La ricerca dottorale che ho sviluppato si propone di analizzare il percorso di valutazione della genitorialità recentemente delineato dai servizi sociali territoriali della provincia di Bologna attraverso la sperimentazione di strumenti per la diagnosi sociale, valutando gli esiti dell’applicazione, anche in confronto all’utilizzo di metodi tradizionali. Il progetto ha il suo fulcro tematico, nel qualificare le pratiche professionali, con il fine ultimo di giungere ad un percorso di diagnosi sociale scientificamente fondato. Il mio obbiettivo quindi non è stato analizzare in termini astratti e idealtipici le metodologie professionali di riferimento per i 32 operatori coinvolti, quanto piuttosto di formarli all’utilizzo di una serie di strumenti elaborati nelle fasi precedenti del progetto, e condurre una ricerca empirica su un numero, sufficientemente ampio, di “casi concreti” costituito da nuclei familiari in carico ai servizi sociali. Più precisamente l'ambito privilegiato d’analisi è stato individuato nel rapporto tra operatore ed utente, allo scopo di evidenziare pregi e difetti dell'utilizzo di strumenti professionali finalizzati alla diagnosi sociale. Inoltre va sottolineato che l’analisi della letteratura sul tema ha evidenziato l’esistenza di un numero molto limitato di studi empirici sulle metodologie di servizio sociale, condotti peraltro su un numero di casi e di variabili molto ristretto, e per lo più in territorio statunitense, dove il sistema di Welfare si caratterizza per una impostazione tale da rendere veramente difficile la comparazione con la realtà italiana. Quello a cui ho inteso pervenire non è un giudizio ultimo sull’efficacia di questa specifica metodologia tout-court, ma piuttosto analizzare, attraverso l’utilizzo di materiali quanti-qualitativi derivati dalla sperimentazione, quelle che sono le condizioni che si vengono a determinare quando, nel percorso di presa in carico sociale, vengono introdotti strumenti specifici, metodologia chiara e alto coinvolgimento delle componenti relazionali del processo: operatori, familiari e rete sociale allargata.

The aim of this dissertation is to provide a translation from English into Italian of a highly specialized scientific article published by the online journal ALTEX. In this text, the authors propose a roadmap for how to overcome the acknowledged scientific gaps for the full replacement of systemic toxicity testing using animals. The main reasons behind this particular choice are my personal interest in specialized translation of scientific texts and in the alternatives to animal testing. Moreover, this translation has been directly requested by the Italian molecular biologist and clinical biochemist Candida Nastrucci. It was not possible to translate the whole article in this project, for this reason, I decided to translate only the introduction, the chapter about skin sensitization, and the conclusion. I intend to use the resources that were created for this project to translate the rest of the article in the near future. In this study, I will show how a translator can translate such a specialized text with the help of a field expert using CAT Tools and a specialized corpus. I will also discuss whether machine translation can prove useful to translate this type of document. This work is divided into six chapters. The first one introduces the main topic of the article and explains my reasons for choosing this text; the second one contains an analysis of the text type, focusing on the differences and similarities between Italian and English conventions. The third chapter provides a description of the resources that were used to translate this text, i.e. the corpus and the CAT Tools. The fourth one contains the actual translation, side-by-side with the original text, while the fifth one provides a general comment on the translation difficulties, an analysis of my translation choices and strategies, and a comment about the relationship between the field expert and the translator. Finally, the last chapter shows whether machine translation and post-editing can be an advantageous strategy to translate this type of document. The project also contains two appendixes. The first one includes 54 complex terminological sheets, while the second one includes 188 simple terminological sheets.

Using bioinformatics tools, we searched the predicted Theileria annulata and T. parva proteomes for putative schizont surface proteins. This led to the identification of gp34, a GPI-anchored protein that is stage-specifically expressed by schizonts of both Theileria species and is downregulated upon induction of merogony. Transfection experiments in HeLa cells showed that the gp34 signal peptide and GPI anchor signal are also functional in higher eukaryotes. Epitope-tagged Tp-gp34, but not Ta-gp34, expressed in the cytosol of COS-7 cells was found to localise to the central spindle and midbody. Overexpression of Tp-gp34 and Ta-gp34 induced cytokinetic defects and resulted in accumulation of binucleated cells. These findings suggest that gp34 could contribute to important parasite-host interactions during host cell division.

