Six Holstein cows fitted with ruminal cannulas and permanent indwelling catheters in the portal vein, hepatic vein, mesenteric vein, and an artery were used to study the effects of abomasal glucose infusion on splanchnic plasma concentrations of gut peptides. The experimental design was a randomized block design with repeated measurements. Cows were assigned to one of 2 treatments: control or infusion of 1,500 g of glucose/d into the abomasum from the day of parturition to 29 d in milk. Cows were sampled 12 ± 6 d prepartum and at 4, 15, and 29 d in milk. Concentrations of glucose-dependent insulinotropic polypeptide, glucagon-like peptide 1(7–36) amide, and oxyntomodulin were measured in pooled samples within cow and sampling day, whereas active ghrelin was measured in samples obtained 30 min before and after feeding at 0800 h. Postpartum, dry matter intake increased at a lower rate with infusion compared with the control. Arterial, portal venous, and hepatic venous plasma concentrations of the measured gut peptides were unaffected by abomasal glucose infusion. The arterial, portal venous, and hepatic venous plasma concentrations of glucose-dependent insulinotropic polypeptide and glucagon-like peptide 1(7–36) amide increased linearly from 12 d prepartum to 29 d postpartum. Plasma concentrations of oxyntomodulin were unaffected by day relative to parturition. Arterial and portal venous plasma concentrations of ghrelin were lower postfeeding compared with prefeeding concentrations. Arterial plasma concentrations of ghrelin were greatest prepartum and lowest at 4 d postpartum, giving a quadratic pattern of change over the transition period. Positive portal venous-arterial and hepatic venous–arterial concentration differences were observed for glucagon-like peptide 1(7–36) amide. A negative portal venous–arterial concentration difference was observed for ghrelin pre-feeding. The remaining portal venous–arterial and hepatic venous–arterial concentration differences of gut peptides did not differ from zero. In conclusion, increased postruminal glucose supply to postpartum transition dairy cows reduced feed intake relative to control cows, but did not affect arterial, portal venous, or hepatic venous plasma concentrations of gut peptide hormones. Instead, gut peptide plasma concentrations increased as lactation progressed. Thus, the lower feed intake of postpartum dairy cows receiving abomasal glucose infusion was not attributable to changes in gut peptide concentrations.

Grasslands restoration is a key management tool contributing to the long-term maintenance of insect populations, providing functional connectivity and mitigating against extinction debt across landscapes. As knowledge of grassland insect communities is limited, the lag between the initiation of restoration and the ability of these new habitats to contribute to such processes is unclear. Using ten data sets, ranging from 3 to 14 years, we investigate the lag between restoration and the establishment of phytophagous beetle assemblages typical of species rich grasslands. We used traits and ecological characteristics to determine factors limiting beetle colonisation, and also considered how food-web structure changed during restoration. For sites where seed addition of host-plants occurred the success in replicating beetle assemblages increased over time following a negative exponential function. Extrapolation beyond the existing data set tentatively suggested that success would plateau after 20 years, representing a c. 60% increase in assemblage similarity to target grasslands. In the absence of seed addition, similarity to the target grasslands showed no increase over time. Where seed addition was used the connectance of plant-herbivore food webs decreased over time, approaching values typical of species rich grasslands after c. 7 years. This trend was, however, dependent on the inclusion of a single site containing data in excess of 6 years of restoration management. Beetles not capable of flight, those showing high degrees of host-plant specialisation and species feeding on nationally rare host plants take between 1 and 3 years longer to colonise. Successful grassland restoration is underpinned by the establishment of host-plants, although individual species traits compound the effects of poor host-plant establishment to slow colonisation. The use of pro-active grassland restoration to mitigate against future environmental change should account for lag periods in excess of 10 years if the value of these habitats is to be fully realised.

