Las masas forestales tienen una importancia colosal para nuestra sociedad y el conjunto de la biosfera. Estudios recientes a escala mundial indican que la sequía es el factor abiótico que más afecta a su crecimiento y supervivencia, seguida por las temperaturas extremas y la salinidad. Aunque comprender los mecanismos con que las especies arbóreas toleran estas formas de estrés tiene un interés aplicado evidente, dichos mecanismos se han estudiado mucho más en especies herbáceas modelo o de interés agronómico. Existen sin embargo diferencias notables entre ellas, como se demuestra en esta tesis y en otros trabajos recientes. Nuestro estudio se centra concretamente en la respuesta molecular del chopo –el sistema modelo forestal más desarrollado– al estrés abiótico, con particular énfasis en la sequía. Utilizando una estrategia proteómica y tratamientos controlados, hemos identificado componentes mayoritarios de dicha respuesta. Su participación en la misma se ha validado mediante análisis transcripcionales detallados utilizando tecnología qRT-PCR (PCR cuantitativa en tiempo real). Hemos identificado proteínas cuyo nexo funcional con mecanismos de tolerancia ya era conocido, como chaperonas moleculares sHSP o enzimas que atenúan el estrés oxidativo, pero también proteínas cuya relación funcional con el estrés es menos clara o incluso novedosa, como polifenol oxidasas (PPO), deshidrogenasas/reductasas de cadena corta (SDR), o bicupinas (BIC), entre otras. El cuerpo central de la tesis consiste en la caracterización detallada de una PPO inusual, cuya inducción por estrés hídrico se describe por vez primera. Estas enzimas están ampliamente distribuidas en plantas, si bien su número es muy variable de unas especies a otras. Algunas, como nogal, tienen un único gen, mientras que Arabidopsis no tiene ninguno. En la última versión del genoma de chopo hemos identificado un total de 12 miembros bona fide, corrigiendo trabajos previos, y hemos caracterizado su expresión individual ante diferentes situaciones de estrés controlado y tratamientos hormonales. La isoforma antedicha es el único miembro de la familia que responde claramente a la deshidratación. También responde a salinidad y a la mayor parte de tratamientos hormonales ensayados, pero no a daño mecánico o tratamientos con metil jasmonato. Esto la diferencia de enzimas homólogas presentes en otras especies de plantas, que se han relacionado experimentalmente con estrés biótico. Los patrones de acumulación de transcritos en árboles adultos son compatibles con un papel protector frente a la sequía. La integración de nuestros estudios funcionales y filogenéticos sugiere que la familia ha sufrido un proceso reciente de diversificación y neofuncionalización, siendo la protección frente a deshidratación su papel primigenio. Aunque se conoce la actividad bioquímica in vitro de este tipo de enzimas, sus sustratos naturales son esencialmente una incógnita. Mediante expresión heteróloga en Escherichia coli BL21(DE3) hemos detectado que la enzima de chopo es capaz de oxidar L-DOPA a dopaquinona, siendo menos activa frente a otros sustratos. Por otra parte, hemos demostrado su localización cloroplástica mediante transformación transitoria de protoplastos con fusiones a la proteína fluorescente YFP. Mediante la obtención de plantas transgénicas de A. thaliana hemos demostrado que la enzima de chopo aumenta considerablemente la tolerancia in vivo frente a la deshidratación y al estrés salino. El análisis fenotípico detallado de las líneas transgénicas, combinando múltiples metodologías, nos ha permitido sustanciar que la tolerancia tiene una base compleja. Esta incluye una mayor protección del sistema fotosintético, una capacidad antioxidante muy incrementada y la acumulación de solutos osmoprotectores como la prolina. Los análisis metabolómicos nos han permitido asociar la expresión de la proteína a la síntesis de un flavano no descrito previamente en A. thaliana, vinculando la enzima de chopo con la síntesis de fenilpropanoides. También hemos observado alteraciones en los niveles hormonales que podrían subyacer a efectos pleiotrópicos con interés aplicado, como un aumento consistente del tamaño de la planta o el acortamiento del ciclo de crecimiento. Además de aportar datos novedosos sobre la funcionalidad in vivo de esta familia de oxidasas, los resultados de esta tesis