Cryopreservation is a process where cells or biological tissues are preserved by freezing at very low temperatures and aims to cease reversibly, in a controlled manner, all the biological functions of living tissues, i.e., maintain cell preservation so that it can recover with high degree of viability and functional integrity. This study aimed to evaluate the influence of cryopreservation on the mesenchymal stem cells originating from the periodontal ligament of human third molars by in vitro experiments. Six healthy teeth were removed and the periodontal cells grown in culture medium containing α-MEM supplemented with antibiotics and 15% FBS in a humidified atmosphere with 5% CO2 at 37° C. Cells isolated from each sample were divided into two groups: Group I - immediate cell culture (not fresh cryopreserved cells) and Group II - cell cryopreservation, during a period of 30 days. Analyses of rates of cell adhesion and proliferation in different groups were performed by counting the cells adhered to the wells, in intervals of 24, 48 and 72 hours after the start of cultivation. The number of cells in each well was obtained by counting viable cells with the use of hemocytometer and the method of exclusion of cells stained by trypan blue. The difference between groups for each of the times was analyzed by Wilcoxon test. Regarding the temporal evolution for each group, analysis was done by Friedman's test to verify the existence of differences between times and, when it existed, the Wilcoxon penalty was applied. The results showed no statistically significant difference between the two groups analyzed in this study. Therefore, we conclude that the cryopreservation process, after a period of 30 days, did not influence the cell type studied, and there was no difference in growth capacity in vitro between the groups

The shrimp Litopenaeus vannamei has been grown in highly variable environments, especially in relation to salinity and water temperature. The adjustment to such conditions mainly involves changes in behavior, physiology, particularly in the immune response. This may consequently reduce the welfare of these animals. Despite the widespread farming of the species, little is known about their behavioral and physiological responses under stressful conditions. Thus, the objective of this study was to assess the influence of different salinities and temperatures in the behavior of the marine shrimp L. vannamei, and its relation to the total hemocytes count. In the laboratory, juvenile shrimp were kept in glass aquaria with a closed water recirculation system, continuous aeration and filtration, and under a 12:12 h light/dark cycle. Behavioral observations occurred 1, 4, 7 and 10 h after the start of each phase (light or dark). To assess the influence of salinity, shrimp were first acclimated and then observed at 2, 30 or 50 ppm salinity water, while temperatures tested were 18, 28 and 33 ° C. At the end of each experiment (30 days), shrimp hemolymph was collected for subsequent total hemocytes count (THC), a parameter used to assess stress. In general, feeding behavior was modified under lower salinity and temperature, with reduced values in feeding, exploration and digestive tract filling. Inactivity and burrowing were prevalent under extreme conditions water salinity and temperature, respectively: 2 and 50 ppm and 18 and 33 ° C; crawling was also less frequent under these conditions. In regards to light/dark cycle, shrimp were more active during the dark phase (crawling and swimming), while burrowing was higher during the light phase, regardless of salinity or temperature of the water. Inactivity behavior did not vary according to the light/dark cycle. Moreover, the total hemocytes count (THC) was reduced under 2 and 50 ppm salinity and 18 ° C temperature. Farming of L. vannamei under extremely low or high salinities and low temperatures is harmful. This suggests the species must be cultivated in salinities closer to those of the sea as well as at high temperatures, which seems to be ideal for a management focused on animal welfare, therefore, producing healthier shrimp

