Single crystals of a recent ferroelectric material, glycine phosphite were grown from aqueous solution employing the techniques of slow cooling and controlled evaporation. Powder X-ray diffraction studies as well as thermal analysis were carried out on the grown crystals. The morphology of the crystal has been determined using contact and optical goniometry. The mechanical hardness of the crystal was evaluated by Vickers indentation method. Thickness dependence of the dielectric properties has been investigated and the results can be interpreted in terms of a surface layer of lower dielectric constant.

Mycobacterium tuberculosis, the causative agent of tuberculosis, is at increased risk of accumulating damaged guanine nucleotides such as 8-oxo-dGTP and 8-oxo-GTP because of its residency in the oxidative environment of the host macrophages. By hydrolyzing the oxidized guanine nucleotides before their incorporation into nucleic acids, MutT proteins play a critical role in allowing organisms to avoid their deleterious effects. Mycobacteria possess several MutT proteins. Here, we purified recombinant M. tuberculosis MutT2 (MtuMutT2) and M. smegmatis MutT2 (MsmMutT2) proteins from M. tuberculosis (a slow grower) and M. smegmatis (fast growing model mycobacteria), respectively, for their biochemical characterization. Distinct from the Escherichia coli MutT, which hydrolyzes 8-oxo-dGTP and 8-oxo-GTP, the mycobacterial proteins hydrolyze not only 8-oxo-dGTP and 8-oxo-GTP but also dCTP and 5-methyl-dCTP. Determination of kinetic parameters (K-m and V-max) revealed that while MtuMutT2 hydrolyzes dCTP nearly four times better than it does 8-oxo-dGTP, MsmMutT2 hydrolyzes them nearly equally. Also, MsmMutT2 is about 14 times more efficient than MtuMutT2 in its catalytic activity of hydrolyzing 8-oxo-dGTP. Consistent with these observations, MsmMutT2 but not MtuMutT2 rescues E. coli for MutT deficiency by decreasing both the mutation frequency and A-to-C mutations (a hallmark of MutT deficiency). We discuss these findings in the context of the physiological significance of MutT proteins.

Increasing nitrate concentrations in ground water is deleterious to human health as ingestion of such water can cause methemoglobinemia in infants and even cancer in adults (desirable limit for nitrate as NO3 - 45 mg/L, IS code 10500-1991). Excess nitrate concentrations in ground water is contributed by reason being disposal of sewage and excessive use of fertilizers. Though numerous technologies such as reverse osmosis, ion exchange, electro-dialysis, permeable reactive barriers using zerovalent iron etc exists, nitrate removal continues to be one of challenging issue as nitrate ion is highly mobile within the soil strata. The tapping the denitrification potential of soil denitrifiers which are inherently available in the soil matrix is the most sustainable approach to mitigate accumulation of nitrate in ground water. The insitu denitrification of sand and bentonite enhanced sand (bentonite content = 5%) in presence of easily assimilable organic carbon such as ethanol was studied. Batch studies showed that nitrate reduction by sand follows first order kinetics with a rate constant 5.3x10(-2) hr(-1) and rate constant 4.3 x 10(-2) hr(-1) was obtained for bentonite-enhanced sand (BS) at 25 degrees C. Filter columns (height = 5 cm and diameter = 8.2 cm) were constructed using sand and bentonite-enhanced sand as filter media. The filtration rate through both the filter columns was maintained at average value of 2.60 cm/h. The nitrate removal rates through both the filter media was assessed for solution containing 22.6 mg NO3-N/L concentrations while keeping C/N mass ratio as 3. For sand filter column, the nitrate removal efficiency reached the average value of 97.6% after passing 50 pore volumes of the nitrate solution. For bentonite-enhanced sand filter column, the average nitrate removal efficiency was 83.5%. The time required for effective operation for sand filter bed was 100 hours, while bentonite-enhanced sand filter bed did not require any maturation period as that of sand filter bed for effective performance because the presence of micropores in bentonite increases the hydraulic retention time of the solution inside the filter bed.

