MVA ist ein attenuiertes Vakziniavirus, das durch wiederholte Passagierung von Chorioallantoisvirus Ankara auf Hühnerembryofibroblasten gewonnen wurde. Es ist bis auf wenige Ausnahmen nicht mehr in der Lage, in Säugerzellen zu replizieren, zeigt aber dennoch eine vollständige virale Proteinexpression und induziert nach Immunisierung eine zu VACV vergleichbare Immunantwort. Aus diesem Grund wurde es bereits als Impfstoff gegen die menschliche Pockenerkrankung eingesetzt, ohne hierbei die bei den klassischen Impfviren beobachtbaren Nebenwirkungen hervorzurufen. Das Genom von MVA enthält jedoch noch Immunmodulatoren, deren Deletion Ansatzpunkt für die weitere Verbesserung des Impfstoffes sein kann. Im Rahmen der vorliegenden Arbeit wurde eine Deletionsmutante untersucht, bei der das Gen für den Interleukin-1β Rezeptor (IL-1βR) deletiert ist (MVA ΔIL-1βR). Es konnte nachgewiesen werden, dass der IL-1βR in murinen als auch in humanen Zellen exprimiertes IL-1β bindet und somit inaktiviert. Des Weiteren wurde gefunden, dass MVA in der Lage ist, IL-1β in antigenpräsentierenden Zellen zu induzieren, welches aber durch die Neutralisierung aufgrund des IL-1βR nur transient nachzuweisen war. Im Gegensatz dazu induzierte MVA ΔIL-1βR eine anhaltende und um ein Vielfaches erhöhte Sekretion an IL-1β. Untersuchungen in antigenpräsentierenden Zellen aus knock-out Mäusen, die verschiedene Defizienzen in den Signalwegen zur IL-1β-Induktion trugen, zeigten, dass die Sekretion von IL-1β von Caspase-1 abhängig war, welches wahrscheinlich aus der vorgeschalteten Aktivierung der NLRP3- und/oder AIM2-Inflammasomen resultierte. Interessanterweise wurden auch Caspase-1 unabhängige Mechanismen beobachtet, die auf eine Inflammasom-unabhängige IL-1β-Induktion hinweisen könnten. In Bezug auf die Immunaktivierung führte die vermehrte Sekretion von IL-1β durch MVA ΔIL-1βR vermutlich zu einer verbesserten Antigenpräsentation, die die nachfolgende T-Zellantwort beeinflusste. In Übereinstimmung mit bereits veröffentlichten Daten wurde nach Immunisierung mit MVA ΔIL-1βR eine effektivere Gedächtnis-T-Zellantwort festgestellt, deren Charakteristika und zu Grunde liegenden Mechanismen hier untersucht wurden. Jedoch konnten weder Unterschiede in weiteren pro-inflammatorischen Zytokinmustern noch im Verlauf insbesondere der CD8+ T-Zell-Aktivierung und -erhaltung zwischen MVA und MVA ΔIL-1βR beobachtet werden. Als mögliche weitere Ursache für die veränderte Gedächtnis-T-Zellantwort könnte daher eine vermehrte Stimulation durch antigenpräsentierende Zellen und eine IL-21-vermittelte bessere Unterstützungsfunktion der CD8+ Gedächtnis-T-Zellen durch CD4+ T-Zellen in Frage kommen. Zusammenfassend konnten hier neue molekulare Mechanismen, die zur Induktion von IL-1β nach einer MVA-Infektion führen, aufgedeckt werden. Darüber hinaus existieren bereits erste Hinweise auf einen Vorteil der Deletion des IL-1βR für MVA-basierte Vektorimpfstoffe. Die vorliegende Arbeit hat weitere Daten erhoben, die das Erzielen verbesserter Immunantworten nach Immunisierung mit MVA ΔIL-1βR unterstützen, woraus sich neue Ansätze für die Entwicklung MVA-basierter Impfstoffe ergeben könnten.