The purpose of this paper is to describe the benefits of sugar cane ethanol in Brazil, appointing the productivity of this type of fuel based on hectares of plantation, its carbon dioxide cycle and the contribution to reduce the greenhouse effect. In the following step the uses of ethanol for hydrogen production by steam reforming is analyzed and some comparison with natural gas steam reforming is performed. The sugar cane industry in Brazil, in a near future, in the hydrogen era, could be modified according to our purpose, since besides the production of sugar, and ethylic and anhydric alcohol, Brazilian sugar cane industry will also be able to produce biohydrogen.Fuel cells appear like a promising technology for energy generation. Among several technologies in the present, the PEMFC (proton exchange membrane fuel cell) is the most appropriate for vehicles application, because it combines durability, high power density, high efficiency, good response and it works at relatively low temperatures. Besides that it is easy to turn it on and off and it is able to support present vibration in vehicles. A PEMFC's problem is the need of noble catalysts like platinum. Another problem is that CO needs to be in low concentration, requiring a more clean hydrogen to avoid fuel cell deterioration.One part of this paper was developed in Stockholm, where there are some buses within the CUTE (clean urban transport for Europe) project that has been in operation with FC since January 2004. Another part was developed in Guaratingueta, Brazil. Brazil intends to start up a program of FC buses. As conclusion, this paper shows the economical analysis comparing buses moved by fuel cells using hydrogen by different kinds of production. Different compositions of Ni0,5-xCuxZn0,5Fe2O4 and Ni0,5-xCoxZn0,5Fe2O4 0 ≤ x ≤ 0.3 were synthesized ferrite y the citrate precursor method. The stoichiometric compositions were calcined in air at 350°C and then pressed into pellets and toroids. The pressed samples were sintered at temperatures of 1000, 1050 and 1100°C/3h in air control at the speed of heating and cooling. The calcined powders were characterized by XRD, TGA / DTG, FTIR, SEM and vibrating sample magnetometry (VSM) and the sintered samples by XRD, SEM, MAV, density and measurements of permeability and magnetic losses. There was pure phase formation ferrimagnetism applied at all temperatures except for A-I composition at all sintering temperatures and A-II only at a temperature of 1100°C. Crystallite sizes were obtained by Rietveld analysis, nanometer size from 11 to 20 nm for the calcined powders. For SEM, the sintered samples showed grain size between 1 and 10 micrometers. Bulk density (ρ) of sintered material presented to the Families almost linear behavior with increasing temperature and a tendency to decrease with increasing concentration of copper, different behavior of the B Family, where the increase in temperature decreased the density. The magnetic measurements revealed the powder characteristics of a soft ferrimagnetic material. Two processes of magnetization were considered, the superparamagnetism at low temperatures (350°C) and the formation of magnetic domains at higher temperatures. Obtaining the best parameters for P and B-II magnetic ferrites at high temperatures. The sintered material at 1000°C showed a relative permeability (μ) from 50 to 800 for the A Family and from 10 to 600 for the B Family. The samples sintered at 1100°C, B Family showed a variation from 10 to 1000 and the magnetic loss (tan δ) of A and B Families, around of 1. The frequency response of the toroidal core is in the range of 0.3 kHz. Several factors contribute to the behavior of microstructure considering the quantities μ and tan δ, such as the grain size, inter-and intragranular porosity, amount of grain boundary and the aspects of the dynamics of domain walls at high frequencies.

Biodiesel production has increased over the last decade because of the benefits associated with this fuel, including renewability, domestic feedstock, lower toxicity, and biodegradability. From 2008, the use of beef tallow as a feedstock for biodiesel production in Brazil has increased in significance, representing the second largest source of biodiesel, after soybeans. However, the performance of biodiesel in cold weather conditions is worse than diesel because of deposition of insoluble at low temperatures, accelerating the plugging of fuel filters and injectors of the vehicle engine. Studies have been conducted on beef tallow biodiesel, mostly related to the properties of thermal and oxidative stability. However, few studies have described the nature of the precipitate formed and its influence on product quality. Research suggests that the cause of deposition is related to the nature of saturated esters and monoacylglycerols as inducing agents. This study monitored the levels of mono-, diand triacylglycerols, the oxidation stability and the cold filter plugging point (CFPP) in beef tallow biodiesel samples from two commercial producers in Brazil for a period of twelve months. Filtered precipitates were analyzed by comparative techniques of GCFID, HPLC-UV/VIS, HPLC-MS-IT-TOF and TG to verify the nature, using monopalmitin and monostearin as reference standards. The formation of precipitate reduced the levels of monoacylglycerols in the beef tallow biodiesel. GC-FID and LCMS- IT-TOF results confirmed the nature of the deposit as saturated monoacylglycerols, predominantly monostearin and monopalmitin as the second major component. Moreover the TG analysis of the residue indicated similar thermal decomposition of the reference standards. The precipitate did not affect the oxidation stability of beef tallow biodiesel and the CFPP characteristic of blends up B60. However, the presence of iron reduced significantly the oxidation stability of biodiesel

