower in gastric cancer samples when compared with paired nonneoplastic tissues (n = 26) and 15-PGDH protein was lost in 65% of gastric adenocarcinomas (n = 210). CONCLUSIONS: 15-PGDH is down-regulated in gastric cancer, which could potentially lead to accelerated tumor progression. Importantly, our data indicate that a proinflammatory cytokine linked to gastric carcinogenesis, IL-1beta, suppresses 15-PGDH expression at least partially by inhibiting promoter activity of the 15-PGDH gene.

NovoTTF-100A (TTF) is a portable device delivering low-intensity, intermediate-frequency, alternating electric fields using noninvasive, disposable scalp electrodes. TTF interferes with tumor cell division, and it has been approved by the US Food and Drug Administration (FDA) for the treatment of recurrent glioblastoma (rGBM) based on data from a phase III trial. This presentation describes the updated survival data 2 years after completing recruitment. Adults with rGBM (KPS ≥ 70) were randomized (stratified by surgery and center) to either continuous TTF (20-24 h/day, 7 days/week) or efficacious chemotherapy based on best physician choice (BPC). The primary endpoint was overall survival (OS), and secondary endpoints were PFS6, 1-year survival, and QOL. Patients were randomized (28 US and European centers) to either TTF alone (n ¼ 120) or BPC (n ¼ 117). Patient characteristics were balanced, median age was 54 years (range, 23-80 years), and median KPS was 80 (range, 50-100). One quarter of the patients had debulking surgery, and over half of the patients were at their second or later recurrence. OS in the intent-to-treat (ITT) population was equivalent in TTF versus BPC patients (median OS, 6.6vs. 6.0 months; n ¼ 237; p ¼ 0.26; HR ¼ 0.86). With a median follow-up of 33.6 months, long-term survival in the TTF group was higher than that in the BPC group at 2, 3, and 4 years of follow-up (9.3% vs. 6.6%; 8.4% vs. 1.4%; 8.4% vs. 0.0%, respectively). Analysis of patients who received at least one treatment course demonstrated a survival benefit for TTF patients compared to BPC patients (median OS, 7.8 vs. 6.0 months; n ¼ 93 vs. n ¼ 117; p ¼ 0.012; HR ¼ 0.69). In this group, 1-year survival was 28% vs. 20%, and PFS6 was 26.2% vs. 15.2% (p ¼ 0.034). TTF, a noninvasive, novel cancer treatment modality shows significant therapeutic efficacy with promising long-term survival results. The impact of TTF was more pronounced when comparing only patients who received the minimal treatment course. A large-scale phase III trial in newly diagnosed GBM is ongoing.

The aim of the study was to determine the prevalence and variables associated with the pattern of risky health behavior (PRHB) among adolescent students in Cartagena, Colombia. A cross-sectional study was designed to investigate PRHB in a random cluster sample of students from middle and high schools. The associations were adjusted by logistic regression. A total of 2,625 students participated in this research, with ages from 10 to 20 years, mean=13.8 years (SD=2.0), and 54.3% were women. A total of 332 students reported PRHB (12.7%, 95%CI 11.4R11;14.0). Age over 15 years (OR=2.19, 95%CI 1.72R11;2.79), not being heterosexual (OR=1.98, 95%CI 1.36-2.87), poor/mediocre academic performance (OR=1.87, 95%CI 1.47R11;2.38), family dysfunction (OR=1.78, 95%CI 1.40R11;2.28) and male gender (OR=1.58, 95%CI 1.24R11;2.01) were associated with PRHB. One in every eight students presented a PRHB. It is important to pay greater attention to students who are over 15 years of age, male, not heterosexual, with a poor/mediocre academic performance and a dysfunctional family.

Human cytosolic thymidine kinase (hTK1) has proven to be a suitable target for the noninvasive imaging of cancer cell proliferation using radiolabeled thymidine analogues such as [(18)F]3'-fluoro-3'-deoxythymidine ([(18)F]FLT). A thymidine analogue for single photon emission computed tomography (SPECT), which incorporates the readily available and inexpensive nuclide technetium-99m, would be of considerable practical interest. hTK1 is known to accommodate modification of the structure of the natural substrate thymidine at the positions N3 and C3' and, to a lesser extent, C5. In this work, we used the copper-catalyzed azide-alkyne cycloaddition to synthesize two series of derivatives in which thymidine is functionalized at either the C3' or N3 position with chelating systems suitable for the M(CO)(3) core (M = (99m)Tc, Re). The click chemistry approach enabled complexes with different structures and overall charges to be synthesized from a common precursor. Using this strategy, the first organometallic hTK1 substrates in which thymidine is modified at the C3' position were identified. Phosphorylation of the organometallic derivatives was measured relative to thymidine. We have shown that the influence of the overall charge of the derivatives is dependent on the position of functionalization. In the case of the C3'-functionalized derivatives, neutral and anionic substrates were most readily phosphorylated (20-28% of the value for the parent ligand thymidine), whereas for the N3-functionalized derivatives, cationic and neutral complexes were apparently better substrates for the enzyme (14-18%) than anionic derivatives (9%).

Switzerland has the lowest adolescent fertility rate in Western Europe. According to data collected in 1993 as part of the Swiss Multicentre Adolescent Survey on Health, 5% of 1,726 sexually active adolescents in a group of 3,993 15-20-year-old women enrolled in academic or vocational classes had ever been pregnant; most of these women (80%) had terminated their pregnancy. Adolescents who had ever been pregnant did not differ significantly from those who had not by demographic characteristics. Multiple logistic regression analysis identified seven factors associated with pregnancy: having had four or more sexual partners; not having used contraceptives at first intercourse; ever use of less-effective contraceptive methods; having used illicit drugs during the last 30 days; living apart from one's parents; recently experiencing stress; and perceiving a lack of future prospects.

