This paper discusses the development of modular solutions for eco low-cost houses based on a pre-fabricated modular wall system environmentally sustainable, socioeconomically competitive and geared towards developing African nations with a housing deficit. The key point to the research of a modular wall solution is a structural layer complemented with local and materials made by non-specialized workforce. This wall also meets also hydrothermal acoustic and mechanical properties. Thus,the solution also offers good safety and interior comfort conditions to its users while maintaining the flexibility to expand the size of the house. Parameters as dimensions, materials and constructive processes of the existing housing stock were studied. Features such as the family size, typology, different uses, common materials, existing regulations, minimal living conditions, safety and comfort have also been considered to achieve the most efficient solution.

A análise fenotípica de 255 amostras do gênero Listeria isoladas de material clínico humano, tanto de indivíduos doentes (220-86,3%), como de aparentemente normais (35-13,7%) de várias regiões do país e colecionadas no período de 1969 a 2000, permitiu caracterizar a distribuição de sorovares de Listeria monocytogenes. Nas faixas etárias de 0 a 10 e de 41 a 60 anos, predominaram os isolamentos de líquido cefalorraquidiano sobre os de sangue, incluindo dos transplantados renais. Somente dos hemocultivos foi possível detectar os sete sorovares de Listeria monocytogenes. No cômputo geral, o sorovar 4b foi o mais incidente (154-60,3%) secundado por ¹/2 a (74-29%) nos três decênios considerados, além de ocorrerem em quase todas as regiões do país. Os dados deste estudo evidenciaram a circulação de L. monocytogenes na espécie humana, provocando quadros graves de meningite e septicemia, bem como, revelando a figura do portador assintomático, razão pela qual são recomendadas novas investigações bacteriológicas, subsidiadas por análises clínico-patológicas e epidemiológicas.

We present the case of a 62-year-old woman with abdominal segmental paresis consequent to radiculopathy caused by zoster, which was confirmed by electroneuromyography. The paresis resolved completely within three months. Recognition of this complication caused by zoster, which is easily misdiagnosed as abdominal herniation, is important for diagnosing this self-limited condition and avoiding unnecessary procedures.

Seismic events are a major factor to consider in structural design of buildings in many countries. With the purpose of saving lives, most of the design codes lead to structural solutions that withstand large seismic actions without collapsing, but without taking into account a possible usage of the structures after the earthquake. As a result, it is necessary to consider the time needed to repair/retrofit the damaged structures (i.e. the downtime) since this period of inactivity may result in huge financial implications for the occupants of the buildings. In order to minimise the damages and simplify repair operations, structural solutions with rocking systems and negligible residual displacements have been developed during the last two decades. Systems with precast concrete rocking walls were studied with the aim of investigat- ing suitable and convenient structural alternatives to minimise the damage in case of an earthquake. Experimental, numerical and analytical analyses on post-tensioned solutions, with and without energy dissipation devices, were carried out in this research. The energy dissipation devices were made from steel angles that were further developed during the research. Different solutions for these devices were experimentally tested under cyclic loading and the results are presented. Numerical and analytical work on steel angles was also carried out. Regarding the concrete rocking wall systems, two concrete rocking wall systems were studied: post-tensioned walls and post-tensioned walls with energy dissipation devices. In the latter, the solution was to fix them externally to the wall, allowing their easy replacement after an earthquake. It is shown that the dissipaters are a viable solution for use in precast concrete rocking wall systems. A building case study is presented. The comparison between a traditional monolithic system and a hybrid solution was carried out, allowing the evaluation of the efficiency of the solution that was developed.

A presente dissertação reflecte a compilação de um estudo essencialmente bibliográfico e iconográfico de doze peças de mobiliário do antigo Egipto, Império Novo. É primeiramente apresentado um breve estudo sobre materiais usados para a construção dos diversos objectos produzidos no referido período histórico e analisados os seus aspectos simbólicos. São também apresentadas sucintamente algumas técnicas de construção adoptadas durante as dinastias XVIII-XX para a construção de mobiliário doméstico e ritual. De seguida estes objectos são analisados material e simbolicamente em dois capítulos tendo como base a investigação efectuada. Colocam-se ao longo deste trabalho diversas questões, sendo as mais prementes “se terá havido transferência de mobiliário doméstico para o contexto funerário” e “Terão tido os espólios funerários mobiliário feito exclusivamente para esse fim”. As conclusões revelam dados comparativos do universo de peças de mobiliário recolhidas e apresentam algumas hipóteses explicativas sobre o assunto abordado.