We experimentally demonstrate photobleaching (PB) in Ge22As22Se56 thin films, when illuminated with a diode pumped solid state laser (DPSSL) of wavelength 671 nm, which is far below the optical bandgap of the sample. Interestingly, we found that PB is a slow process and occurs even at moderate pump beam intensity of 0.2 W/cm(2), however the kinetics remain rather different.

The steady-state negative supercoiling of eubacterial genomes is maintained by the action of DNA topoisomerases. Topoisomerase distribution varies in different species of mycobacteria. While Mycobacterium tuberculosis (Mtb) contains a single type I (Topol) and a single type II (Gyrase) enzyme, Mycobacterium smegmatis (Msm) and other members harbour additional relaxases. Topol is essential for Mtb survival. However, the necessity of Topol or other relaxases in Msm has not been investigated. To recognize the importance of Topol for growth, physiology and gene expression of Msm, we have developed a conditional knock-down strain of Topol in Msm. The Topol-depleted strain exhibited extremely slow growth and drastic changes in phenotypic characteristics. The cessation of growth indicates the essential requirement of the enzyme for the organism in spite of having additional DNA relaxation enzymes in the cell. Notably, the imbalance in Topol level led to the altered expression of topology modulatory proteins, resulting in a diffused nucleoid architecture. Proteomic and transcript analysis of the mutant indicated reduced expression of the genes involved in central metabolic pathways and core DNA transaction processes. RNA polymerase (RNAP) distribution on the transcription units was affected in the Topol-depleted cells, suggesting global alteration in transcription. The study thus highlights the essential requirement of Topol in the maintenance of cellular phenotype, growth characteristics and gene expression in mycobacteria. A decrease in Topol level led to altered RNAP occupancy and impaired transcription elongation, causing severe downstream effects.

The central problem in the study of glass-forming liquids and other glassy systems is the understanding of the complex structural relaxation and rapid growth of relaxation times seen on approaching the glass transition. A central conceptual question is whether one can identify one or more growing length scale(s) associated with this behavior. Given the diversity of molecular glass-formers and a vast body of experimental, computational and theoretical work addressing glassy behavior, a number of ideas and observations pertaining to growing length scales have been presented over the past few decades, but there is as yet no consensus view on this question. In this review, we will summarize the salient results and the state of our understanding of length scales associated with dynamical slow down. After a review of slow dynamics and the glass transition, pertinent theories of the glass transition will be summarized and a survey of ideas relating to length scales in glassy systems will be presented. A number of studies have focused on the emergence of preferred packing arrangements and discussed their role in glassy dynamics. More recently, a central object of attention has been the study of spatially correlated, heterogeneous dynamics and the associated length scale, studied in computer simulations and theoretical analysis such as inhomogeneous mode coupling theory. A number of static length scales have been proposed and studied recently, such as the mosaic length scale discussed in the random first-order transition theory and the related point-to-set correlation length. We will discuss these, elaborating on key results, along with a critical appraisal of the state of the art. Finally we will discuss length scales in driven soft matter, granular fluids and amorphous solids, and give a brief description of length scales in aging systems. Possible relations of these length scales with those in glass-forming liquids will be discussed.

