RATIONALE: Dopamine D2 receptors are the main target of antipsychotic drugs. In the brain, D2 receptors coexpress with adenosine A2A and CB1 cannabinoid receptors, leading to functional interactions. OBJECTIVES: The protein and messenger RNA (mRNA) contents of A2A, D2, and CB1 receptors were quantified in postmortem prefrontal cortex of subjects with schizophrenia. MATERIALS AND METHODS: The study was performed in subjects suffering schizophrenia (n=31) who mainly died by suicide, matched with non-schizophrenia suicide victims (n=13) and non-suicide controls (n=33). The density of receptor proteins was evaluated by immunodetection techniques, and their relative mRNA expression was quantified by quantitative real-time polymerase chain reaction. RESULTS: In schizophrenia, the densities of A2A (90+/-6%, n=24) and D2-like receptors (95+/-5%, n=22) did not differ from those in controls (100%). Antipsychotic treatment did not induce changes in the protein expression. In contrast, the immunodensity of CB1 receptors was significantly decreased (71+/-7%, n=11; p<0.05) in antipsychotic-treated subjects with schizophrenia but not in drug-free subjects (104+/-13%, n=11). The relative mRNA amounts encoding for A2A, D2, and CB1 receptors were similar in brains of drug-free, antipsychotic-treated subjects with schizophrenia and controls. CONCLUSIONS: The findings suggest that antipsychotics induce down-regulation of CB1 receptors in brain. Since A2A, D2, and CB1 receptors coexpress on brain GABAergic neurons and reductions in markers of GABA neurotransmission have been identified in schizophrenia, a lower density of CB1 receptor induced by antipsychotics could represent an adaptative mechanism that reduces the endocannabinoid-mediated suppression of GABA release, contributing to the normalization of cognitive functions in the disorder.

BACKGROUND AND PURPOSE: Beyond the Framingham Stroke Risk Score, prediction of future stroke may improve with a genetic risk score (GRS) based on single-nucleotide polymorphisms associated with stroke and its risk factors. METHODS: The study includes 4 population-based cohorts with 2047 first incident strokes from 22,720 initially stroke-free European origin participants aged ≥55 years, who were followed for up to 20 years. GRSs were constructed with 324 single-nucleotide polymorphisms implicated in stroke and 9 risk factors. The association of the GRS to first incident stroke was tested using Cox regression; the GRS predictive properties were assessed with area under the curve statistics comparing the GRS with age and sex, Framingham Stroke Risk Score models, and reclassification statistics. These analyses were performed per cohort and in a meta-analysis of pooled data. Replication was sought in a case-control study of ischemic stroke. RESULTS: In the meta-analysis, adding the GRS to the Framingham Stroke Risk Score, age and sex model resulted in a significant improvement in discrimination (all stroke: Δjoint area under the curve=0.016, P=2.3×10(-6); ischemic stroke: Δjoint area under the curve=0.021, P=3.7×10(-7)), although the overall area under the curve remained low. In all the studies, there was a highly significantly improved net reclassification index (P<10(-4)). CONCLUSIONS: The single-nucleotide polymorphisms associated with stroke and its risk factors result only in a small improvement in prediction of future stroke compared with the classical epidemiological risk factors for stroke.

Contexte : Les aidants naturels sont des personnes bénévoles qui dispensent des soins physiques et, ou un soutien émotionnel à l'un(e) de leur proche ou ami(e). Ceci peut générer un stress intense tant physique que psychique. Des interventions visant principalement à augmenter la qualité de vie et à soutenir la vie affective des aidants naturels ont été développées. Para aumentar a precisão nas análises químicas de fertilidade do solo e dosar simultaneamente vários elementos, alguns laboratórios vêm optando pelo uso da técnica da espectrofotometria de emissão ótica em plasma induzido (ICP), em detrimento da técnica da espectrofotometria de absorção atômica (EAA), hoje comumente utilizada nos laboratórios de análise de solos. Este trabalho, além de comparar as duas técnicas de dosagem quanto à precisão, à reprodutibilidade e à magnitude dos teores dos micronutrientes Fe, Zn, Cu e Mn, extraídos por Mehlich-1, Mehlich-3 e DTPA-TEA, objetivou, também, selecionar os comprimentos de onda que apresentam menores interferências espectrais no ICP. Foram utilizadas 36 amostras (0 a 0,2 m) de solos coletadas nos Estados de Minas Gerais e Bahia, com ampla variação nos teores de micronutrientes, sendo selecionados três solos para definir os comprimentos de onda do ICP e avaliar a precisão e a reprodutibilidade dos métodos de dosagem. Os comprimentos de onda com menores interferências espectrais no ICP foram: 259,939 nm para Fe em Mehlich-1 e DTPA-TEA e 234,349 nm em Mehlich-3; 213,857 nm para Zn e 324,752 nm para Cu nos três extratores; e 259,372 nm para Mn em Mehlich-1 e DTPA-TEA e 260,568 nm em Mehlich-3. Tanto o ICP quanto o EAA foram precisos e reprodutíveis nas dosagens de Fe e Mn, sendo o ICP, em virtude do seu menor limite de detecção, mais preciso e reprodutível nas dosagens de Zn e Cu. Os métodos de dosagem diferiram estatisticamente (p < 0,01) pelo teste de identidade aplicado, para as dosagens de Fe, Zn, Cu e Mn, utilizando Mehlich-1, Mehlich-3 e DTPA-TEA, comprometendo assim a interpretação dos resultados gerados pelo ICP, com base nos níveis críticos gerados a partir do EAA.