Due to their exposure to environmental conditions, outer coatings composed by render and painting system are usually the first construction elements to deteriorate and require intervention. A correct conservation and rehabilitation of these materials is fundamental once they provide protection to other façade materials. It is known that old mortar renders were essentially air lime based mortars. To maintain the integrity of the whole wall-render elements, the image of the building and to avoid accelerated degradation, conservation and rehabilitation must be implemented with compatible mortars. As that, lime based mortars would be preferable. It was also common, in ancient renders, the incorporation of ceramic residues, which is, nowadays, an abundant material, especially in Central Region of Portugal. The reuse of these materials has great relevance once their landfilling causes serious environmental issues. In an attempt to combine the environmental and technical advantages of the use of ceramic waste in mortars’ production for rehabilitation purposes, a research has been developed at the University of Coimbra, in cooperation with Nova University of Lisbon, on the long term behaviour of air lime mortars with ceramic residues. In this paper the most significant up to one year results of an experimental campaign with air lime mortars with 1:3 and 1:2 volumetric proportions and ceramic residues are presented.

Neste trabalho foram produzidas matrizes de nanofibras de gelatina de porco e de gelatina de peixe através da técnica de electrofiação. Estas matrizes têm uma estrutura morfológica e química interessantes para o desenvolvimento de scaffolds no âmbito da Engenharia de Tecidos mas a sua instabilidade em água compromete essa aplicação. Assim, as matrizes foram reticuladas recorrendo a dois processos diferentes: um físico (tratamento desidrotérmico - DHT) e outro químico (exposição a vapor de glutaraldeído - GTA). Os resultados obtidos permitiram verificar que a gelatina de porco reticulou mais facilmente que a gelatina de peixe, provavelmente devido a uma maior presença de grupos funcionais capazes de reagir com os processos de reticulação. Foram analisados possíveis efeitos citotóxicos das matrizes reticuladas em culturas de células Vero. Os extratos das matrizes não revelaram ser tóxicos mas o tratamento com GTA comprometeu a adesão das células às matrizes indicando acarretar um certo risco de toxicidade. O uso da glicina revelou-se eficaz na redução dessa toxicidade. Para além das matrizes de fibras sem orientação preferencial (depositadas num coletor plano), foram depositadas fibras paralelamente alinhadas recorrendo a um coletor cilíndrico rotatório. Marcações fluorescentes de células semeadas nos dois tipos de fibras revelaram que o citoesqueleto das células semeadas nas fibras alinhadas se organiza na direção do alinhamento.

Three different treatments were applied on several specimens of dolomitic and calcitic marble, properly stained with rust to mimic real situations (the stone specimens were exposed to the natural environment for about six months in contact with rusted iron). Thirty six marble specimens, eighteen calcitic and eighteen dolomitic, were characterized before and after treatment and monitored throughout the cleaning tests. The specimens were characterized by SEM-EDS (Scanning Electron Microscopy coupled with Energy Dispersion System), XRD (XRay Diffraction), XRF (X-Ray Fluorescence), FTIR (Fourier Transform Infrared Spectroscopy) and color measurements. It was also made a microscopic and macroscopic analysis of the stone surface along with the tests of short and long term capillary absorption. A series of test trials were conducted in order to understand which concentrations and contact times best suits to this purpose, to confirm what had been written to date in the literature. We sought to develop new methods of treatment application, skipping the usual methods of applying chemical treatments on stone substrates, with the use of cellulose poultice, resorting to the agar, a gel already used in many other areas, being something new in this area, which possesses great applicability in the field of conservation of stone materials. After the application of the best methodology for cleaning, specimens were characterized again in order to understand which treatment was more effective and less harmful, both for the operator and the stone material. Very briefly conclusions were that for a very intense and deep penetration into the stone, a solution of 3.5% of SDT buffered with ammonium carbonate to pH around 7 applied with agar support would be indicated. For rust stains in its initial state, the use of Ammonium citrate at a concentration of 5% buffered with ammonium to pH 7 could be applied more than once until satisfactory results appear.

