A novel protein, named BAS-AH, was purified and characterized from the skin of the toad Bufo andrewsi. BAS-AH is a single chain protein and the apparent molecular weight is about 63 kDa as judged by SDS-PAGE. BAS-AH was determined to bind heme (0.89 mol heme/mol protein) as determined by pyridine haemochrome analysis. Fifty percentage cytotoxic concentration (CC50) of BAS-AH on C8166 cells was 9.5 mu M. However, at concentrations that showed little effect oil cell viability, BAS-AH displayed dose dependent inhibition oil HIV-1 infection and replication. The antiviral selectivity indexes corresponding to the measurements of syncytium formation and HIV-1 p24 (CC50/EC50) were 14.4 and 11.4, respectively, corresponding to the . BAS-AH also showed an inhibitory effect on the activity of recombinant HIV-1 reverse transcriptase (IC50 = 1.32 mu M). The N-terminal sequence of BAS-AH was determined to be NAKXKADVIGKISILLGQDNLSNIVAM, which exhibited little identity with other known anti-HIV-1 proteins. Amphibian skin secretions are rich in antimicrobial peptides acting as important components of innate defense system against invading microorganisms. A novel type of peptide, designated as maximin S, was deduced by random sequencing of 793 clones from a constructed Bombina maxima skin cDNA library. The putative primary structures of maximin S peptides can be grouped into five species, in which maximin S I has 14 amino acid residues and the rest of maximin S peptides (S2-S5) all have 18 amino acid residues. Unlike most of the amphibian antimicrobial peptides so far identified, the newly characterized four maximin S precursors are composed of maximin S I and different combinations of tandem repeated maximin S2-S5 linked by internal peptides. Except maximin S I, the predicted secondary structures of maximin S2-S5 show a similar amphipathic alpha-helical structure. MALDI-TOF mass spectrometry analysis of partially isolated skin secretions of the toad indicates that most of the deduced maximin S peptides are expressed. Two deduced maximin S peptides (S1, S4) were synthesized and their antimicrobial activities were tested. Maximin S4 only had an antibiotic activity against mycoplasma and had no antibacterial or antifungal activity toward tested strains. Maximin S1 had no activity under the same conditions. Crude mucus and its partially purified fractions from two marine catfish from Mumbai, Arius dussumieri and Osteogeneiosus militaris were assayed for their crinotoxicity through assays for hemolysis and haemagglutination of chicken erythrocytes, formation of paw edema in mice, and antibacterial activity against one gram-positive and four gram-negative bacteria. Assays were also done to block the edema using Phineramine maleate, Piroxicam, and Atropine sulfate. Crude toxin as well as their fractions from both the fishes exhibited haemolytic and haemagglutinating activities on chicken blood, besides edematous activity in mice models. The edematous activity was blocked by Phineramine maleate and Piroxicam but enhanced by Atropine sulfate; however, all these activities, either blocking or enhancing, were statistically insignificant. Antibacterial activity was absent in all the extracts tested.

The role of Vibrio parahaemolyticus in food borne gastroenteritis outbreaks associated primarily with the consumption of contaminated seafoods has been well documented. Information pertaining to various aspects of its occurrence in seafoods, procedures for isolation and identification, generation time and inactivation profiles is discussed. Emphasis has been given to the response of V. parahaemolyticus to low temperatures, heating and antibacterial agents. The public health hazard posed by the pathogen is outlined and the guidelines for control are reviewed in detail.

目的 体外测定两种AZT-氟喹诺酮偶联物SRLZ和SROZ的抗HIV-1活性及抗菌活性.方法 通过合胞体抑制、HIV-1感染细胞保护、HIV-1 p24抗原测定等方法检测急性感染中化合物对HIV-1实验株、临床分离株的抑制作用和对慢性感染细胞中的病毒复制影响;通过体外金黄色葡萄球菌的抑制实验检测化合物的抗菌活性.结果 AZT-氟喹诺酮偶联物SRLZ和SROZ能抑制HIV-1实验株诱导的合胞体形成,减少病毒感染细胞的死亡和抑制病毒在细胞内的复制;SRLZ和SROZ对HIV-1临床分离株也有较好的抑制作用;SRLZ和SROZ对HIV-1的抑制活性与AZT相近;AZT-氟喹诺酮偶联物也能抑制金黄色葡萄球菌,其MIC值与其相应的阳性药物相近.结论 SRLZ和SROZ有很好的抗HIV-1活性和抗菌活性.

