World population is increasing at an alarming rate while food productivity is decreasing due to the effect of various abiotic stresses. Soil salinity is one of the most important abiotic stress and a limiting factor for worldwide plant production. In addition to its important effects on yield, salt stress affects numerous cellular activities, including cell wall composition, photosynthesis, protein synthesis, ions and organic solutes. Up to 20% of the irrigated arable land in arid and semiarid regions is already salt affected and is still expanding. Improving salt tolerant varieties is of major importance, and efforts should be focused on finding adaptive mechanisms which are involved in salinity tolerance. In this study, several spelt wheat (Triticum aestivum var. Spelta) genotypes and one cultivar of modern bread wheat were used to screen them for salt tolerance. Spelt is an old-European cereal crop currently attracting renewed interest as a food grain because it is said to be harder than wheat and requires less fertilizer. Spelt wheat is also becoming very attractive genetic source by plant breeders due to its wide adaptation ability to various stressful conditions such as soil salinity. In this study morphological parameters (e.g., leaf appearance; shoot elongation), dry matter production, mineral nutrients (especially Na and K), and activity of antioxidative enzymes were measured to select superior genotypes of spelt for salt tolerance. The results showed that Spelt genotype Sp41 is a salt sensitive genotype and genotypes Sp69, Sp96 and Sp912 are good candidates for salt tolerant genotypes.

Grapevine (Vitis vinifera) is one of most agro-economically important fruit crops worldwide, with a special relevance in Portugal where over 300 varieties are used for wine production. Due to global warming, temperature stress is currently a serious issue affecting crop production especially in temperate climates. Mobile genetic elements such as retrotransposons have been shown to be involved in environmental stress induced genetic and epigenetic modifications. In this study, sequences related to Grapevine Retrotransposon 1 (Gret1) were utilized to determine heat induced genomic and transcriptomic modifications in Touriga Nacional, a traditional Portuguese grapevine variety. For this purpose, growing canes were treated to 42 oC for four hours and leaf genomic DNA and RNA was utilized for various techniques to observe possible genomic alterations and variation in transcription levels of coding and non-coding sequences between non-treated plants and treated plants immediately after heat stress (HS-0 h) or after a 24 hour recovery period (HS-24 h). Heat stress was found to induce a significant decrease in Gret1 related sequences in HS-24 h leaves, indicating an effect of heat stress on genomic structure. In order to identify putative heat induced DNA modifications, genome wide approaches such as Amplified Fragment Length Polymorphism were utilized. This resulted in the identification of a polymorphic DNA fragment in HS-0 h and HS-24 h leaves whose sequence mapped to a genomic region flanking a house keeping gene (NADH) that is represented in multiple copies in the Vitis vinifera genome. Heat stress was also found to affect the transcript levels of various non-coding and gene coding sequences. Accordingly, quantitative real time PCR results established that Gret1 related sequences are up regulated immediately after heat stress whereas the level of transcript of genes involved in identification and repair of double strand breaks are significantly down regulated in HS-0 h plants. Taken together, the results of this work demonstrated heat stress affects both genomic integrity and transcription levels.

Organisms that thrive optimally at temperatures above 80°C are called hyperthermophiles. These prokaryotes have been isolated from a variety of hot environments, such as marine geothermal areas, hence they are usually slightly halophilic. Like other halophiles, marine hyperthermophiles have to cope with fluctuations in the salinity of the external medium and generally use low-molecular mass organic compounds to adjust cell turgor pressure. These compounds can accumulate to high levels without interfering with cell metabolism, thereby deserving the designation of compatible solutes. Curiously, the accumulation of compatible solutes also occurs in response to supraoptimal temperatures.(...)

The purpose of the project is to measure the impact of fiscal policy on the Portuguese GDP and how it may vary according to the state of the financial market. A Threshold VAR model is presented in which the two regimes are found using a financial stress index that divides the economy into a situation of financial stress and financial stability.

INTRODUCTION: The capacity to overcome the oxidative stress imposed by phagocytes seems to be critical for Candida species to cause invasive candidiasis. METHODS: To better characterize the oxidative stress response (OSR) of 8 clinically relevant Candida sp., glutathione, a vital component of the intracellular redox balance, was measured using the 5,5'-dithiobis-(2-nitrobenzoic acid (DTNB)-glutathione disulfide (GSSG) reductase reconversion method; the total antioxidant capacity (TAC) was measured using a modified method based on the decolorization of the 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulfonic) acid radical cation (ABTS*+). Both methods were used with cellular Candida sp. extracts treated or not with hydrogen peroxide (0.5 mM). RESULTS: Oxidative stress induced by hydrogen peroxide clearly reduced intracellular glutathione levels. This depletion was stronger in Candida albicans and the levels of glutathione in untreated cells were also higher in this species. The TAC demonstrated intra-specific variation. CONCLUSIONS: Glutathione levels did not correlate with the measured TAC values, despite this being the most important non-enzymatic intracellular antioxidant molecule. The results indicate that the isolated measurement of TAC does not give a clear picture of the ability of a given Candida sp. to respond to oxidative stress.