Summary: The offshore shelf and canyon habitats of the OCNMS (Fig. 1) are areas of high primary productivity and biodiversity that support extensive groundfish fisheries. Recent acoustic surveys conducted in these waters have indicated the presence of hard-bottom substrates believed to harbor unique deep-sea coral and sponge assemblages. Such fauna are often associated with shallow tropical waters, however an increasing number of studies around the world have recorded them in deeper, cold-water habitats in both northern and southern latitudes. These habitats are of tremendous value as sites of recruitment for commercially important fishes. Yet, ironically, studies have shown how the gear used in offshore demersal fishing, as well as other commercial operations on the seafloor, can cause severe physical disturbances to resident benthic fauna. Due to their exposed structure, slow growth and recruitment rates, and long life spans, deep-sea corals and sponges may be especially vulnerable to such disturbances, requiring very long periods to recover. Potential effects of fishing and other commercial operations in such critical habitats, and the need to define appropriate strategies for the protection of these resources, have been identified as a high-priority management issue for the sanctuary. To begin addressing this issue, an initial pilot survey was conducted June 1-12, 2004 at six sites in offshore waters of the OCNMS (Fig. 2, average depths of 147-265 m) to explore for the presence of deep-sea coral/sponge assemblages and to look for evidence of potential anthropogenic impacts in these critical habitats. The survey was conducted on the NOAA Ship McARTHUR-II using the Navy’s Phantom DHD2+2 remotely operated vehicle (ROV), which was equipped with a video camera, lasers, and a manipulator arm for the collection of voucher specimens. At each site, a 0.1-m2 grab sampler also was used to collect samples of sediments for the analysis of macroinfauna (> 1.0 mm), total organic carbon (TOC), grain size, and chemical contaminants. Vertical profiles of salinity, dissolved oxygen (DO), temperature, and pressure were recorded at each site with a small SeaCat conductivity-temperature-depth (CTD) profiler. Niskin bottles attached to the CTD also obtained near-bottom water samples in support of a companion study of microbial indicators of coral health and general ecological condition across these sites. All samples except the sediment-contaminant samples are being analyzed with present project funds. Original cruise plans included a total of 12 candidate stations to investigate (Fig. 3). However, inclement weather and equipment failures restricted the sampling to half of these sites. In spite of the limited sampling, the work completed was sufficient to address key project objectives and included several significant scientific observations. Foremost, the cruise was successful in demonstrating the presence of target deepwater coral species in these waters. Patches of the rare stony coral Lophelia pertusa, more characteristic of deepwater coral/sponge assemblages in the North Atlantic, were observed for the first time in OCNMS at a site in 271 meters of water. A large proportion of these corals consisted of dead and broken skeletal remains, and a broken gorgonian (soft coral) also was observed nearby. The source of these disturbances is not known. However, observations from several sites included evidence of bottom trawl marks in the sediment and derelict fishing gear (long lines). Preliminary results also support the view that these areas are important reservoirs of marine biodiversity and of value as habitat for demersal fishes. For example, onboard examination of 18 bottom-sediment grabs revealed benthic infaunal species representative of 14 different invertebrate phyla. Twenty-eight species of fishes from 11 families, including 11 (possibly 12) species of ommercially important rockfishes, also were identified from ROV video footage. These initial discoveries have sparked considerable interests in follow-up studies to learn more about the spatial extent of these assemblages and magnitude of potential impacts from commercial-fishing and other anthropogenic activities in the area. It is essential to expand our knowledge of these deep-sea communities and their vulnerability to potential environmental risks in order to determine the most appropriate management strategies. The survey was conducted under a partnership between NOAA’s National Centers for Coastal Ocean Science (NCCOS) and National Marine Sanctuary Program (NMSP) and included scientists from NCCOS, OCNMS, and several other west-coast State, academic, private, and tribal research institutions (see Section 4 for a complete listing of participating scientists). Este trabalho iniciou-se a partir das inquietações que surgiram por ocasião da primeira visita ao município de Itacaré, no litoral sul da Bahia, onde trabalhamos com alfabetização de jovens e adultos, em função das muitas vozes que se fizeram ouvir. Itacaré, rica em belezas naturais, com praias, rios e cachoeiras lindíssimas, vem sofrendo, segundo a população nativa da região, muitas transformações em nome do progresso e da globalização, via processo de turistificação. Precisamente, nos últimos cinco anos as mudanças são tantas, com a chegada dos turistas das mais variadas regiões, que moradores já se percebem desterritorializados, bem como já se torna visível por intermédio das vozes ouvidas, que o processo de desconstrução de identidades é uma realidade inconteste. Desta forma, tentando entender o processo de desterritorialização que se generaliza, como também o de desconstrução de identidades, nos propusemos a entender as especificidades deste município permeadas pelos desdobramentos do turismo e suas consequências, desvelando as memórias coletivas e as representações sociais, através das histórias da população local que apontam, de certa forma, a usurpação daquilo que, segundo eles, são os seus maiores bens: a dignidade e o respeito aos filhos da terra, pois que boa parcela da população local está perdendo o seu espaço na medida em que as preocupações do poder público local se solidificam nos privilégios aos estrangeiros. Buscou-se o Serviço Social como o lugar da discussão na medida em que neste vislumbramos de forma mais adensada a preocupação com políticas sociais que deem conta de grande parcela da população excluída ou incluída perversamente.