Biodegradable protein-based film was developed by incorporating cinnamon essential oil (CEO) into whey protein concentrate (WPC) at level of 0.8% and 1.5% v/v. Then physical and mechanical properties of the films were evaluated. Adding CEO to the WPC matrix decreased the water vapour permeability of the films and water solubility. Films containing CEO showed significant antibacterial activity both gram-positive and gram-negative strains and exhibited significant inhibitory effect on the studied fungi. In continue, the effect of whey coating and whey coating incorporated with 1.5% CEO on quality and shelf life of Huso huso fillet during refregrated (4±1°C) storage period were also investigated. The control and treated fish samples were analyzed for microbiological (total viable count, psychrophilic counts), chemical (PV, TBA, FFA, pH, TVB-N), and sensory characteristics in 4-day intervals up of microbial, chmical and sensoy analyses indicated lower levels of PV, TBA, FFA, pH, TVB-N in coasted sampels and specially, those with CEO while were kept in refrigerator. Based on results, whey protein edible coating contain 1.5% cinnamon essential oil could enhance preserving ability Huso huso during storage cold.

Fish are an important part of a healthy diet since they contain high quality protein, but typically present a low fat percent when compared to other meats. Fish is an extremely perishable food commodity. On the other hand, food borne diseases are still a major problem in the world, even in well-developed countries. The increasing incidence of food borne diseases coupled with the resultant social and economic implications means there is a constant striving to produce safer food and to develop new antimicrobial agents concerns over the safety of some chemical preservatives and negative consumer reactions to preservatives they perceive as chemical and artificial, have prompted on increased interest in more ‘‘naturalgreen’’ alternatives for the maintenance or extension of product shelf-life. Particular interest has focused on the potential applications of plant essential oils. However, to establish the usefulness of natural antimicrobial preservatives, they must be evaluated alone and in combination with other preservation factors to determine whether there are synergistic effects and multiple hurdles can be devised. In this study, were evaluated the effects of different concentrations of Rosmarinus officinalis and nisin and storage time (15 days) on growth of Streptococcus iniae GQ850377 in a lab conditions and a food model system (fillets of rainbow trout) in 4 and 8 °C. In addition, we also studied multi factorial effects of four different concentration of rosemary, three different concentrations of nisin, two different levels of pH in 3 temperature 4,15 and 37 °C on log% of S.iniae during 43 days in BHI broth. The results on growth of S. iniae were evaluated using SPSS 20.0 statistical software and analyzed the logarithm of total count of the bacterial by Tukey Test. Results were considered statistically significant when P<0.05. MIC and MBC values of rosemary and nisin were 0.03, 0.075 % and 5, 40 μg/mL, respectively. The growth of S. iniae was effected significantly (P<0.05) by rosemary and nisin and also combination of rosemary and nisin in 4 and 8 °C. Samples treated with 0.135 and 0.405 % of rosemary showed a significant decrease on the growth of the bacteria compared with control sample(P<0.05). The most ١٤٦ inhibitory effects were seen in samples treated with 0.135 and 0.405% of rosemary until 9 days after storage. Also, the synergism effects of rosemary and nisin on the growth rate of bacteria was significant (P<0.05) compared with untreated samples and samples treated with the rosemary or nisin, only. Synergistic effects was observed at concentration of 0.405% rosemary and 0.75 μg/mL nisin in both temprature. Results of this study showed that different concentration of rosemary a significant inhibitory effect (P<0.05) on log% of S. iniae, in BHI broth in pH 5.5 and 7 in 4,15 and 37 °C during 43 days. In concentration of 0% rosemary (control) in pH 5.5 and 7 and 37°C, log% were 1.099 and 3.15, whereas in concentration of 0.015% rosemary were -4/241 and 1.454, respectively. The use of essential oils may improve food safety and overall microbial quality. If essential oils were to be more widely applied as antibacterials in foods, the organoleptic impact would be important. In addition, it is recommended to apply essential oils or their compounds as part of a hurdle system and to use it as an antimicrobial component along with other preservation techniques. Thus essential of R. officinalis with high antibacterial activity selected in this study could be a potential source for inhibitory substances against some food-borne pathogens and they may be candidates for using in foods or food-processing systems.