O eucalipto apresenta ótimo crescimento em solo com baixa fertilidade, mas pouco se sabe sobre a participação das ectomicorrizas e de ácidos orgânicos na aquisição e no acúmulo de nutrientes pela planta em campo. A produção de ácido oxálico e sua relação com as concentrações de P, Ca, Mg e K foram avaliadas em ectomicorrizas e raízes laterais finas de híbrido de Eucalyptus grandis x Eucalyptus urophylla, de 2,5 anos de idade, na região de Viçosa, MG. A área de estudo apresenta topografia típica em meia laranja, de vertente côncavo-convexa. Foram também avaliadas as concentrações desse composto no solo rizosférico, não rizosférico e ectomicorrizosférico. As maiores percentagens de colonização micorrízica foram observadas na área de encosta, onde havia limitada disponibilidade de nutrientes e alta saturação de Al. As concentrações de ácido oxálico + oxalato corresponderam, em mg kg-1: folhas, 324,6; ectomicorrizas, 208,3; raízes laterais finas não colonizadas, 183,1. Já no solo, as concentrações foram maiores no solo ectomicorrizosférico, com 183,7 mg kg-1, seguido pelo solo rizosférico, com 134,3 mg kg-1, e pelo solo não rizosférico, com 76,0 mg kg-1. As maiores concentrações de ácido oxálico e P (p < 0,05) nas ectomicorrizas da área do topo, 117,3 mg kg-1 e 6,3 g kg-1, respectivamente, sugerem que as populações de fungos ectomicorrízicos nesta área têm papel importante na solubilização e disponibilização de nutrientes para o hospedeiro. Não foram observadas correlações positivas significativas entre a produção de ácido oxálico e as concentrações de Ca nas raízes laterais finas e nas ectomicorrizas de eucalipto.

OBJECTIVES: Women with a history of preeclampsia (PE) are at increased risk of long term cardiovascular and end-stage renal diseases. However, follow up of preeclamptic women is often omitted, mainly due to a weakness of knowledge of maternal caregivers and lack of comprehensive guidelines. The aim of this study was to define the prevalence of albuminuria, high blood pressure, and renal dysfunction 6 weeks after a preeclampsia. METHODS: This is a prospective case-control study comparing women presenting with preeclampsia to an unmatched control group of women with no hypertensive disorders of pregnancy. A complete medical assessment was performed at 6 weeks post-partum. Recruitment started in June 2010. RESULTS: 324 women were included in the PE group and 50 in the control one. Characteristics of both groups and results of the medical work-up at 6 weeks post-partum are presented in Table 1. Women with preeclampsia presented with a higher BMI, higher prevalence of office high blood pressure, pathological albuminuria and renal hyper-filtration than women in the control group. CONCLUSIONS: Prevalence of post-partum hypertension, and renal dysfunction is higher in women with PE than in uncomplicated pregnancies. Systematic assessment of renal risk factors 6 weeks after preeclampsia allows identification of high-risk women and early implementation of preventive and therapeutic strategies. DISCLOSURES: A. The Department identified 2,706 potential retail locations at which motor fuel may be sold. Reporting forms were mailed to 1,675 locations identified using Department of Agriculture and Land Stewardship motor fuel license information and to 305 locations identified through the Department of Revenue motor fuel tax database. In addition, 726 locations were identified for four large motor fuel retailers that submitted their annual sales data electronically. The Department received either forms or electronic files representing 2,324 (85.9%) of the locations. Replies for 115 of the locations indicated that no retail sales of any type of motor fuel were made during 2010. 2,209 locations reported retail motor fuel sales. This report’s primary focus is on biofuel sales. Of the reporting retail locations, 2,075 (93.9%) reported selling E10 blend motor fuel, 165 (7.5%) reported selling the E85 blend motor fuel, and 239 (10.8%) reported selling various blends of biodiesel.