In four chapters various aspects of earthquake source are studied.

Chapter I

Surface displacements that followed the Parkfield, 1966, earthquakes were measured for two years with six small-scale geodetic networks straddling the fault trace. The logarithmic rate and the periodic nature of the creep displacement recorded on a strain meter made it possible to predict creep episodes on the San Andreas fault. Some individual earthquakes were related directly to surface displacement, while in general, slow creep and aftershock activity were found to occur independently. The Parkfield earthquake is interpreted as a buried dislocation.

Chapter II

The source parameters of earthquakes between magnitude 1 and 6 were studied using field observations, fault plane solutions, and surface wave and S-wave spectral analysis. The seismic moment, MO, was found to be related to local magnitude, ML, by log MO = 1.7 ML + 15.1. The source length vs magnitude relation for the San Andreas system found to be: ML = 1.9 log L - 6.7. The surface wave envelope parameter AR gives the moment according to log MO = log AR300 + 30.1, and the stress drop, τ, was found to be related to the magnitude by τ = 0.54 M - 2.58. The relation between surface wave magnitude MS and ML is proposed to be MS = 1.7 ML - 4.1. It is proposed to estimate the relative stress level (and possibly the strength) of a source-region by the amplitude ratio of high-frequency to low-frequency waves. An apparent stress map for Southern California is presented.

Chapter III

Seismic triggering and seismic shaking are proposed as two closely related mechanisms of strain release which explain observations of the character of the P wave generated by the Alaskan earthquake of 1964, and distant fault slippage observed after the Borrego Mountain, California earthquake of 1968. The Alaska, 1964, earthquake is shown to be adequately described as a series of individual rupture events. The first of these events had a body wave magnitude of 6.6 and is considered to have initiated or triggered the whole sequence. The propagation velocity of the disturbance is estimated to be 3.5 km/sec. On the basis of circumstantial evidence it is proposed that the Borrego Mountain, 1968, earthquake caused release of tectonic strain along three active faults at distances of 45 to 75 km from the epicenter. It is suggested that this mechanism of strain release is best described as "seismic shaking."

Chapter IV

The changes of apparent stress with depth are studied in the South American deep seismic zone. For shallow earthquakes the apparent stress is 20 bars on the average, the same as for earthquakes in the Aleutians and on Oceanic Ridges. At depths between 50 and 150 km the apparent stresses are relatively high, approximately 380 bars, and around 600 km depth they are again near 20 bars. The seismic efficiency is estimated to be 0.1. This suggests that the true stress is obtained by multiplying the apparent stress by ten. The variation of apparent stress with depth is explained in terms of the hypothesis of ocean floor consumption.

Esta dissertação versa sobre o Museu de Favela (MUF), organização não-governamental fundada em 2008 durante as obras do Programa de Aceleração do Crescimento (PAC) nas favelas do Pavão, Pavãozinho e Cantagalo, na Zona Sul da cidade do Rio de Janeiro. As principais propostas do MUF são a patrimonialização da favela/seus elementos culturais, bem como o desenvolvimento turístico dessas favelas. A partir de um trabalho de campo, do tipo observação participante junto à diretoria do MUF, analiso a construção de sua proposta de turismo em oposição à favela da Rocinha, caso emblemático do turismo em favelas. Mostro a presença de duas ideias de turismo a propósito do caso estudado: o que chamo de turismo vilão, representado pelo contexto da Rocinha na visão dos promotores do MUF; e o que chamo de turismo solução, representado na mesma visão pelo turismo organizado pelos moradores para benefício da população local e solução dos problemas das favelas. Analiso ainda a proposta do MUF, sua organização como ONG e seus diretores. Por fim, focalizo o Museu de Favela e o Estado como atores sociais básicos envolvidos na questão analisada, mostrando como turismo e patrimônio aparecem como ferramentas para ambas as instituições, que guardam semelhanças e diferenças no que se refere a dois processos em andamento nas favelas do Pavão, Pavãozinho e Cantagalo: o processo de patrimonialização e o processo de turismização.

