Role of nitric oxide and prostaglandin in the maintenance of cortical and renal medullary blood flow

This study was undertaken in anesthetized dogs to evaluate the relative participation of prostaglandins (PGs) and nitric oxide (NO) in the maintenance of total renal blood flow (TRBF), and renal medullary blood flow (RMBF). It was hypothesized that the inhibition of NO should impair cortical and medullary circulation because of the synthesis of this compound in the endothelial cells of these two territories. In contrast, under normal conditions of perfusion pressure PG synthesis is confined to the renal medulla. Hence PG inhibition should predominantly impair the medullary circulation. The initial administration of 25 µM kg-1 min-1 NG-nitro-L-arginine methyl ester produced a significant 26% decrease in TRBF and a concomitant 34% fall in RMBF, while the subsequent inhibition of PGs with 5 mg/kg meclofenamate further reduced TRBF by 33% and RMBF by 89%. In contrast, the initial administration of meclofenamate failed to change TRBF, while decreasing RMBF by 49%. The subsequent blockade of NO decreased TRBF by 35% without further altering RMBF. These results indicate that initial PG synthesis inhibition predominantly alters the medullary circulation, whereas NO inhibition decreases both cortical and medullary flow. This latter change induced by NO renders cortical and RMBF susceptible to a further decrease by PG inhibition. However, the decrease in medullary circulation produced by NO inhibition is not further enhanced by subsequent PG inhibition.

We investigated the involvement of GABAergic mechanisms of the central amygdaloid nucleus (CeA) in unanesthetized rats subjected to acute isotonic or hypertonic blood volume expansion (BVE). Male Wistar rats bearing cannulas unilaterally implanted in the CeA were treated with vehicle, muscimol (0.2 nmol/0.2 µL) or bicuculline (1.6 nmol/0.2 µL) in the CeA, followed by isotonic or hypertonic BVE (0.15 or 0.3 M NaCl, 2 mL/100 g body weight over 1 min). The vehicle-treated group showed an increase in sodium excretion, urinary volume, plasma oxytocin (OT), and atrial natriuretic peptide (ANP) levels compared to control rats. Muscimol reduced the effects of BVE on sodium excretion (isotonic: 2.4 ± 0.3 vs vehicle: 4.8 ± 0.2 and hypertonic: 4.0 ± 0.7 vs vehicle: 8.7 ± 0.6 µEq·100 g-1·40 min-1); urinary volume after hypertonic BVE (83.8 ± 10 vs vehicle: 255.6 ± 16.5 µL·100 g-1·40 min-1); plasma OT levels (isotonic: 15.3 ± 0.6 vs vehicle: 19.3 ± 1 and hypertonic: 26.5 ± 2.6 vs vehicle: 48 ± 3 pg/mL), and ANP levels (isotonic: 97 ± 12.8 vs vehicle: 258.3 ± 28.1 and hypertonic: 160 ± 14.6 vs vehicle: 318 ± 16.3 pg/mL). Bicuculline reduced the effects of isotonic or hypertonic BVE on urinary volume and ANP levels compared to vehicle-treated rats. However, bicuculline enhanced the effects of hypertonic BVE on plasma OT levels. These data suggest that CeA GABAergic mechanisms are involved in the control of ANP and OT secretion, as well as in sodium and water excretion in response to isotonic or hypertonic blood volume expansion.

This study examined the effects of pre-exercise carbohydrate availability on the time to exhaustion for moderate and heavy exercise. Seven men participated in a randomized order in two diet and exercise regimens each lasting 3 days with a 1-week interval for washout. The tests were performed at 50% of the difference between the first (LT1) and second (LT2) lactate breakpoint for moderate exercise (below LT2) and at 25% of the difference between the maximal load and LT2 for heavy exercise (above LT2) until exhaustion. Forty-eight hours before each experimental session, subjects performed a 90-min cycling exercise followed by 5-min rest periods and a subsequent 1-min cycling bout at 125% VO2max/1-min rest periods until exhaustion to deplete muscle glycogen. A diet providing 10% (CHOlow) or 65% (CHOmod) energy as carbohydrates was consumed for 2 days until the day of the experimental test. In the exercise below LT2, time to exhaustion did not differ between the CHOmod and the CHOlow diets (57.22 ± 24.24 vs 57.16 ± 25.24 min). In the exercise above LT2, time to exhaustion decreased significantly from 23.16 ± 8.76 min on the CHOmod diet to 18.30 ± 5.86 min on the CHOlow diet (P < 0.05). The rate of carbohydrate oxidation, respiratory exchange ratio and blood lactate concentration were reduced for CHOlow only during exercise above LT2. These results suggest that muscle glycogen depletion followed by a period of a low carbohydrate diet impairs high-intensity exercise performance.