INTRODUCTION: This study evaluated the intracellular profile of interleukin-2 (IL-2), interleukin-4 (IL-4), interleukin-10 (IL-10) and interferon-γ (IFN-γ) in peripheral blood mononuclear cells (PBMCs) from leprosy patients based on oral infections presence to determine whether these coinfections could be associated with pro-inflammatory activity in leprosy. METHODS: Leprosy patients regardless of clinical form and specific leprosy treatment (n=38) were divided into two groups: Group I - leprosy patients with oral infections (n=19), and Group II - leprosy patients without oral infections (n=19). Non-leprosy patients presenting oral infections were assigned to the control Group (n=10). Intracellular IL-2, IL-4, IL-10 and IFN-γ production was evaluated by flow cytometry (FACS) before and 7 days after controlling the oral infection in the Group I, before and 7 days after dental prophylaxis in the Group II, and during oral infection process in control Group. RESULTS: Low percentages of CD3+ lymphocytes bearing IL-2, IL-10 and IFN-γ were observed in the Group I and Group II at baseline and 7 days after therapy or prophylaxis compared to controls. Group I showed reduced percentages of IL-4 at baseline and 7 days after therapy compared to controls, or at baseline of Group II, and the Group II showed reduced percentages of CD3+ cells bearing IL-4 compared to control. An increase of the percentages of CD3+cells bearing IL-4 was observed in the Group I after the oral infections treatment. CONCLUSIONS: The occurrence of oral infections favors the intracellular cytokines expression and, probably, the inflammatory reaction operating as a stimulatory signal triggering the leprosy reactions.

RESUMO - O tabagismo surge como a primeira causa evitável de doença, incapacidade e morte prematura em países desenvolvidos. Dentre os fatores influenciadores do comportamento face ao consumo de tabaco, a influência dos fatores de stress no local de trabalho foi alvo de diversos estudos com resultados mistos, revelando uma evidência empírica inconclusiva. Foi feito um estudo exploratório, transversal, que visou verificar a existência de associação entre algum dos fatores de stress no trabalho, nomeadamente 1) requisitos no trabalho, 2) autoridade decisória, 3) discriminação de tarefas, 4) condições de emprego, 5) apoio dos chefes e colegas, 6) número de horas de trabalho e 7) satisfação com o trabalho; com a motivação para a cessação tabágica. A amostra contou com 95 vendedores de bens de consumo alimentar, fumadores. Foi aplicado um questionário com variáveis sociodemográficas, variáveis de caracterização do comportamento face ao consumo de tabaco, o Teste de Richmond, variáveis de caracterização profissional e o Questionário Sobre Stress no Local de Trabalho. O Questionário Sobre o Stress no Local de Trabalho revelou uma validade insuficiente para a caracterização da maioria dos fatores de stress em estudo, manifestada por α de Cronbach muito baixos, pelo que a análise de associação apenas foi realizada entre os fatores de stress 1) apoio dos chefes e colegas, 2) número de horas de trabalho e 3) satisfação com o trabalho; e a motivação para a cessação tabágica. Para níveis de significância de 10%, o número de horas de trabalho apresentou uma associação positiva estatisticamente significativa com a motivação para a cessação tabágica, duplicando a probabilidade da sua ocorrência [OR = 2,429 (IC95%: 0,945 – 6,240)]. Em toda a análise de dados realizada foi prevalente a baixa motivação para a cessação tabágica dos vendedores, independentemente da variação dos fatores de stress relacionados com o trabalho.

A cross-sectional study of 120 subjects was performed with the purpose of evaluating stress hormones and emotional stress (anxiety) in outpatient and hospitalized subjects. The aims were to determine the degree of objective stress, as well as to correlate this finding with subjective findings, estimated using Beck's Anxiety Inventory.. METHOD: Three populations were investigated, namely outpatient clinical cases (Group I, n = 30), hospitalized clinical individuals (Group II, n = 30), and hospitalized surgical candidates (Group III, n = 30). Controls (Group IV, n = 30) were healthy volunteers who were health-care professionals and students. To avoid hormone interactions, only men were enrolled in all groups. All hospitalized subjects were tested on admission and before therapeutic interventions. Fasting epinephrine, norepinephrine, and cortisol were measured in the morning, and Beck's Anxiety Inventory was adminstered by a trained psychologist. RESULTS: The 3 patient groups displayed higher anxiety levels than the controls. Hormone concentrations did not present remarkable changes and did not correlate with subjective stress (anxiety). CONCLUSIONS: 1) Subjective disorders (as determined with Beck's Anxiety Inventory ) were a common finding in both outpatient and hospitalized populations, without differences between the various groups; 2) Objective stress (as determined by elevated hormone levels) was more difficult to confirm-findings rarely exceeded the reference range; 3) Correlation between the two variables could not be demonstrated; 4) Further studies are necessary to define stress quantification and interpretation in patient populations, especially in relationship with nutritional diagnosis and dietetic prescription.