Interleukin (IL)-22 ist ein Effektorzytokin, das von Zellen des Immunsystems produziert wird und auf Epithelzellen wirkt. Es nimmt eine duale Rolle ein, indem es abhängig vom Gewebe und Zytokinmilieu entweder entzündungsfördernden oder entzündungshemmenden Einfluss ausübt. Über seine Wirkung bei Asthma bronchiale ist bislang noch wenig bekannt. In der vorliegenden Arbeit konnten in einem murinen Modell der allergischen Atemwegsentzündung lymphoide Zellen des angeborenen Immunsystems als Hauptproduzenten für IL-22 detektiert werden, die bislang noch nicht im Zusammenhang mit Asthma bronchiale beschrieben wurden. Es konnte gezeigt werden, dass IL-22- defiziente Mäuse eine verstärkte Atemwegsentzündung entwickelten und sich die IL-22-Defizienz in diesem Modell entzündungsfördernd auf die induzierte Atemwegsentzündung auswirkte. Mit Hilfe einer murinen bronchialen Epithelzelllinie wurden die Mechanismen des IL-22 und die Expression Asthma-relevanter Mediatoren untersucht. Der beobachtete inhibierende IL-22-Effekt ließ sich mit Hilfe seines natürlichen Antagonisten IL-22BP neutralisieren. Diese entzündungshemmende Wirkung des IL-22 konnte ebenfalls in Wildtyp-Mäusen, denen rekombinantes IL-22 intratracheal verabreicht worden war, bestätigt werden.

It is well known that ageing and cancer have common origins due to internal and environmental stress and share some common hallmarks such as genomic instability, epigenetic alteration, aberrant telomeres, inflammation and immune injury. Moreover, ageing is involved in a number of events responsible for carcinogenesis and cancer development at the molecular, cellular, and tissue levels. Ageing could represent a “blockbuster” market because the target patient group includes potentially every person; at the same time, oncology has become the largest therapeutic area in the pharmaceutical industry in terms of the number of projects, clinical trials and research and development (R&D) spending, but cancer remains one of the leading causes of mortality worldwide. The overall aim of the work presented in this thesis was the rational design of new compounds able to modulate activity of relevant targets involved in cancer and aging-related pathologies, namely proteasome and immunoproteasome, sirtuins and interleukin 6. These three targets play different roles in human cells, but the modulation of its activity using small molecules could have beneficial effects on one or more aging-related diseases and cancer. We identified new moderately active and selective non-peptidic compounds able to inhibit the activity of both standard and immunoproteasome, as well as novel and selective scaffolds that would bind and inhibit SIRT6 selectively and can be used to sensitize tumor cells to commonly used anticancer agents such gemcitabine and olaparib. Moreover, our virtual screening approach led us also to the discovery of new putative modulators of SIRT3 with interesting in-vitro and cellular activity. Although the selectivity and potency of the identified chemical scaffolds are susceptible to be further improved, these compounds can be considered as highly promising leads for the development of future therapeutics.

The aim of this randomized, single-blinded cross-over study was to investigate the response of interleukin-6 (IL-6) during moderate aerobic exercise in stable euglycaemia and hyperglycaemia in seven male patients with type 1 diabetes mellitus. IL-6 increased significantly over the entire study period in euglycaemia, but not in hyperglycaemia.

The production of epithelial neutrophil activating peptide-78 (NA-78) and the interleukins IL-8 and IL-6 by endometrial stromal cells is stimulated by pro-inflammatory interleukin-1 (IL-1) and tumour necrosis factor-α (TNF-α). IL-8 is suggested to play a role in the pathogenesis of endometriosis, and in these women the peritoneal fluid concentrations of ENA-78 and IL-8 are increased. TNF-α has been tested together with interferon-γ because of their cooperative stimulation of IL-6. The release of IL-8, however, is inhibited with increasing interferon levels. The aim of the study was the analysis of the production of ENA-78, IL-6 and IL-8 by cultured human endometrial stromal cells in the presence of varying concentrations of IL-1β, TNF-α, and interferon-γ.