demuestran que los árboles son sistemas de estudio interesantes para caracterizar nuevas estrategias de tolerancia al estrés abiótico con potencial aplicado. ABSTRACT Forests masses have an extraordinary importance for our society and the biosphere. Recent worldwide studies indicate that drought is the abiotic factor that affects more their growing and survival, followed by extreme temperatures and salinity. The understanding of how the arboreal species tolerate the stress has an evident practical interest, but their mechanisms have been studied much more in herbaceous species or with agronomic interest. However, considerable differences exist between them, as this thesis and recent studies show. Our study is focused on the molecular response of the poplar –the more developed forestry model system- to abiotic stress, specifically focused in the drought. Using a proteomic strategy and controlled treatments, we have identified main components in such response. Its participation has been validated through transcriptional analysis using qRT-PCR technology. We have identified proteins whose functional connection with tolerance mechanisms were already known, as molecular chaperones sHSP or enzymes that attenuate the oxidative stress, but also some proteins whose functional relationship with the stress is less clear or even novel, as polifenol oxidases (PPO), short chain deshidrogenases/reductases (SDR), or bicupines (BIC), among others. The central body of the thesis consists of the detailed characterization of an unsual PPO, whose induction due to drought stress is first described. These enzymes are thoroughly distributed in plants, but their number of members is very variable among species. Some of them, as the walnut tree, have a single gene, while Arabidopsis has none. We have identified a total of 12 members in the last version of the poplar genome, correcting previous works, and have characterized their individual expression against different situations of controlled stress and hormone treatments. The aforementioned isoform is the only member of the family that responds clearly to the drought. It also reacts to salinity and the majority of hormonal treatments tested, but it does not respond to mechanical damage or treatments with methyl jasmonate. This is the difference with homologue enzymes present in other plant species, which have been related experimentally with abiotic stress. The accumulation patterns of transcripts in adult trees are compatible with a protector role against drought. The integration of our functional and phylogenetic studies suggests that the family has suffered a recent process of diversification and neofunctionalization, being the protection against drought their original role. Although the in vitro biochemistry activity of this kind of enzymes is already known, their natural substracts are essentially a mystery. By means of heterologous expression of Escherichia coli BL21(DE3) we have detected that the enzyme of poplar is able to oxidize L-DOPA to dopaquinone, being less active against other substrates. Additionally, we have proven its chloroplastic location with transitory transformation of protoplasts with YFP protein fusion. By means of getting transgenic plants of A. thaliana, we have demonstrated that the poplar enzyme increases notably the in vivo tolerance against the drought and salinity stresses. The phenotypic analysis of the transgenic lines, and the use of multiple methodologies, allowed us to test the complexity of the tolerance. This includes a major protection of the photosynthetic system, a very increased antioxidant capacity and the accumulation of osmoprotectant solutes as the proline. The metabolic analysis has allowed to associate the protein expression with the synthesis of a Flavan non described previously in A. thalaiana, linking the enzyme of poplar with the synthesis of phenylpropanoids. We have observed alterations in the hormonal levels that could underlie pleiotropic effects with applied interest, as a consistent increase of the size of the plant and the reduction of the growth cycle. The results of this thesis, in addition to provide novel data about the in vivo functionality of the oxidase family, demonstrate that the trees are interesting systems of study to characterize new strategies of tolerance against abiotic stress with applied potential.