We investigate the role of the anthropogenic heat flux on the urban heat island of London. To do this, the time-varying anthropogenic heat flux is added to an urban surface-energy balance parametrization, the Met Office–Reading Urban Surface Exchange Scheme (MORUSES), implemented in a 1 km resolution version of the UK Met Office Unified Model. The anthropogenic heat flux is derived from energy-demand data for London and is specified on the model's 1 km grid; it includes variations on diurnal and seasonal time-scales. We contrast a spring case with a winter case, to illustrate the effects of the larger anthropogenic heat flux in winter and the different roles played by thermodynamics in the different seasons. The surface-energy balance channels the anthropogenic heat into heating the urban surface, which warms slowly because of the large heat capacity of the urban surface. About one third of this additional warming goes into increasing the outgoing long-wave radiation and only about two thirds goes into increasing the sensible heat flux that warms the atmosphere. The anthropogenic heat flux has a larger effect on screen-level temperatures in the winter case, partly because the anthropogenic flux is larger then and partly because the boundary layer is shallower in winter. For the specific winter case studied here, the anthropogenic heat flux maintains a well-mixed boundary layer through the whole night over London, whereas the surrounding rural boundary layer becomes strongly stably stratified. This finding is likely to have important implications for air quality in winter. On the whole, inclusion of the anthropogenic heat flux improves the comparison between model simulations and measurements of screen-level temperature slightly and indicates that the anthropogenic heat flux is beginning to be an important factor in the London urban heat island.

Bacterial soft rot is a globally significant plant disease that causes major losses in the production of many popular crops, such as potato. Little is known about the dispersal and ecology of soft-rot enterobacteria, and few animals have been identified as vectors for these pathogens. This study investigates whether soil-living and bacterial-feeding nematodes could act as vectors for the dispersal of soft-rot enterobacteria to plants. Soft-rot enterobacteria associated with nematodes were quantified and visualized through bacterial enumeration, GFP-tagging, and confocal and electron scanning microscopy. Soft-rot enterobacteria were able to withstand nematode grazing, colonize the gut of Caenorhabditis elegans and subsequently disperse to plant material while remaining virulent. Two nematode species were also isolated from a rotten potato sample obtained from a potato storage facility in Finland. Furthermore, one of these isolates (Pristionchus sp. FIN-1) was shown to be able to disperse soft-rot enterobacteria to plant material. The interaction of nematodes and soft-rot enterobacteria seems to be more mutualistic rather than pathogenic, but more research is needed to explain how soft-rot enterobacteria remain viable inside nematodes.

The potential risk of agricultural pesticides to mammals typically depends on internal concentrations within individuals, and these are determined by the amount ingested and by absorption, distribution, metabolism, and excretion (ADME). Pesticide residues ingested depend, amongst other things, on individual spatial choices which determine how much and when feeding sites and areas of pesticide application overlap, and can be calculated using individual-based models (IBMs). Internal concentrations can be calculated using toxicokinetic (TK) models, which are quantitative representations of ADME processes. Here we provide a population model for the wood mouse (Apodemus sylvaticus) in which TK submodels were incorporated into an IBM representation of individuals making choices about where to feed. This allows us to estimate the contribution of individual spatial choice and TK processes to risk. We compared the risk predicted by four IBMs: (i) “AllExposed-NonTK”: assuming no spatial choice so all mice have 100% exposure, no TK, (ii) “AllExposed-TK”: identical to (i) except that the TK processes are included where individuals vary because they have different temporal patterns of ingestion in the IBM, (iii) “Spatial-NonTK”: individual spatial choice, no TK, and (iv) “Spatial-TK”: individual spatial choice and with TK. The TK parameters for hypothetical pesticides used in this study were selected such that a conventional risk assessment would fail. Exposures were standardised using risk quotients (RQ; exposure divided by LD50 or LC50). We found that for the exposed sub-population including either spatial choice or TK reduced the RQ by 37–85%, and for the total population the reduction was 37–94%. However spatial choice and TK together had little further effect in reducing RQ. The reasons for this are that when the proportion of time spent in treated crop (PT) approaches 1, TK processes dominate and spatial choice has very little effect, and conversely if PT is small spatial choice dominates and TK makes little contribution to exposure reduction. The latter situation means that a short time spent in the pesticide-treated field mimics exposure from a small gavage dose, but TK only makes a substantial difference when the dose was consumed over a longer period. We concluded that a combined TK-IBM is most likely to bring added value to the risk assessment process when the temporal pattern of feeding, time spent in exposed area and TK parameters are at an intermediate level; for instance wood mice in foliar spray scenarios spending more time in crop fields because of better plant cover.

