The gene Pi-ar confers resistance to Pyricularia grisea race IB-45 in a somaclone derived from immature panicles of the susceptible rice (Oryza sativa) cultivar Araguaia. RAPD technique was used to identify molecular markers linked to this gene utilizing bulked segregant analysis. Initially, the two parental DNAs from the resistant donor SC09 and 'Araguaia' were analyzed using random primers. Of the 240 primers tested, 203 produced amplification products. The two parental DNAs along with the resistant and susceptible bulks of F2 population were screened using 48 primers that differentiated resistant and susceptible parents. Even though eight primers differentiated the resistant bulk from the susceptible bulk, as well as somaclone SC09 and 'Araguaia', only one primer, OPC02 ('GTGAGGCGTC'), was found to be tightly linked (1.7cM) to the resistance gene of somaclone SC09.

Thirty-nine rice (Oryza sativa) hybrids and their restorers were assessed for vertical resistance to Pyricularia grisea in the rice blast nursery, and in artificial inoculation tests with two pathotypes, under controlled greenhouse conditions. The hybrids were developed from cytoplasmic genetic male sterile lines 046I and IR 58025A, derived from WA cytoplasm. In the rice blast nursery all hybrids showed susceptible reaction varying from 5 to 9. Compatible and incompatible leaf blast reactions of hybrids to two pathotypes, IC-1 and IB-45, were observed in inoculation tests. A majority of the hybrids were resistant when the restorer was resistant. However, seven of the 25 F1 hybrids exhibited susceptible reactions even when one of the parents was resistant to a pathotype. The partial resistance of 11 hybrids and their parents that showed compatible reactions to two pathotypes was analyzed. Differential interaction between isolates and genotypes was observed for partial resistance in relation to both disease severity and lesion number indicating the specific nature of partial resistance.

A study was undertaken to examine the pathogenic diversity of Pyricularia grisea isolates retrieved from 14 upland rice (Oryza sativa) cultivars in experimental plots during a period of five years. Inoculations were performed on 32 genotypes with 85 monoconidial isolates under controlled greenhouse conditions. Based on the reaction pattern of eight international differentials, eleven pathotypes of P. grisea were identified. The predominant international races or pathotypes were IB-9 (56.4%), IB-1 (16.4%) and IB-41 (11.8%). A set of eight commercial upland rice cultivars ('Carajás', 'Confiança', 'Maravilha', 'Primavera', 'Progresso', 'Caiapó', 'IAC-47', 'IAC-201') was utilized as additional differentials for describing the virulence pattern of P. grisea. Twenty-six Brazilian pathotypes were identified on the basis of disease reaction on these differentials, in contrast to the 11 international pathotypes. The most predominant Brazilian pathotypes, BB-21 and BB-41 were represented by 28.2% and 17.6% of the isolates tested, respectively. Isolates virulent and avirulent to cultivar 'Primavera' were encountered within the pathotype IB-1. Utilizing Brazilian cultivars as differentials, the 14 isolates of the pathotype IB-1could be further classified into eight local pathotypes, BB-41, BB-13, BB-21, BB-9, BB-29, BB-61, BD-9 and BG-1. Virulence to improved rice cultivars 'Canastra', 'Confiança', 'Carisma', 'Maravilha', 'Primavera' and 'Bonança' was frequent in pathogen population. Some of the Brazilian pathotypes that showed differential reaction on commercial rice cultivars could be utilized for incorporating resistance genes in susceptible cultivars improved for grain quality, by conventional breeding methods.

An epidemic of rice (Oryza sativa) blast occurred on cultivars Epagri 108 and 109 in the municipalities of Lagoa da Confusão and Duerê in the State of Tocantins, during the rice-growing season 1998-99. DNA fingerprinting and virulence phenotype analysis were utilized to determine the diversity of Pyricularia grisea isolates collected from these cultivars in one epidemic year. Rep-PCR analysis of isolates was done by using two primer sequences from Pot2. Two distinct fingerprint groups or lineages were identified among 53 isolates collected from nine different commercial fields. The virulence pattern of isolates retrieved from these two cultivars was analyzed in artificial inoculation tests utilizing 32 genotypes in the greenhouse. A dendrogram constructed from virulence phenotype data showed a single group considering 77% similarity level. The predominant pathotype IB-45 was represented by 47 of the 53 isolates corresponding to 83%. Four other pathotypes (IB-1, IB-9, IB-13 and IB-41) were identified at random among the isolates from these cultivars. There was no relation between rep-PCR grouping and pathotypes. The results showed that the isolates of P. grisea recovered from cultivars Epagri108 and 109 in farmers' fields had narrow phenotypic and genetic diversity. The blast outbreak on these two cultivars one year after their introduction could be attributed to the new pathotype IB-45 or its increase, which was hitherto existing in low frequency.

