Objective. Formocresol, paramonochlorophenol, or calcium hydroxide have been widely used in dental practice to eradicate bacteria and consequently to produce root canal disinfection. Taking into consideration strong evidence for a relationship between DNA damage and carcinogenesis, the purpose of the present study was to evaluate the genotoxic effects of antimicrobial endodontic compounds in human peripheral lymphocytes by single-cell gel ( comet) assay. This technique detects DNA strand breaks in individual cells.Study design. A total of 10 mu L of the tested substance solution (formocreso1, paramonochlorofeno1, and calcium hydroxide at 100-mu g/mL concentration) was added to human peripheral lymphocytes from 10 volunteers for 1 hour at 37 degrees C. The negative control group was treated with vehicle control (PBS) for 1 hour at 37 degrees C, as well. For the positive control group, lymphocytes were exposed to hydrogen peroxide at 100 mu M during 5 minutes on ice.Results. No DNA breakage was detected after a treatment of peripheral lymphocytes by formocresol, paramonochlorophenol, or calcium hydroxide at 100 mu g/mL.Conclusions. In summary, our results indicate that exposure to formocresol, paramonochlorophenol, or calcium hydroxide may not be a factor that increases the level of DNA lesions in human peripheral lymphocytes as detected by single-cell gel (comet) assay.

The objective of this study is to translate to Brazilian Portuguese, and validate (criterion) it to the Brazilian population, the Women's Health Questionnaire (WHQ), questionnaire was developed by Hunter (1992) in order to evaluate physical and mental symptoms, which were experienced by women, along their climacteric transition. A sample of 63 women, from 45 to 65 years old, was selected from the HC-UNESP staff and HC-Community Health School Center users. The WHQ and the General Health Questionnaire - GHQ (Goldberg-1972 - 60 items) were answered by all of them. The GHQ was used as reference. Pearson correlation coefficient, sensitivity and specificity were calculated for assessing the validity of the questionnaire. Cut-off points were determined in the WHQ by linear regressions (the reference was GHQ's cut-off points - higher than 2). Outcomes highlighted a general index of sensitivity of 0.83, specificity of 0.87, and correlation of - 0.82 and reliability of 0.89. The study sample does not portray the general population because the persons interviewed had high level of education. As the WHQ is easy to be applied and analyzed, it could be used in general, gynecologic, oncology and psychiatric practice.
VEGF-C expression in oral cancer by neurotransmitter-induced activation of beta-adrenergic receptors

The aim of this study was to investigate the expression of vascular endothelial growth factor type C (VEGF-C) in oral squamous cell carcinoma (OSCC) cell lines through norepinephrine-induced activation of beta-adrenergic receptors. Human OSCC cell lines (SCC-9 and SCC-25) expressing beta-adrenergic receptors were stimulated with different concentrations of norepinephrine (0.1, 1, and 10 μM) and 1 μM of propranolol, and analyzed after 1, 6, and 24 h. VEGF-C gene expression and VEGF-C production in the cell supernatant were evaluated by real-time PCR and by ELISA, respectively. The results showed that beta-adrenergic receptor stimulation by different concentrations of norepinephrine or blocking by propranolol did not markedly alter VEGF-C expression by SCC-9 and SCC-25 cells. VEGF-C protein levels produced by oral malignant cell lines after stimulation with different norepinephrine concentrations or blocking with propranolol was statistically similar (p > 0.05) to those of the control group (nonstimulated OSCC cell lines). Our findings suggest that stimulation of beta-adrenergic receptors by means of norepinephrine does not seem to modulate the VEGF-C expression in OSCC cell lines. These findings reinforce the need for further studies in order to understand the responsiveness of oral cancer to beta-adrenergic receptor stimulation or blockage, especially with regard to VEGF-C production. © 2012 International Society of Oncology and BioMarkers (ISOBM).

Antioxidants are compounds responsible for free radical scavenging in the body. They protect the organism from oxidative modification of cells and tissues. These modifications have been associated with degenerative diseases, atherosclerosis and carcinogenesis. Punica granatum displays high antioxidant potential due to the presence of phenolic compounds, which are capable of disease prevention. The present study showed the highest antioxidant activity in pomegranate peel than in seeds and pulp. Based on these results, pomegranate peel was used to produce dried extract that was added to commercial tomato juice and orange juice with strawberries. Analysis to determine the content of phenolic compounds and antioxidant activity was performed on pomegranate pulp, seeds and peel and in juices enriched with dried extract of pomegranate peel. The dried extract was responsible for a significant increase in antioxidant activity of the juices, proportional to the concentrations added. However, although both flavors of enriched juices displayed high antioxidant levels, the samples with higher dried extract concentrations received the lowest scores from sensory analysis participants due to the characteristic astringent flavor of pomegranate peels. Therefore, to obtain greater acceptance in the consumer market, we concluded that the maximum addition of dried pomegranate peel extract is 0.5% in tomato juice and orange juice with strawberries.

