In the past, protease-substrate finding proved to be rather haphazard and was executed by in vitro cleavage assays using singly selected targets. In the present study, we report the first protease proteomic approach applied to meprin, an astacin-like metalloendopeptidase, to determine physiological substrates in a cell-based system of Madin-Darby canine kidney epithelial cells. A simple 2D IEF/SDS/PAGE-based image analysis procedure was designed to find candidate substrates in conditioned media of Madin-Darby canine kidney cells expressing meprin in zymogen or in active form. The method enabled the discovery of hitherto unknown meprin substrates with shortened (non-trypsin-generated) N- and C-terminally truncated cleavage products in peptide fragments upon LC-MS/MS analysis. Of 22 (17 nonredundant) candidate substrates identified, the proteolytic processing of vinculin, lysyl oxidase, collagen type V and annexin A1 was analysed by means of immunoblotting validation experiments. The classification of substrates into functional groups may propose new functions for meprins in the regulation of cell homeostasis and the extracellular environment, and in innate immunity, respectively.

Comprehending speech is one of the most important human behaviors, but we are only beginning to understand how the brain accomplishes this difficult task. One key to speech perception seems to be that the brain integrates the independent sources of information available in the auditory and visual modalities in a process known as multisensory integration. This allows speech perception to be accurate, even in environments in which one modality or the other is ambiguous in the context of noise. Previous electrophysiological and functional magnetic resonance imaging (fMRI) experiments have implicated the posterior superior temporal sulcus (STS) in auditory-visual integration of both speech and non-speech stimuli. While evidence from prior imaging studies have found increases in STS activity for audiovisual speech compared with unisensory auditory or visual speech, these studies do not provide a clear mechanism as to how the STS communicates with early sensory areas to integrate the two streams of information into a coherent audiovisual percept. Furthermore, it is currently unknown if the activity within the STS is directly correlated with strength of audiovisual perception. In order to better understand the cortical mechanisms that underlie audiovisual speech perception, we first studied the STS activity and connectivity during the perception of speech with auditory and visual components of varying intelligibility. By studying fMRI activity during these noisy audiovisual speech stimuli, we found that STS connectivity with auditory and visual cortical areas mirrored perception; when the information from one modality is unreliable and noisy, the STS interacts less with the cortex processing that modality and more with the cortex processing the reliable information. We next characterized the role of STS activity during a striking audiovisual speech illusion, the McGurk effect, to determine if activity within the STS predicts how strongly a person integrates auditory and visual speech information. Subjects with greater susceptibility to the McGurk effect exhibited stronger fMRI activation of the STS during perception of McGurk syllables, implying a direct correlation between strength of audiovisual integration of speech and activity within an the multisensory STS.

Clearance of allergic inflammatory cells from the lung through matrix metalloproteinases (MMPs) is necessary to prevent lethal asphyxiation, but mechanistic insight into this essential homeostatic process is lacking. In this study, we have used a proteomics approach to determine how MMPs promote egression of lung inflammatory cells through the airway. MMP2- and MMP9-dependent cleavage of individual Th2 chemokines modulated their chemotactic activity; however, the net effect of complementing bronchoalveolar lavage fluid of allergen-challenged MMP2(-/-)/MMP9(-/-) mice with active MMP2 and MMP9 was to markedly enhance its overall chemotactic activity. In the bronchoalveolar fluid of MMP2(-/-)/MMP9(-/-) allergic mice, we identified several chemotactic molecules that possessed putative MMP2 and MMP9 cleavage sites and were present as higher molecular mass species. In vitro cleavage assays and mass spectroscopy confirmed that three of the identified proteins, Ym1, S100A8, and S100A9, were substrates of MMP2, MMP9, or both. Function-blocking Abs to S100 proteins significantly altered allergic inflammatory cell migration into the alveolar space. Thus, an important effect of MMPs is to differentially modify chemotactic bioactivity through proteolytic processing of proteins present in the airway. These findings provide a molecular mechanism to explain the enhanced clearance of lung inflammatory cells through the airway and reveal a novel approach to target new therapies for asthma.

