Le [6]-gingérol est un analogue structurel de la capsaïcine, une molécule agoniste au récepteurs TRPV1 et ayant des propriétés thérapeutiques connues dans le traitement de la douleur. Deux objectifs principaux ont été poursuivis lors de la réalisation de ce projet de recherche. D’abord, établir une meilleure caractérisation du métabolisme du [6]-gingérol chez le rat. Pour ce faire, une méthode sensible et spécifique pour la quantification du [6]-gingérol et ses métabolites par HPLC-ESI/MS/MS a été développée. Une étude de stabilité métabolique in vitro utilisant des microsomes hépatiques de rats a ensuite été réalisée. Les résultats démontrent une dégradation lente avec un temps de demi-vie de 163 minutes et une clairance intrinsèque relativement basse de 0.0043 mL/min. D’autres analyses ont ensuite été performées pour caractériser les métabolites in vitro et in vivo. Trois principaux métabolites de phase I et quatre métabolites de phase II ont été identifiés par HPLC-MS/MS et HPLC-MSD TOF. Les résultats suggèrent que le principal métabolite excrété dans l’urine est un glucuronide du [6]-gingérol hydroxylé. Le second objectif de ce projet était de déterminer l’effet central du [6]-gingérol sur la douleur neuropathique lorsqu’injecté par voie intrathécale. La distribution de la molécule a d’abord été évaluée suite à une administration intra-péritonéale de 40 mg/kg de [6]-gingérol et les ratios des concentrations cerveau-plasma et moelle épinière-plasma (0.73 et 1.7, respectivement) suggèrent que le [6]-gingérol se distribue efficacement au niveau du système nerveux central. Une injection intrathécale de 10 μg de [6]-gingérol à été performée chez les rats suite à l’induction de douleur par la pose de ligatures au niveau du nerf sciatique. Les résultats suggèrent une réduction significative de l’allodynie mécanique et de l’hyperalgésie thermique à 30 min, 2 h et 4 h suivant l’injection (p < 0.05, p < 0.01 et p < 0.001). Le [6]-gingérol se distribue donc adéquatement au niveau du système nerveux central des rats, permettant une action au niveau des récepteurs TRPV1. Ainsi, le [6]-gingérol pourrait soulager la douleur neuropathique en agissant centralement au niveau de la moelle épinière.

Este recurso ayudará a los maestros a supervisar y registrar el progreso de sus alumnos en ortografía. Los alumnos cuentan con una tira de palabras que tienen que aprender en la semana, que en la mayoría de los casos se entregan como tarea. En la parte inferior de cada prueba de ortografía se proporcionan notas al profesor con indicaciones y frases para dictar a los niños. La prueba de ortografía, entonces toma la forma de las frases dictadas o de los párrafos leídos en voz alta en el CD de audio; esto ayuda a los alumnos a la comprensión de las palabras a medida que se establecen dentro de un contexto. Utilizando los resultados de las pruebas, los profesores pueden crear un perfil claro de los niveles de la ortografía de los alumnos y supervisar sus progresos.

OBJECTIVE: The present study was carried out to investigate effects of meals, rich in either saturated fatty acids (SFA), or n-6 or n-3 fatty acids, on postprandial plasma lipid and hormone concentrations as well as post-heparin plasma lipoprotein lipase (LPL) activity. DESIGN: The study was a randomized single-blind study comparing responses to three test meals. SETTING: The volunteers attended the Clinical Investigation Unit of the Royal Surrey County Hospital on three separate occasions in order to consume the meals. SUBJECTS: Twelve male volunteers with an average age of 22.5 +/- 1.4 years (mean +/- SD), were selected from the University of Surrey student population; one subject dropped out of the study because he found the test meal unpalatable. INTERVENTIONS: Three meals were given in the early evening and postprandial responses were followed overnight for 11h. The oils used to prepare each of the three test meals were: a mixed oil rich in saturated fatty acids (SFA) which mimicked the fatty acid composition of the current UK diet, corn oil, rich in n-6 fatty acids and a fish oil concentrate (MaxEPA) rich in n-3 fatty acids. The oil under investigation (40 g) was incorporated into the test meals which were otherwise identical [208 g carbohydrates, 35 g protein, 5.65 MJ (1350 kcal) energy]. Postprandial plasma triacylglycerol (TAG), gastric inhibitory polypeptide (GIP), and insulin responses, as well as post-heparin LPL activity (measured at 12 h postprandially only) were investigated. RESULTS: Fatty acids of the n-3 series significantly reduced plasma TAG responses compared to the mixed oil meal (P < 0.05) and increased post-heparin LPL activity 15 min after the injection of heparin (P < 0.01). A biphasic response was observed in TAG, with peak responses occurring at 1 h and between 3-7 h postprandially. GIP and insulin showed similar responses to the three test meals and no significant differences were observed. CONCLUSION: We conclude that fish oils can decrease postprandial plasma TAG levels partly through an increase in post-heparin LPL activity, which however, is not due to increased GIP or insulin concentrations.