El bosque tropical de montaña, es considerado zona de mega diversidad y de alto grado de endemismo, por las diferentes zonas ecológicas que presenta. Durante las últimas décadas estos bosques han recibido mayor atención por parte de investigadores, sin embargo, aún existe poca investigación en determinar cuáles son las respuestas de los bosques a los cambios ambientales a los que son sometidos. Estos bosques están sufriendo serias amenazas como pérdida de cobertura vegetal y cambios en los ciclos de nutrientes. El trabajo se dividió en cuatro objetivos específicos. i) Caracterización y análisis de patrones altitudinales de la riqueza de especies en el bosque tropical de montaña en el sur del Ecuador; con el fin de conocer cómo varía la diversidad de especies riqueza lo largo de un gradiente altitudinal. ii) Conocer los patrones espaciales del crecimiento en tres remanentes boscosos de un bosque tropical de montaña para determinar cómo la vecindad y la semejanza funcional de ésta influyen en el crecimiento forestal. iii) Conocer los efectos de la fertilización en el crecimiento diamétrico de especies arbóreas, en el bosque tropical de montaña; se analizó cómo reaccionan los árboles a la adición de nutrientes N y P en tres tipos de bosque. iv) Saber la respuesta de la comunidad de árboles a la adición de nutrientes en el bosque montano andino; este objetivo se basó con el supuesto de la deficiencia de tres tipos de nutrientes N, P y Ca, en esta formación boscosa y cómo reaccionan los árboles a la adición de nutrientes. El presente trabajo se llevó a cabo, en el bosque tropical de montaña que se encuentra localizada en la parte adyacente del Parque Nacional Podocarpus (PNP) en la cordillera del Consuelo, forma parte de la cadena oriental de los Andes del sur del Ecuador El trabajo de desarrollo entre los años 2008 y 2014. Para abordar el primer objetivo se establecieron 54 parcelas ubicadas aleatoriamente a lo largo de un gradiente altitudinal (3 niveles de altitud) y se e midieron e identificaron todos los individuos mayores a 5cm de DAP. Se construyó una filogenia con Phylocom y se calcularon diferentes componentes de diversidad para cada parcela ( riqueza taxonómica, diversidad filogenética y edad media de las especies). Ajustando modelos lineares se contrastó el efecto de la altitud sobre dichos componentes y se vio que la riqueza taxonómica y la edad media de las especies aumentaron con la altitud, en sentido contrario a las predicciones de la "hipótesis del conservadurismo tropical" (Tropical Conservatism Hypothesis). Para abordar el segundo objetivo se realizó una remedición de todos los árboles cartografiados en tres parcelas permanentes de alrededor de 5000 m2 cada una, representativas de tres estados diferentes de la sucesión del bosque montano. A partir de las coordenadas y de los datos de registrados, y empleando diferentes funciones de correlación de marca se analizó la distribución espacial del tamaño y del crecimiento relativo y del tamaño. Se constató que mientras que el tamaño de los árboles presentó una correlación espacial negativa, el crecimiento presentó correlación espacial positiva, en ambos casos a distancias cortas. El rango y la magnitud de ambas correlaciones aumentaron al avanzar la sucesión. La distribución espacial del crecimiento mostró una correlación negativa con la distribución espacial de tamaños. Por otro lado, la distribución espacial del crecimiento mostró una correlación negativa para árboles semejantes funcionalmente y positiva cuando se calculó entre árboles con diferente estrategia funcional. En conjunto, los resultados obtenidos señalan un aumento de la importancia de procesos competitivos y una mayor estructuración espacial del crecimiento y de la distribución de tamaños al avanzar la sucesión. Para el tercer y cuarto objetivo se instalaron 52 parcelas distribuidas en bloques donde se fertilizaron dos veces al año durante 6,4 años, se identificaron todos los individuos mayores a 10 cm de DAP, y se midió el crecimiento diamétrico durante estos años Con la adición de nutrientes realizada a los diferentes tipos de bosque en la gradiente altitudinal, encontramos que el efecto sobre el crecimiento diamétrico en la comunidad varia con el rango altitudinal, y el tipo de nutriente, analizando a nivel de las especies, en la mayoría de los casos las especies comunes no tuvieron cambios significativos a la adición de nutrientes. Los resultados de este estudio aportan nuevas evidencias para el entendimiento de la diversidad, estructura y dinámica de los bosques tropicales de montaña. ABSTRACT The montane tropical forest is considered a megadiverse habitat that harbor an enormous degree of endemism. This is mainly due to the high degree of environmental heterogeneity found and the presence of different well defined ecological areas. These forests have received more attention during the last decades, however, the information regarding the responses of these forests to environmental change, is still scarce. These forests are seriously endangered and are suffering serious threats, such as loss of vegetative cover, changes in the nutrient cycles. The work was divided in four specific