Inappropriate activation of the renin-angiotensin system (RAS) contributes to many CKDs. However, the role of the RAS in modulating AKI requires elucidation, particularly because stimulating type 1 angiotensin II (AT1) receptors in the kidney or circulating inflammatory cells can have opposing effects on the generation of inflammatory mediators that underpin the pathogenesis of AKI. For example, TNF-α is a fundamental driver of cisplatin nephrotoxicity, and generation of TNF-α is suppressed or enhanced by AT1 receptor signaling in T lymphocytes or the distal nephron, respectively. In this study, cell tracking experiments with CD4-Cre mT/mG reporter mice revealed robust infiltration of T lymphocytes into the kidney after cisplatin injection. Notably, knockout of AT1 receptors on T lymphocytes exacerbated the severity of cisplatin-induced AKI and enhanced the cisplatin-induced increase in TNF-α levels locally within the kidney and in the systemic circulation. In contrast, knockout of AT1 receptors on kidney epithelial cells ameliorated the severity of AKI and suppressed local and systemic TNF-α production induced by cisplatin. Finally, disrupting TNF-α production specifically within the renal tubular epithelium attenuated the AKI and the increase in circulating TNF-α levels induced by cisplatin. These results illustrate discrepant tissue-specific effects of RAS stimulation on cisplatin nephrotoxicity and raise the concern that inflammatory mediators produced by renal parenchymal cells may influence the function of remote organs by altering systemic cytokine levels. Our findings suggest selective inhibition of AT1 receptors within the nephron as a promising intervention for protecting patients from cisplatin-induced nephrotoxicity.

The Hyades stream has long been thought to be a dispersed vestige of the Hyades cluster. However, recent analyses of the parallax distribution, of the mass function, and of the action-space distribution of stream stars have shown it to be rather composed of orbits trapped at a resonance of a density disturbance. This resonant scenario should leave a clearly different signature in the element abundances of stream stars than the dispersed cluster scenario, since the Hyades cluster is chemically homogeneous. Here, we study the metallicity as well as the element abundances of Li, Na, Mg, Fe, Zr, Ba, La, Ce, Nd and Eu for a random sample of stars belonging to the Hyades stream, and compare them with those of stars from the Hyades cluster. From this analysis: (i) we independently confirm that the Hyades stream cannot be solely composed of stars originating in the Hyades cluster; (ii) we show thatsomestars (namely 2/21) from the Hyades stream nevertheless have abundances compatible with an origin in the cluster; (iii) we emphasize that the use of Li as a chemical tag of the cluster origin of main-sequence stars is very efficient in the range 5500K ≤Teff≤ 6200K, since the Li sequence in the Hyades cluster is very tight, while at the same time spanning a large abundance range; (iv) we show that, while this evaporated population has a metallicity excess of ~0.2 dex with respect to the local thin-disc population, identical to that of the Hyades cluster, the remainder of the Hyades stream population has still a metallicity excess of ~0.06-0.15 dex, consistent with an origin in the inner Galaxy and (v) we show that the Hyades stream can be interpreted as an inner 4:1 resonance of the spiral pattern: this then also reproduces an orbital family compatible with the Sirius stream, and places the origin of the Hyades stream up to 1kpc inwards from the solar radius, which might explain the observed metallicity excess of the stream population. © 2011 The Authors Monthly Notices of the Royal Astronomical Society © 2011 RAS.

En sistemas lecheros pastoriles la estacionalidad y la variación interanual de la productividad forrajera constituyen la principal restricción tanto biofísica como económica. La medición de biomasa aérea permite estimar el forraje disponible y la productividad forrajera. Para medir la biomasa aérea de un recurso el productor puede utilizar tanto métodos directos como indirectos. El presente trabajo estudia estos métodos de estimación de biomasa aérea en una pastura mixta (trébol blanco, trébol rojo y cebadilla) de un establecimiento lechero ubicado en Suipacha, Pcia. de Buenos Aires (Argentina). Entre 2007 y 2008 tres observadores recorrieron un potrero cada tres semanas y estimaron la biomasa aérea presente en 12 unidades de muestreo de 0,09 m². Luego, se cortó al ras del suelo el material presente en cada unidad, se recolectó y se llevó al laboratorio para su procesamiento. Los métodos indirectos utilizados fueron la estimación visual, el pasturómetro y la regla graduada. Se generaron ecuaciones de calibración para cada método a distintas escalas temporales (Capítulo 2). Los resultados sugieren que los métodos indirectos son buenos estimadores de la biomasa aérea obtenida por cortes, tanto a escala anual como a escala estacional. En algunos casos, la precisión de la calibración del método estuvo afectada por la estación considerada. La validación de los modelos se realizó a dos niveles de análisis (Capítulo 3). La validación con una ecuación por estación sugiere que el método más preciso fue la estimación visual. En cambio, la validación con una ecuación para todo el período sugiere que todos los métodos fueron poco precisos. Algunos de los métodos estudiados en este trabajo, como la estimación visual y el pasturómetro, se presentan como promisorios. El uso de métodos sencillos, económicos y prácticos como estos permitiría mejorar el manejo y la eficiencia de los sistemas de producción de leche de base pastoril.