One of the best examinations used routinely to detect gastro-oesophageal reflux (GOR) is scintigraphy and attempts are continuously being made to improve the performance of the test. We have tested a new manoeuvre to improve the results. Three hundred and twenty eight patients (190 males and 138 females, aged 1 month to 84 years, average 4.47 years) were studied after a 12 h fast period with 100 MBq of Tc-99(m)-Phytate, 50 ml volume, directly delivered into the stomach in a mixture of orange juice. All patients were strongly suspected of having GOR. Conventional acquisition was done for 20 min, at a rate of one frame every 20 s. in the middle of the examination, small children were put in the upright position and held there for a few seconds or released in a way that allowed movement; adults were asked to stand up and walk a few metres. The test was then continued for the remaining 10 min. The global index of positivity was 64.6% (76 positive cases in the first half of the examination and 134 positive cases only after the manoeuvre). The mean of GOR episodes observed in the positive cases without the manoeuvre was 0.5 per patient, significantly different from the mean of 1.59 noted after the manoeuvre (P<0.01). In 28 cases the manoeuvre increased either the intensity or the frequency of reflux previously detected without it. We believe that this manoeuvre should be performed in all scintigraphic tests aiming to detect GOR. In the present series, the manoeuvre increased the frequency of GOR episodes. Four groups of horses of Brasileiro de Hipismo bred were submitted to fasting for 24 and 48 hours in order to study the absorption capacity of the small intestine. Two groups were fed with coast cross grass (Cynodon dactylon) and the other two groups with coast cross pasture and grains. At the end of the fasting periods, the groups received 1g of glucose/kg of body weight in a 20% solution through a nasogastric tube. Blood samples were collected immediately before and 70, 60, 120, 180, 240, 300, and 360 minutes after glucose administration. Glycemia was determined by the orthotoluidine method and insulin by radioimmunoassay. The animals which received grains showed larger increase in glycemia and insulinemia than those maintained on pasture regimen alone. The 48-hour fasting period induced higher glycemia and insulinemia levels than those observed after 24-hour fasting.

The present work aimed at studying the influence of the estrous cycle in the forced swim test, an animal model of depression. For this, 44 male and female Wistar rats were divided into five groups according to the hormonal state in the first day of the study: metaestrus (N = 12), diestrus (N = 8), proestrus (N = 7), estrous (N = 6) and males (N = 11). They were housed in groups of five, with water and food ad libitum under a 12/12 h light/dark cycle. Females were screened daily for the estrous cycle. The animals were subjected to two swimming sessions in a glass cylinder with water up to 15 cm at 28±2º C. The data of the first five minutes of a 15-min first session were compared to those of a 5-min second session 24 h later. The results indicate that the latency to the first immobility was substantially reduced in the second session and was longer for females in diestrus and proestrus in the first session. The results also indicate that females in diestrus and proestrus exhibited less immobility than males in the first session; females in diestrus also exhibited less immobility than females in metaestrus. Females in metaestrus and diestrus, as well as males, did not present the decrease in total immobility times in the second session. The present results are analyzed in terms of differential effects of progesterone and estrogen on a learning component and an affective component.

OBJECTIVES: The effects of the orthostatic test on the cardiovascular system have been investigated, but there are no data on the behavior of the geometric indices of heart rate variability. We evaluated the effects of the active orthostatic test on the geometric indices of heart rate variability in women. METHOD: The study was performed on 12 healthy women between ages 18 and 30. For the analysis of heart rate variability, heart rate was recorded beat-to-beat. The RR intervals were transformed into geometric figures, and from these we calculated the triangular index, the triangular interpolation of RR intervals, the SD1, SD2 indexes and the SD1/SD2 ratio, which were obtained from the Poincaré plot. Visual analysis of the plot was also performed. For the test, participants quickly stood up from a seated position in 3 seconds or less, and remained standing still for 15 minutes. Heart rate variability samples were collected at four moments: rest, 0-5 minutes, 5-10 minutes and 10-15 minutes at standing position. RESULTS: The triangular index, the triangular interpolation of RR intervals, the SD1 and SD2 indices were reduced (p < 0.05) at 10-15 minutes after the volunteers stood up from seated position. The SD1/SD2 ratio was decreased at 0-15 minutes after the subjects changed from seated to orthostatic position.
