Since ancient times, the utilization of yeasts by the man has a great impact on the socio-economic development. After the advent of the technology of recombinant DNA, great advances have occurred due to the acquisition of strains of mutant yeasts in the field of applied research, and Saccharomyces cerevisiae has soon been outstanding as an interesting candidate for the expression of heterologous proteins of biotechnological interest. As the time goes by other alternative systems of expression have been shown because they have advantages over Saccharomyces cerevisiae. Among those new systems, Pichia pastoris is outstanding as methylotrophic yeast capable of growing in a culture medium containing methanol as the only source of carbon and energy. The induction of production of glycerol-3-phosphate dehydrogenase (GPD, NAD(+): oxido-redutase EC 1.1. 1.8) by Pichia pastoris was accomplished in the medium containing methanol. One of the most important key parameters in Pichia pastoris expression system is the methanol concentration. Bibliographic reviews on the Pichia pastoris production system have shown that the best culture conditions vary according to the strain used and/or kind of heterologous protein desired to be expressed. Therefore, we have sought to develop a system, involving expression of glycerol-3-phosphate dehydrogenase in the yeast Pichia pastoris, for generating sufficient quantities of the enzyme in order to asses its potential value for use in various food bioanalytical determination. Dehydrogenases have been widely used in the enzymatic assays of diverse composites of industrial interest, being enclosed among them glycerol and a number of important bioanalytical applications.

The acute, subchronic and chronic toxicities of 2,4- dichlorophenoxyacetic acid (2,4-D) were studied in rats. Animals were exposed acutely (600 mg/kg), subchronically (200 ppm for 30 d) and chronically (200 ppm for 180 d) to 2,4-D by the oral route. Clinical, laboratory and histopathological methods were used as indicators of toxicity. After acute exposure, the herbicide decreased locomotor activity and induced ataxia, sedation, muscular weakness (mainly of the hind quarters) and gasping for breath; increased aspartate aminotransferase (AST), alanine aminotransferase (ALT), lactate dehydrogenase (LDH), alkaline phosphatase (AP), amylase activities and creatinine levels; decreased total protein (TP) and glucose levels; and increased hematocrit values. Subchronic and chronic 2,4-D exposures did not induce overt clinical signs or symptoms of intoxication. However, subchronic herbicide exposure increased AST activity and albumin and hematocrit values, and chronic exposure increased AST, AP and LDH activities, decreased amylase and glucose levels, but did not change hematocrit values. Chromatographic analysis of the serum of chronically exposed rats showed the presence of the herbicide; the amount found (3.76 ± 1.16 mg/ml) suggested the absence of 2,4-D accumulation within the body. Although macroscopic or histopathological lesions were not observed in acutely, subchronically or chronically 2,4-D exposed rats, the laboratory data obtained suggest tissue injuries after dosing, since the results are considered early indicators of primarily hepatic and muscle tissue damage.

Pyrophosphate-dependent phosphofructokinase (PPi-PFK) has been detected in several types of plant cells, but the gene has not been reported in sugar cane. Using Citrux paradixi PPi-PFK gene (AF095520 and AF095521) sequences to search the sugar cane EST database, we have identified both the α and β subunits of this enzyme. The deduced amino acid sequences showed 76 and 80% similarity with the corresponding α and β subunits of C. paradisi. A high degree of similarity was also observed among the PFK β subunits when the alignment of the sugar cane sequences was compared to those of Ricinus communis and Solanum tuberosum, it appears that α and β are two distinct subunits; they were found at different concentrations in several sugar cane tissues. It remains to be determined if the different gene expression levels have some physiological importance and how they affect sucrose synthesis, export, and storage in vacuoles. A comparison between the amino acid sequences of β PFKs from a variety of organisms allowed us to identify the two critical Asp residues typical of this enzyme's activity site and the other binding sites; these residues are tightly conserved in all members of this protein family. Apparently, there are catalytic residues on the β subunit of the pyrophosphate-dependent enzyme.

