Reactive oxygen species (ROS) are reactive molecules containing oxygen, that form as byproducts of aerobic metabolism, including immune system processes. Too much ROS may cause oxidative stress. In this study, we examined whether it can also limit the production of immune system compounds. To assess the relationship between antioxidant status and immunity we evaluated the effect of dietary supplementation with organic selenium, given at various levels for 10 days, on the antioxidant and immune system of the pacu fish (Piaractus mesopotamicus). Fish fed a diet containing 0.6 mg Se-yeast kg(-1) showed significant improvement in antioxidant status, as well as in hematological and immunological profiles. Specifically, they had the highest counts for catalase (CAT), glutathione peroxidase (GPx), glutathione S-transferase (GST), red blood cells, and thrombocytes; the highest leukocyte count (particularly for monocytes); and the highest serum lysozyme activity. There was also a positive correlation between GPx and lysozyme in this group of fish. These findings indicate that short-term supplementation with 0.6 mg Se-yeast kg(-1) reestablished the antioxidative status, allowing the production of innate components which can boost immunity without the risk of oxidative stress. This study shows a relationship between oxidative stress and immunity, and, from a practical perspective, shows that improving immunity and health in pacu through the administration of selenium could improve their growth performance.

Cat's claw (Uncaria tomentosa) is an Amazon herb using in native cultures in Peru. In mammals, it has been described several effects of this herb. However, this is the first report of its use on the diet of fish. The aim of this study was to determinate the effect of this plant on the growth and immune activity in Oreochromis niloticus. Nile tilapia (81.3 ± 4.5 g) were distributed into 5 groups and supplemented with 0 (non-supplement fish), 75, 150, 300, and 450 mg of U. tomentosa.kg(-1) of diet for a period of 28 days. Fish were inoculated in the swim bladder with inactivated Streptococcus agalactiae and samples were taken at 6, 24, and 48 h post inoculation (HPI). Dose dependent increases were noted in some of the evaluated times of thrombocytes and white blood cells counts (WBC) in blood and exudate, burst respiratory activity, lysozyme activity, melanomacrophage centers count (MMCs), villi length, IgM by immunohistochemistry in splenic tissue, and unexpectedly on growth parameters. However, dietary supplementation of this herb did not affect red blood cells count (RBC), hemoglobin, and there were no observed histological lesions in gills, intestine, spleen, and liver. The current results demonstrate for the first time that U. tomentosa can stimulate fish immunity and improve growth performance in Nile tilapia.

The course of leprosy depends of the host immune response which ranges from the lepromatous pole (LL) to the tuberculoid pole (TT). A comparative study was conducted in 60 patients with the LL and TT The results showed a mean expression of TGF-beta of 339 +/- 99.4 cells/field for TT and of 519.2 +/- 68.2 cells/field for LL. Frequency of apoptosis was 6.3 +/- 1.8 in TT and 14.0 +/- 6.1 in LL. A correlation (p = 0.0251) between TGF-beta and caspase-3 in the LL was found. Objective: NALP3-inflammasome is an innate mechanism, alternative to type-1 interferon, which is able to recognize nucleic acids and viruses in the cytoplasm and to induce pro-inflammatory response. Here, we hypothesized the involvement of inflammasome in the early defense against HIV-1 and in the full maturation of dendritic cells: for this, we evaluated the response of dendritic cells pulsed with HIV-1 in terms of inflammasome activation in healthy donors. Moreover, inflammasome response to HIV was evaluated in HIV-infected individuals. Design and methods: Monocyte-derived dendritic cells isolated from 20 healthy individuals (HC-DC) and 20 HIV-1-infected patients (HIV-DC) were pulsed with alditrithiol-2-inactivated HIV-1. We then analyzed inflammasome genes expression and interleukin-1 beta (IL-1 beta) secretion. Results: In HC-DC, HIV-1 induced higher NLRP3/NALP3 mRNA expression compared with other inflammasome genes such as NALP1/NLRP1 or IPAF/NLRC4 (P < 0.001). This augmented expression was accompanied by CASP1-increased and IL1B-increased mRNA levels and by a significant increment of IL-1b secretion (P < 0.05). Otherwise, HIV-1 failed to activate inflammasome and cytokine production in HIV-DC. HIV-DC showed an increased NLRP3/NALP3 basal expression, suggesting a chronic inflammatory profile of patients' immune cells. Conclusion: HIV-1 was able to induce a NALP3-inflammasome response in healthy individuals, indicating that this inflammasome could play a role in the first steps of HIV-1 infection; the consequent inflammatory process may be important for directing host immune response against the virus and/or disease progression. HIV-DC seemed to