objectives: i) Characterization and analysis of the species richness altitudinal patterns in the montane tropical forest of south Ecuador. Specifically, how species diversity changes along altitudinal gradients. ii) Exploring the spatial patterns of tree growth in three remnants of a montane tropical forest, and analyze how tree neighborhood and functional similarity among trees influence tree growth. Tropical Conservatism Hypothesis iii) Understanding the effects of fertilization in arboreal species growth (increase in diameter) of the montane tropical forest. Specifically we studied the effects of P and N addition on three different forests across an altitudinal gradient. iv) Know the response of the community of trees to the addition of nutrients in the Andean montane forest; this objective was based on the supposition of deficiency of three types of nutrients: P, N and Ca in this forest all formation and how the trees react to the addition of these nutrients. The present work was carried out in the montane tropical forest located in Bombuscaro, San Francisco and Cajanuma close to Podocarpus National Park (PNP) on Consuelo mountain range (Andean oriental range) at South of Ecuador. Field work was carried out during 2008 and 2014. To address the first objective, we randomly placed 54 plots along an altitudinal gradient. In these plots, every individual larger than 5 cm of DBH was measured and identified. A phylogeny was build with Phylocom and different diversity components (taxonomic richness, phylogenetic diversity and average species age) were computed for each plot. Linear models were used to test the effects of altitude on the diversity components. Our results showed that, contrary to the Tropical Conservatism Hypothesis, both taxonomic richness and average species age increased with altitude. To address our second objective, all mapped trees in three successional permanent plots (around ~5000 m2 each) were re-measured. Using different mark correlation functions, we analyzed the spatial distribution of tree-size and tree relative growth rate. Whereas tree size showed negative spatial correlation at fine spatial scales, relative growth rate showed positive correlation at the same scales. The range and magnitude of those correlations increased along successional stage. The spatial distribution of the relative growth rate was negatively correlated with the spatial distribution of tree sizes. Additionally, we found that the spatial correlation of the relative growth rate was negative for functionally similar trees and positive when computed for functionally different trees. In synthesis, our results point to an increase of competitive processes and strong spatial structure of relative growth rate and tree size along succession. For the third and fourth objectives, 52 plots were placed in a block design and were fertilized twice a year for 6,4 years. In these plots all the individuals with DBH > 10 cm were identified, and the diametrical growth was measured during these years. The nutrient addition at the three different altitude forests, revealed that the effect on the diametrical growth in the community varied with the altitudinal range. When analyzed at species level, the addition of nutrients was no significant in most cases. These results represent new evidences that will improved our understanding of diversity patterns and structure, and the dynamics of tropical montane forests.
Phalangeal curvature and positional behavior in extinct sloth lemurs (Primates, Palaeopropithecidae)

Recent paleontological discoveries in Madagascar document the existence of a diverse clade of palaeopropithecids or “sloth lemurs”: Mesopropithecus (three species), Babakotia (one species), Palaeopropithecus (three species), and Archaeoindris (one species). This mini-radiation of now extinct (“subfossil”) lemurs is most closely related to the living indrids (Indri, Propithecus, and Avahi). Whereas the extant indrids are known for their leaping acrobatics, the palaeopropithecids (except perhaps for the poorly known giant Archaeoindris) exhibit numerous skeletal design features for antipronograde or suspensory positional behaviors (e.g., high intermembral indices and mobile joints). Here we analyze the curvature of the proximal phalanges of the hands and feet. Computed as the included angle (θ), phalangeal curvature develops in response to mechanical use and is known to be correlated in primates with hand and foot function in different habitats; terrestrial species have straighter phalanges than their arboreal counterparts, and highly suspensory forms such as the orangutan possess the most curved phalanges. Sloth lemurs as a group are characterized by very curved proximal phalanges, exceeding those seen in spider monkeys and siamangs, and approaching that of orangutans. Indrids have curvatures roughly half that of sloth lemurs, and the more terrestrial, subfossil Archaeolemur possesses the least curved phalanges of all the indroids. Taken together with many other derived aspects of their postcranial anatomy, phalangeal curvature indicates that the sloth lemurs are one of the most suspensory clades of mammals ever to evolve.