A new image-guided microscope using augmented reality overlays has been developed. Unlike other systems, the novelty of our design consists in mounting a precise mini and low-cost tracker directly on the microscope to track the motion of the surgical tools and the patient. Correctly scaled cut-views of the pre-operative computed tomography (CT) stack can be displayed on the overlay, orthogonal to the optical view or even including the direction of a clinical tool. Moreover, the system can manage three-dimensional models for tumours or bone structures and allows interaction with them using virtual tools, showing trajectories and distances. The mean error of the overlay was 0.7 mm. Clinical accuracy has shown results of 1.1-1.8 mm.

We propose a novel class of models for functional data exhibiting skewness or other shape characteristics that vary with spatial or temporal location. We use copulas so that the marginal distributions and the dependence structure can be modeled independently. Dependence is modeled with a Gaussian or t-copula, so that there is an underlying latent Gaussian process. We model the marginal distributions using the skew t family. The mean, variance, and shape parameters are modeled nonparametrically as functions of location. A computationally tractable inferential framework for estimating heterogeneous asymmetric or heavy-tailed marginal distributions is introduced. This framework provides a new set of tools for increasingly complex data collected in medical and public health studies. Our methods were motivated by and are illustrated with a state-of-the-art study of neuronal tracts in multiple sclerosis patients and healthy controls. Using the tools we have developed, we were able to find those locations along the tract most affected by the disease. However, our methods are general and highly relevant to many functional data sets. In addition to the application to one-dimensional tract profiles illustrated here, higher-dimensional extensions of the methodology could have direct applications to other biological data including functional and structural MRI.

OBJECTIVE: In search of an optimal compression therapy for venous leg ulcers, a systematic review and meta-analysis was performed of randomized controlled trials (RCT) comparing compression systems based on stockings (MCS) with divers bandages. METHODS: RCT were retrieved from six sources and reviewed independently. The primary endpoint, completion of healing within a defined time frame, and the secondary endpoints, time to healing, and pain were entered into a meta-analysis using the tools of the Cochrane Collaboration. Additional subjective endpoints were summarized. RESULTS: Eight RCT (published 1985-2008) fulfilled the predefined criteria. Data presentation was adequate and showed moderate heterogeneity. The studies included 692 patients (21-178/study, mean age 61 years, 56% women). Analyzed were 688 ulcerated legs, present for 1 week to 9 years, sizing 1 to 210 cm(2). The observation period ranged from 12 to 78 weeks. Patient and ulcer characteristics were evenly distributed in three studies, favored the stocking groups in four, and the bandage group in one. Data on the pressure exerted by stockings and bandages were reported in seven and two studies, amounting to 31-56 and 27-49 mm Hg, respectively. The proportion of ulcers healed was greater with stockings than with bandages (62.7% vs 46.6%; P < .00001). The average time to healing (seven studies, 535 patients) was 3 weeks shorter with stockings (P = .0002). In no study performed bandages better than MCS. Pain was assessed in three studies (219 patients) revealing an important advantage of stockings (P < .0001). Other subjective parameters and issues of nursing revealed an advantage of MCS as well. CONCLUSIONS: Leg compression with stockings is clearly better than compression with bandages, has a positive impact on pain, and is easier to use.

BACKGROUND: The mollicute Mycoplasma conjunctivae is the etiological agent leading to infectious keratoconjunctivitis (IKC) in domestic sheep and wild caprinae. Although this pathogen is relatively benign for domestic animals treated by antibiotics, it can lead wild animals to blindness and death. This is a major cause of death in the protected species in the Alps (e.g., Capra ibex, Rupicapra rupicapra). METHODS: The genome was sequenced using a combined technique of GS-FLX (454) and Sanger sequencing, and annotated by an automatic pipeline that we designed using several tools interconnected via PERL scripts. The resulting annotations are stored in a MySQL database. RESULTS: The annotated sequence is deposited in the EMBL database (FM864216) and uploaded into the mollicutes database MolliGen http://cbi.labri.fr/outils/molligen/ allowing for comparative genomics. CONCLUSION: We show that our automatic pipeline allows for annotating a complete mycoplasma genome and present several examples of analysis in search for biological targets (e.g., pathogenic proteins).