Levantamentos periódicos da freqüência de virulência do patógeno causador de oídio (Blumeria graminis f. sp. tritici) em trigo (Triticum aestivum)auxiliam na escolha de fontes de resistência para uso em cruzamentos. Este trabalho relata resultados de cinco anos de verificação de efetividade de genes de resistência para populações patogênicas de B. graminis f. sp. tritici oriundas do Brasil e do Chile. Amostras de oídio foram recebidas pela Embrapa Trigo, Passo Fundo, RS, em 1995, 1996, 1997, 1999 e 2000. Inoculou-se cada isolado monopustular em plantas de trigo da série diferencial para raças. Foram identificadas 90 combinações diferentes de genes efetivos e inefetivos para resistência. Em 1995 e 1997, a maioria dos isolados foram virulentos com relação aos genes Pm3a, Pm3c, Pm8, Pm1+... e PmD1; em 1999, para Pm3a, Pm3c, Pm4b, Pm8, PmD1 e PmD2; e, em 2000, para Pm3a, Pm3c, Pm4b, Pm6, Pm8, PmD1 e PmD2. Em todos os anos analisados, permaneceram efetivos a todos os isolados os genes Pm2 e Pm4a+.... O número de genes inefetivos por isolado variou entre um e nove, no total de 13 genes ou combinações de genes testados. Isolados apresentando virulência a cinco genes ou combinações foram mais freqüentes em 1995 e em 1997, a seis em 1999 e a sete em 2000, o que pode significar aumento na complexidade da composição racial do patógeno a cada ano.

O efeito de diferentes temperaturas no solo e de diversas palhas sobre o solo na viabilidade dos escleródios de Sclerotinia sclerotiorum foi estudado em estufa. Em três ensaios de campo, estudou-se a solarização do solo associada à presença de palha de milho (Zea mays) sobre o solo na viabilidade de escleródios, durante três meses. Ensaios de campo foram realizados em Piracicaba e em Brasília. Escleródios foram produzidos em meio cenoura+fubá, e incorporados ao solo (ensaios em estufa), ou acondicionados em invólucros, e enterrados no solo a 5, 10 e 30 cm (ensaios no campo). Os tratamentos de solo no campo foram: solarizado (S), não solarizado (NS) e solarizado com adição de palha de milho (PS). Foram feitas avaliações a cada 30 dias, em meio NEON, observando a viabilidade e a presença de contaminantes nos escleródios. O aquecimento do solo em estufa a 50 e 60 ºC com diversas palhas inativou os escleródios, que tiveram maior incidência de contaminantes. No campo, o efeito da solarização do solo foi significativo, inviabilizando os escleródios enterrados a diferentes profundidades: em S após 90 dias, nas três profundidades, e em PS, após 60 dias, a 5 e 10 cm de profundidade. A incidência de escleródios contaminados em solos solarizados foi maior em S, seguido de PS. A maior variabilidade de contaminantes, porém, foi observada em PS. As temperaturas do solo em PS foram maiores quando comparadas aos outros tratamentos na mesma profundidade. Este fator proporcionou a redução do tempo da inativação dos escleródios de 90 dias em S, para 60 dias em PS.

A podridão vermelha da raiz, causada por Fusarium solani f. sp. glycines, é considerada uma das doenças mais severas de soja (Glycine max) no Sul do Brasil. Este trabalho avaliou a reação de 30 genótipos de soja em experimentos conduzidos em câmara de crescimento, na Embrapa Trigo, Passo Fundo, RS. Inoculou-se o fungo nos genótipos pelo método "palito-de-dente", através da introdução de uma ponta de palito colonizada pelo fungo no hipocótilo de cada plântula e pelo método "grão de sorgo", em que o inóculo, constituído de grãos de sorgo (Sorghum bicolor) colonizados, foi colocado ao redor do colo da planta. Avaliaram-se os sintomas na parte aérea e no sistema radicular. Para ambos métodos de inoculação, houve diferenças entre genótipos e a amplitude de reação variou de resistência moderada à alta suscetibilidade. Os genótipos BRS 66, BRS 137 e BRS 138, pelo método "palito-de-dente", e IAS 5 e BRS 137, pelo método "grão de sorgo", foram considerados moderadamente resistentes. Os genótipos Embrapa 59, CEP 12-Cambará, Ipagro 21, FT-Guaíra, FT-Abyara, BR-4 e FT-2003 foram moderadamente suscetíveis (MS) pelo método "palito-de-dente", enquanto Ivorá, RS 7-Jacuí, Fepagro RS-10, BR-16, CD 203, BR-4, CEP 20-Guajuvira, BRS 154, BRS 138 e Cobb foram MS pelo método "grão de sorgo". Por outro lado, Bragg, CD 205, RS 5-Esmeralda, RS 9- Itaúba, IAS 4, Ocepar 14, CD 201, FT 2011, FT-Saray, BRS 153 e FT-2004 foram suscetíveis em ambos os métodos usados.

