Mannose-binding lectin (MBL) is an innate immune system pattern recognition molecule that kills a wide range of pathogens via the lectin complement pathway. MBL deficiency is associated with severe infection but the best measure of this deficiency is undecided. We investigated the influence of MBL functional deficiency on the development of sepsis in 195 adult patients, 166 of whom had bloodstream infection and 35 had pneumonia. Results were compared with 236 blood donor controls. MBL function (C4b deposition) and levels were measured by enzyme-linked immunosorbent assay. Using receiver-operator characteristics of MBL function in healthy controls, we identified a level of < 0.2 U mu L-1 as a highly discriminative marker of low MBL2 genotypes. Median MBL function was lower in sepsis patients (0.18 U mu L-1) than in controls (0.48 U mu L-1, P < 0.001). MBL functional deficiency was more common in sepsis patients than controls (P < 0.001). MBL functional deficient patients had significantly higher sequential organ failure assessment (SOFA) scores and higher MBL function and levels were found in patients with SOFA scores predictive of good outcome. Deficiency of MBL function appears to be associated with bloodstream infection and the development of septic shock. O objetivo deste trabalho foi analisar a expressão dos torques dentários em pacientes tratados com aparelhos ortodônticos fixos com bráquetes autoligados, utilizando tomografias computadorizadas. Para este estudo, foi selecionada uma amostra clínica de 10 pacientes, seguindo como critérios de inclusão, indivíduos que apresentavam dentição permanente e todos os dentes presentes, com apinhamento superior ou igual a 4 mm, tratados sem extração. Todos os pacientes foram tratados na clínica da pós-graduação em Ortodontia da Universidade Metodista de São Paulo, utilizando-se bráquetes autoligados Damon 2 ORMCO na prescrição padrão. Foram realizadas medições das inclinações dos dentes anteriores, de canino a canino, superiores e inferiores, realizadas por meio de imagens tomográficas obtidas em um tomógrafo computadorizado volumétrico NewTom, em seu modelo DVT-9000 (NIM Verona - Itália), obtidas em dois tempos: antes do início do tratamento ortodôntico (denominado de T1) e depois do tratamento ortodôntico, após a inserção do último fio de nivelamento, de calibre 0,019 x 0,025 de aço inoxidável(denominado de T2). Para auxílio destas mensurações, foi utilizado o software QR-DVT 9000 e após análise dos resultados foram aplicados testes estatíscos (testes "t" pareado e Dalberg) e observou-se que as inclinações dos dentes do segmento anterior aumentaram, principalmente, nos caninos e incisivos laterais superiores, incisivos centrais e laterais inferiores. Os dentes apresentaram valores de inclinação diferentes da prescrição, tanto no início quanto no final do tratamento, denotando a incapacidade do fio 0,019 x 0,025 de aço inoxidável em reproduzir os torques indicados na prescrição padrão utilizada neste presente estudo.(AU)

Este trabalho teve como objetivo principal avaliar as inclinações dentárias e o perfil facial de pacientes tratados ortodonticamente com braquete autoligado Damon 2, prescrição padrão. Para este estudo, foi selecionada uma amostra de 18 indivíduos, sendo 12 do sexo masculino e 6 do feminino, com idades que variaram de 12 a 20 anos (idade média de 15 anos) e que apresentavam discrepância de modelo negativa (de 4 mm a 15 mm). Como critério de inclusão, os pacientes deveriam apresentar dentição permanente completa até os segundos molares, má oclusão de Classe I de Angle, telerradiografias em norma lateral do início e final do tratamento ortodôntico e modelos de estudo em gesso dos arcos dentários superiores e inferiores do início e final do tratamento ortodôntico. As medições das inclinações ântero-posteriores dos incisivos centrais superiores e incisivos centrais inferiores e alterações do perfil facial foram realizadas por meio das telerradiografias em norma lateral. As distâncias transversais das regiões de cúspide e cervical inter-caninos, primeiros e segundos pré-molares e primeiros molares, foram obtidas medindo-se os modelos de gesso. Todas as mensurações foram realizadas nas telerradiografias e modelos de gesso obtidos antes do início do tratamento ortodôntico (T1) e no final do tratamento (T2). Após a coleta de todos os dados aplicou-se o teste t de Student para comparar as medidas dos dois tempos avaliados neste estudo. Verificou-se diferença estatisticamente significante (p<0,05) na inclinação dos incisivos centrais inferiores e nas dimensões transversais inter-cúspides e inter-cervicais dos primeiros e segundos pré-molares e primeiros molares na maxila. Na mandíbula todas as distâncias transversais (inter cúspides e cervicais) de caninos, primeiros e segundos pré-molares e primeiros molares apresentaram significância estatística (p<0,05), exceto a distância inter-cervical dos caninos inferiores. Concluímos que o tratamento ortodôntico sem extrações de pacientes com má oclusão de Classe I realizados com os braquetes Damon 2, prescrição padrão, não promoveu alteração na inclinação vestíbulo-lingual dos incisivos centrais superiores, aumentou a inclinação vestibular dos incisivos centrais inferiores, não alterou o perfil facial e aumentou de modo significativo a dimensão transversal da maxila e da mandíbula por meio de inclinação vestibular de coroa.(AU)