be chronically activated, but unresponsive against pathogens. Our findings could be of interest considering the ongoing research about dendritic cell manipulation and therapeutic strategies for AIDS involving dendritic cell-based immune-vaccines. The expression of Langerhans cell (LC) and dermal dendritic cell (dDC) as well as T CD4+ and CD8+ immune responses was evaluated in the skin of BALB/c mice experimentally infected by L. (L.) amazonensis (La) and L. (V.) braziliensis (Lb). At 4th and 8th weeks post infection (PI), skin biopsies were collected to determine the parasite load and CD207+, CD11c+, CD4+, CD8+, iNOS+ cellular densities. Cytokine (IFN-?, IL-4 and IL-10) profiles were also analysed in draining lymph node. At 4th week, the densities of CD207+ and CD11c+ were higher in the La infection, while in the Lb infection, these markers revealed a significant increase at 8th week. At 4th week, CD4+ and CD8+ were higher in the La infection, but at 8th week, there was a substantial increase in both markers in the Lb infection. iNOS+ was higher in the Lb infection at 4th and 8th weeks. In contrast, the parasite load was higher in the La infection at 4th and 8th weeks. The concentration of IFN-? was higher in the Lb infection, but IL-4 and IL-10 were higher in the La infection at 4th and 8th weeks. These results confirm the role of the Leishmania species in the BALB/c mice disease characterized by differences in the expression of dendritic cells and cellular immune response.

The dengue virus non-structural 1 (NS1) protein contributes to evasion of host immune defenses and represents a target for immune responses. Evidences generated in experimental models, as well as the immune responses elicited by infected individuals, showed that induction of anti-NS1 immunity correlates with protective immunity but may also result in the generation of cross-reactive antibodies that recognize platelets and proteins involved in the coagulation cascade. In the present work, we evaluated the immune responses, protection to type 2 dengue virus (DENV2) challenges and safety parameters in BALB/c mice vaccinated with a recombinant NS1 protein in combination with three different adjuvants: aluminum hydroxide (alum), Freund's adjuvant (FA) or a genetically detoxified derivative of the heat-labile toxin (LTG33D), originally produced by some enterotoxigenic Escherichia coil (ETEC) strains. Mice were subcutaneously (s.c.) immunized with different vaccine formulations and the induced NS1-specific responses, including serum antibodies and T cell responses, were measured. Mice were also subjected to lethal challenges with the DENV2 NGC strain. The results showed that maximal protective immunity (50%) was achieved in mice vaccinated with NS1 in combination with LIG33D. Analyses of the NS1-specific immune responses showed that the anti-virus protection correlated mainly with the serum anti-NS1 antibody responses including higher avidity to the target antigen. Mice immunized with LTG33D elicited a prevailing IgG2a subclass response and generated antibodies with stronger affinity to the antigen than those generated in mice immunized with the other vaccine formulations. The vaccine formulations were also evaluated regarding induction of deleterious side effects and, in contrast to mice immunized with the FA-adjuvanted vaccine, no significant hepatic damage or enhanced C-reactive protein levels were detected in mice immunized with NS1 and LTG33D. Similarly, no detectable alterations in bleeding time and hematological parameters were detected in mice vaccinated with NS1 and LTG33D. Altogether, these results indicate that the combination of a purified recombinant NS1 and a nontoxic LT derivative is a promising alternative for the generation of safe and effective protein-based anti-dengue vaccine. Mygalin is an antibacterial molecule isolated froth the hemocytes of the spider Acanthoscurria gomesiana. It was identified as bis-acylpolyamine spermidine. We evaluated the modulator effects of synthetic Mygalin in the innate immune response. We demonstrate that Mygalin induces IFN-gamma synthesis by splenocytes increasing the nitrite secretion by splenocytes and macrophages. A specific inhibitor of iNOS abrogated Mygalin-induced nitrite production in macrophages independent of IFN-gamma activation. In addition, Mygalin-activated macrophages produced TNF-alpha but not IL-1 beta, demonstrating that Mygalin does not act directly on the inflammasome. Furthermore, this compound did not affect spontaneous or Concanavalin A-induced proliferative responses by murine splenocytes and did not induce IL-5 or apoptosis of splenocytes or bone marrow-derived macrophages. These data provide evidence that Mygalin modulates the innate immune response by inducing IFN-gamma and NO synthesis. The combined immune regulatory and antibacterial qualities of Mygalin should be explored as a strategy to enhance immune responses in infection. (C) 2012 Elsevier Inc. All rights reserved.