Many studies evaluating model boundary-layer schemes focus either on near-surface parameters or on short-term observational campaigns. This reflects the observational datasets that are widely available for use in model evaluation. In this paper we show how surface and long-term Doppler lidar observations, combined in a way to match model representation of the boundary layer as closely as possible, can be used to evaluate the skill of boundary-layer forecasts. We use a 2-year observational dataset from a rural site in the UK to evaluate a climatology of boundary layer type forecast by the UK Met Office Unified Model. In addition, we demonstrate the use of a binary skill score (Symmetric Extremal Dependence Index) to investigate the dependence of forecast skill on season, horizontal resolution and forecast leadtime. A clear diurnal and seasonal cycle can be seen in the climatology of both the model and observations, with the main discrepancies being the model overpredicting cumulus capped and decoupled stratocumulus capped boundary-layers and underpredicting well mixed boundary-layers. Using the SEDI skill score the model is most skillful at predicting the surface stability. The skill of the model in predicting cumulus capped and stratocumulus capped stable boundary layer forecasts is low but greater than a 24 hr persistence forecast. In contrast, the prediction of decoupled boundary-layers and boundary-layers with multiple cloud layers is lower than persistence. This process based evaluation approach has the potential to be applied to other boundary-layer parameterisation schemes with similar decision structures.

A first case of subsociality is reported for the genus Latrodectus. Individuals were found sharing the same web and feeding together. In captivity they showed mutual tolerance and communal feeding. This finding is remarkable for two reasons. First, widow spiders, even compared with other spiders, are famously aggressive and cannibalistic so that social behavior in the genus was unexpected. Second, the genus nests outside the ""Anelosimus + lost colulus"" clade where all the other social theridiids are found.

Syftet var att beskriva omvårdnaden kring nyfödda barns tillmatning på svenska BB-avdelningar, om föreskrifter följdes och om hänsyn togs till eventuell allergihereditet. Metod: Deskriptiv tvärsnittsstudie med kvantitativ ansats Resultat: Av alla nyfödda barn vid 26 barnkliniker i Sverige vecka 26 år 2009 (n=849) tillmatades 18 % . Moderns bröstmjölk gavs till 16% medan 78 % tillmatades med komjölksbaserad modersmjölksersättning. Den vanligaste orsaken till tillmatning av ersättning var att mammans egen mjölkproduktion inte hade kommit igång (24 %). Andra vanligt förekommande anledningar till tillmatning var lågt blodsocker (18 %) samt sugproblem hos barnen (16 %). Sexton barn tillmatades trots ett blodsockervärde motsvarande >2.2mmol/l. Vad gäller allergiförekomst tillfrågades endast 37 % av barnens föräldrar före tillmatning. Dokumentation om tillmatning fanns noterat i 80 % av barnens journaler. Motsvarande siffra för mammornas journaler var 34 %. Konklusion: Bristfälliga omvårdnadsåtgärder vidtas i samband med tillmatning av nyfödda spädbarn på BB-avdelningar, där allergifrågor ställs i för låg utsträckning. BB-kliniker har varierande referensvärde för vad som räknas som lågt blodsocker. Omvårdnadsbehov för nyfödda barn och deras föräldrar, där förmedlad kunskap om amningens betydelse, tas inte på allvar och oklara riktlinjer skapar förvirring. Föreskrifter och riktlinjer bör följas i större utsträckning an vad denna studie visar.