O objetivo do presente estudo consistiu em comparar in vitro o atrito produzido por braquetes estéticos, convencionais e autoligados quando inseridos fios retangulares de aço inoxidável de diferentes dimensões, nas angulações de zero e três graus entre os braquetes. Foram utilizados 120 braquetes de 6 marcas comerciais (Gemini, Clarity convencional, Transcend, Inspire, Clarity autoligado, Damon autoligado), sendo 20 braquetes de cada marca comercial. Para os ensaios laboratoriais, foram colados dois pares de braquetes de cada marca comercial em uma placa metálica, com uma angulação de zero e três graus entre os braquetes. Foram empregados os fios retangulares de aço inoxidável 0,017 x 0,025 , 0,019 x 0,025 e 0,021 x0,025 em uma máquina de ensaios universal Instron . Para a comparação entre os braquetes nos diferentes fios e angulações, foi utilizada a análise de variância e o teste de Tukey (p<0,05). Os resultados demonstraram que na angulação de zero grau, os braquetes autoligados apresentaram menor atrito em relação aos convencionais em todos os fios avaliados, sendo que, o braquete Clarity autoligado apresentou menor atrito estatisticamente significante que o Damon, exceto no fio 0,021 x 0,025 , em que ambos os braquetes autoligados apresentaram atrito estatisticamente semelhante. O braquete Inspire demonstrou o maior atrito em relação aos demais braquetes nos fios 0,019 x0,025 e 0,021 x0,025 . Na angulação de três graus, observou-se resultados semelhantes aos braquetes posicionados sem angulação comparando os braquetes autoligados em relação aos convencionais. Contudo, verificou-se que o atrito entre os braquetes convencionais ocorreu em ordem crescente sendo braquetes Gemini, Clarity convencional, Transcend e Inspire. Pôde-se concluir que a angulação entre os braquetes aumenta consideravelmente o atrito. Além disso, em ambas as angulações, os braquetes autoligados demonstraram menor atrito em relação aos convencionais, sendo que o braquete Clarity autoligado promoveu menor atrito que o Damon estético, exceto no fio 0,021 x 0,025 .(AU)

This chapter introduces activity theory as an approach for studying strategy as practice. Activity theory conceptualizes the ongoing construction of activity as a product of activity systems, comprising the actor, the community with which that actor interacts and those symbolic and material tools that mediate between actors, their community and their pursuit of activity. The focus on the mediating role of tools and cultural artefacts in human activity seems especially promising for advancing the strategy-as-practice agenda, for example as a theoretical resource for the growing interest in sociomateriality and the role of tools and artefacts in (strategy) practice (for example, Balogun et al. 2014; Lanzara 2009; Nicolini 2009; Spee and Jarzabkowski 2009; Stetsenko 2005). Despite its potential, in a recent review Vaara and Whittington (2012) identified only three strategy-as-practice articles explicitly applying an activity theory lens. In the wider area of practice-based studies in organizations, activity theory has been slightly more popular (for example, Blackler 1993; 1995; Blackler, Crump and McDonald 2000; Engeström, Kerosuo and Kajamaa 2007; Groleau 2006; Holt 2008; Miettinen and Virkkunen 2005). It still lags behind its potential, however, primarily because of its origins as a social psychology theory developed in Russia with little initial recognition outside the Russian context, particularly in the area of strategy and organization theory, until recently (Miettinen, Samra-Fredericks and Yanow 2009). This chapter explores activity theory as a resource for studying strategy as practice as it is socially accomplished by individuals in interaction with their wider social group and the artefacts of interaction. In particular, activity theory’s focus on actors as social individuals provides a conceptual basis for studying the core question in strategy-as-practice research: what strategy practitioners do. The chapter is structured in three parts. First, an overview of activity theory is provided. Second, activity theory as a practice-based approach to studying organizational action is introduced and an activity system conceptual framework is developed. Third, the elements of the activity system are explained in more detail and explicitly linked to each of the core SAP concepts: practitioners, practices and praxis. In doing so, links are made to existing strategy-as-practice research, with brief empirical examples of topics that might be addressed using activity theory. Throughout the chapter, we introduce key authors in the development of activity theory and its use in management and adjacent disciplinary fields, as further resources for those wishing to make greater use of activity theory.

