Automated Production Systems Development involves aspects concerning the integration of technological components that exist on the market, such as: Programmable Logic Controllers (PLC), robot manipulators, various sensors and actuators, image processing systems, communication networks and collaborative supervisory systems; all integrated into a single application. This paper proposes an automated platform for experimentation, implemented through typical architecture for Automated Production Systems, which integrates the technological components described above, in order to allow researchers and students to carry out practical laboratory activities. These activities will complement the theoretical knowledge acquired by the students in the classroom, thus improving their training and professional skills. A platform designed using this generic structure will allow users to work within an educational environment that reflects most aspects found in Industrial Automated Manufacturing Systems, such as technology integration, communication networks, process control and production management. In addition, this platform offers the possibility complete automated process of control and supervision via remote connection through the internet (WebLab), enabling knowledge sharing between different teaching and research groups.

The use of alternative energy systems in the current days is an urgent necessity due to the problems that the planet is facing as the heating and loss of ozone layer. The scarcity of conventional energy is another problem that must be solved for the future of humanity. It must be considered that the people are inhabiting places moved away not always with available energy. The application of technologies as automation and control can help us to solve this problem. Therefore, this work aimed at apply an equipment of industrial usage, the Programmable Logical Controller, PLC, in alternative energies systems, as eolic generation and fotovoltaic generation used for water pumping, aiming the automatic control and the efficiency in the places where it has simultaneous availability of these sources, based in criterion of priority that previously established itself between them. It was made a hydraulic and energetic evaluation of the energy system, eolic and fotovoltaic, used in the automatic control system of pumping, in the place of accomplishment of the experiment, according to previously established physical conditions. The results have shown that the control system using the PLC is practicable and has trustworthiness. The program developed can be adapted for the use with several power plants in a specific application place. The fotovoltaic system of pumping, using a polycrystalline of 70 Watts connected to a pump Shurflo 8000, showed to be efficient with significant flows in almost all the months. The eolic system of pumping, using an eolic generator of 400 Watts assembled in place of experiment, did not demonstrate energetic capacity for use in this specific type of application.

This work studies and develops a machine automation for tyre truing. It is discussed industry development, industrial processes automation, tyre-recycling process, advantages of tyre reuse for preservation of the environment and probable gains from the automation of part of the tyre-recycling process. In this text, it is detailed the work done to configure, program and optimize the truing process through automation components as CNC, PLC and drives. Tests and simulations are performed to determine the payback necessary period in productivity and profit gains

Desde a antiguidade, enxertos e membranas são estudados para promoverem um reparo ósseo otimizado. O reparo ósseo consiste na reabsorção do tecido necrosado e de seu coágulo, juntamente com um processo inflamatório que libera fatores de crescimento que irão reparar o osso danificado. Em algumas situações, o osso lesionado não tem a capacidade de se auto reparar, portanto, são necessárias intervenções cirúrgicas para inserir um enxerto ósseo. Entretanto, há uma grande dificuldade em se encontrar um material que forneça os fatores necessários para o crescimento ósseo. Para isso, foram confeccionadas membranas à base de ácido polilático e poli-ε-caprolactona (PLC) (Purasorb: PLC 7015 - Purac, Holanda), com a incorporação de fosfato de lantânio (PLC/LaPO4) e oxiapatita dopada com 20% de lantânio (PLC/La20OAP) pelo Instituto de Química de Araraquara, UNESP. Assim, o objetivo do presente estudo foi avaliar a citotoxicidade desses materiais por meio dos testes XTT e sobrevivência clonogênica. Os eluatos foram preparados com as membranas citadas de acordo com a ISO 10993-12. O cloridrato de doxorrubicina foi utilizado como controle positivo para ambos os testes. Como Controle Negativo (CN) foram utilizadas somente as células CHO-K1 (sem a ação de qualquer tratamento) por 24 horas. Os eluatos foram mantidos em contato com células CHO-K1 por 24 horas. Como os dados apresentaram aderência à curva normal, foi aplicada a análise de variância (ANOVA) one-way, seguido dos testes de Tukey e Dunnett (p<0,05). Verificou-se que os materiais testados não demonstraram absorbância estatisticamente diferente em relação ao CN (p>0,05; Dunnett - XTT) e também não causaram comprometimento na capacidade proliferativa das células (p>0,05; Dunnett - Sobrevivência clonogênica). Assim, pode-se concluir que as amostras de PLC, PLC/LaPO4 e PLC/La20OAP não apresentaram citotoxicidade em células CHO-K1.

This monograph presents the main objective of analyzing the redundancy protection systems of protection for redundancy and block of energies against accident in presses and rotating equipment. After understanding two specific goals, on this case, show the systems of protection against accidents in rotating equipments, presses and similar; and discuss the vulnerabilities of current systems of protection against accidents in presses and similar, we will propose a system of autonomous redundancy of Programmable Logic Controller (PLC) operating simultaneously taking online in the event of failure of one of the two. The methodology was worked through a revision of a variety of bibliography, and interpretation of national and international standards as well as access to research on systems, practices used in industrial and companies supplying products and of companies and energy blockages solutions

The neutron-rich lead isotopes, up to Pb-216, have been studied for the first time, exploiting the fragmentation of a primary uranium beam at the FRS-RISING setup at GSI. The observed isomeric states exhibit electromagnetic transition strengths which deviate from state-of-the-art shell-model calculations. It is shown that their complete description demands the introduction of effective three-body interactions and two-body transition operators in the conventional neutron valence space beyond Pb-208.

