The objective was to evaluate the chemical composition and in vitro digestibility of sugarcane hydrolysed with increasing doses of calcium oxide and varying air exposure times. A completely randomised, split plot design was used; the doses were allocated to the plots, and the air exposure times were allocated to the subplots, with four repetitions. The data underwent analysis of variance and were laid out according to the effect of the treatment on the components of polynomial regressions, and evaluated at the 5% probability. The increase in the dosage negatively affected the quantities of neutral-detergent fibre (NDF), acid-detergent fibre (ADF), lignin (LIG), total carbohydrates (TC), cellulose (CEL), crude protein (CP), and ether extract (EE); and positively affected the quantities of non-fibrous carbohydrates (NFC) and mineral matter (MM). The addition of calcium oxide improved the in vitro digestible dry matter (IVDMD) coefficients and was able to keep up to 72 hours. The in vitro digestibility of the neutral-detergent fibre (IVDNDF) and of the acid-detergent fibre (IVDADF) coefficients decreased when calcium oxide was added. Calcium oxide has the ability to hydrolyse the fibrous fraction and conserve chopped sugarcane. Doses of 0.5 and 1.0% lime exhibited similar results to those achieved at higher doses; therefore, higher doses are not required in the hydrolyses of sugarcane. Over time, the sugarcane deteriorates, but this deterioration is reduced by the addition of calcium oxide.

The aim of this study was to verify the effects of running overtraining protocols performed in downhill, uphill, and without inclination on the proteins related to hypertrophy signaling pathway in extensor digitorum longus (EDL) and soleus of C57BL/6 mice. We also performed histological and stereological analyses. Rodents were divided into control (CT; sedentary mice), overtrained by downhill running (OTR/down), overtrained by uphill running (OTR/up), and overtrained by running without inclination (OTR). The incremental load, exhaustive, and grip force tests were used as performance evaluation parameters. 36 h after the grip force test, EDL and soleus were removed and immediately used for immunoblotting analysis or stored at -80°C for histological and stereological analyses. For EDL, OTR/down decreased the protein kinase B (Akt) and tuberous sclerosis protein 2 (TSC2) phosphorylation (p), and increased myostatin, receptor-activated Smads (pSMAD2-3), and insulin receptor substrate-1 (pIRS-1; Ser307/636). OTR/down also presented low and high relative proportions of cytoplasm and connective tissue, respectively. OTR/up increased the mammalian target of rapamycin (pmTOR), 70-kDa ribosomal protein S6 kinase 1 (pS6K1) and pSMAD2-3, and decreased pTSC2. OTR decreased pTSC2 and increased pIRS-1 (Ser636). For soleus, OTR/down increased S6 ribosomal protein (pS6RP) and pSMAD2-3, and decreased pIRS-1 (Ser639). OTR/up decreased pS6K1, pS6RP and pIRS-1 (Ser639), and increased pTSC2 (Ser939), and pSMAD2-3. OTR increased pS6RP, 4E-binding protein-1 (p4E-BP1), pTSC2 (Ser939), and pSMAD2-3, and decreased pIRS-1 (Ser639). In summary, OTR/down inhibited the skeletal muscle hypertrophy with concomitant signs of atrophy in EDL. The effects of OTR/up and OTR depended on the analyzed skeletal muscle type. J. Cell. Aquafeed production faces global issues related to availability of feed ingredients. Feed manufacturers require greater flexibility in order to develop nutritional and cost-effective formulations that take into account nutrient content and availability of ingredients. The search for appropriate ingredients requires detailed screening of their potential nutritional value and variability at the industrial level. In vitro digestion of feedstuffs by enzymes extracted from the target species has been correlated with apparent protein digestibility (APD) in fish and shrimp species. The present study verified the relationship between APD and in vitro degree of protein hydrolysis (DH) with Litopenaeus vannamei hepatopancreas enzymes in several different ingredients (n = 26): blood meals, casein, corn gluten meal, crab meal, distiller`s dried grains with solubles, feather meal, fish meals, gelatin, krill meals, poultry by-product meal, soybean meals, squid meals and wheat gluten. The relationship between APD and DH was further verified in diets formulated with these ingredients at 30% inclusion into a reference diet. APD was determined in vivo (30.1 +/- 0.5 degrees C, 32.2 +/- 0.4%.) with juvenile L vannamei (9 to 12 g) after placement of test ingredients into a reference diet (35 g kg(-1) CP: 8.03 g kg(-1) lipid; 2.01 kcal g(-1)) with chromic oxide as the inert marker. In vitro DH was assessed in ingredients and diets with standardized