Polymer implants are interesting alternatives to the contemporary load-bearing implants made from metals. Polyetheretherketone (PEEK), a well-established biomaterial for example, is not only iso-elastic to bone but also permits investigating the surrounding soft tissues using magnetic resonance imaging or computed tomography, which is particularly important for cancer patients. The commercially available PEEK bone implants, however, require costly coatings, which restricts their usage. As an alternative to coatings, plasma activation can be applied. The present paper shows the plasma-induced preparation of nanostructures on polymer films and on injection-molded micro-cantilever arrays and the associated chemical modifications of the surface. In vitro cell experiments indicate the suitability of the activation process. In addition, we show that microstructures such as micro-grooves 1 μm deep and 20 μm wide cause cell alignment. The combination of micro-injection molding, simultaneous microstructuring using inserts/bioreplica and plasma treatments permits the preparation of polymer implants with nature-analogue, anisotropic micro- and nanostructures.

The evolutionary conservation of the core NMD factors UPF1, UPF2 and UPF3 would imply a similar basic mechanism of PTC recognition in all eukaryotes. However, unlike NMD in yeast, which targets PTC-containing mRNAs irrespectively of whether their 5' cap is bound by the cap-binding complex (CBC) or by the eukaryotic initiation factor 4E (eIF4E), mammalian NMD has been claimed to be restricted to CBC-bound mRNAs during the pioneer round of translation. In our recent study we compared decay kinetics of two NMD reporter systems in mRNA fractions bound to either CBC or eIF4E in human cells. Our findings reveal that NMD destabilizes eIF4E bound transcripts as efficiently as those associated with CBC. These results corroborate an emerging unified model for NMD substrate recognition, according to which NMD can ensue at every aberrant translation termination event. Additionally, our results indicate that the closed loop structure of mRNA forms only after the replacement of CBC with eIF4E at the 5' cap.

Rutile (TiO2) is an important host phase for high field strength elements (HFSE) such as Nb in metamorphic and subduction zone environments. The observed depletion of Nb in arc rocks is often explained by the hypothesis that rutile sequesters HFSE in the subducted slab and overlying sediment, and is chemically inert with respect to aqueous fluids evolved during prograde metamorphism in the forearc to subarc environment. However, field observations of exhumed terranes, and experimental studies, indicate that HFSE may be soluble in complex aqueous fluids at high pressure (i.e., >0.5 GPa) and moderate to high temperature (i.e., >300 degrees C). In this study, we investigated experimentally the mobility of Nb in NaCl- and NaF-bearing aqueous fluids in equilibrium with Nb-bearing rutile at pressure-temperature conditions applicable to fluid evolution in arc environments. Niobium concentrations in aqueous fluid at rutile saturation were measured directly by using a hydrothermal diamond-anvil cell (HDAC) and synchrotron X-ray fluorescence (SXRF) at 2.1 to 6.5 GPa and 300-500 degrees C, and indirectly by performing mass loss experiments in a piston-cylinder (PC) apparatus at similar to 1 GPa and 700-800 degrees C. The concentration of Nb in a 10 wt% NaCl aqueous fluid increases from 6 to 11 mu g/g as temperature increases from 300 to 500 degrees C, over a pressure range from 2.1 to 2.8 GPa, consistent with a positive temperature dependence. The concentration of Nb in a 20 wt% NaCl aqueous fluid varies from 55 to 150 mu g/g at 300 to 500 degrees C, over a pressure range from 1.8 to 6.4 GPa; however, there is no discernible temperature or pressure dependence. The Nb concentration in a 4 wt% NaF-bearing aqueous fluid increases from 180 to 910 mu g/g as temperature increases from 300 to 500 degrees C over the pressure range 2.1 to 6.5 GPa. The data for the F-bearing fluid indicate that the Nb content of the fluid exhibits a dependence on temperature between 300 and 500 degrees C at >= 2 GPa, but there is no observed dependence on pressure. Together, the data demonstrate that the hydrothermal mobility of Nb is strongly controlled by the composition of the fluid, consistent with published data for Ti. At all experimental conditions, however, the concentration of Nb in the fluid is always lower than coexisting rutile, consistent with a role for rutile in moderating the Nb budget of arc rocks.