Rupture of aortic aneurysms (AA) is a major cause of death in the Western world. Currently, clinical decision upon surgical intervention is based on the diameter of the aneurysm. However, this method is not fully adequate. Noninvasive assessment of the elastic properties of the arterial wall can be a better predictor for AA growth and rupture risk. The purpose of this study is to estimate mechanical properties of the aortic wall using in vitro inflation testing and 2D ultrasound (US) elastography, and investigate the performance of the proposed methodology for physiological conditions. Two different inflation experiments were performed on twelve porcine aortas: 1) a static experiment for a large pressure range (0 – 140 mmHg); 2) a dynamic experiment closely mimicking the in vivo hemodynamics at physiological pressures (70 – 130 mmHg). 2D raw radiofrequency (RF) US datasets were acquired for one longitudinal and two cross-sectional imaging planes, for both experiments. The RF-data were manually segmented and a 2D vessel wall displacement tracking algorithm was applied to obtain the aortic diameter–time behavior. The shear modulus G was estimated assuming a Neo-Hookean material model. In addition, an incremental study based on the static data was performed to: 1) investigate the changes in G for increasing mean arterial pressure (MAP), for a certain pressure difference (30, 40, 50 and 60 mmHg); 2) compare the results with those from the dynamic experiment, for the same pressure range. The resulting shear modulus G was 94 ± 16 kPa for the static experiment, which is in agreement with literature. A linear dependency on MAP was found for G, yet the effect of the pressure difference was negligible. The dynamic data revealed a G of 250 ± 20 kPa. For the same pressure range, the incremental shear modulus (Ginc) was 240 ± 39 kPa, which is in agreement with the former. In general, for all experiments, no significant differences in the values of G were found between different image planes. This study shows that 2D US elastography of aortas during inflation testing is feasible under controlled and physiological circumstances. In future studies, the in vivo, dynamic experiment should be repeated for a range of MAPs and pathological vessels should be examined. Furthermore, the use of more complex material models needs to be considered to describe the non-linear behavior of the vascular tissue.

A incorporação de materiais reciclados na construção de estradas, como substituição de materiais virgens é percebida como uma oportunidade de economizar recursos, e evitar o impacto associado à sua extração e transporte. Desta forma, a incorporação de material fresado (do termo inglês RAP - Reclaimed Asphalt Pavement) em misturas betuminosas produzidas a quente em central tem sido uma alternativa satisfatória. Neste trabalho, foram estudadas três misturas betuminosas do tipo macadame betuminoso (AC 20 base 35/50), tradicional e com taxas de incorporação de RAP de 10% e 20%, com o objetivo de efetuar uma análise comparativa do seu comportamento. Começou-se por realizar um estudo Marshall de forma a determinar a percentagem ótima de betume a usar em cada mistura e, posteriormente, a composição foi transposta para central para o seu fabrico. No final foram realizados ensaios de caracterização mecânica sobre as misturas para avaliar o seu desempenho, através de ensaios de sensibilidade à água, módulo de rigidez, resistência à fadiga e à deformação permanente. Esta dissertação teve o apoio da empresa Tecnovia – Sociedade de Empreitadas S.A., com a disponibilização de instalações e materiais para o trabalho experimental. De acordo com os resultados obtidos, é possível concluir que as misturas recicladas apresentaram um comportamento satisfatório, quando comparado com as misturas tradicionais.