Neurons obtained directly from human somatic cells hold great promise for disease modeling and drug screening. Available protocols rely on overexpression of transcription factors using integrative vectors and are often slow, complex, and inefficient. We report a fast and efficient approach for generating induced neural cells (iNCs) directly from human hematopoietic cells using Sendai virus. Upon SOX2 and c-MYC expression, CD133-positive cord blood cells rapidly adopt a neuroepithelial morphology and exhibit high expansion capacity. Under defined neurogenic culture conditions, they express mature neuronal markers and fire spontaneous action potentials that can be modulated with neurotransmitters. SOX2 and c-MYC are also sufficient to convert peripheral blood mononuclear cells into iNCs. However, the conversion process is less efficient and resulting iNCs have limited expansion capacity and electrophysiological activity upon differentiation. Our study demonstrates rapid and efficient generation of iNCs from hematopoietic cells while underscoring the impact of target cells on conversion efficiency.

A geografia da religião no Brasil pós 1990, vem apresentando temas que auxiliam os geógrafos no entendimento do espaço. Para o geógrafo da religião, os estudos têm por análise a dimensão espacial da fé, no tempo e no espaço, trata-se da análise de duas categorias: o sagrado e o profano. Esta pesquisa visa como objetivo, o estudo da comunidade católica, a Canção Nova, inserida em um movimento renovador na Igreja Católica Apostólica Romana denominada Renovação Carismática Católica (RCC) em seu espaço e no tempo de fé. Torna-se necessário desenvolver a importância do sagrado no Vale do Paraíba Paulista em seus sucessivos (re) arranjos espaciais na hierópolis de Cachoeira Paulista. A interpretação dessas organizações espaciais ocorreu ao analisar os possíveis agentes modeladores no espaço: peregrinos, turistas, visitantes, comerciantes e moradores. O Espaço Sagrado e seus respectivos espaços vinculados representaram a contribuição geográfica dessa dissertação e facilitou a compreensão do homem com o divino e sua relação na dimensão econômica, a dimensão política e a dimensão do lugar.

Phosphoglucose isomerase (PGI) catalyzes the reversible isomerization of glucose-6-phosphate and fructose-6-phosphate. It is involved in glycolysis and in the regeneration of glucose-6-P molecules in the oxidative pentose phosphate pathway (OPPP). In chloroplasts of illuminated mesophyll cells PGI also connects the Calvin-Benson cycle with the starch biosynthetic pathway. In this work we isolated pgi1-3, a mutant totally lacking pPGI activity as a consequence of aberrant intron splicing of the pPGI encoding gene, PGI1. Starch content in pgi1-3 source leaves was ca. 10-15% of that of wild type (WT) leaves, which was similar to that of leaves of pgi1-2, a T-DNA insertion pPGI null mutant. Starch deficiency of pgi1 leaves could be reverted by the introduction of a sex1 null mutation impeding beta-amylolytic starch breakdown. Although previous studies showed that starch granules of pgi1-2 leaves are restricted to both bundle sheath cells adjacent to the mesophyll and stomata guard cells, microscopy analyses carried out in this work revealed the presence of starch granules in the chloroplasts of pgi1-2 and pgi1-3 mesophyll cells. RT-PCR analyses showed high expression levels of plastidic and extra-plastidic beta-amylase encoding genes in pgi1 leaves, which was accompanied by increased beta-amylase activity. Both pgi1-2 and pgi1-3 mutants displayed slow growth and reduced photosynthetic capacity phenotypes even under continuous light conditions. Metabolic analyses revealed that the adenylate energy charge and the NAD(P) H/NAD(P) ratios in pgi1 leaves were lower than those of WT leaves. These analyses also revealed that the content of plastidic 2-C-methyl-D-erythritol 4-phosphate (MEP)-pathway derived cytokinins (CKs) in pgi1 leaves were exceedingly lower than in WT leaves. Noteworthy, exogenous application of CKs largely reverted the low starch content phenotype of pgi1 leaves. The overall data show that pPGI is an important determinant of photosynthesis, energy status, growth and starch accumulation in mesophyll cells likely as a consequence of its involvement in the production of OPPP/glycolysis intermediates necessary for the synthesis of plastidic MEP-pathway derived hormones such as CKs.