Stretching has been widely used to increase the range of motion. We assessed the effects of a stretching program on muscle-tendon length, flexibility, torque, and activities of daily living of institutionalized older women. Inclusion/exclusion criteria were according to Mini-Mental State Examination (MMSE) (>13), Barthel Index (>13) and Lysholm Scoring Scale (>84). Seventeen 67 ± 9 year-old elderly women from a nursing home were divided into 2 groups at random: the control group (CG, N = 9) participated in enjoyable cultural activities; the stretching group (SG, N = 8) performed active stretching of hamstrings, 4 bouts of 1 min each. Both groups were supervised three times per week over a period of 8 weeks. Peak torque was assessed by an isokinetic method. Both groups were evaluated by a photogrammetric method to assess muscle-tendon length of uni- and biarticular hip flexors and hamstring flexibility. All measurements were analyzed before and after 8 weeks by two-way ANOVA with the level of significance set at 5%. Hamstring flexibility increased by 30% in the SG group compared to pre-training (76.5 ± 13.0° vs 59.5 ± 9.0°, P = 0.0002) and by 9.2% compared to the CG group (76.5 ± 13.0° vs 64.0 ± 12.0°, P = 0.0018). Muscle-tendon lengths of hip biarticular flexor muscles (124 ± 6.8° vs 118.3 ± 7.6°, 5.0 ± 7.0%, P = 0.031) and eccentric knee extensor peak torque were decreased in the CG group compared to pre-test values (-49.4 ± 16.8 vs -60.5 ± 18.9 Nm, -15.7 ± 20%, P = 0.048). The stretching program was sufficient to increase hamstring flexibility and a lack of stretching can cause reduction of muscle performance.

We studied the effect of pulsed ultrasound therapy (UST) and antibothropic polyvalent antivenom (PAV) on the regeneration of mouse extensor digitorum longus muscle following damage by Bothrops jararacussu venom. Animals (Swiss male and female mice weighing 25.0 ± 5.0 g; 5 animals per group) received a perimuscular injection of venom (1 mg/kg) and treatment with UST was started 1 h later (1 min/day, 3 MHz, 0.3 W/cm², pulsed mode). Three and 28 days after injection, muscles were dissected and processed for light microscopy. The venom caused complete degeneration of muscle fibers. UST alone and combined with PAV (1.0 mL/kg) partially protected these fibers, whereas muscles receiving no treatment showed disorganized fascicules and fibers with reduced diameter. Treatment with UST and PAV decreased the effects of the venom on creatine kinase content and motor activity (approximately 75 and 48%, respectively). Sonication of the venom solution immediately before application decreased the in vivo and ex vivo myotoxic activities (approximately 60 and 50%, respectively). The present data show that UST counteracts some effects of B. jararacussu venom, causing structural and functional improvement of the regenerated muscle after venom injury.

Myoclonus induced by etomidate during induction of general anesthesia is undesirable. This study evaluated the effect of dexmedetomidine (DEX) pretreatment on the incidence and severity of etomidate-induced myoclonus. Ninety patients undergoing elective surgical procedures were randomly allocated to three groups (n=30 each) for intravenous administration of 10 mL isotonic saline (group I), 0.5 µg/kg DEX in 10 mL isotonic saline (group II), or 1.0 µg/kg DEX in 10 mL isotonic saline (group III) over 10 min. All groups subsequently received 0.3 mg/kg etomidate by intravenous push injection. The incidence and severity of myoclonus were recorded for 1 min after etomidate administration and the incidence of cardiovascular adverse events that occurred between the administration of the DEX infusion and 1 min after tracheal intubation was recorded. The incidence of myoclonus was significantly reduced in groups II and III (30.0 and 36.7%), compared with group I (63.3%). The incidence of severe sinus bradycardia was significantly increased in group III compared with group I (P<0.05), but there was no significant difference in heart rate in groups I and II. There were no significant differences in the incidence of low blood pressure among the 3 groups. Pretreatment with 0.5 and 1.0 µg/kg DEX significantly reduced the incidence of etomidate-induced myoclonus during anesthetic induction; however, 0.5 µg/kg DEX is recommended because it had fewer side effects.