Aboveground net primary production (ANPP) by the dominant macrophyte and plant community composition are related to the changing hydrologic environment and to salinity in the southern Everglades, FL, USA. We present a new non-destructive ANPP technique that is applicable to any continuously growing herbaceous system. Data from 16 sites, collected from 1998 to 2004, were used to investigate how hydrology and salinity controlled sawgrass (Cladium jamaicense Crantz.) ANPP. Sawgrass live biomass showed little seasonal variation and annual means ranged from 89 to 639 gdw m)2. Mortality rates were 20–35% of live biomass per 2 month sampling interval, for biomass turnover rates of 1.3–2.5 per year. Production by C. jamaicense was manifest primarily as biomass turnover, not as biomass accumulation. Rates typically ranged from 300 to 750 gdw m)2 year)1, but exceeded 1000 gdw m)2 year)1 at one site and were as high as 750 gdw m)2 year)1 at estuarine ecotone sites. Production was negatively related to mean annual water depth, hydroperiod, and to a variable combining the two (depth-days). As water depths and hydroperiods increased in our southern Everglades study area, sawgrass ANPP declined. Because a primary restoration goal is to increase water depths and hydroperiods for some regions of the Everglades, we investigated how the plant community responded to this decline in sawgrass ANPP. Spikerush (Eleocharis sp.) was the next most prominent component of this community at our sites, and 39% of the variability in sawgrass ANPP was explained by a negative relationship with mean annual water depth, hydroperiod, and Eleocharis sp. density the following year. Sawgrass ANPP at estuarine ecotone sites responded negatively to salinity, and rates of production were slow to recover after high salinity years. Our results suggest that ecologists, managers, and the public should not necessarily interpret a decline in sawgrass that may result from hydrologic restoration as a negative phenomenon.

Everglades periphyton mats are tightly-coupled autotrophic (algae and cyanobacteria) and heterotrophic (eubacteria, fungi and microinvertebrates) microbial assemblages. We investigated the effect of water column total phosphorus and nitrogen concentrations, water depth and hydroperiod on periphyton of net production, respiration, nutrient content, and biomass. Our study sites were located along four transects that extended southward with freshwater sheetflow through sawgrass-dominated marsh. The water source for two of the transects were canal-driven and anchored at canal inputs. The two other transects were rain-driven (ombrotrophic) and began in sawgrass-dominated marsh. Periphyton dynamics were examined for upstream and downstream effects within and across the four transects. Although all study sites were characterized as short hydroperiod and phosphorus-limited oligotrophic, they represent gradients of hydrologic regime, water source and water quality of the southern Everglades. Average periphyton net production of 1.08 mg C AFDW−1 h−1 and periphyton whole system respiration of 0.38 mg C AFDW−1 h−1 rates were net autotrophic. Biomass was generally highest at ombrotrophic sites and sites downstream of canal inputs. Mean biomass over all our study sites was high, 1517.30 g AFDW m−2. Periphyton was phosphorus-limited. Average periphyton total phosphorus content was 137.15 μg P g−1 and average periphyton total N:P ratio was 192:1. Periphyton N:P was a sensitive indicator of water source. Even at extremely low mean water total phosphorus concentrations ( ≤ 0.21 μmol l−1), we found canal source effects on periphyton dynamics at sites adjacent to canal inputs, but not downstream of inflows. These canal source effects were most pronounced at the onset of wet season with initial rewetting. Spatial and temporal variability in periphyton dynamics could not solely be ascribed to water quality, but was often associated with both hydrology and water source.