Speciation is a fundamental evolutionary process, the knowledge of which is crucial for understanding the origins of biodiversity. Genomic approaches are an increasingly important aspect of this research field. We review current understanding of genome-wide effects of accumulating reproductive isolation and of genomic properties that influence the process of speciation. Building on this work, we identify emergent trends and gaps in our understanding, propose new approaches to more fully integrate genomics into speciation research, translate speciation theory into hypotheses that are testable using genomic tools and provide an integrative definition of the field of speciation genomics

Gap junctions between neurons form the structural substrate for electrical synapses. Connexin 36 (Cx36, and its non-mammalian ortholog connexin 35) is the major neuronal gap junction protein in the central nervous system (CNS), and contributes to several important neuronal functions including neuronal synchronization, signal averaging, network oscillations, and motor learning. Connexin 36 is strongly expressed in the retina, where it is an obligatory component of the high-sensitivity rod photoreceptor pathway. A fundamental requirement of the retina is to adapt to broadly varying inputs in order to maintain a dynamic range of signaling output. Modulation of the strength of electrical coupling between networks of retinal neurons, including the Cx36-coupled AII amacrine cell in the primary rod circuit, is a hallmark of retinal luminance adaptation. However, very little is known about the mechanisms regulating dynamic modulation of Cx36-mediated coupling. The primary goal of this work was to understand how cellular signaling mechanisms regulate coupling through Cx36 gap junctions. We began by developing and characterizing phospho-specific antibodies against key regulatory phosphorylation sites on Cx36. Using these tools we showed that phosphorylation of Cx35 in fish models varies with light adaptation state, and is modulated by acute changes in background illumination. We next turned our focus to the well-studied and readily identifiable AII amacrine cell in mammalian retina. Using this model we showed that increased phosphorylation of Cx36 is directly related to increased coupling through these gap junctions, and that the dopamine-stimulated uncoupling of the AII network is mediated by dephosphorylation of Cx36 via protein kinase A-stimulated protein phosphatase 2A activity. We then showed that increased phosphorylation of Cx36 on the AII amacrine network is driven by depolarization of presynaptic ON-type bipolar cells as well as background light increments. This increase in phosphorylation is mediated by activation of extrasynaptic NMDA receptors associated with Cx36 gap junctions on AII amacrine cells and by Ca2+-calmodulin-dependent protein kinase II activation. Finally, these studies indicated that coupling is regulated locally at individual gap junction plaques. This work provides a framework for future study of regulation of Cx36-mediated coupling, in which increased phosphorylation of Cx36 indicates increased neuronal coupling.

Rapid-eye movement (REM) sleep correlates with neuronal activity in the brainstem, basal forebrain and lateral hypothalamus. Lateral hypothalamus melanin-concentrating hormone (MCH)-expressing neurons are active during sleep, but their effects on REM sleep remain unclear. Using optogenetic tools in newly generated Tg(Pmch-cre) mice, we found that acute activation of MCH neurons (ChETA, SSFO) at the onset of REM sleep extended the duration of REM, but not non-REM, sleep episodes. In contrast, their acute silencing (eNpHR3.0, archaerhodopsin) reduced the frequency and amplitude of hippocampal theta rhythm without affecting REM sleep duration. In vitro activation of MCH neuron terminals induced GABAA-mediated inhibitory postsynaptic currents in wake-promoting histaminergic neurons of the tuberomammillary nucleus (TMN), and in vivo activation of MCH neuron terminals in TMN or medial septum also prolonged REM sleep episodes. Collectively, these results suggest that activation of MCH neurons maintains REM sleep, possibly through inhibition of arousal circuits in the mammalian brain.