The aim of this study was to analyze the morphoquantitative features of the nitrergic myenteric neurons from the body of the stomach of diabetic rats. The body of the stomach of five normoglicemic rats and of five diabetic rats were prepared as whole-mounts stained by the histochemical technique of NADPH-diaphorase. Decreased body weight and increased daily ingestion of water, fast glicemia and glycated hemoglobin were verified in diabetic animals. According to the data obtained, significant difference in the density of nitrergic neurons was not observed between the two groups, but the areas of the neuronal cell body profiles in the diabetic rats were significantly larger. Results showed that the streptozotocin that induced diabetes does not accelerate the death of the nitrergic neurons, but increases the expression of these cells.

The present study examines the effects of a hypercaloric diet on hepatic glucose metabolism of young rats, with and without monosodium glutamate (MSG) administration, and the association of these treatments with evaluating markers of oxidative stress. Male weaned Wistar rats (21 days old) from mothers fed with a hypercaloric diet or a normal diet, were divided into four groups (n=6): control (C) fed with control diet; (MSG) treated with MSG (4 mg/g) and control diet; (HD) fed with hypercaloric diet and (MSG-HD) treated with MSG and HD. Rats were sacrificed after the oral glucose tolerance test (OGTT), at 45 days of treatments. Serum was used for insulin determination. Glycogen, hexokinase(HK), glucose-6-phosphatase(G6PH), lipid hydroperoxide, superoxide dismutase(SOD) and glutathione peroxidase(GSH-Px) were determined in liver. HD rats showed hypoglycemia, hyperinsulinemia, and high hepatic glycogen, HK and decreased G6PH. MSG and MSG-HD had hyperinsulinemia, hyperglycemia, decreased HK and increased G6PH in hepatic tissue. These animals had impaired OGTT. HD, MSG and MSG-HD groups had increased lipid hydroperoxide and decreased SOD in hepatic tissue. Hypercaloric diet and monosodium glutamate administration induced alterations in metabolic rate of glucose utilization and decreased antioxidant defenses. Therefore, the hepatic glucose metabolic shifting induced by HD intake and MSG administration were associated with oxidative stress in hepatic tissue.

It is now commonly accepted that chronic inflammation associated with obesity during aging induces insulin resistance in the liver. In the present study, we investigated whether the improvement in insulin sensitivity and insulin signaling, mediated by acute exercise, could be associated with modulation of protein-tyrosine phosphatase 1B (PTP-1B) in the liver of old rats. Aging rats were subjected to swimming for two 1.5-h long bouts, separated by a 45 min rest period. Sixteen hours after the exercise, the rats were sacrificed and proteins from the insulin signaling pathway were analyzed by immunoblotting. Our results show that the fat mass was increased in old rats. The reduction in glucose disappearance rate (Kitt) observed in aged rats was restored 16 h after exercise. Aging increased the content of PTP-1B and attenuated insulin signaling in the liver of rats, a phenomenon that was reversed by exercise. Aging rats also increased the IRβ/PTP-1B and IRS-1/PTP-1B association in the liver when compared with young rats. Conversely, in the liver of exercised old rats, IRβ/PTP-1B and IRS-1/PTP-1B association was markedly decreased. Moreover, in the hepatic tissue of old rats, the insulin signalling was decreased and PEPCK and G6Pase levels were increased when compared with young rats. Interestingly, 16 h after acute exercise, the PEPCK and G6Pase protein level were decreased in the old exercised group. These results provide new insights into the mechanisms by which exercise restores insulin signalling in liver during aging. © 2013 Moura et al; licensee BioMed Central Ltd.

