Virgin queens of the fire ant,Solenopsis invicta Buren, that were removed from the influence of the inhibitory queen pheromone, dealated more readily in the presence of workers than in their absence. During 72 hours after disinhibition, a significantly greater number of overwintered virgin queens than spring-reared virgin queens dealated when they were isolated, but the numbers that dealated in the presence of workers were very similar. Some sexually immature virgin queens dealated after disinhibition. Virgin dealates were found to be capable of preventing other virgin queens from dealating. The various factors that influence dealation by virgin queens were used to develop a bioassay for the inhibitory queen pheromone ofS. invicta. Lorsque des reines vierges de la fourmi de feu sont soustraites à l'influence de la phéromone inhibitrice produite par la reine, elles perdent leurs ailes plus facilement en présence d'ouvrières qu'en leur absence. Lorsqu'elles sont isolées pendant 72 heures après la levée de l'inhibition, un nombre significativement plus grand de reines vierges ayant hiverné perdent leurs ailes, par rapport aux reines vierges élevées au printemps. Toutefois, les nombres d'individus perdant leurs ailes en présence d'ouvrières sont similaires. Après désinhibition, quelques reines vierges immatures perdent leurs ailes. Les sexués vierges désailés sont capables de prévenir la perte des ailes chez d'autres sexués vierges. Les divers facteurs influençant la déalation chez les reines vierges ont été utilisés afin de développer un essai biologique pour la phéromone inhibitrice produite par la reine deS. invicta.

1. Accumulating evidence indicates that plant resistance against above-ground herbivores can be affected by the presence of arbuscular mycorrhizal fungi (AMF) in association with the host plant. Little is known, however, about how AMF composition can influence herbivore choice to feed on a particular plant. 2. Unravelling the preference-performance hypothesis in a multitrophic context is needed to expand our knowledge of complex multitrophic interactions in natural systems. If given mycorrhizal fungal genotypes increase attractiveness for a herbivore (reduced plant resistance), then the benefits of increased unpalatability provided by the mycorrhizal fungi (increased plant resistance) might be outweighed by the increased herbivore recruitment. 3. This was addressed by designing three experiments to test the effects of different AMF genotypes, inoculated either alone or in combination, to measure intraspecific AMF effects on plant resistance and insect herbivore preference. Using strawberry (Fragaria vesca L.) plants that were colonised by eight different combinations of Rhizophagus irregularis isolates, we measured effects on plant growth, insect growth and survival, as well as feeding preferences of a generalist herbivore caterpillar (Spodoptera littoralis Boisduval). 4. Overall, it was found that: (i) AMF influenced plant resistance in an AMF genotype-specific manner; (ii) some AMF inoculations decreased insect performance; (iii) insects preferentially chose to feed more on leaves originating from non-mycorrhizal plants; but also that (iv) in a whole plant bioassay, insects preferentially chose the biggest plant, regardless of their mycorrhizal status. 5. Therefore, AMF-mediated trade-offs between growth and resistance against herbivores have been shown. Such trade-offs, particularly driven by plant attractiveness to herbivores, buffer the positive effects of the mycorrhizal symbiosis on enhanced plant growth.

L’objectiu d’aquest estudi ha estat avaluar l’efecte que té sobre algunes propietats fisicoquímiques del sòl i el creixement de plantes, l’adició del producte compostat obtingut a partir de la fracció orgànica dels residus sòlids urbans de recollida no selectiva, material anomenat bioestabilitzat. S’ha dut a terme un bioassaig amb blat (Triticum aestivum), utilitzant bioestabilitzat procedent de dues plantes de tractament mecànic i biològic, Vacarisses i Mataró, que s’ha aplicat a diferents dosis en un sòl de baixa qualitat. S’ha observat una relació positiva entre el creixement del blat i la dosis de bioestabilitzat aplicada. Pel que fa a la germinació, ha estat lleugerament estimulada per les dosis intermèdies de bioestabilitzat assajades. En quant a les propietats fisicoquímiques del sòl, s’ha observat un increment de la salinitat en funció de la dosis de bioestabilitzat aplicada, que en les més elevades podria ser problemàtica. També, el contingut de matèria orgànica ha augmentat de manera proporcional a la dosis, cosa que ha millorat les propietats físiques del sòl. Per altra banda, s’han trobat continguts d’impropis (vidre, plàstic ...) i concentracions d’alguns metalls pesants bastant elevades, així com una considerable variabilitat entre les partides de bioestabilitzat. Finalment, es conclou que abans de permetre l’aplicació de bioestabilitzat per a la rehabilitació de sòls degradats, cal aprofundir més en l’estudi d’aquest material.