A global dataset of in situ particulate absorption spectra has been decomposed into component functions representing absorption by phytoplankton pigments and non-algal particles. The magnitudes of component Gaussian functions, used to represent absorption by individual or groups of pigments, are well correlated with pigment concentrations determined using High Performance Liquid Chromatography. We are able to predict the presence of chlorophylls a,ba,b, and cc, as well as two different groups of summed carotenoid pigments with percent errors between 30% and 57%. Existing methods of analysis of particulate absorption spectra measured in situ provide for only chlorophyll aa; the method presented here, using high spectral resolution particulate absorption, shows the ability to obtain the concentrations of additional pigments, allowing for more detailed studies of phytoplankton ecology than currently possible with in-situ spectroscopy.

Several lines of evidence indicate that altered expression of SEPT9 is seen in human neoplasia. In particular there is evidence of altered expression of the SEPT9_v4 isoform. The functional consequences of this remain unclear. We have studied the expression of wild-type- and GTP-binding mutants (G144V and S148N) of the SEPT9_v4 isoform in the MCF7 cell line as a model for its deregulation in neoplasia. We find that SEPT9_v4 expression induces dramatic actin cytoskeletal reorganization with the formation of processes around the cell periphery. Expression of the SEPT9_v4 isoform and a G144V mutant cause delocalization of endogenous SEPT9 from filamentous structures but the S148N mutant does not have this effect. In addition SEPT9_v4 isoform expression enhances cell motility and is associated with perturbation of directional movement. Expression of SEPT9_v4 GTP binding mutants also has potent effects on morphology and motility and causes loss of normal polarity, as judged by Golgi reorientation assays. The phenotypes induced by expression of the SEPT9_v4 isoform and the GTP mutants provide an insight into possible mechanisms of SEPT9_v4 function and suggest that the GTPase functions have both ras- and rab-like features. We propose a model in which overexpression of the SEPT9_v4 isoform in neoplasia is associated with perturbation of SEPT9 complexes, leading to phenotypes associated with neoplasia. Atheroma formation involves the movement of vascular smooth muscle cells (VSMC) into the subendothelial space. The aim of this study was to determine the involvement of PI3K and MAPK pathways and the importance of cross-talk between these pathways, in glucose-potentiated VSMC chemotaxis to serum factors. VSMC chemotaxis occurred in a serum gradient in 25 mmol/L glucose (but not in 5 mmol/L glucose) in association with increased phosphorylation (activation) of Akt and ERK1/2 in PI3K and MAPK pathways, respectively. Inhibitors of these pathways blocked chemotaxis, as did an mTOR inhibitor. VSMC expressed all class IA PI3K isoforms, but microinjection experiments demonstrated that only the p110beta isoform was involved in chemotaxis. ERK1/2 phosphorylation was reduced not only by MAPK pathway inhibitors but also by PI3K and mTOR inhibitors; when PI3K was inhibited, ERK phosphorylation could be induced by microinjected activated Akt, indicating important cross-talk between the PI3K and ERK1/2 pathways. Glucose-potentiated phosphorylation of molecules in the p38 and JNK MAPK pathways inhibited these pathways but did not affect chemotaxis. The statin, mevinolin, blocked chemotaxis through its effects on the MAPK pathway. Mevinolin-inhibited chemotaxis was restored by farnesylpyrophosphate but not by geranylgeranylpyrophosphate; in the absence of mevinolin, inhibition of farnesyltransferase reduced ERK phosphorylation and blocked chemotaxis, indicating a role for the Ras family of GTPases (MAPK pathway) under these conditions. In conclusion, glucose sensitizes VSMC to serum, inducing chemotaxis via pathways involving p110beta-PI3K, Akt, mTOR, and ERK1/2 MAPK. Cross-talk between the PI3K and MAPK pathways is necessary for VSMC chemotaxis under these conditions.