Vinal (Prosopis ruscifolia G.) es una especie de importancia forestal, nativa de la Región Fitogeográfica del Chaco Occidental, altamente tolerante al estrés salino. El objetivo de este trabajo fue poner a prueba la hipótesis de que la suplementación con CaSO4 disminuye la concentración de iones tóxicos en plántulas de vinal, e incrementa la concentración de prolina, permitiendo un ajuste osmótico. Las plántulas se cultivaron en solución nutritiva de Hoagland al 25%, con o sin la adición de 0,4 mol L-1 de NaCl, y con o sin suplementación de 5 ó 10 mmol L-1 de CaSO4. Se determinó la materia seca, composición mineral, contenido relativo de agua y concentraciones de prolina y azúcares solubles. Se utilizó un diseño experimental completamente aleatorizado con cinco repeticiones, y los resultados se analizaron con ANOVA y test de Tukey. Los resultados obtenidos en este trabajo confirman el rol protector del Ca+2, a través de la manutención de las concentraciones de Ca+2, K+ y Mg+2 en los tejidos, la inhibición de la absorción de Na+ y el ajuste osmótico mediante la síntesis de solutos osmocompatibles.

El objetivo fue evaluar la supervivencia al riego con soluciones de NaCl en 28 genotipos del género Vitis para identificar su grado de tolerancia salina. Se estudiaron 11 híbridos de Vitis americanos y 17 variedades de Vitis vinifera. Plantas de 1 y 2 años se regaron durante 66 días con una solución 100 mM de NaCl. Para clasificar las variedades de acuerdo con su grado de tolerancia a la salinidad, se calculó el día en que el 25% de la población había muerto (1er cuartil de la muestra). Se consideraron como genotipos sensibles aquellos en que al menos 25% había muerto antes del día 30 (3309 Couderc, 161-49 Couderc, Fercal, Freedom, 1103 Paulsen, 99 Richter, SO4, Torrontés Sanjuanino), como poco tolerantes cuando al menos 25% murió entre los días 30 y 60 (Cereza, Colombard, Criollas Blanca y Ballista, Palomino, 110 Richter, 140 Ruggeri, Syrah, Torrontés Riojano) y como tolerantes cuando el 75% o menos sobrevivió más de 60 días (101-14 Millardet-Grasset, 196-17 Castel, Criollas Chica y Sanjuanina, Moscatel de Alejandría, Pedro Giménez). Aun en los genotipos más tolerantes cuando las plantas crecieron bajo salinidad, la integridad de membranas celulares se redujo un 17% y el contenido de clorofila total disminuyó un 52%.

The influence of microhabitat type on the diversity and community structure of the harpacticoid copepod fauna associated with a cold-water coral degradation zone was investigated in the Porcupine Seabight (North-East Atlantic). Three substrate types were distinguished: dead fragments of the cold-water coral Lophelia pertusa, skeletons of the glass sponge Aphrocallistes bocagei and the underlying sediment. At the family level, it appears that coral fragments and underlying sediment do not harbour distinctly diVerent assemblages, with Ectinosomatidae, Ameiridae, Pseudotachidiidae, Argestidae and Miraciidae as most abundant. Conclusions on assemblage structure and diversity of the sponge skeletons are limited as only two samples were available. Similarity analysis at species level showed a strong variation in the sediment samples, which did not harbour a distinctly different assemblage in opposition to the coral and sponge samples. Several factors (sediment infill on the hard substrates, mobility of the copepods, limited sample sizes) are proposed to explain this apparent lack of a distinct difference between the microhabitats. Coral fragments and sediment were both characterised by high species diversity and low species dominance, which might indicate that copepod diversity is not substantially influenced by hydrodynamic stress. The additive partitioning of species diversity showed that by adding locations species richness was greatly enhanced. The harpacticoid community in the cold-water coral degradation zone is highly diverse and includes 157 species, 62 genera and 19 families. Information from neighbouring soft-bottom regions is necessary to assess whether total species diversity is increased by the presence of these complex habitatproviding substrates.