Breast cancer is the most common type of cancer among women worldwide. Research using breast cancer cell lines derived from primary tumors may provide valuable additional knowledge regarding this type of cancer. Therefore, the aim of this study was to investigate the phenotypic profiles of MACL-1 and MGSO-3, the only Brazilian breast cancer cell lines available for comparative studies. We evaluated the presence of hormone receptors, proliferation, differentiation and stem cell markers, using immunohistochemical staining of the primary tumor, cultured cells and xenografts implanted in immunodeficient mice. We also investigated the ability of the cell lines to form colonies and copy number alterations by array comparative genomic hybridization. Histopathological analysis showed that the invasive primary tumor from which the MACL-1 cell line was derived, was a luminal A subtype carcinoma, while the ductal carcinoma in situ (DCIS) that gave rise to the MGSO-3 cell line was a HER2 subtype tumor, both showing different proliferation levels. The cell lines and the tumor xenografts in mice preserved their high proliferative potential, but did not maintain the expression of the other markers assessed. This shift in expression may be due to the selection of an 'establishment' phenotype in vitro. Whole-genome DNA evaluation showed a large amount of copy number alterations (CNAs) in the two cell lines. These findings render MACL-1 and MGSO-3 the first characterized Brazilian breast cancer cell lines to be potentially used for comparative research. © 2013 Spandidos Publications Ltd. O objetivo desse trabalho foi analisar a ativação e o crescimento de folículos préantrais de Sapajus apella, submetidos a um sistema de cultivo in vitro em curto-prazo. Nesse sentido, dois experimentos foram realizados neste trabalho. Experimento I: exposição à fresco e à crioprotetores de biopsias de fragmentos do córtex ovariano. Ambos os fragmentos ovarianos foram submetidos à extração total de RNA e síntese de cDNA. Após a amplificação do cDNA por PCR em tempo-real (RT-PCR), os software GeNorm, bestkeeper e Normfinder foram usados para avaliar a estabilidade dos genes GAPDH (glyceraldehyde-2-phosphate dehydrogenase), HPRT1 (hypoxanthine phosphoribosyltransferase 1) e TBP (TATA-binding protein). Experimento II: o tecido do córtex ovariano de quatro fêmeas foi oletado e dividido em nove pedaços de 1 mm³. Um fragmento ovariano (grupo controle) foi imediatamente dividido em dois pedaços, os quais foram destinados para análise da viabilidade ou por RT-PCR. Os 8 fragmentos restantes foram individualmente cultivados in vitro em um meio constituído de TCM suplementado com 100 ng/mL EGF (T1), com adição de 10 μM de BME (T2), 100 ng/mL de BMP4 (T3), 25 IU de PMSG (T4), 10 μM de BME e 100 ng/mL de BMP4 (T5), 10 μM de BME, 25 IU de PMSG (T6), 100 ng/mL de BMP4, 25 IU de PMSG (T7) ou 10 μM de BME, 100 ng/mL de BMP4, 25 IU de PMSG (T8). Os resultados demonstraram que no tecido do córtex ovariano de S. apella, os genes HPRT1 e TBP foram os mais apropriados como genes de referência, podendo ser usados como parâmetro para normalizar dados em estudos futuros. Ao contrário, o GAPDH se apresentou como menos estável dos genes de referencia testados. Após o cultivo in vitro, todos os tratamentos alcançaram percentuais similares da viabilidade de folículos pré-antrais. O tecido ovariano cultivado na presença de GF+BME/BMP4/PMSG resultou no aumento da taxa de ativação e crescimento folicular, assim como no aumento da expressão de AMH, BMP15 e GDF9, genes conhecidos como indicadores específicos de desenvolvimento folicular. Dessa forma, o HPRT1 e TBP são os genes de referência mais estáveis, na exposição à crioprotetores, a fresco e no o cultivo de tecidos de córtex ovariano de S. apella. Os folículos pré-antrais são capazes de desenvolverem-se in vitro quando cultivados em meio suplementado com PMSG, BME e BMP4. A viabilidade folicular, entretanto, permaneceu independentemente do meio de cultivo in vitro e o uso de fatores de crescimento, como marcadores de desenvolvimento folicular, foi crucial para identificar o melhor meio de cultivo in vitro.