An efficient screening strategy for the identification of potentially interesting low-abundance antifungal natural products in crude extracts that combines both a sensitive bioautography assay and high performance liquid chromatography (HPLC) microfractionation was developed. This method relies on high performance thin layer chromatography (HPTLC) bioautography with a hypersusceptible engineered strain of Candida albicans (DSY2621) for bioactivity detection, followed by the evaluation of wild type strains in standard microdilution antifungal assays. Active extracts were microfractionated by HPLC in 96-well plates, and the fractions were subsequently submitted to the bioassay. This procedure enabled precise localisation of the antifungal compounds directly in the HPLC chromatograms of the crude extracts. HPLC-PDA-mass spectrometry (MS) data obtained in parallel to the HPLC antifungal profiles provided a first chemical screening about the bioactive constituents. Transposition of the HPLC analytical conditions to medium-pressure liquid chromatography (MPLC) allowed the efficient isolation of the active constituents in mg amounts for structure confirmation and more extensive characterisation of their biological activities. The antifungal properties of the isolated natural products were evaluated by their minimum inhibitory concentration (MIC) in a dilution assay against both wild type and engineered strains of C. albicans. The biological activity of the most promising agents was further evaluated in vitro by electron microscopy and in vivo in a Galleria mellonella model of C. albicans infection. The overall procedure represents a rational and comprehensive means of evaluating antifungal activity from various perspectives for the selection of initial hits that can be explored in more in-depth mode-of-action studies. This strategy is illustrated by the identification and bioactivity evaluation of a series of antifungal compounds from the methanolic extract of a Rubiaceae plant, Morinda tomentosa, which was used as a model in these studies.

Blue light mediates the phosphorylation of a membrane protein in seedlings from several plant species. When crude microsomal membrane proteins from dark-grown pea (Pisum sativum L.), sunflower (Helianthus annuus L.), zucchini (Cucurbita pepo L.), Arabidopsis (Arabidopsis thaliana L.), or tomato (Lycopersicon esculentum L.) stem segments, or from maize (Zea mays L.), barley (Hordeum vulgare L.), oat (Avena sativa L.), wheat (Triticum aestivum L.), or sorghum (Sorghum bicolor L.) coleoptiles are illuminated and incubated in vitro with [gamma-(32)P]ATP, a protein of apparent molecular mass from 114 to 130 kD is rapidly phosphorylated. Hence, this system is probably ubiquitous in higher plants. Solubilized maize membranes exposed to blue light and added to unirradiated solubilized maize membranes show a higher level of phosphorylation of the light-affected protein than irradiated membrane proteins alone, suggesting that an unirradiated substrate is phosphorylated by a light-activated kinase. This finding is further demonstrated with membrane proteins from two different species, where the phosphorylated proteins are of different sizes and, hence, unambiguously distinguishable on gel electrophoresis. When solubilized membrane proteins from one species are irradiated and added to unirradiated membrane proteins from another species, the unirradiated protein becomes phosphorylated. These experiments indicate that the irradiated fraction can store the light signal for subsequent phosphorylation in the dark. They also support the hypothesis that light activates a specific kinase and that the systems share a close functional homology among different higher plants.

The objective of this work was to analyze the agronomic performance and chromosomal stability of transgenic homozygous progenies of soybean [Glycine max (L.) Merrill.], and to confirm the resistance of these plants against Anticarsia gemmatalis. Eleven progenies expressing cry1Ac, hpt and gusA genes were evaluated for agronomic characteristics in relation to the nontransformed parent IAS 5 cultivar. Cytogenetical analysis was carried out on transgenic and nontransgenic plants. Two out of the 11 transgenic progenies were also evaluated, in vitro and in vivo, for resistance to A. gemmatalis. Two negative controls were used in resistance bioassays: a transgenic homozygous line, containing only the gusA reporter gene, and nontransgenic 'IAS 5' plants. The presence of cry1Ac transgene affected neither the development nor the yield of plants. Cytogenetical analysis showed that transgenic plants presented normal karyotype. In detached-leaf bioassay, cry1Ac plants exhibited complete efficacy against A. gemmatalis, whereas negative controls were significantly damaged. Whole-plant feeding assay confirmed a very high protection of cry1Ac against velvetbean caterpillar, while nontransgenic 'IAS 5' plants and homozygous gusA line exhibited 56.5 and 71.5% defoliation, respectively. The presence of cry1Ac transgene doesn't affect the majority of agronomic traits (including yield) of soybean and grants high protection against A. gemmatalis.