BACKGROUND: Although Temozolomide is effective against glioblastoma, the prognosis remains dismal and new regimens with synergistic activity are sought for. METHODS: In this phase-I/II trial, pegylated liposomal doxorubicin (Caelyx, PEG-Dox) and prolonged administration of Temozolomide in addition to radiotherapy was investigated in 63 patients with newly diagnosed glioblastoma. In phase-I, PEG-Dox was administered in a 3-by-3 dose-escalation regimen. In phase-II, 20 mg/m2 PEG-Dox was given once prior to radiotherapy and on days 1 and 15 of each 28-day cycle starting 4 weeks after radiotherapy. Temozolomide was given in a dose of 75 mg/m2 daily during radiotherapy (60 Gy) and 150-200 mg/m2 on days 1-5 of each 28-day cycle for 12 cycles or until disease progression. RESULTS: The toxicity of the combination of PEG-Dox, prolonged administration of Temozolomide, and radiotherapy was tolerable. The progression free survival after 12 months (PFS-12) was 30.2%, the median overall survival was 17.6 months in all patients including the ones from Phase-I. None of the endpoints differed significantly from the EORTC26981/NCIC-CE.3 data in a post-hoc statistical comparison. CONCLUSION: Together, the investigated combination is tolerable and feasible. Neither the addition of PEG-Dox nor the prolonged administration of Temozolomide resulted in a meaningful improvement of the patient's outcome as compared to the EORTC26981/NCIC-CE.3 data.

Em solos que apresentam características naturais favoráveis ao cultivo, o preparo convencional degrada as propriedades físicas, pois o revolvimento rompe os agregados, compacta o solo abaixo da camada preparada e o deixa descoberto. A semeadura direta, em virtude da pequena mobilização do solo, preserva os agregados e a cobertura do solo, porém consolida a camada superficial. Este trabalho foi desenvolvido em um Cambissolo Húmico alumínico léptico, de maio de 1995 a abril de 2001. Os tratamentos de manejo do solo foram: preparo convencional com uma aração+duas gradagens em rotação de culturas (PCR) e em sucessão (PCS), semeadura direta em rotação de culturas (SDR) e em sucessão (SDS) e campo nativo (CN). A seqüência de cultivo nos tratamentos foi: PCR - feijão/pousio/milho/pousio/soja/pousio; PCS - milho/pousio/milho/pousio/milho/pousio; SDR - feijão/aveia/milho/nabo/soja/ervilhaca e, na SDS, milho/ervilhaca/milho/ervilhaca/milho/ervilhaca. No CN, a área foi pastejada até o momento da instalação do experimento, permanecendo em pousio com roçadas periódicas depois disso. Em abril de 2001, foram avaliados a densidade do solo, o volume de macroporos, microporos e total de poros, o teor de carbono orgânico e a estabilidade de agregados em água, nas profundidades de 0-2,5; 2,5-5; 5-10; 10-15; 15-20 e 20-30 cm. Na camada de 0-10 cm, a densidade do solo foi maior na SDS e SDR e igual no PCS e PCR, enquanto o volume total de poros e de macroporos foi igual na SDS e SDR, em relação ao CN. Nessa camada, a relação volume de macroporos/total de poros foi maior no PCS e PCR do que na SDS, SDR e CN. Na média das camadas estudadas, o teor de carbono orgânico e a estabilidade dos agregados foram maiores na SDS, SDR e CN do que no PCS e PCR. No PCS e PCR, o índice de sensibilidade para estabilidade de agregados foi expressivamente menor do que a unidade, enquanto, na SDS e SDR, os valores foram próximos à unidade, indicando a sustentabilidade da semeadura direta no que se refere à estabilidade dos agregados em água. Os sistemas de cultivo não influenciaram as propriedades físicas do solo.

A suinocultura tem como inconveniente a grande produção de dejetos, os quais são reutilizados como fertilizantes agrícolas, porém, se utilizados em doses excessivas, tornam-se severos poluentes ambientais. O objetivo deste trabalho foi avaliar as formas de acumulação de P em um solo submetido a sucessivas aplicações de doses de dejeto líquido de suínos. O experimento foi instalado em 1995, em uma área cultivada com pastagem natural, que recebeu doses de 0, 20 e 40 m³ ha-1 de dejeto líquido de suínos (DLS), aplicadas em intervalos de 45 a 60 dias durante cinco anos. Em 1999, após 28 aplicações de DLS, que totalizaram 0, 560 e 1.120 m³ ha-1, foram coletadas amostras de solo nas profundidades de 0-2,5, 2,5-5, 5-10, 10-15 e 15-20 cm. A amostra foi seca, moída, passada em peneira de malha de 2 mm e submetida ao fracionamento químico do P pelo método de Hedley. No tratamento testemunha, foi determinada a capacidade máxima de adsorção de fósforo (CMAP), utilizando-se isotermas de adsorção e ajuste matemático pela equação de Langmuir. Os resultados mostraram que o P adicionado ao solo via DLS é acumulado essencialmente em formas inorgânicas, sendo as principais as extraídas por resina trocadora de ânions, NaHCO3 0,5 mol L-1, NaOH 0,1 mol L-1 e HCl 1,0 mol L-1.