UVA light (320–400 nm) represents approximately 95% of the total solar UV radiation that reaches the Earth’s surface. UVA light induces oxidative stress and the formation of DNA photoproducts in skin cells. These photoproducts such as pyrimidine dimers (cyclobutane pyrimidine dimers, CPDs, and pyrimidine (6-4) pyrimidone photoproducts, 6-4PPs) are removed by nucleotide excision repair (NER). In this repair pathway, the XPA protein is recruited to the damage removal site; therefore, cells deficient in this protein are unable to repair the photoproducts. The aim of this study was to investigate the involvement of oxidative stress and the formation of DNA photoproducts in UVA-induced cell death. In fact, similar levels of oxidative stress and oxidised bases were detected in XP-A and NER-proficient cells exposed to UVA light. Interestingly, CPDs were detected in both cell lines; however, 6-4PPs were detected only in DNA repairdeficient cells. XP-A cells were also observed to be significantly more sensitive to UVA light compared to NER-proficient cells, with an increased induction of apoptosis, while necrosis was similarly observed in both cell lines. The induction of apoptosis and necrosis in XP-A cells using adenovirus-mediated transduction of specific photolyases was investigated and we confirm that both types of photoproducts are the primary lesions responsible for inducing cell death in XP-A cells and may trigger the skin-damaging effects of UVA light, particularly skin ageing and carcinogenesis.

To perform a systematic review of the utility of the Beck Depression Inventory for detecting depression in medical settings, this article focuses on the revised version of the scale (Beck Depression Inventory-II), which was reformulated according to the DSM-IV criteria for major depression. We examined relevant investigations with the Beck Depression Inventory-II for measuring depression in medical settings to provide guidelines for practicing clinicians. Considering the inclusion and exclusion criteria seventy articles were retained. Validation studies of the Beck Depression Inventory-II, in both primary care and hospital settings, were found for clinics of cardiology, neurology, obstetrics, brain injury, nephrology, chronic pain, chronic fatigue, oncology, and infectious disease. The Beck Depression Inventory-II showed high reliability and good correlation with measures of depression and anxiety. Its threshold for detecting depression varied according to the type of patients, suggesting the need for adjusted cut-off points. The somatic and cognitive-affective dimension described the latent structure of the instrument. The Beck Depression Inventory-II can be easily adapted in most clinical conditions for detecting major depression and recommending an appropriate intervention. Although this scale represents a sound path for detecting depression in patients with medical conditions, the clinician should seek evidence for how to interpret the score before using the Beck Depression Inventory-II to make clinical decisions

Im Rahmen dieser Arbeit wurde der Einfluss zweier möglicher Biomarker auf die Atherosklerose untersucht.rnMilk fat globule-EGF factor 8 (MFG-E8, Lactadherin) ist ein Glycoprotein, das vornehmlich von Makrophagen, glatten Muskelzellen und Endothelzellen sezerniert wird. MFG-E8-/--Mäuse zeigen vermehrt apoptotische Zellen in der atherosklerotischen Plaque, verstärkte Inflammationszeichen und vergrößerte Läsionen. In situ-Hybridisierung und Immunfluoreszenz zeigen eine starke Lactadherin-Expression in den Schaumzellen atherosklerotischer Plaques von Apo E-/-, Apo E-/-/GPx 1-/-und LDLR-/- Mäusen, vor allem in der Nähe des Lipid Core. Dort kolokalisiert Lactadherin mit dem Makrophagenmarker CD 68 und dem Chemokin Fraktalkin, das die MFG-E8 Sekretion stimuliert und so die Phagocytose forciert. Untersuchungen mittels RTD-PCR ergaben, dass Peritonealmakrophagen der Genotypen Apo E-/-, Apo E-/-/GPx 1-/- und GPx 1-/-, deren Gemeinsamkeit eine höhere Empfindlichkeit gegenüberrnoxidativem Stress ist, mehr Lactadherin exprimieren als andere Genotypen (B6, LDLR-/-). Die Inkubation muriner oder humaner Makrophagen mit oxLDL und eLDL hat keinen Einfluss auf die Expression der MFG-E8 mRNA. Der Kontakt mit apoptotischer Zellen hingegen erhöht die Expression signifikant. Lactadherin ist entscheidend für die effektive Phagozytose apoptotischer Zellen in der atherosklerotischen Läsion. Seine Expression wird vermutlich durch die Apoptose in der Nähe liegender Zellen und das verstärkte Vorkommen von ROS reguliert. Macrophage stimulating protein (MSP) übt Einfluss auf Migration, Proliferation und Phagocytose von Makrophagen aus. Seine Beteiligung an inflammatorischen Vorgängen und der Karzinogenese ist intensiv untersucht worden, nicht jedoch der Einfluss auf die Atherosklerose. Es ist bekannt, dass der SNP rs3197999 mit chronisch entzündlichen Darmerkrankungen (CED) assoziiert ist. Zudem geht er vermutlich mit einem erniedrigten Atheroskleroserisiko einher. Der Polymorphismus c2078t hat den Aminosäureaustausch R689C zur Folge. Rekombinant erzeugtes, mutantes und wildtypisches MSP induziert Migration und Proliferation bei THP-1-Makrophagen. MSPmut vermittelt dies jedoch wesentliche effektiver als MSPwt. Apoptose hingegen wird durch keine der Formen induziert. R689C führt zu einem “gain of function” des MSP-Proteins in Bezug auf die Proliferations- und Migrationsfähigkeit von Makrophagen und verändert vermutlich deren Cytokinfreisetzung. Dies führt möglicherweise zu einer erhöhten Phagocytoseeffizienz in der atherosklerotischen Läsion (erniedrigtes Atherosklerose-Risiko), und zu einer aberranten immunologischen Reaktion im Rahmen der CED (erhöhtes CED-Risiko).