A obtenção de novas fontes de resistência à brusone é requerimento básico para melhoramento do arroz (Oryza sativa). O objetivo do trabalho foi avaliar, para resistência à brusone, 39 somaclones da cultivar CICA-8, desenvolvidos a partir de calos de panículas imaturas. Os somaclones, nas gerações avançadas de R5 a R7, foram avaliados sob condições naturais de infecção e em testes de inoculação artificial em casa de vegetação, utilizando cinco isolados, pertencentes a quatro patótipos (ID-14, II-1, IB-1 e IB-45) de Pyricularia grisea. No viveiro de brusone os somaclones apresentaram diferentes reações à doença. Nos testes de inoculação os somaclones mostraram interação diferencial com os isolados do patógeno. Dois isolados altamente virulentos a cinco somaclones foram selecionados para determinar a resistência parcial. Não houve interação significativa entre genótipos e isolados para o índice de resistência parcial determinado com base no número de lesões/cm² de folha. Quatro somaclones mostraram graus significativamente maiores de resistência parcial à brusone quando comparados à cultivar parental CICA-8.

Wheat (Triticum aestivum) powdery mildew, caused by the biotrophic fungus Blumeria graminis f. sp. tritici, is one of the most severe foliar diseases attacking this crop, reducing grain yields by 10% to 62% in Brazil. The disease can be controlled by genetic resistance of the host, but the pathogen has physiological specialization, which enables it to infect wheat cultivars that have remained resistant for years. The objective of this work was to evaluate the variability of pathogenic strains of B. graminis f. sp. tritici collected in Brazil and the effectiveness of wheat resistance genes to powdery mildew in the 2003 crop season. Plants of a differential series were inoculated with each monopustular isolate. Thirty-one combinations of effective and ineffective resistance genes were identified. Only the gene Pm4a+... remained totally effective to all isolates, and gene Pm6 was highly effective (below 10% of susceptibility), whereas genes Pm3a and Pm8 were totally ineffective (susceptible to all isolates). Genes Pm3c, D1, and D2 showed low effectiveness (above 50% of susceptibility), and genes Pm1, 2, 4a, 1+?, and 2+Mld had mean effective results to most strains (susceptibility between 10% and 49%). The virulence formula Pm1, 3c, 4a, 6, 1+?, 2+Mld, 4a+..., D2 (effective genes) / 2, 3a, 8, D1 (ineffective genes) was most frequently found, accounting for 15% of the occurrences. The most frequent number of ineffective genes was seven, ranging from three to ten.

Cercospora kikuchii, involved with the defoliation of soybean (Glycine max) plants, is normally associated with Septoria glycines in late season. Seventy-two isolates from different regions in Brazil, obtained mainly from purple stained seeds, showed phenotypic variation. Cercosporin content and rate of colony growth was higly variable among isolates. A strong correlation between cercosporin content and virulence was identified. Genetic variation among and within populations was evaluated based on 86 RAPD loci. The RAPD analysis clustered all isolates into seven groups. No relationship was observed between RAPD groups and geographic origin or cercosporin content. The sequence of the internal spacer regions (ITS1-5.8S-ITS2) from 13 isolates chosen according to the previous RAPD and clustering analysis showed high similarity (97%-100%) to the GenBank sequences of C. kikuchii (AY266160, AY266161, AY152577 and AF291708). It is clear from this work that Brazilian isolates of C. kikuchii from different geographic regions, are variable in relation to virulence, RAPD profiles and cercosporin content. Cercosporin content could be a good parameter for choosing an adequate isolate for screening resistant or tolerant cultivars. Considering that this pathogen is easily seed-borne, findings are expected to show the same haplotypes in different regions. Migration could be favoured by infected seeds as demonstrated by RAPD analysis.