A Bacia Hidrográfica Lagos São João, localizada no sudoeste do Estado do Rio de Janeiro, abrange 13 municípios que abrigam cerca de 520 mil habitantes. Na temporada de férias esse número sobe para mais de 1 milhão de pessoas. A pastagem constitui o principal tipo de uso do solo, em seguida vem as áreas urbanas e as salinas. A partir da década de 1960 essa região passou a receber maior contingente populacional, tanto de veranistas quanto de moradores fixos, beneficiados pela implantação de novas vias de acesso, como a Ponte Rio-Niteroi e pela construção da represa de Juturnaíba, que ampliou o abastecimento de água dos municípios. Surge neste contexto a especulação imobiliária, que acelera a ocupação das terras próximas a Lagoa de Araruama. Rapidamente essas terras foram loteadas e o setor da construção civil foi ganhando força. Entretanto, a região não contou com adequado planejamento, e os investimentos em saneamento básico e outras infraestruturas urbanas não acompanharam o ritmo da construção civil, que cada vez mais investia em casas, prédios e condomínios, que ampliaram consideravelmente a área urbana e a ocupação da zona costeira. Sendo assim, ficou visível o aumento da malha urbana e a ocupação de áreas impróprias, como as margens dos corpos hídricos, os manguezais, dunas e restingas, além da redução da cobertura vegetal. Dessa forma, foi substancial a perda de qualidade ambiental na região, sobretudo, com relação a água da lagoa e dos rios, que passaram a receber maior volume de efluentes sem tratamento. O potencial turístico da região tem sido explorado e provocado altos investimentos dos agentes de especulação imobiliária, entretanto além de promover a ocupação em áreas irregulares, leva a privatização de espaços públicos e incentiva o fenômeno da segunda residência. A chegada de novos turistas iniciou o processo de desenvolvimento do turismo e, consequentemente, a redução da produção salineira. Com isso, o espaço local ganhou novos significados, inseridos pela lógica da urbanização turística. Foi essa nova lógica transformadora que, gradativamente, valorizou a paisagem local, ampliando e encarecendo o seu consumo. Além de ampliar as transformações espaciais, tendo em vista a expansão da malha urbana verificada nas imagens de satélites, atuais, que foram comparadas com fotografias aéreas de décadas anteriores. Todas as transformações ocorridas na região apresentam alguma relação com o desencadeamento de novos problemas ambientais identificados nos seus ecossistemas, sobretudo a Lagoa de Araruama, ou a ampliação de problemas anteriormente existentes.

Trawling and dredging on Georges Bank (northwest Atlantic Ocean) have altered the cover of colonial epifauna, as surveyed through in situ photography. A total of 454 photographs were analyzed from areas with gravel substrate between 1994 and 2000 at depths of 40–50 m and 80–90 m. The cover of hydroids, bushy bryozoans, sponges, and tubeworms was generally higher at sites undisturbed by fishing than at sites classified as disturbed. The magnitude and significance of this effect depended on depth and year. Encrusting bryozoans were the only type of colonial epifauna positively affected by bottom fishing. Species richness of noncolonial epifauna declined with increased bottom fishing, but Simpson’s index of diversity typically peaked at intermediate levels of habitat disturbance. Species that were more abundant at undisturbed sites possessed characteristics that made them vulnerable to bottom fishing. These characteristics include emergent growth forms, soft body parts, low motility, use of complex microhabitats, long life spans, slow growth, and larval dispersal over short distances. After the prohibition of bottom fishing at one site, both colonial and noncolonial species increased in abundance. Populations of most taxa took two years or more to increase after the fishing closure. This finding indicates that bottom fishing needs to be reduced to infrequent intervals to sustain the benthic species composition of Georges Bank at a high level of biodiversity and abundance.