Este estudo examinou as respostas de estresse de juvenis de pirarucu transportados em sistema fechado. Pirarucu (Arapaima gigas) é um peixe nativo da bacia Amazônica, da família Osteoglossidae que possui respiração aérea obrigatória. Foi realizado um transporte de curta duração (6 h) em sacos de polietileno inflados com ar atmosférico (grupo ar) ou com oxigênio puro (grupo oxi). O oxigênio dissolvido foi o único parâmetro de qualidade da água que apresentou diferença estatística entre os grupos, e como esperado, o oxigênio estava supersaturado para o grupo oxi. Não houve mortalidade após o transporte em ambos os grupos. Os peixes se alimentaram 36 h após o transporte e apresentaram um consumo de ração habitual após 72 h. As respostas fisiológicas foram semelhantes nos dois grupos. O cortisol não apresentou mudança significativa durante o período de amostragem. Ao contrário da maioria das espécies, os valores de cortisol se apresentaram inalterados nos dois grupos durante a amostragem, enquanto a glicose teve um aumento significativo até 12 h após o transporte. Os resultados mostram que o transporte de pirarucu em sacos de polietileno pode ser realizado com ar atmosférico ou oxigênio puro, uma vez que as respostas de estresse, a qualidade da água e o comportamento alimentar após 36 h foram similar entre os grupos.

The present study aimed to compare the main zootechnical and economic indicators of farming tilapia in superintensive system developed in two different net cages dimensions: 6 and 18 m(3). Were used 12,960 juveniles male reversed Nile tilapia (Oreochromis niloticus), strain supreme, with an initial average weight of 32.0 g (+/- 0.85) for 6 m(3) net cages and 88.5 g (+/- 0.96) for 18 m(3) net cages. The fish were fed three times a day with commercial extruded ration, crude protein levels from 40 to 32%, particle size from 2 to 8 mm and daily feeding rate starting with 8% of live weight (LW), decreasing up to 1% LW. It was conclued that, although no statistical difference in most indicators zootechnical (P>0.05), the production of tilapias in bigger net cages resulted in lower average total cost and higher rentability per kg of fish produced, 34.4% higher, resulting in better profitability.

Objetivou-se estudar em laboratório a biologia de Dione juno juno (Cramer), determinando-se os efeitos da densidade larval e da alimentação das larvas com folhas de nove genótipos de maracujazeiro. Foram analisados a duração, o peso e a viabilidade das fases larval e pupal e a longevidade dos adultos sem alimento. Verificou-se que a densidade de cinco lagartas por recipiente proporcionou melhor desenvolvimento do inseto do que uma e dez lagartas. Passiflora alata e P. setacea apresentaram resistência do tipo não-preferência para alimentação e/ou antibiose, enquanto o híbrido P. alata2 x P. macrocarpa apresentou não-preferência para alimentação, possivelmente associada à presença de compostos químicos com alto grau de repelência a D. juno juno ou a supressores de alimentação.

Os efeitos de extratos aquosos de amêndoas de Azadirachta indica e frutos de Sapindus saponaria, aplicados em discos de folhas de couve (Brassica oleracea var. acephala) nas concentrações de 0,0117% e 1,0342% (p/v), respectivamente, foram estudados sobre a alimentação das lagartas de Ascia monuste orseis. Avaliou-se a atratividade e o consumo de área foliar de lagartas de primeiro e terceiro ínstar durante 24 horas, em condições de laboratório (T = 25 ± 2ºC, UR = 60 ± 10% e fotofase = 12 horas). Nos testes com e sem chance de escolha, para lagartas de primeiro ínstar e teste sem chance de escolha para lagartas de terceiro ínstar, não houve diferença quanto à atratividade das lagartas. No teste com chance de escolha para lagartas de terceiro ínstar, houve menor atratividade das lagartas pelos discos de folhas tratadas com S. saponaria, diferindo da testemunha. No decorrer de 24 horas de avaliações, pode-se observar 58,3% de lagartas atraídas na testemunha, não diferindo de A. indica e diferindo de S. saponaria, com 39,3% e 2,4% das lagartas atraídas, respectivamente. Quanto ao consumo de área foliar, o extrato de S. saponaria diminuiu o consumo de lagartas, em todos os testes realizados. Quando não tinham opção de escolha para se alimentar de folhas sem os extratos, as lagartas consumiram as folhas tratadas nas concentrações testadas, porém em menor quantidade. Os extratos testados neste experimento demonstram ter efeitos sobre a alimentação das lagartas de A. monuste orseis, possivelmente com propriedades deterrentes e/ou supressoras de alimentação.