We report a theoretical investigation of thermal hysteresis in magnetic nanoelements. Thermal hysteresis originates in the existence of meta-stable states in temperature intervals which may be tuned by small values of the external magnetic field, and are controlled by the systems geometric dimensions as well as the composition. Two systems have been investigated. The first system is a trilayer consisting of one antiferromagnetic MnF2 film, exchange coupled with two Fe lms. At low temperatures the ferromagnetic layers are oriented in opposite directions. By heating in the presence of an external magnetic field, the Zeeman energy induces a gradual orientation of the ferromagnets with the external field and the nucleation of spin- op-like states in the antiferromagnetic layer, leading eventually, in temperatures close to the Neel temperature, to full alignment of the ferromagnetic films and the formation of frustrated exchange bonds in the center of the antiferromagnetic layer. By cooling down to low temperatures, the system follows a different sequence of states, due to the anisotropy barriers of both materials. The width of the thermal hysteresis loop depends on the thicknesses of the FM and AFM layers as well as on the strength of the external field. The second system consists in Fe and Permalloy ferromagnetic nanoelements exchange coupled to a NiO uncompensated substrate. Este trabalho visou a relacionar variáveis ambientais em instalação para criação de rãs, com cobertura de polietileno e baias construídas usando material alternativo, com o desempenho de rãs-touro (Rana catesbeiana). No interior das baias, foram medidas as temperaturas do piso, do ar ambiente (bulbo seco), de bulbo úmido, globo negro e da água do reservatório. Foram utilizados 60 animais por baia e três baias por galpão. As variáveis de desempenho estudadas foram peso vivo, ganho de peso e conversão alimentar. Nas condições experimentais, quando a temperatura do ar atingiu valores abaixo de 10 ºC ou superiores a 40 ºC, houve diminuição no consumo de ração pelos animais. Concluiu-se que o estresse predominante, neste tipo de estrutura, para as condições climáticas do período experimental, foi devido, principalmente, às baixas temperaturas. Concluiu-se, ainda, que o uso do Índice de Temperatura e Umidade (THI), na estimativa de variáveis de desempenho, melhorou a precisão da estimativa em relação ao uso exclusivo da temperatura do ar, embora valores desse índice, considerados estressantes para animais superiores, não o tenham sido para as rãs.

The aim was to study the effect of different storage temperatures on quality of red mombin fruit. The red mombin fruits were obtained from the Company CEAGESP/São Paulo/Brazil and transported in cool boxes to the laboratory, where they were selected on the base of appearance, maturity lack of physical damage, sanitized in 50 ppm chlorine-free solution and packaged in polystyrene trays wrapped with film of polyvinyl chloride (PVC). The experiment was a completely randomized design with three temperatures (4, 8 and 25 degrees C) and 5 time intervals (0, 2, 4, 6 and 8 days after the experiment installation). In each survey firmness, titratable acidity, soluble solids, ascorbic acid content, the skin color and also the release of CO(2) by the fruit over time were evaluated. It was observed that low temperatures prolong the fruits shelf life and the storage temperature influences the characteristics, the temperature of 8 degrees C was most suitable for the storage of red mombin fruits. Besides, the fruit color was a good indicator of changes in the pulp during storage.

In adult mammals, severe hypothermia leads to respiratory and cardiac arrest, followed by death. Neonatal rats and hamsters can survive much lower body temperatures and, upon artificial rewarming, spontaneously recover from respiratory arrest (autoresuscitate), typically suffering no long-term effects. To determine developmental and species differences in cold tolerance (defined here as the temperature of respiratory arrest) and its relation to the ability to autoresuscitate, we cooled neonatal and juvenile Sprague-Dawley rats and Syrian hamsters until respiration ceased, followed by rewarming. Ventilation and heartbeat were continuously monitored. In rats, cold tolerance did not change throughout development, however the ability to autoresuscitate from hypothermic respiratory arrest did (lost between postnatal days, P, 14 and 20), suggesting that the mechanisms for maintaining breathing at low temperatures was retained throughout development while those initiating breathing on rewarming were altered. Hamsters, however, showed increased cold tolerance until P26-28 and were able to autoresuscitate into adulthood (provided the heart kept beating throughout respiratory arrest). Also, hamsters were more cold tolerant than rats. We saw no evidence of gasping to initiate breathing following respiratory arrest, contributing to the hypothesis that hypothermic respiratory arrest does not lead to anoxia. (C) 2012 Elsevier B.V.