The virulence pattern of the isolates of Pyricularia grisea from commercial fields of the upland rice (Oryza sativa) cultivars 'Primavera' and 'BRS Bonança' was analyzed. A hundred and seventy monoconidial isolates of the pathogen virulent to 'Primavera' and 139 to 'BRS Bonança' collected from eight fields, during two years (2001-2003) were tested, under greenhouse conditions, on six newly released rice cultivars. Differences in virulence pattern were observed in pathogenic populations of 'Primavera' and 'BRS Bonança'. Isolates with virulence to improved cultivars were common in samples from farmers' fields in the absence of aloinfection. The virulence frequency of P. grisea isolates collected from 'Primavera'' to cultivars 'BRS Vencedora', 'BRS Colosso', 'BRS Liderança', 'BRS Soberana', 'BRS Curinga' and 'BRS Talento', was high in descending order. On the other hand, in the fungus population of 'BRS BRS Bonança' virulence frequency was high in 'BRS Talento', followed by 'BRS Curinga', 'BRS Vencedora', 'BRS Liderança', 'BRS Colosso' and 'BRS Soberana'. While virulence to 'BRS Talento' was rare among isolates from 'Primavera', it was most frequent in isolates of 'BRS Bonança'. The six improved rice cultivars permitted to differentiating agriculturally important virulences in the pathogen population which can be utilized in selecting breeding lines for specific resistance, in rice blast improvement program.

O efeito da queima-da-bainha em alguns componentes da produtividade do arroz (Oryza sativa) irrigado foi estimado utilizando o método de perfilhos únicos como unidade experimental. As epidemias artificiais foram desenvolvidas através de inoculação de campo com inóculo de Rhizoctonia solani multiplicado em casca e grão de arroz. A severidade da doença foi avaliada em sete somaclones de Metica-1 e na cultivar Metica-1, considerando a altura da lesão em 50 perfilhos como variável. As relações entre o comprimento da panícula e a severidade da queima-da-bainha foram lineares e negativas para os genótipos (r = -0.65 P<0.01 ) e os de ciclo médio (r =-0.62 P<0.01). A queima-da-bainha afetou o comprimento da panícula em 8,83% e 6,26% nos genótipos de ciclo precoce e médio, respectivamente. Os danos em porcentagem de espiguetas vazias para os genótipos de ciclo precoce e médio foram de 10,94% e 12,20%, respectivamente. A queima-da-bainha afetou mais a massa de grãos/panícula do que o enchimento das espiguetas e o comprimento da panícula, sendo de 32,75% para os genótipos de ciclo precoce e de 30,99% para os genótipos de ciclo médio. O método de perfilhos únicos pode ser utilizado para estimar com precisão os danos potenciais causados pela queima-da-bainha em diferentes genótipos.

The phenotypic diversity of Magnaporthe grisea was evaluated based on leaf samples with blast lesions collected from eight commercial fields of the upland rice cultivars 'BRS Primavera' and 'BRS Bonança', during the growing seasons of 2001/2002 and 2002/2003, in Goias State. The number of M. grisea isolates from each field utilized for virulence testing varied from 28 to 47. Three different indices were used based on reaction type in the eight standard international differentials and eight Brazilian differentials. The M. grisea subpopulations of ´Primavera' and 'Bonança', as measured by Simpson, Shannon and Gleason indices, showed similar phenotypic diversities. The Simpson index was more sensitive relation than those of Shannon and Gleason for pathotype number and standard deviation utilizing Brazilian differentials. However, the Gleason index was sensitive to standard deviation for international differentials. The sample size did not significantly influence the diversity index. The two sets of differential cultivars used in this study distinguished phenotypic diversity in different ways in all of the eight subpopulations analyzed. The phenotypic diversity determined based on eight differential Brazilian cultivars was lower in commercial rice fields of 'Primavera' than in the fields of 'Bonança,' independent of the diversity index utilized, year and location. Considering the Brazilian differentials, the four subpopulations of 'BRS Primavera' did not show evenness in distribution and only one pathotype dominated in the populations. The even distribution of pathotype was observed in three subpopulations of 'BRS Bonança'. The pathotype diversity of M. grisea was determined with more precision using Brazilian differentials and Simpson index.

The phenotypic and genetic diversity of 77 isolates of Pyricularia grisea collected from two upland rice cultivars, Maravilha and Primavera, was studied. Isolates exhibiting compatible reaction to cv.Primavera were incompatible to cv.Maravilha and vice versa, with the exception of six isolates that were compatible to both cultivars. The virulence of isolates from cv. Maravilha on 32 test genotypes of rice was significantly higher (t = 9.09, p < 0.0001) than the isolates from cv.Primavera. A phenogram constructed from virulence data showed two main groups, one constituted mainly of isolates from cv.Primavera (97.6%) and the other of isolates from cv.Maravilha (91.17%). Rep-PCR analysis of isolates using two primers designed from sequences of Pot2 showed that isolates could be clustered broadly into two groups. The average similarity within a cluster of isolates from cv.Primavera was significantly greater than the average similarity among the isolates of cv.Maravilha (t = 5.37, p < 0.0001). There was close correspondence between clusters based on PCR and virulence data (r = 0.48, p < 0.011). The results showed that isolates of P. grisea were cultivar specific and had low phenotypic and genetic diversity.