The objective of this work was to evaluate the effect of organic compounds from plant extracts of six species and phosphate fertilization on soil phosphorus availability. Pots of 30 cm height and 5 cm diameter were filled with Typic Hapludox. Each pot constituted a plot of a completely randomized design, in a 7x2 factorial arrangement, with four replicates. Aqueous extracts of black oat (Avena strigosa), radish (Raphanus sativus), corn (Zea mays), millet (Pennisetum glaucum), soybean (Glycine max), sorghum (Sorghum bicolor), and water, as control, were added in each plot, with or without soluble phosphate fertilization. After seven days of incubation, soil samples were taken from soil layers at various depths, and labile, moderately labile and nonlabile P fractions in the soil were analysed. Plant extracts led to an accumulation of inorganic phosphorus in labile and moderately labile fractions, mainly in the soil surface layer (0-5 cm). Radish, with a higher amount of malic acid and higher P content than other species, was the most efficient in increasing soil P availability.

The objective of this work was to evaluate the occurrence of Aceria tosichella Keifer (Prostigmata: Eriophyidae), the wheat curl mite, in Rio Grande do Sul, Brazil. Samples to detect A. tosichella specimens were collected in wheat, corn, oat crops and potential host grasses in 46 localities, in October 2006, August and October 2007. Samples of wheat were also collected in experimental greenhouses at Embrapa Trigo, Passo Fundo, RS, Brazil. A. tosichella specimens were found in wheat samples from Passo Fundo, Palmeira das Missões, São Luís Gonzaga, and Santo Antônio das Missões, RS, Brazil. Symptoms due to A. tosichella infestations were observed only in greenhouse conditions. This is the first report of A. tosichella in Brazil and the second in South America.

The objective of this work was to evaluate the effects of chlorpyrifos on earthworms and on soil functional parameters. An integrated laboratory-field study was performed in a wheat field in Argentina, sprayed with chlorpyrifos at two recommended application rates (240 or 960 g ha-1 style='vertical-align:baseline'> a.i.). Laboratory tests included neutral red retention time, comet assay (single cell gel electrophoresis), and avoidance behavior, each using the earthworm Eisenia andrei exposed in soil collected 1 or 14 days after pesticide application, and the bait-lamina test. Field tests assessed organic matter breakdown using the litterbag and bait-lamina assays. Earthworm populations in the field were assessed using formalin application and hand-sorting. The neutral red retention time and comet assays were sensitive biomarkers to the effects of chlorpyrifos on the earthworm E. andrei; however, the earthworm avoidance test was not sufficiently robust to assess these effects. Feeding activity of soil biota, assessed by the bait lamina test, was significantly inhibited by chlorpyrifos after 97 days, but recovered by the 118th day of the test. Litterbag test showed no significant differences in comparison to controls. Earthworm abundance in the field was too low to adequately test the sensitivity of this assessment endpoint.

The objective of this work was to establish and test the induced-feeding bioassay in order to detect Bacillus thuringiensis insecticidal activity against Epilachna paenulata (Coleoptera: Coccinellidae). Larvae were induced to swallow high concentrations of spore-crystal suspensions of seven exotic and 30 Argentine B. thuringiensis strains. The great majority of strains showed no toxicity to E. paenulata larvae, and observed mortality was lower than 30%. Induced-feeding bioassay is feasible, and should be used for prospecting strains that produce right combinations of Cry proteins needed to an efficient pest control.

The objective of this work was to evaluate the effect of winter land use on the amount of residual straw, the physical soil properties and grain yields of maize, common bean and soybean summer crops cultivated in succession. The experiment was carried out in the North Plateau of Santa Catarina state, Brazil, from May 2006 to April 2010. Five strategies of land use in winter were evaluated: intercropping with black oat + ryegrass + vetch, without grazing and nitrogen (N) fertilization (intercropping cover); the same intercropping, with grazing and 100 kg ha-1 of N per year topdressing (pasture with N); the same intercropping, with grazing and without nitrogen fertilization (pasture without N); oilseed radish, without grazing and nitrogen fertilization (oilseed radish); and natural vegetation, without grazing and nitrogen fertilization (fallow). Intercropping cover produces a greater amount of biomass in the system and, consequently, a greater accumulation of total and particulate organic carbon on the surface soil layer. However, land use in winter does not significantly affect soil physical properties related to soil compaction, nor the grain yield of maize, soybean and common bean cultivated in succession.