Guanylyl cyclase C (GCC) has been detected only in intestinal mucosa and colon carcinoma cells of placental mammals. However, this receptor has been identified in several tissues in marsupials, and its expression has been suggested in tissues other than intestine in placental mammals. Selective expression of GCC by colorectal tumor cells in extraintestinal tissues would permit this receptor to be employed as a selective marker for metastatic disease. Thus, expression of GCC was examined in human tissues and tumors, correlating receptor function with detection by PCR. GCC was detected by ligand binding and catalytic activation in normal intestine and primary and metastatic colorectal tumors, but not in extraintestinal tissues or tumors. Similarly, PCR yielded GCC-specific amplification products with specimens from normal intestine and primary and metastatic colorectal tumors, but not from extraintestinal tissues or tumors. Northern blot analysis employing GCC-specific probes revealed an ≈4-kb transcript, corresponding to recombinant GCC, in normal intestine and primary and metastatic colorectal tumors, but not in extraintestinal tissues. Thus, GCC is selectively expressed in intestine and colorectal tumors in humans and appears to be a relatively specific marker for metastatic cancer cells in normal tissues. Indeed, PCR of GCC detected tumor cells in blood from some patients with Dukes B colorectal cancer and all patients examined with Dukes C and D colorectal cancer, but not in that from normal subjects or patients with Dukes A colon carcinoma or other nonmalignant intestinal pathologies.

Mutations in the ATRX gene on the human X chromosome cause X-linked α-thalassemia and mental retardation. XY patients with deletions or mutations in this gene display varying degrees of sex reversal, implicating ATRX in the development of the human testis. To explore further the role of ATRX in mammalian sex differentiation, the homologous gene was cloned and characterized in a marsupial. Surprisingly, active homologues of ATRX were detected on the marsupial Y as well as the X chromosome. The Y-borne copy (ATRY) displays testis-specific expression. This, as well as the sex reversal of ATRX patients, suggests that ATRY is involved in testis development in marsupials and may represent an ancestral testis-determining mechanism that predated the evolution of SRY as the primary mammalian male sex-determining gene. There is no evidence for a Y-borne ATRX homologue in mouse or human, implying that this gene has been lost in eutherians and its role supplanted by the evolution of SRY from SOX3 as the dominant determiner of male differentiation.

The subclass Theria of Mammalia includes marsupials (infraclass Metatheria) and placentals (infraclass Eutheria). Within each group, interordinal relationships remain unclear. One limitation of many studies is incomplete ordinal representation. Here, we analyze DNA sequences for part of exon 1 of the interphotoreceptor retinoid binding protein gene, including 10 that are newly reported, for representatives of all therian orders. Among placentals, the most robust clades are Cetartiodactyla, Paenungulata, and an expanded African clade that includes paenungulates, tubulidentates, and macroscelideans. Anagalida, Archonta, Altungulata, Hyracoidea + Perissodactyla, Ungulata, and the “flying primate” hypothesis are rejected by statistical tests. Among marsupials, the most robust clade includes all orders except Didelphimorphia. The phylogenetic placement of the monito del monte and the marsupial mole remains unclear. However, the marsupial mole sequence contains three frameshift indels and numerous stop codons in all three reading frames. Given that the interphotoreceptor retinoid binding protein gene is a single-copy gene that functions in the visual cycle and that the marsupial mole is blind with degenerate eyes, this finding suggests that phenotypic degeneration of the eyes is accompanied by parallel changes at the molecular level as a result of relaxed selective constraints.

Fossorial salamanders typically have elongate and attenuated heads and bodies, diminutive limbs, hands and feet, and extremely elongate tails. Batrachoseps from California, Lineatriton from eastern México, and Oedipina from southern México to Ecuador, all members of the family Plethodontidae, tribe Bolitoglossini, resemble one another in external morphology, which has evolved independently. Whereas Oedipina and Batrachoseps are elongate because there are more trunk vertebrae, a widespread homoplasy (parallelism) in salamanders, the genus Lineatriton is unique in having evolved convergently by an alternate “giraffe-neck” developmental program. Lineatriton has the same number of trunk vertebrae as related, nonelongated taxa, but individual trunk vertebrae are elongated. A robust phylogenetic hypothesis, based on sequences of three mtDNA genes, finds Lineatriton to be deeply nested within a clade characterized by generalized ecology and morphology. Lineatriton lineolus, the only currently recognized taxon in the genus, shows unanticipated genetic diversity. Surprisingly, geographically separated populations of L. lineolus are not monophyletic, but are sister taxa of different species of the morphologically generalized genus Pseudoeurycea. Lineatriton, long thought to be a unique monospecific lineage, is polyphyletic. Accordingly, the specialized morphology of Lineatriton displays homoplasy at two hierarchical levels: (i) with respect to other elongate lineages in the family (convergence), and (ii) within what is currently recognized as a single taxon (parallelism). These evolutionary events are of adaptive significance because to invade the lowland tropics salamanders must be either arboreal or fossorial; the repeated evolution of elongation and attenuation has led to multiple lowland invasions.