In this project sampling was done from 9 stations in 3 depths during 5 seasons to separate, identify and examine the biodiversity of cyanobacteria. Another sampling also has been done to analyse all physical and physicochemical parameters, primary production and polluting agents such as heavy metals and oil hydrocarbons in water and sediments. Along with optimization of Oscillatoria to study the ability of producing natural substances, these cyanobacteria were analysed from the point of antimicrobial and mutagenic effects. To examine the relationships among analysed parameters, the regression test, analysis of varian and Post Hoc were used. As the result of this study 48 genus of cyanobacteria were pecognised among which 35.5% were croco ccales and 64.4% were Hermogonals. Oscillatoria was one of the Filamentous cyanobacteria which had antibacterial and mutagenic effects. The results of multicommunity consistency varians test, post Hoc and homogenous subsets show significant difference between biodiversity of cyanotbacteria in coral ecosystem, Mangrove and transite. The linear correlation coefficient between biodiversity of cyanobacteria and bioenvironmental agents were examined, but there was no continuous relation between these factors and biological biodiversity. In Surface layer there was a significant correlation coefficent at 0.048 and probability at 95% confidence interval. Also, the biodiversity is depended on oil pollution and heavy metals such as copper (Cu) and chromium (Cr).

Defensins are a group of cationic antimicrobial peptides which play an important role in the innate immune system by exerting their antimicrobial activity against pathogens. In this study, we cloned a novel beta-defensin cDNA from medaka (Oryzias latipes) by rapid amplification of cDNA ends (RACE) technique. The full-length cDNA consists of 480 bp, and the open reading frame (CRF) of 189 bp encodes a polypeptide of 63 amino acids (aa) with a predicted molecular weight of 7.44 kDa. Its genomic organization was analyzed, and Southern blot detection confirmed that only one copy of beta-defensin exists in the medaka HNI strain. RT-PCR, Western blot and immunohistochemistry detections showed that the beta-defensin transcript and protein could be detected in eyes, liver, kidney, blood, spleen and gill, and obviously prevalent expression was found in eyes. Antimicrobial activity of the medaka beta-defensin was evaluated, and the antibacterial activity-specific to Gram-negative bacteria was revealed. Furthermore, the lipopolysaccharide (LPS), a major component of the outer membrane of Gram-negative bacteria, was demonstrated to be able to induce about 13-fol up-regulation of the beta-defensin within first 12 h. In addition, promoter and promoter mutagenesis analysis were performed in the medaka beta-defensin. A proximal 100 base pair(bp) sequence (+26 to -73)and the next 1700 bp sequence (-73 to -1755) were demonstrated to be responsible for the basal promoter activity and for the transcription regulation. Three nuclear factor kappa B (NF-kappa B) cis-elements and a Sp1 cis-element were revealed by mutagenesis analysis to exist in the 5' flanking sequence, and they were confirmed to be responsible for the up-regulation of medaka beta-defensin stimulated by LPS. And, the Sp1 cis-element was further revealed to be related to the basal promoter activity, and transcriptional factor II D (TFIID) was found to be in charge of the gene transcription initiation. All the obtained data suggested that the novel medaka beta-defensin should have antimicrobial activity-specific to Gram-negative bacteria, and the antibacterial immune function should be modulated by NF-kappa B and Sp1. The antibacterial drug furazolidone belonging to the group of nitrofuran antibacterial agents has been widely used as an antibacterial and antiprotozoal feed additive for poultry, cattle, and farmed fish in China. During application a large proportion of the administered drug may reach the environment directly or via feces. Although the use of furazolidone is prohibited in numerous countries, there are indications of its illegal use. It is known that furazolidone can be rapidly metabolized to 3-amino-2-oxazolidinone (AOZ) in the body of the target organism. In this study, a total of 21 fish feed samples, including 17 commercial fish feeds from local markets in China (representing 15 different formulations) and 4 fish feeds obtained from Germany and Turkey, respectively, are analyzed to determine whether the drug is still illegally used or commercially available feeds are contaminated by this drug. High-performance liquid chromatography (HPLC) and liquid chromatography-electrospray ionization tandem mass spectrometry (LC-ESI-MS/MS) methods have been implemented to determine furazolidone and its metabolite AOZ in fish feeds containing animal protein, respectively. An efficient and convenient cleanup method for the determination of furazolidone in fish feeds is developed, and a simple cleanup method for the determination of AOZ is used. Method recoveries for samples used were determined as 87.7-98.3% for furazolidone at two spike levels of 2.0 and 5.0 ng g(-1) and as 95.6-102.8% for AOZ at spike levels of 0.4 and 0.8 ng g(-1). Limits of detections were 0.4 ng g(-1) for furazolidone and 0.05 ng g(-1) for AOZ. The established methods are therefore suitable for the determination of furazolidone and its metabolite AOZ in fish feeds at trace contamination levels. Using the established methods, all fish feed samples have been proved to be furazolidone negative; however, AOZ is tested in 16 of 17 fish feeds obtained from local markets in the Hubei province of China, with a positive rate as high as 94.1%.

