The purpose of this study was to evaluate the effectiveness of glycolic propolis (PRO) and ginger (GIN) extracts, calcium hydroxide (CH), chlorhexidine (CLX) gel and their combinations as ICMs (ICMs) against Candida albicans, Enterococcus faecalis, Escherichia coli and endotoxins in root canals. Material and Methods: After 28 days of contamination with microorganisms, the canals were instrumented and then divided according to the ICM: CH+saline; CLX, CH+CLX, PRO, PRO+CH; GIN; GIN+CH; saline. The antimicrobial activity and quantification of endotoxins by the chromogenic test of Limulus amebocyte lysate were evaluated after contamination and instrumentation at 14 days of ICM application and 7 days after ICM removal. Results and Conclusion: After analysis of results and application ofthe Kruskal-Wallis and Dunn statistical tests at 5% significance level, it was concluded that all ICMs were able to eliminate the microorganisms in the root canals and reduce their amount of endotoxins; however, CH was more effective in neutralizing endotoxins and less effective against C. albicans and E. faecalis, requiring the use of medication combinations to obtain higher success.

In this study, curved maxillary molar root canals were instrumented with RaCe rotary system to evaluate: 1. the occurrence of canal transportation using a radiographic platform; 2. the action of the instruments on the dentin walls, centering ability and canal enlargement by analysis of digital images; and the percentage of regular dentin surfaces and debris within the canal by histological analysis. Ten mesiobuccal roots of extracted human maxillary molars were embedded in acrylic resin and sectioned at the middle and apical thirds. Root canal shaping was performed using the RaCe rotary system at 250 rpm and 1 Ncm torque. Each instrument set was used five times according to a crowndown technique in the following sequence: 40/0.10, 35/0.08, 25/0.06, 25/0.04, 25/0.02 (working length - WL), 30/0.02 (WL) and 35/0.02 (WL). Each instrument was inserted until resistance was felt and then pulled back, followed by brushing movements towards all canal walls. Each specimen was assessed by three study methods: radiographic platform, digitized image assessment and histological analysis. The radiographic platform showed lack of apical transportation. No statistically significant difference (Wilcoxon test, p>0.05) was found between the middle and apical thirds regarding instrument action on dentin walls, centering ability, area of root canal enlargement, percentage of regular dentin surfaces and debris within the root canal. It may be concluded that RaCe system is a suitable method for the preparation of curved root canals, regarding the maintenance of root canal original path, action on dentin walls, canal enlargement and removal of debris from the root canal lumen.

The aim of this study was to evaluate the effect of final irrigation protocols (17% EDTA, BioPure MTAD, SmearClear, and QMiX) on microhardness and erosion of root canal dentin. Fifty roots were sectioned transversely at the cement-enamel junction and each root was sectioned horizontally into 4-mm-thick slices. The samples were divided into five groups (n=10) according to the final irrigation protocol: G1: distilled water (control group); G2: 17% EDTA; G3: BioPure MTAD; G4: SmearClear; and G5: QMiX. The dentin microhardness was then measured with a load of 25 g for 10 s. Initially, the reference microhardness values were obtained for the samples without any etching. The same samples were then submitted to the final irrigation protocols. A new measure was realized and the difference between before and after the procedures was the dentin microhardness reduction. In sequence, the specimens were submitted to SEM analysis to verify the dentinal erosion. The Kruskal Wallis and Dunn tests (α=5%) were used to compare the results. The dentin microhardness decreased for all final irrigation protocols. There was no significant difference between groups 2, 3, 4, and 5 (P>0.05), but this groups presented significant dentin microhardness reduction than G1 (P<0.05). In G2, occurred the highest incidence of dentinal erosion (P<0.05). 17% EDTA, BioPure MTAD, SmearClear, and QMiX promoted significant dentin microhardness reduction. © 2013 Wiley Periodicals, Inc.

Com o objetivo de avaliar a superfície dentinária apical, após a remoção de diferentes pastas de hidróxido de cálcio (Ca(OH)2), foram selecionados 68 dentes unirradiculares, preparados por um único operador, valendo-se da técnica regressiva. Os canais foram irrigados com 5ml de hipoclorito de sódio (NaOCl) a 1%, seguido de irrigação final com 10 ml de ácido etilenodiaminotetracético associado ao tergentol (EDTA-T) a 17%. As amostras foram divididas, aleatoriamente, em 4 grupos (n=15), de acordo com o tipo de associação de Ca(OH)2: associado à solução de clorexidina 0.2% (Grupo 1); ao propilenoglicol (Grupo 2); à ciprofloxacina, metronidazol e água destilada (Grupo 3); à ciprofloxacina, metronidazol e propilenoglicol (Grupo 4). Os dentes controle negativos (n=4) não foram preenchidos com Ca(OH)2, e nos controle positivo, (n=4) não foi feita a remoção do Ca(OH)2. As amostras foram armazenadas à 37ºC, em 100% de umidade relativa, por 21 dias. A remoção da medicação foi realizada com: irrigação com 5ml de NaOCl 1%; limagem circunferencial com instrumento memória; 5ml de NaOCl 1%; patência com lima K #10; ultra-som com lima K #15; e 10ml de EDTA-T a 17%. Os espécimes foram analisados em microscópio eletrônico de varredura e, após a aplicação do teste de Kruskal-Wallis (α = 5%), observou-se que não houve diferença entre os grupos em relação à remoção do Ca(OH)2 (p = 0.0951). O teste Qui-quadrado (α = 5%) indicou a predominância de Ca(OH)2 obstruindo os túbulos dentinários, independente do veículo utilizado. Após análise química pontual (espectrometria por dispersão de raios-x EDS) e, aplicação do teste de análise das variâncias e Tukey (α = 5%), pôde-se observar que houve diferença na quantidade de íons cálcio (Ca) remanescente entre os grupos 1 e 4 (p<0.01). Com base na metodologia empregada, concluiu-se que, independente do tipo de veículo utilizado, a superfície dentinária apical permaneceu igualmente coberta por Ca (OH)2.

An increase in dentin roughness, associated with surface composition, contributes to bacterial adherence in recontaminations. Surface roughness is also important for micromechanical interlocking of dental materials to dentin, and understanding the characteristics of the surface is essential to obtain the adhesion of root canal sealers that have different physico-chemical characteristics.

O objetivo deste estudo foi avaliar a resistência à fadiga cíclica de instrumentos rotatórios de níquel-titânio após simulação de uso clinico em canais curvos (curvatura de 40° e raio de 5 mm). Trinta e seis instrumentos, calibre n° 25 conicidade 0,04, foram divididos em três grupos: o Grupo A com um ciclo de uso; Grupo B, três ciclos de uso e grupo C, cinco ciclos de uso. Um cronômetro digital aferiu em segundos o tempo até a fratura do instrumento que, posteriormente, foi convertido em número de ciclos para fratura. Os dados foram analisados por ANOVA e teste de Tukey (p<0,05). O grupo que utilizou o instrumento por cinco ciclos (grupo C) atingiu significativamente menores números de ciclos antes da fratura (média = 197,5 ciclos) quando comparado com os instrumentos utilizados em um ciclo (média = 309,2) e três ciclos (média = 287,5). Os resultados mostraram que o número de uso de instrumentos RaCe para modelar canais curvos afeta negativamente a resistência à fadiga cíclica dos instrumentos após cinco usos.