hepatopancreas enzymes extracted from pond-reared shrimp. The DH of ingredients was determined under different assay conditions to check for the most suitable in vitro protocol for APD prediction: different batches of enzyme extracts (HPf5 or HPf6), temperatures (25 or 30 degrees C) and enzyme activity (azocasein): crude protein ratios (4 U: 80 mg CP or 4 U: 40 mg CP). DH was not affected by ingredient proximate composition. APD was significantly correlated to DH in regressions considering either ingredients or diets. The relationships between APD and DH of the ingredients could be suitably adjusted to a Rational Function (y = (a + bx)/(1 + cx + dx2), n = 26. Best in vitro APD predictions were obtained at 25 degrees C, 4 U: 80 mg CP both for ingredients (R(2) = 0.86: P = 0.001) and test diets (R(2) = 0.96; P = 0.007). The regression model including all 26 ingredients generated higher prediction residuals (i.e., predicted APD - determined APD) for corn gluten meal, feather meal. poultry by-product meal and krill flour. The remaining test ingredients presented mean prediction residuals of 3.5 points. A model including only ingredients with APD>80% showed higher prediction precision (R(2) = 0.98: P = 0.000004; n = 20) with average residual of 1.8 points. Predictive models including only ingredients from the same origin (e.g., marine-based, R(2) = 0.98; P = 0.033) also displayed low residuals. Since in vitro techniques have been usually validated through regressions against in vivo APD, the DH predictive capacity may depend on the consistency of the in vivo methodology. Regressions between APD and DH suggested a close relationship between peptide bond breakage by hepatopancreas digestive proteases and the apparent nitrogen assimilation in shrimp, and this may be a useful tool to provide rapid nutritional information. Aims: Arthrospira platensis has been studied for single-cell protein production because of its biomass composition and its ability of growing in alternative media. This work evaluated the effects of different dilution rates (D) and urea concentrations (N0) on A.similar to platensis continuous culture, in terms of growth, kinetic parameters, biomass composition and nitrogen removal. Methods and results: Arthrospira platensis was continuously cultivated in a glass-made vertical column photobioreactor agitated with Rushton turbines. There were used different dilution rates (0.040.44 day-1) and urea concentrations (0.5 and 5 mmol l-1). With N0 = 5 mmol l-1, the maximum steady-state biomass concentration was1415 mg l-1, achieved with D = 0.04 day-1, but the highest protein content (71.9%) was obtained by applying D = 0.12 day-1, attaining a protein productivity of 106.41 mg l-1 day-1. Nitrogen removal reached 99% on steady-state conditions. Conclusions: The best results were achieved by applying N0 = 5 mmol l-1; however, urea led to inhibitory conditions at D = 0.16 day-1, inducing the system wash-out. The agitation afforded satisfactory mixture and did not harm the trichomes structure. Significance and Impact of the Study: These results can enhance the basis for the continuous removal of nitrogenous wastewater pollutants using cyanobacteria, with an easily assembled photobioreactor.

The hydroponic cultivation of vegetables has increased markedly in recent years, however, little is known about its chemical composition, which is of extreme importance in view of changing food habits of a considerable fraction of the population seeking food quality. For this reason, cultivation of watercress, chicory, rocket and lettuce American (Lucy Brown) and smooth (cv. Regina) were grown in hydroponics, NFT system, aiming to evaluate the yield, nitrate content and chemical composition. The experiment was conducted in a greenhouse at the Universidade Federal de Santa Catarina, Florianopolis city in the Santa Catarina State (Brazil), from may to july 2004 under a completely randomized design with four replications. The spacing used was 25 x 25 cm for the crops of watercress, chicory and lettuce (cv. Lucy Brown and cv. Regina) and 5 x 5 cm to the rocket. The traits were: number of leaves, nitrate content and chemical composition of vegetables. The lettuce cv. Regina and chicory had higher number of leaves per plant. The watercress had lower water content and higher dry mass of shoots. The largest increase in fresh weight was obtained in chicory, lettuce, cv. Regina. Higher levels of lipids, protein, ash, carbohydrates, calories, fiber and nitrate were obtained from the watercress. The rocket had lower values for the variables fresh and dries the whole plant, shoot and root and leaf number per plant. All cultures showed good visual appearance, low calorie and nitrate levels suitable for human consumption.