We report the cloning of a novel antimicrobial peptide gene, termed rtCATH_1, found in the rainbow trout, Oncorhynchus mykiss. The predicted 216-residue rtCATH_1 prepropeptide consists of three domains: a 22-residue signal peptide, a 128-residue cathelin-like region containing two identifiable cathelicidin family signatures, and a predicted 66-residue C-terminal cationic antimicrobial peptide. This predicted mature peptide was unique in possessing features of different known (mammalian) cathelicidin subgroups, such as the cysteine-bridged family and the specific amino-acid-rich family. The rtCATH_1 gene comprises four exons, as seen in all known mammalian cathelicidin genes, and several transcription factor binding sites known to be of relevance to host defenses were identified in the 5' flanking region. By Northern blot analysis, the expression of rtCATH_1 was detected in gill, head kidney, and spleen of bacterially challenged fish. Primary cultures of head kidney leukocytes from rainbow trout stimulated with lipopolysaccharide or poly(I (.) C) also expressed riCATH_1. A 36-residue peptide corresponding to the core part of the fish cathelicidin was chemically synthesized and shown to exhibit potent antimicrobial activity and a low hemolytic effect. Thus, rtCATH_1 represents a novel antimicrobial peptide gene belonging to the cathelicidin family and may play an important role in the innate immunity of rainbow trout.

由于近年来有害微生物的肆虐和人们防病抗菌意识的增强,社会对各种新型抗菌材料的需求越来越迫切。抗菌不锈钢是一种兼具良好机械和抗菌性能的新型功能材料,具有广泛的应用前景。开发和推广抗菌不锈钢,具有重要的经济和社会意义。 本文以中科院金属研究所研制的两种抗菌不锈钢为研究对象,对其抗菌性能和抗菌机理进行了研究,以期为抗菌不锈钢的深入研发和推广提供理论依据。 本文获得了如下主要结论: 1.采用覆膜法研究了抗菌不锈钢的抗菌广谱性,铁素体和奥氏体抗菌不锈钢对供试的17株菌(除产气肠杆菌外),均显示了较强的杀灭作用,证明其具有抗菌广谱性。两种材料对非孢子(或芽孢)产生菌的杀灭作用强于孢子(或芽孢)产生菌。但是,两种材料对产气肠杆菌均无杀灭作用。 2.考察了铁素体和奥氏体的杀菌率随时间和菌浓度的变化情况。两种材料分别在作用3h时和9h时,对菌体浓度为1×107CFU/mL的大肠杆菌(ATCC25922)的杀菌率均达到99.5%。对菌体浓度≤1×107CFU/mL的大肠杆菌在24h内均可全部杀灭。 3.随打磨次数的增加,抗菌率没有变化,表明打磨不影响铁素体和奥氏体抗菌不锈钢的抗菌性能;抗菌率不受温度的影响,0~37℃之间,未观察到抗菌率受温度变化的明显影响。 4.从抗菌不锈钢中溶出的铜在杀菌过程中可能起重要作用。铁素体和奥氏体抗菌不锈钢与添加了终浓度为1mmol/L的EDTA的菌液作用后,杀菌率分别降为46.8%和38.8%,因为EDTA络合了溶液中的金属离子,间接表明铜离子在杀菌过程中起重要作用。 5.两种抗菌不锈钢中O2-的产生速率分别为0.117 nmol•min-1•g-1和0.107nmol•min-1•g-1,显著高于普通不锈钢(0.062 nmol•min-1•g-1)。细胞膜不饱和脂肪酸过氧化的终产物丙二醛含量分别为1.96 nmol/g和1.93 nmol/g。显著高于普通对照的1.08 nmol/g,表明存在脂肪酸被氧化的现象。这些结果为抗菌过程中存在O2-的氧化杀菌作用提供了间接证据。 6.通过透射电镜和原子力显微镜观察,与抗菌不锈钢作用后的细胞外形多发生变化,细胞壁和细胞膜破裂,细胞内容物漏出,有的菌体完全溶解为碎片,显示两种不锈钢对细胞膜和细胞壁形态的显著影响。随作用时间延长,菌液中钾离子浓度亦显著升高,在3h时,分别达到2.57mg/L和2.79mg/L(普通对照为1.61mg/L)。 7.在巯基氧化实验中,添加终浓度为5 mmol/L的L-cys,作用8h,铁素体的抗菌率降为11.1%,薄层层析结果显示,半胱氨酸可能被氧化成胱氨酸,表明抗菌不锈钢可能对还原性酶具有氧化作用。 8.提取与抗菌不锈钢作用后菌的DNA,凝胶电泳观察并未出现smear现象。表明在本实验条件下尚不能检测到抗菌不锈钢对细菌遗传物质的氧化切割作用。 关键词: 抗菌材料 抗菌不锈钢 抗菌性能 抗菌机理 O2-