The objective of this work was to evaluate the microbiological and chemical attributes of a soil with a seven‑year history of urea and swine manure application. In the period from October 2008 to October 2009, soil samples were collected in the 0-10 cm layer and were subjected to the treatments: control, without application of urea or manure; and with the application of urea, pig slurry, and deep pig litter in two doses, in order to supply one or two times the recommended N doses for the maize (Zea mays)/black oat (Avena strigosa) crop succession. The carbon of the microbial biomass (MB‑C) and the basal respiration (C‑CO2) were analyzed, and the metabolic (qCO2) and microbial quotient (qmic) were calculated with the obtained data. Organic matter, pH in water, available P and K, and exchangeable Ca and Mg were also determined. The application of twice the dose of deep pig litter increases the MB‑C and C‑CO2 values. The qmic and qCO2 are little affected by the application of swine manure. The application of twice the dose of deep pig litter increases the values of pH in water and the contents of available P and of exchangeable Ca and Mg in the soil.

The objective of this work was to evaluate the effectiveness of ruzigrass (Urochloaruziziensis) in enhancing soil-P availability in areas fertilized with soluble or reactive rock phosphates. The area had been cropped for five years under no-till, in a system involving soybean, triticale/black-oat, and pearl millet. Previously to the five-year cultivation period, corrective phosphorus fertilization was applied once on soil surface, at 0.0 and 80 kg ha-1 P2O5, as triple superphosphate or Arad rock phosphate. After this five-year period, plots received the same corrective P fertilization as before and ruzigrass was introduced to the cropping system in the stead of the other cover crops. Soil samples were taken (0-10 cm) after ruzigrass cultivation and subjected to soil-P fractionation. Soybean was grown thereafter without P application to seed furrow. Phosphorus availability in plots with ruzigrass was compared to the ones with spontaneous vegetation for two years. Ruzigrass cultivation increased inorganic (resin-extracted) and organic (NaHCO3) soil P, as well as P concentration in soybean leaves, regardless of the P source. However, soybean yield did not increase significantly due to ruzigrass introduction to the cropping system. Soil-P availability did not differ between soluble and reactive P sources. Ruzigrass increases soil-P availability, especially where corrective P fertilization is performed.

O objetivo deste trabalho foi avaliar o efeito de extratos de capeba (Piper marginatum) e nim (Azadirachta indica) sobre o fungo Colletotrichum scovillei e determinar o componente mais ativo no controle pós-colheita da antracnose em pimentão. A atividade de extratos metanólicos das folhas (P.marginatum e A. indica) e das sementes (A. indica) a 0, 125, 250, 500, 1.000 e 2.000 ppm, na inibição do crescimento micelial de C. scovillei in vitro foram avaliadas. O extrato metanólico de folhas de P.marginatum foi o mais ativo e, consequentemente, foi submetido ao fracionamento biomonitorado ("bioassay-guided fractionation"). Esse processo rendeu 10 frações majoritárias, obtidas por cromatografia líquida de alta performance, das quais a fração à concentração de 1,5 ppm inibiu o desenvolvimento de C. scovillei de forma mais eficiente do que o fungicida mancozeb.

Bacterial bioreporters have substantial potential for contaminant assessment but their real world application is currently impaired by a lack of sensitivity. Here, we exploit the bioconcentration of chemicals in the urine of animals to facilitate pollutant detection. The shore crab Carcinus maenas was exposed to the organic contaminant 2-hydroxybiphenyl, and urine was screened using an Escherichia coli-based luciferase gene (luxAB) reporter assay specific to this compound. Bioassay measurements differentiated between the original contaminant and its metabolites, quantifying bioconcentration factors of up to one hundred-fold in crab urine. Our results reveal the substantial potential of using bacterial bioreporter assays in real-time monitoring of biological matricesto determine exposure histories, with wide ranging potential for the in situ measurement of xenobiotics in risk assessments and epidemiology.