The humoral immune response is dependent on the formation of antibodies. Antibodies are produced by terminally differentiated B cells, plasma cells. Plasma cells are generated either directly from antigen challenged B cells, memory cells or from cells that have undergone the germinal center (GC) reaction. The GC is the main site for class switch, somatic hypermutation and generation of memory cells. Different factors, both internal and external, shape the outcome of the immune response. In this thesis, we have studied a few factors that influence the maturation of the humoral response. We have studied how age affects the response, and we show that responses against thymus dependent antigens (TD) are more affected than responses to thymus independent (TI) antigens, in concordance with the view that the T cell compartment is more affected by age than the B cell compartment. Furthermore, we demonstrate that priming early in life have a big influence on the immune response in the aged individual. Priming with a TI form of the carbohydrate dextran B512 (Dx) induces a reduction of IgG levels in later TD responses against Dx. We have evaluated possible mechanisms for this reduction. The reduction does not seem to be caused by clonal exhaustion or antibody mediated mechanisms. We also showed that the reduced TD response after TI priming can be induced against another molecule than Dx. With the hypothesis that TI antigens induce a plasma cell biased maturation of the responding B cells, we examined the presence of Blimp-1, a master regulator of plasma cell differentiation, in GCs induced by TD and TI antigen. Blimp-1 was found earlier in GCs induced by TI antigen and the staining intensity in these GCs was stronger than in TD antigen induced GCs, indicating that plasma cells might be continuously recruited from these GCs. B cells undergoing the GC reaction are thought to be under a strict selection pressure that removes cells with low affinity for the antigen and also cells that have acquired self-reactivity. We investigated the effect of apoptotic deficiencies on the accumulation of somatic mutations in GC B cells. In mice lacking the death receptor Fas, lpr mice, the frequency of mutations was increased but the pattern of the mutations did not differ from wild type mice. In contrast, mice over-expressing the anti-apoptotic protein Bcl-2, had a lowered frequency of mutations and the mutations introduced had other characteristics.

Das Humane Cytomegalovirus (HCMV) ist ein Erreger von großer klinischer Relevanz. Die HCMV-Infektion, die insbesondere bei immunsupprimierten Patienten mit hoher Morbidität und Mortalität assoziiert ist, wird vorwiegend durch CD8+-zytotoxische T-Lymphozyten (CTL) kontrolliert. Das Tegumentprotein pp65 und das immediate early 1-Protein (IE1) waren als die dominanten CTL-Antigene bekannt. Ziel dieser Arbeit war es, die zur Immundominanz des pp65 führenden molekularen Mechanismen aufzuklären und die Grundlagen für die Analyse der IE1-spezifischen Immunantwort zu erarbeiten. Durch Peptidimmunisierung HLA-A2-transgener Mäuse wurden hochaffine pp65-spezifische CTL-Klone generiert. Für die Generierung ähnlicher CTL-Klone gegen IE1 konnte erstmals ein konserviertes HLA-A2-bindendes Peptid identifiziert werden. Mit Hilfe der pp65-spezifischen CTL-Klone konnte gezeigt werden, dass das durch Viruspartikel in die Zelle eingebrachte pp65 die Erkennung infizierter Zellen durch CD8+-CTL vermittelt. Durch den Nachweis der außergewöhnlichen Stabilität von pp65 in der Zelle gelang es, eine hohe metabolische Umsatzrate als eine Ursache von Immundominanz auszuschließen. Dagegen hob die Blockierung des CRM1-vermittelten nukleären Exportweges durch Zugabe von Hemmstoffen oder Zutransfektion kompetitiver Inhibitoren die Erkennung des pp65 nahezu auf. Hiermit wurde erstmalig eine Abhängigkeit der Präsentation eines immundominanten nukleären Proteins vom nukleozytoplasmatischen Transport nachgewiesen. Die Erkenntnisse dieser Arbeit stellen die Grundlage für die detaillierte Analyse der Zusammenhänge zwischen nukleärem Export und Antigenpräsentation dar.