链霉菌是十分重要的一类放线菌,绝大多数的抗生素都由该类细菌产生。毛壳属真菌是一类重要的丝状真菌,从中也发现有很多结构新颖、活性独特的活性物质。因此本论文对两株链霉菌的活性成分及一株金毛壳菌的次生代谢产物进行了研究。 1.从吸水链霉菌(Streptomyces hygroscopicus 1.358)液态发酵产物(乙酸乙酯提取物)中分离得到3个化合物,通过波谱方法鉴定为RK955A (1)、Nigericin(2)、Elaiophylin(3)。以青霉素耐药-金黄色葡萄球菌作为指示菌的抗菌活性测定表明,三者均具有较强抗菌活性。 2.通过抗肿瘤体外活性筛选模型筛选得到得到一株链霉属土壤放线菌,从中分离得到六个化合物:苯乙酰胺(4)、苯丙酰胺(5)、肉桂酰胺(6)、3-(N-(甲酰胺基)乙酰基)吲哚(7)、鸟苷磷酸(8)、鸟苷(9)。 3.从金色毛壳菌(Chaetomium aureus)的固态培养物中分离得到13个化合物,利用波谱方法将其鉴定为:金毛壳菌素A(10)、金毛壳菌素B(11)、Eugenetin(12)、Eugenitol(13)、Chaetoquadrin A(14)、Chaetoquadrin B(15)、Chaetoquadrin G(16)、Chaetoquadrin H(17)、Chaetochromin A(18)、Sterigmatocystin(19)、O-methylsterigmatocystin(20)、3β-羟基-麦角甾-5,7,22-三烯(21)和过氧麦角甾醇(22)。 4.综述了聚醚类抗生素的结构、生物合成、生物活性及作用机理。 The genus Streptomyces (Actinomycetes) is an important group of microbe. Most antibiotics known nowdays are discovered from species of Streptomyces. The fungi of the genus Chaetomium have attracted much attention because various kinds of secondary metabolites with diverse bioactivities have been found from them. Thus, the bioactive compounds from two strains of Streptomyces and the secondary metabolites of Chaetomium aureus were investigated. 1. Three compounds were isolated from the ethyl acetate extract of the fermentation broth of Streptomyces hygroscopicus. They are identified to be elaiophylin (1), nigericin (2), and antibiotic RK955A (3) on the basis of their spectroscopic data. Compounds 1-3 possess antibacterial activities against Staphyloccocus aureus. 2. It was found that the extract of the fermented broth of a strain of Actinomycetes could inhibit some tumor cel lines. Separation of the bioactive fraction led to the isolation of six compounds. They were characterized to be phenylacetamide (4), phenylpropylamide (5), trans-cinnamamide (6), 3- (N- (formylmethyl) acetamide) indole (7), guanylicacid (8), and guanosine (9). 3. From the fermented broth of Chaetomium aureus, 13 compounds were isolated for the first time. They were determined to be chaetomiumycin A (10), chaetomiumycin B (11), eugenetin (12), eugenitol (13), chaetoquadrin A (14), chaetoquadrin B (15), chaetoquadrin G (16), chaetoquadrin H (17), chaetochromin A (18), sterigmatocystin (19), O-methylsterigmatocystin (20), 3β-hydroxyergosta-5, 7, 22-triene (21) and peroxy-ergosterol (22). Compounds 10 and 11 are new ones. 4. Structure, biosynthesis, biological activity, and mechanisms of polyether antibiotics were reviewed.