A three-level satellite to ground monitoring scheme for conservation easement monitoring has been implemented in which high-resolution imagery serves as an intermediate step for inspecting high priority sites. A digital vertical aerial camera system was developed to fulfill the need for an economical source of imagery for this intermediate step. A method for attaching the camera system to small aircraft was designed, and the camera system was calibrated and tested. To ensure that the images obtained were of suitable quality for use in Level 2 inspections, rectified imagery was required to provide positional accuracy of 5 meters or less to be comparable to current commercially available high-resolution satellite imagery. Focal length calibration was performed to discover the infinity focal length at two lens settings (24mm and 35mm) with a precision of O.1mm. Known focal length is required for creation of navigation points representing locations to be photographed (waypoints). Photographing an object of known size at distances on a test range allowed estimates of focal lengths of 25.lmm and 35.4mm for the 24mm and 35mm lens settings, respectively. Constants required for distortion removal procedures were obtained using analytical plumb-line calibration procedures for both lens settings, with mild distortion at the 24mm setting and virtually no distortion found at the 35mm setting. The system was designed to operate in a series of stages: mission planning, mission execution, and post-mission processing. During mission planning, waypoints were created using custom tools in geographic information system (GIs) software. During mission execution, the camera is connected to a laptop computer with a global positioning system (GPS) receiver attached. Customized mobile GIs software accepts position information from the GPS receiver, provides information for navigation, and automatically triggers the camera upon reaching the desired location. Post-mission processing (rectification) of imagery for removal of lens distortion effects, correction of imagery for horizontal displacement due to terrain variations (relief displacement), and relating the images to ground coordinates were performed with no more than a second-order polynomial warping function. Accuracy testing was performed to verify the positional accuracy capabilities of the system in an ideal-case scenario as well as a real-world case. Using many welldistributed and highly accurate control points on flat terrain, the rectified images yielded median positional accuracy of 0.3 meters. Imagery captured over commercial forestland with varying terrain in eastern Maine, rectified to digital orthophoto quadrangles, yielded median positional accuracies of 2.3 meters with accuracies of 3.1 meters or better in 75 percent of measurements made. These accuracies were well within performance requirements. The images from the digital camera system are of high quality, displaying significant detail at common flying heights. At common flying heights the ground resolution of the camera system ranges between 0.07 meters and 0.67 meters per pixel, satisfying the requirement that imagery be of comparable resolution to current highresolution satellite imagery. Due to the high resolution of the imagery, the positional accuracy attainable, and the convenience with which it is operated, the digital aerial camera system developed is a potentially cost-effective solution for use in the intermediate step of a satellite to ground conservation easement monitoring scheme.

La producción de Solanum tuberosum L., Lycopersicum esculentum Mill. y Physalis ixocarpa Brot. (Solanales: Solanaceae) ha sufrido fuertes pérdidas económicas por la presencia de Bactericera cockerelli Sulc. (Hemiptera: Triozidae) al asociarse con las enfermedades punta morada o “zebra chip", además de ser el transmisor de Candidatus Liberibacter solanacearum. Las alternativas de control utilizadas han carecido de eficacia por desconocer la distribución espacial del insecto dentro de la parcela. Conocer dicho comportamiento permitiría focalizar las alternativas de control, haciéndolas más eficaces. Este trabajo tuvo por objetivo modelizar la distribución espacial de los estadíos de huevo, ninfa y adulto de B. cockerelli obtenidos en muestreos por transectos en un cultivo de papa, utilizando herramientas geoestadísticas. Los resultados indican que la distribución espacial de las poblaciones de huevos, ninfas y adultos de B. cockerelli fue de tipo agregada en cada fecha de muestreo. La validación cruzada de los semivariogramas obtenidos corrobora la distribución agregada en las poblaciones de B. cockerelli. Por su parte, los mapas elaborados permiten observar la estructura agregada de las poblaciones del insecto, permitiendo identificar áreas infestadas y áreas libres. Se encontró estabilidad espacio temporal para los tres estadios del insecto.

Pese la importancia de la extensión rural en toda estrategia orientada el desarrollo de los sectores de productores rurales pequeños y medianos, no existen en la Argentina trabajos académicos que reconstruyan de manera completa y sistemática el perfil de quienes trabajan en el país en extensión rural. Por esta razón, se realizó una investigación cuantitativa descriptiva de carácter transversal que permitió reconstruir el perfil de los extensionistas rurales que trabajan en el sistema de extensión público argentino. Para este fin se realizó una encuesta (enviada vía email) a extensionistas del INTA, ProHuerta y de la Subsecretaría de Agricultura Familiar. Los resultados de esta investigación permitieron aportar, a partir del uso de herramientas estadísticas, a la descripción del perfil de los extensionistas rurales que trabajan en la Argentina, así como a la del perfil diferencial de hombres y mujeres y de quienes trabajan en las diferentes instituciones