The effect of prolonged electroporation-mediated human interleukin-10 (hIL-10) overexpression 24 hours before transplantation, combined with sequential human hepatocyte growth factor (HGF) overexpression into skeletal muscle on day 5, on rat lung allograft rejection was evaluated. Left lung allotransplantation was performed from Brown-Norway to Fischer-F344 rats. Gene transfer into skeletal muscle was enhanced by electroporation. Three groups were studied: group I animals (n = 5) received 2.5 μg pCIK-hIL-10 (hIL-10/CMV [cytomegalovirus] early promoter enhancer) on day -1 and 80 μg pCIK-HGF (HGF/CMV early promoter enhancer) on day 5. Group II animals (n = 4) received 2.5 μg pCIK-hIL-10 and pUbC-hIL-10 (hIL-10/pUbC promoter) on day -1. Control group III animals (n = 4) were treated by sham electroporation on days -1 and 5. All animals received daily nontherapeutic intraperitoneal dose of cyclosporin A (2.5 mg/kg) and were sacrificed on day 15. Graft oxygenation and allograft rejection were evaluated. Significant differences were found between study groups in graft oxygenation (Pao(2)) (P = .0028; group I vs. groups II and III, P < .01 each). Pao(2) was low in group II (31 ± 1 mm Hg) and in group III controls (34 ± 10 mm Hg), without statistically significant difference between these 2 groups (P = .54). In contrast, in group I, Pao(2) of recipients sequentially transduced with IL-10 and HGF plasmids was much improved, with 112 ± 39 mm Hg (vs. groups II and III; P < .01 each), paralleled by reduced vascular and bronchial rejection (group I vs. groups II and III, P < .021 each). Sequential overexpression of anti-inflammatory cytokine IL-10, followed by sequential and overlapping HGF overexpression on day 5, preserves lung function and reduces acute lung allograft rejection up to day 15 post transplant as compared to prolonged IL-10 overexpression alone.

The IABP-SHOCK-trial was a morbidity-based randomized controlled trial in patients with infarction-related cardiogenic shock (CS), which used the change of the quantified degree of multiorgan failure as determined by APACHE II score over a 4-day period as primary outcome measure. The prospective hypothesis was that adding IABP therapy to "standard care" would improve CS-triggered multi organ dysfunction syndrome (MODS). The primary endpoint showed no difference between conventionally managed cardiogenic shock patients and those with IABP support. In an inflammatory marker substudy, we analysed the prognostic value of interleukin (IL)-1β, -6, -7, -8, and -10 in patients with acute myocardial infarction complicated by cardiogenic shock.

Systemic concentrations of interleukin-6 (IL-6) are elevated in patients with liver cirrhosis, and impaired hepatic uptake of IL-6 was suggested to contribute to higher levels in these patients. To test this hypothesis IL-6 was measured in portal venous serum (PVS), hepatic venous serum (HVS) and systemic venous serum (SVS) of 41 patients with liver cirrhosis and four patients with normal liver function. IL-6 was higher in PVS than HVS of all blood donors and about 43% of portal vein derived IL-6 was extracted by the healthy liver, and 6.3% by the cirrhotic liver demonstrating markedly impaired removal of IL-6 by the latter. Whereas in patients with CHILD-PUGH stage A IL-6 in HVS was almost 25% lower than in PVS, in patients with CHILD-PUGH stage C IL-6 was similarly abundant in the two blood compartments. Ascites is a common complication in cirrhotic patients and was associated with higher IL-6 levels in all blood compartments without significant differences in hepatic excretion. Hepatic venous pressure gradient did not correlate with the degree of hepatic IL-6 removal excluding hepatic shunting as the principal cause of impaired IL-6 uptake. Furthermore, patients with alcoholic liver cirrhosis had higher IL-6 in all blood compartments than patients with cryptogenic liver cirrhosis. Aetiology of liver cirrhosis did not affect hepatic removal rate indicating higher IL-6 synthesis in patients with alcoholic liver cirrhosis. In summary, the current data provide evidence that impaired hepatic removal of IL-6 is explained by hepatic shunting and liver dysfunction in patients with liver cirrhosis partly explaining higher systemic levels.