DNA was extracted from the remains of 35 ground sloths from various parts of North and South America. Two specimens of Mylodon darwinii, a species that went extinct at the end of the last glaciation, yielded amplifiable DNA. However, of the total DNA extracted, only approximately 1/1000 originated from the sloth, whereas a substantial part of the remainder was of bacterial and fungal origin. In spite of this, > 1100 bp of sloth mitochondrial rDNA sequences could be reconstructed from short amplification products. Phylogenetic analyses using homologous sequences from all extant edentate groups suggest that Mylodon darwinii was more closely related to the two-toed than the three-toed sloths and, thus, that an arboreal life-style has evolved at least twice among sloths. The divergence of Mylodon and the two-toed sloth furthermore allows a date for the radiation of armadillos, anteaters, and sloths to be estimated. This result shows that the edentates differ from other mammalian orders in that they contain lineages that diverged before the end of the Cretaceous Period.

The life histories of many animals are characterized by niche shifts, the timing of which can strongly affect fitness. In the tree frog Agalychnis callidryas, which has arboreal eggs, there is a trade-off between predation risks before and after hatching. When eggs are attacked by snakes, tadpoles escape by hatching rapidly and falling into the water below. Eggs not attacked by snakes hatch later, when newly emerged tadpoles are less vulnerable to aquatic predators. Plasticity in hatching allows embryos to use immediate, local information on risk of mortality to make instantaneous behavioral decisions about hatching and the accompanying shift from arboreal to aquatic habitats.

A diversidade de espécies e fenotípica pode variar consideravelmente entre grupos taxonômicos e ao longo do tempo em uma mesma linhagem. O estudo de tais variações tornou-se um dos principais objetivos da biologia evolutiva fornecendo informações importantes a respeito dos possíveis mecanismos que regulam a biodiversidade. Dessa forma, o objetivo geral da presente tese foi investigar os padrões da diversificação de espécies e da morfologia em um grupo cosmopolita de serpentes, a família Viperidae, e os potenciais processos subjacentes. Primeiramente, (1) reconstruímos as relações filogenéticas e estimamos os tempos de divergência entre as linhagens da família Viperidae utilizando uma abordagem Bayesiana. (2) Aplicando um método recentemente desenvolvido (BAMM), exploramos como as taxas de especiação e extinção variaram ao longo da radiação do grupo inferindo os possíveis processos reguladores. Por fim, (3) analisamos se a evolução do tamanho do corpo e as taxas de especiação variam nos diferentes habitats ocupados pelos viperídeos (terrestres vs arborícola). Nesta tese geramos a filogenia molecular de viperídeos mais completa até o momento utilizando sequências para 11 genes mitocondriais e nucleares abrangendo 79% das espécies viventes (264 terminais) e todos com exceção de um gênero. De maneira geral, foi possível obter relações filogenéticas robustas para o grupo com a maioria dos gêneros sendo monofilética. Os tempos de divergência obtidos indicam que os viperídeos começaram a diversificar em meados do Paleoceno tardio/meio do Eoceno inferindo idades um pouco mais tardias que o encontrado em estudos anteriores. Durante a radiação do grupo, um aumento nas taxas de especiação parece ter ocorrido durante a diversificação dos crotalíneos (pit vipers) em decorrência não só da evolução das fossetas loreais mas também como resultado de mudanças geológicas e climáticas na Ásia e da invasão do novo mundo. Após este rápido aumento inicial, as taxas de especiação desaceleraram em direção ao presente. Por fim, os resultados aqui apresentados indicam que apesar dos habitats arborícolas limitarem a evolução morfológica nos viperídeos, a evolução da arborealidade parece não afetar as taxas de especiação que permanecem similares entre linhagens arborícolas e terrestres. Isto sugere dois cenários: (1) a especiação acontece de forma independente das mudanças morfológicas