O arroz (Oryza sativa L.) constitui a principal fonte de alimento para mais de metade da população mundial. O frio tem um grande impacto no desenvolvimento da planta de arroz com efeitos negativos ao nível da produtividade e economia mundial. Os mecanismos epigenéticos associados a alterações estruturais da cromatina estão envolvidos nos mecanismos de resposta das plantas a stresses abióticos. O foco do presente trabalho consistiu na compreensão da influência de fatores epigenéticos, nomeadamente de modificações de histonas, na regulação transcricional de um gene específico (OsDREB1B) envolvido na resposta de arroz ao stress de frio. O estudo da regulação epigenética deste gene envolveu a utilização de plantas de arroz com mutações em enzimas epigenéticas e drogas remodeladoras da cromatina. Neste trabalho mostramos que o gene OsDREB1B é induzido pelo stress de frio (4 °C) em todas os genótipos de arroz analisados, no entanto, a indução do gene em resposta ao stress de frio é maior em Nipponbare que em Dongjin. Nos genótipos que apresentam epimutações, nomeadamente o knockout de osdrm2 (DMT706) e de oshac704 (HAC704), o gene OsDREB1B é mais expresso que no respetivo genótipo selvagem (Dongjin), sendo que a indução da expressão em resposta ao frio foi maior em DMT706 do que em HAC704. A análise do padrão de marcas histónicas ao longo do promotor de OsDREB1B, através da imunoprecipitação da cromatina (ChIP) com anticorpos específicos, revelou que, em resposta ao frio (4ºC), Dongjin apresenta uma menor predominância da marca H3K9ac na região de -519 a -355 pares de bases (antes do ATG), enquanto que Nipponbare possui maior presença de H4K5ac na região de -280 a -121 pares de bases, sugerindo que a regulação epigenética de OsDREB1B seja influenciada pelo genótipo. No caso dos epi-mutantes (HAC704 e DMT706) registou-se um empobrecimento das marcas histónicas testadas sugerindo que a indução da expressão do OsDREB1B em reposta ao stress provocado pelo frio não depende necessariamente da presença de OsDRM2 nem de OsHAC704. Uma visão integrada da paisagem epigenética e do transcriptoma, perspetiva uma melhor compreensão dos mecanismos envolvidos na resposta da planta a condições ambientais adversas, abrindo novas linhas de trabalho no sentido de obter plantas mais tolerantes a fatores de stress.

In the investigation and diagnosis of damages to historical masonry structures, the state of stress of the masonry is an important characteristic that must be determined with as much accuracy as possible. Flat-jack testing is a traditional method used to determine the state of stress in historical masonry structures. However, when irregular masonry is tested the method can cause damage to the masonry units and the accuracy of the method is reduced. An enhanced technique, called tube-jack testing, is being developed at the University of Minho to reduce the damage caused during testing and improve the accuracy when used on irregular masonry. This method uses multiple cylindrical jacks inserted in a line of holes drilled in the mortar joints of the masonry, avoiding damage to the masonry units. Concurrently with the development of tube-jack testing, the effect of stress state on sonic testing is being studied. Sonic testing is often used to determine locations of voids and damage in masonry. The focus of these studies was to determine if the state of stress is influencing the sonic test results. In this paper the results of tube-jack testing and sonic testing on masonry walls, built for the purpose of this study in the laboratory, loaded in compression is presented. The tube-jack testing is used to estimate the state of stress in the masonry and the sonic test results are evaluated based on the effect of the applied load on the wall. Future testing and study are suggested for continued development of these test methods.

Stressed plants are generally more attacked by galling insects. In this study we investigated the relationship between population abundance and species richness of galling insects on the tree Alchornea castaneaefolia A. JUSS. (Euphorbiaceae), submited to stress induced by the hemiparasite Psittacanthus sp. (Loranthaceae) in the Amazon, Brazil. Branches of A. castaneaefolia attacked by the hemiparasite were more heavily infested by galling insects than non-attacked branches. The field observations partially corroborate the hypothesis that there would be an optimal level of host-plant stress for galling insect establishment.

Fish transport is one of the most stressful procedures in aquaculture facilities. The present work evaluated the stress response of matrinxã to transportation procedures, and the use of clove oil as an alternative to reduce the stress response to transport in matrinxã (Brycon cephalus). Clove oil solutions were tested in concentrations of 0, 1, 5 and 10 mg/L during matrinxã transportation in plastic bags, supplied with water and oxygen as the usual field procedures in Brazil. Clove oil reduced some of the physiological stress responses (plasma cortisol, glucose and ions) that we measured. The high energetic cost to matrinxã cope with the transport stress was clear by the decrease of liver glycogen after transport. Our results suggest that clove oil (5 mg/l) can mitigate the stress response in matrinxã subjected to transport.

Conformally flat spacetimes with an elastic stress energy tensor having diagonal trace-free anisotropic pressure are investigated using 1+3 formalism. The 1+3 Bianchi and Jacobi identities and Einstein field equations are written for a particular case with a conformal factor dependent on only one spatial coordinate. Solutions with non null anisotropic pressure are obtained.