Das humane Cytomegalovirus (HCMV) ist ein fakultativ-pathogener Erreger, der bei Patienten mit geschwächter oder unausgereifter Immunabwehr schwerwiegende Erkrankungen hervorrufen kann. Wie alle Herpesviren zeigt das HCMV eine streng koordinierte Expression viraler Gene, die in eine „sehr frühe-“ (IE), „frühe “ (E) und „späte-“ (L) Phase unterteilt werden kann. Die Produkte der IE-Gene IE1 und IE2 sind für die Expression der frühen Gene und somit für die Initiation der viralen DNA-Replikation entscheidend. Sie greifen gleichzeitig in den zellulären Stoffwechsel ein und schaffen damit optimale Vorraussetzungen für die virale Vermehrung. Zu Beginn dieser Arbeit war bekannt, dass HCMV in lytisch infizierten Zellen ein abundantes IE-Transkript von 5 kb (IE4-RNA) exprimierte, dessen Funktion bislang unklar war. Ältere Publikationen deuteten darauf hin, dass die IE4-Genregion an der Transformation eukaryonter Zellen beteiligt sein könnte. Neuere Arbeiten zeigten, dass es sich bei diesem IE4-Transkript um ein metabolisch stabiles Intron handelt. Im Rahmen dieser Arbeit sollte zunächst geklärt werden, ob die IE4-Genregion ein Protein kodiert. In der Folge sollten mit viralen Deletionsmutanten Hinweise auf die biologische Funktion des IE4-Bereichs erarbeitet werden. Durch Northern Blot Analysen und cDNA-Klonierungsexperimente konnte eine Reihe neuer Spleiß-Varianten der IE4-RNA identifiziert werden. Durch Sequenzanalysen wurde gezeigt, dass diese Transkripte keine längeren offenen Leserahmen enthalten. Zusammen mit bereits publizierten Erkenntnissen, kann aus diesen Ergebnissen mit hoher Wahrscheinlichkeit geschlossen werden, dass die IE4 Region nicht für ein Protein kodiert. Zur Analyse der biologischen Funktion der IE4-Region wurde das DNA-Genom des HCMV gezielt mutagenisiert. Eine phänotypische Analyse der entsprechenden Viren mittels Reportergen-Tests und quantitativer RealTime RT-PCR zeigte, dass einige der Mutanten eine verringerte Expression früher Gene aufwiesen, die mit einer Beeinträchtigung ihrer Replikationsfähigkeit in Fibroblastenkulturen korrelierte. Dabei war die Ausbildung eines Phänotyps jedoch von dem genetischen Hintergrund des verwendeten viralen Ausgangsstammes abhängig. Auffällig war, dass phänotypische Veränderungen nur bei solchen Mutanten sichtbar wurden, die auf der Grundlage des Laborstammes Ad169 des HCMV generiert worden waren. Die nachfolgende Analyse der Ausgangsstämme ergab deutliche Unterschiede in der IE-Genexpression. Die Ergebnisse dieser Arbeit zeigen somit, dass die IE4-RNA mit hoher Wahrscheinlichkeit nicht für ein Protein kodiert, aber bei limitierender Expression der essentiellen Regulatoren IE1 und IE2 die frühe lytische Genexpression stimuliert. Die Ergebnisse dieser Arbeit stellen die Grundlage für nachfolgende Untersuchungen zur Aufklärung der molekularen Funktion der IE4-RNA im Rahmen der lytischen Infektion des HCMV dar.