nos viperídeos; ou (2) o isolamento geográfico seria um mecanismo importante na diversificação de linhagens arborícolas contrabalançando decréscimos nas oportunidades de especiação possivelmente relacionados às pressões seletivas impostas pelo ambiente arborícola. A presente tese contribui para entendermos mais sobre como evoluíram os viperídeos ao longo dos seus ∼50 milhões de anos. Além de propor cenários e hipóteses a serem futuramente explorados com os viperídeos, elaboramos uma discussão ampla e conceitual a respeito dos possíveis mecanismos por trás da diversificação de espécies e da morfologia que poderiam também ser contemplados para outros grupos de organismos. Portanto, a presente tese contribui não só para entendermos os mecanismos que geram e mantém a diversidade de serpentes, mas também para enriquecer a discussão dos mecanismos que geram e mantém a biodiversidade como um todo

Os meios urbanos são espaços cada vez mais edificados, compactos e com solos impermeabilizados, onde a ausência ou reduzida presença de espaços verdes é uma realidade. Os elementos arbóreos, qualquer que seja a sua disposição, ganham cada vez mais significado enquanto representantes do meio natural na conservação da qualidade e conforto de vida nas cidades. Este trabalho desenvolveu-se a partir do tema das árvores em meio urbano, tendo por base os benefícios das árvores nas cidades, a sua distribuição espacial e o estado de conservação deste tipo de património. O principal fio condutor passou pela concepção do inventário do Património Arbóreo da cidade de Portalegre, com recurso às novas tecnologias da informação, nomeadamente os Sistemas de Informação Geográfica, capaz de se integrar nos sistemas homólogos do Município de Portalegre. O inventário à escala do perímetro urbano da cidade de Portalegre contabilizou 2087 elementos arbóreos localizados em áreas públicas, dos quais 1500 são árvores de folha caduca e 587 são perenifólias. Registou-se um elevado índice de diversidade para aquela área, cerca de 705 árvores/km2, embora as árvores estejam distribuídas de forma heterogénea pelo território considerado, percebendo-se com isso as zonas de carência de arborização. Com este inventário foi ainda possível conceber uma avaliação económica segundo a Norma de Granada, de 13 árvores isoladas e uma alameda com 38 indivíduos, assim como propor a classificação destes elementos a Árvores de Interesse Público. Anseia-se que este trabalho seja um documento informativo e sensibilizador de todo o público em geral, mas principalmente que se assuma como uma ferramenta útil aos responsáveis pelos espaços verdes da cidade de Portalegre.

The Nachtigall clay pit near Holzminden, northern Germany, is located in a subrosional basin filled with 43 m of interglacial, interstadial and stadial deposits adjacent to the Weser River. The succession separates the Older Middle Terrace from the Younger Middle Terrace of the Weser River. Nachtigall core KB1 (1998) mainly contains silt and clay with intercalated peat layers. The layers of fen peat and intercalated humic silt are between 36 and 22.5 m depth. According to palynological studies, the peat layers and some humic silts were deposited during interglacial and interstadial periods marked by forest vegetation, termed Nachtigall 1 and Nachtigall 2. They are subdivided by a stadial, termed Albaxen. The peat of Nachtigall 1 is interrupted twice by silt and clay strata (Allochthonous Unit I, II) which are reworked sediments of older glacial periods, possibly of late Elsterian or early Holsteinian age. The palynological sequences of Nachtigall and Göttingen/Ottostrasse show the same pattern. Moreover, the contemporaneous pollen profiles of Nachtigall and Göttingen/Ottostrasse can be compared with the Velay pollen sequence (France). The Nachtigall core section 36-26.02 m corresponds to Bouchet 2 - Bonnefond - Bouchet 3 in Velay. The profiles of Velay and Nachtigall are independently correlated to the MIS-timescale and correspond to MIS 7c, 7b, and 7a. TIMS 230Th/U-dating shows ages ranging from 227 + 9/-8 to 201 + 15/-13 ka, which are in good agreement with the inferred MIS 7 age.