Tendo como caso de estudo um conjunto documental do Arquivo Histórico dos Museus da Universidade de Lisboa (AHMUL) – Museu Nacional de História Natural e da Ciência (MUHNAC), anteriormente fazendo parte do antigo Arquivo Histórico do Museu Bocage – fortemente atingido por um incêndio em 1978, o objectivo principal desta investigação é dar início à pesquisa e explorar a possibilidade de aplicação de métodos não invasivos de restauro digital, como a técnica de digitalização volumétrica (volumetric scanning), para a recuperação da informação escrita na parte mais deteriorada do arquivo; e propor métodos de estabilização/recuperação do seu suporte físico, usando técnicas tradicionais de conservação e restauro. Assim, o objectivo deste estudo não é apenas rever métodos de recuperação da informação para estes documentos tidos como perdidos, mas também propor uma forma de recuperar, tanto quanto possível, o seu suporte original. Será realizada a caracterização material da colecção, com recurso a métodos de exame e análise e o diagnóstico de conservação da parte da colecção mais deteriorada, bem como uma contextualização histórica sobre o arquivo e as condições do incêndio. Como conclusões principais salienta-se a confirmação da possibilidade de aplicação de técnicas de digitalização, os resultados promissores de outras técnicas como a fotografia de infravermelho com recurso adicional a software de processamento da imagem e o uso da imagem multiespectral em documentos carbonizados, além do estabelecimento de uma proposta de protocolo de intervenção para documentos queimados, com recurso a dois éteres de celulose, com diferentes solventes (água e etanol) de acordo com a solubilidade dos meios de escrita. Pretendeu-se ainda com este estudo contribuir para a divulgação da existência deste fundo do AHMUL em específico, tendo sido fundamental a realização, após 37 anos, do primeiro registo e compilação de fontes documentais sobre o incêndio de 18 de março de 1978, um facto tão relevante para a Memória do MUHNAC e dos Museus de Portugal.

Mega-organs, primarily in the digestive tract, are well known to occur in chronic Chagas disease. Acute experimental infection with Trypanosoma cruzi results in parasitism of a wide range of cells, tissues, and organs, including the urinary bladder. Infection of BALB/c mice with 100,000 bloodstream forms of the Y strain of T. cruzi induced acute infection with intense parasitism of all layers of the urinary bladder. Parasites were found in the mucosa, lamina propria, muscular, adventitial connective, and fat tissue. Desquamate epithelial cells with amastigotes in the bladder lumen were also found. After 60 days of infection, mice inoculated with 50 bloodstream forms developed dilated, thin-walled bladders that had inflammatory infiltrates and foci of fibrosis replacing areas of damaged muscular layer. These lesions result from direct damage to the muscle fibers by the T. cruzi, leading to myosites, muscle damage, and scarring. Direct damage of paraganglia cells secondary to parasitism, leading to dilatation, damage of muscle fibers, and scarring with replacement of muscular tissue with connective tissue, should also be considered as a cause of functional disturbance of the urinary bladder.

The cell wall of Staphylococcus aureus is a highly complex network mainly composed of highly cross-linked peptidoglycan (PG) and teichoic acids (TAs), both important for the maintenance of the integrity and viability of bacteria. The penicillin binding proteins (PBPs), which catalyse the final stage of PG biosynthesis, are targets of β-lactam antibiotics and have been a key focus of antibacterial research. S. aureus has four native PBPs, PBP1-4 carried by both methicillin-sensitive (MSSA) and –resistant (MRSA) strains. PBP4 is required for the synthesis of the highly cross-linked PG and, as shown in recent studies, is essential for the expression of β-lactam resistance in community-acquired strains (CA-MRSA). This protein has a septal localization that seems to be spatially and temporally regulated by an unknown intermediate of the wall teichoic acids (WTA) biosynthesis pathway. Therefore, if WTA synthesis is compromised, PBP4 becomes dispersed throughout the entire cell membrane. The aim of this project was to identify the WTA precursor responsible for the septal recruitment of PBP4. In order to do so, inducible mutants of tarB and tarL genes in the background of NCTCPBP4-YFP were constructed allowing for the study of PBP4 localization in the presence and absence of these specific tar genes.With this work we were able to show that the absence of TarB or TarL leads to the delocalization of PBP4, indicating that TarL or a protein/WTA precursor whose localization/synthesis is dependent on TarL is responsible for the recruitment of PBP4.