Pathogenic Leptospira is the etiological agent of leptospirosis, a life-threatening disease that affects populations worldwide. Surface proteins have the potential to promote several activities, including adhesion. This work aimed to study the leptospiral coding sequence (CDS) LIC11087, genome annotated as hypothetical outer membrane protein. The LIC11087 gene was cloned and expressed in Escherichia coil BL21 (DE3) strain by using the expression vector pAE. The recombinant protein tagged with N-terminal 6XHis was purified by metal-charged chromatography and characterized by circular dichroism (CD) spectroscopy. The recombinant protein has the ability to mediate attachment to the extracellular matrix (ECM) components, laminin and plasma fibronectin, and was named Lsa30 (Leptospiral surface adhesin of 30 kDa). Lsa30 binds to laminin and to plasma fibronectin in a dose-dependent and saturable manner, with dissociation equilibrium constants (K-D) of 292 +/- 24 nM and 157 +/- 35 nM, respectively. Moreover, the Lsa30 is a plasminogen (PLC) receptor, capable of generating plasmin, in the presence of activator. This protein may interfere with the complement cascade by interacting with C4bp regulator. The Lsa30 is probably a new surface protein of Leptospira as revealed by immunofluorescence assays with living organisms and the reactivity with antibodies present in serum samples of experimentally infected hamsters. Thus, Lsa30 is a novel versatile protein that may play a role in mediating adhesion and may help pathogenic Leptospira to overcome tissue barriers and to escape the immune system. Cannabidiol (CBD) is the most abundant cannabinoid in Cannabis sativa that has no psychoactive properties. CBD has been approved to treat inflammation, pain and spasticity associated with multiple sclerosis (MS), of which demyelination and oligodendrocyte loss are hallmarks. Thus, we investigated the protective effects of CBD against the damage to oligodendrocyte progenitor cells (OPCs) mediated by the immune system. Doses of 1 mu M CBD protect OPCs from oxidative stress by decreasing the production of reactive oxygen species. CBD also protects OPCs from apoptosis induced by LPS/IFN gamma through the decrease of caspase 3 induction via mechanisms that do not involve CB1, CB2, TRPV1 or PPAR gamma receptors. Tunicamycin-induced OPC death was attenuated by CBD, suggesting a role of endoplasmic reticulum (ER) stress in the mode of action of CBD. This protection against ER stress-induced apoptosis was associated with reduced phosphorylation of eiF2 alpha, one of the initiators of the ER stress pathway. Indeed, CBD diminished the phosphorylation of PKR and eiF2 alpha induced by LPS/IFN gamma. The pro-survival effects of CBD in OPCs were accompanied by decreases in the expression of ER apoptotic effectors (CHOP, Bax and caspase 12), and increased expression of the anti-apoptotic Bcl-2. These findings suggest that attenuation of the ER stress pathway is involved in the 'oligoprotective' effects of CBD during inflammation. Exposure of luminal membranes of LLC-PK1 cells to ANG II induces dimerization of AT(1)/AT(2) receptors to activate SERCA and to promote Ca2+ mobilization. Am J Physiol Renal Physiol 302: F875-F883, 2012. First published January 4, 2012; doi:10.1152/ajprenal.00381.2011.-ANG II is secreted into the lumens of proximal tubules where it is also synthesized, thus increasing the local concentration of the peptide to levels of potential physiological relevance. In the present work, we studied the effect of ANG II via the luminal membranes of LLC-PK1 cells on Ca2+-ATPase of the sarco(endo) plasmic reticulum (SERCA) and plasma membrane (PMCA). ANG II (at concentrations found in the lumen) stimulated rapid (30 s) and persistent (30 min) SERCA activity by more than 100% and increased Ca2+ mobilization. Pretreatment with ANG II for 30 min enhanced the ANG II-induced Ca2+ spark, demonstrating a positively self-sustained stimulus of Ca2+ mobilization by ANG II. ANG II in the medium facing the luminal side of the cells decreased with time with no formation of metabolites, indicating peptide internalization. ANG II increased heterodimerization of AT(1) and AT(2) receptors by 140%, and either losartan or PD123319 completely blocked the stimulation of SERCA by ANG II. Using the PLC inhibitor U73122, PMA, and calphostin C, it was possible to demonstrate the involvement of a PLC -> DAG(PMA)-> PKC pathway in the stimulation of SERCA by ANG II with no effect on PMCA. We conclude that ANG II triggers SERCA activation via the luminal membrane, increasing the Ca2+ stock in the reticulum to ensure a more efficient subsequent mobilization of Ca2+. This first report on the regulation of SERCA activity by ANG II shows a new mechanism for Ca2+ homeostasis in renal cells and also for regulation of Ca2+-modulated fluid reabsorption in proximal tubules.

Soybean (Glycine max (Merrill) L) contains high content of aglycone isoflavones, as well as glucoside and malonylconjugates. In this work, the content of isoflavones in defatted soy flour was determined by reversed-phase high-performance liquid chromatography (RPHPLC) after alcoholic extraction in methanol/water mixture in the ratio 80:20 (v/v). It was observed that the heating treatment transformed the malonylglucosides into glucoside isoflavones. After heat treatment at 121 degrees C for 30 min, nearly all malonylisoflavones were converted into glucoside, but acetylisoflavones were not detected via RPHPLC analysis. Electrospray ionization mass spectrometry confirmed the presence of malonylisoflavones in heat-treated defatted soy flour by direct infusion analysis. (c) 2012 Elsevier Ltd. All rights reserved.