We investigated the sedimentary record of Lake Hancza (northeastern Poland) using a multi-proxy approach, focusing on early to mid-Holocene climatic and environmental changes. AMS 14C dating of terrestrial macrofossils and sedimentation rate estimates from occasional varve thickness measurements were used to establish a chronology. The onset of the Holocene at c. 11600 cal. a BP is marked by the decline of Lateglacial shrub vegetation and a shift from clastic-detrital deposition to an autochthonous sedimentation dominated by biochemical calcite precipitation. Between 10000 and 9000 cal. a BP, a further environmental and climatic improvement is indicated by the spread of deciduous forests, an increase in lake organic matter and a 1.7% rise in the oxygen isotope ratios of both endogenic calcite and ostracod valves. Rising d18O values were probably caused by a combination of hydrological and climatic factors. The persistence of relatively cold and dry climate conditions in northeastern Poland during the first one and a half millennia of the Holocene could be related to a regional eastern European atmospheric circulation pattern. Prevailing anticyclonic circulation linked to a high-pressure cell above the retreating Scandinavian Ice Sheet might have blocked the influence of warm and moist Westerlies and attenuated the early Holocene climatic amelioration in the Lake Hancza region until the final decay of the ice sheet.

Excavations were carried out in a Late Palaeolithic site in the community of Bad Buchau-Kappel between 2003 and 2007. Archaeological investigations covered a total of more than 200 m**2. This site is the product of what likely were multiple occupations that occurred during the Late Glacial on the Federsee shore in this location. The site is situated on a mineral ridge that projected into the former Late Glacial lake Federsee. This beach ridge consists of deposits of fine to coarse gravel and sand and was surrounded by open water, except for a connection to the solid shore on the south. A lagoon lay between the hook-shaped ridge and the shore of the Federsee. This exposed location provided optimal access to the water of the lake. In addition, the small lagoon may have served as a natural harbor for landing boats or canoes. Sedimentological and palynological investigations document the dynamic history of the location between 14,500 and 11,600 years before present (cal BP). Evidence of the deposition of sands, gravels and muds since the Bølling Interstadial is provided by stratigraphic and palynological analyses. The major occupation occurred in the second half of the Younger Dryas period. Most of the finds were located on or in the sediments of the ridge; fewer finds occurred in the surrounding mud, which was also deposited during the Younger Dryas. Direct dates on some bone fragments, however, demonstrate that intermittent sporadic occupations also took place during the two millennia of the Meiendorf, Bølling, and Allerød Interstadials. These bones were reworked during the Younger Dryas and redeposited in the mud. A 14C date from one bone of 11,600 years ago (cal BP) places the Late Palaeolithic occupation of the ridge at the very end of the Younger Dryas, which is in agreement with stratigraphic observations. Stone artifacts, numbering 3,281, comprise the majority of finds from the site. These include typical artifacts of the Late Palaeolithic, such as backed points, short scrapers, and small burins. There are no bipointes or Malaurie-Points, which is in accord with the absolute date of the occupation. A majority of the artifacts are made from a brown chert that is obtainable a few kilometers north of the site in sediments of the Graupensandrinne. Other raw materials include red and green radiolarite that occur in the fluvioglacial gravels of Oberschwaben, as well as quartzite and lydite. The only non-local material present is a few artifacts of tabular chert from the region near Kelheim in Bavaria. A unique find consists of two fragments of a double-barbed harpoon made of red deer antler, which was found in the Younger Dryas mud. It is likely, but not certain, that this find belongs to the same assemblage as the numerous stone artifacts. Although not numerous, animal bones were also found in the excavations. Most of them lay in sediments of the Younger Dryas, but several 14C dates place some of these bones in earlier periods, including the Meiendorf, Bølling, and Allerød Interstadials. These bones were reworked by water and redeposited in mud sediments during the Younger Dryas. As a result, it is difficult to attribute individual bones to particular chronological positions without exact dates. Species that could be identified include wild horse (Equus spec.), moose or elk (Alces alces), red deer (Cervus elaphus), roe deer (Capreolus capreolus), aurochs or bison (Bos spec.), wild boar (Sus scrofa), as well as birds and fish, including pike (Esox Lucius).