Toll interleukin-1 receptor (IL-1R) 8 (TIR8), also known as single Ig IL-1 receptor (IL-R)-related molecule, or SIGIRR, is a member of the IL-1R-like family, primarily expressed by epithelial cells. Current evidence suggests that TIR8 plays a nonredundant role as a negative regulator in vivo under different inflammatory conditions that are dependent on IL-R and Toll-like receptor (TLR) activation. In the present study, we examined the role of TIR8 in innate resistance to acute lung infections caused by Pseudomonas aeruginosa, a Gram-negative pathogen responsible for life-threatening infections in immunocompromised individuals and cystic fibrosis patients. We show that Tir8 deficiency in mice was associated with increased susceptibility to acute P. aeruginosa infection, in terms of mortality and bacterial load, and to exacerbated local and systemic production of proinflammatory cytokines (gamma interferon [IFN-γ], tumor necrosis factor alpha [TNF-α], IL-1β, and IL-6) and chemokines (CXCL1, CXCL2, and CCL2). It has been reported that host defense against P. aeruginosa acute lung infection can be improved by blocking IL-1 since exaggerated IL-1β production may be harmful for the host in this infection. In agreement with these data, IL-1RI deficiency rescues the phenotype observed in Tir8-deficient mice: in Tir8-/- IL-1RI-/- double knockout mice we observed higher survival rates, enhanced bacterial clearance, and reduced levels of local and systemic cytokine and chemokine levels than in Tir8-deficient mice. These results suggest that TIR8 has a nonredundant effect in modulating the inflammation caused by P. aeruginosa, in particular, by negatively regulating IL-1RI signaling, which plays a major role in the pathogenesis of this infectious disease.

nterleukins (ILs) are cytokines which are defined by their capability to convey information between leukocytes, in this way directing proliferation, activation, and migration and also regulation of the cells. Data from anti-IL treatments in systemic autoimmune diseases have shown these drugs to be beneficial and to have a satisfactory safety profile and tolerance. Recent publications of small case series suggest that several anti-IL drugs have considerable efficacy in treating otherwise refractory uveitis. Anti-IL therapy, therefore, might constitute an option for the treatment of uveitis resistant to corticosteroids, classical immunosuppressives, or tumor necrosis factor-α inhibitors. However, due to high costs and possible long-term risks, anti-IL agents should currently be reserved to selected uveitis patients and be administered only under close interdisciplinary monitorin

The identification of associations between interleukin-28B (IL-28B) variants and the spontaneous clearance of hepatitis C virus (HCV) raises the issues of causality and the net contribution of host genetics to the trait. To estimate more precisely the net effect of IL-28B genetic variation on HCV clearance, we optimized genotyping and compared the host contributions in multiple- and single-source cohorts to control for viral and demographic effects. The analysis included individuals with chronic or spontaneously cleared HCV infections from a multiple-source cohort (n = 389) and a single-source cohort (n = 71). We performed detailed genotyping in the coding region of IL-28B and searched for copy number variations to identify the genetic variant or haplotype carrying the strongest association with viral clearance. This analysis was used to compare the effects of IL-28B variation in the two cohorts. Haplotypes characterized by carriage of the major alleles at IL-28B single-nucleotide polymorphisms (SNPs) were highly overrepresented in individuals with spontaneous clearance versus those with chronic HCV infections (66.1% versus 38.6%, P = 6 × 10(-9) ). The odds ratios for clearance were 2.1 [95% confidence interval (CI) = 1.6-3.0] and 3.9 (95% CI = 1.5-10.2) in the multiple- and single-source cohorts, respectively. Protective haplotypes were in perfect linkage (r(2) = 1.0) with a nonsynonymous coding variant (rs8103142). Copy number variants were not detected. CONCLUSION: We identified IL-28B haplotypes highly predictive of spontaneous HCV clearance. The high linkage disequilibrium between IL-28B SNPs indicates that association studies need to be complemented by functional experiments to identify single causal variants. The point estimate for the genetic effect was higher in the single-source cohort, which was used to effectively control for viral diversity, sex, and coinfections and, therefore, offered a precise estimate of the net host genetic contribution.

Recurrent airway obstruction (RAO) in horses is the result of an interaction of genetic and environmental factors and shares many characteristics with human asthma. Many studies have suggested that the interleukin-4 receptor gene (IL4R) is associated with this disease, and a QTL region on chromosome 13 containing IL4R was previously detected in one of the two Swiss Warmblood families. We sequenced the entire IL4R gene in this family and detected 93 variants including five non-synonymous protein-coding variants. The allele distribution at these SNPs supported the previously detected QTL signal. Subsequently, we investigated IL4R mRNA expression in bronchoalveolar lavage fluid cells. During exacerbation, IL4R expression was increased in RAO-affected offspring in the implicated family, but not in the other family. These findings support that IL4R plays a role in some cases of RAO.