The aim of this study was to explore the clinical efficacy of a novel retrograde puncture approach to establish a preperitoneal space for laparoscopic direct inguinal hernia repair with inguinal ring suturing. Forty-two patients who underwent laparoscopic inguinal hernia repair with retrograde puncture for preperitoneal space establishment as well as inguinal ring suturing between August 2013 and March 2014 at our hospital were enrolled. Preperitoneal space was successfully established in all patients, with a mean establishment time of 6 min. Laparoscopic repairs were successful in all patients, with a mean surgical time of 26±15.1 min. Mean postoperative hospitalization duration was 3.0±0.7 days. Two patients suffered from postoperative local hematomas, which were relieved after puncturing and drainage. Four patients had short-term local pain. There were no cases of chronic pain. Patients were followed up for 6 months to 1 year, and no recurrence was observed. Our results demonstrate that preperitoneal space established by the retrograde puncture technique can be successfully used in adult laparoscopic hernioplasty to avoid intraoperative mesh fixation, and thus reduce medical costs.

The objective of this study was to evaluate the carcass suspension method concerning quality of sheep meat. Ten discard ewes were used, with approximately 62 kg of body weight. After slaughtering, flaying, evisceration and removal of head and paws, carcasses were longitudinally divided into two parts. Alternated sides of half carcasses were hanged by the tendon of the gastrocnemius (Treatment 1 - T1) and by the pelvic bone (Treatment 2 - T2) in cold store for a 24-hour period. Subsequently, the Semimembranosus muscle was removed from all half carcasses for meat quality analyses. The Semimembranosus muscles from the carcasses hanged by the pelvis suspension method presented higher softness than the same muscles from the carcasses hanged by the tendon of the gastrocnemius, with values of 1.99 kgf.cm-2 and 3.15 kgf.cm-2, respectively. Treatment 2 presented lower meat cooking losses than Treatment 1, with average values of 32.14 and 33.44%, respectively. The remaining meat quality parameters evaluated were not influenced by the carcass suspension method. We concluded that the carcass suspension method influenced meat softness and losses by cooking, with better results for carcasses hanged by the pelvic bone.

Tempeh is a food obtained by fermentation of soybean grains by the fungus Rizophus oligosporus. It is a traditional Indonesian food that presents benefits for human health protecting against diarrhea and chronic diseases. Tempeh processing includes dehulling, cooking, inoculation, and fermentation. In this study, chemical characteristics of tempeh prepared with soybean cultivars specially developed for human consumption (BRS 216, BRS 232, BRS 257, and BRS 267) were investigated. Soybean grains and tempeh obtained from these cultivars were analyzed for oil, protein, antinutrional factors, and isoflavone content. Cultivar BRS 216 presented the highest protein content in the grains (36.81%) and in tempeh (51.99%). On average, the protein content in tempeh increased 16% in relation to that of soybean grains. Isoflavone content was higher in the grains than in tempeh with significant differences among the cultivars. However, the aglycones content increased about 50% in tempeh (49.00 mg.100 g-1 on average) compared to that of raw material (soybean grains - 21.49 mg.100 g-1, on average). The content of Kunitz trypsin inhibitor (KSTI) reduced 83% in tempeh, on average, as compared to the value found in the grains. Phytic acid content was similar in both tempeh and the grains.

Puolustusvoimien runkona toimiva maavoimat on puolustusvoimiemme selkäranka. Nykyaikaisen taistelukentän moninaisuus ei ole vähentänyt hyvin koulutetun jalkaväkisotilaan tarvetta. Teknologian kehityksen myötä sotilaiden kannettava taakka on lisääntynyt merkittävästi. Tämän seurauksena sotilaiden taakankantokyky on merkittävä osa sotilaiden fyysistä toimintakykyä. Tämän tutkimuksen tarkoituksena on tutkia raskaan taakankantamisen fysiologisia vasteita taistelu- ja partiovarustuksella (29 kg ja 45 kg) suoritetuissa taakankantotesteissä. Tutkimukseen osallistui kahdeksan koehenkilöä Porin prikaatin kansainväliseen valmiusjoukkokoulutukseen valituista sotilaista (ikä 20 ±1 v, pituus 1,80±0.10 m, paino 77,9±12,3 kg, painoindeksi 23,9±2,4, maksimaalinen hapenottokyky 51,8± 4,2 ml kg-1∙min-1). Koehenkilöiltä mitattiin ennen taakankantotestejä maksimaalinen hapenottokyky (VO2Maks), maksimaalinen voima, lihaskestävyys sekä kehonkoostumus. Testeinä käytettiin suoraa juoksumattotestiä, isometrisiä maksimivoimatestejä, puolustusvoimien lihaskuntotestejä sekä bioimpedanssimenetelmää kehonkoostumuksen mittaamiseen. Taakankantotestit suoritettiin juoksumatolla siten, että 45 minuuttia käveltiin 4 km∙h-1 nopeudella ja 1,0 asteen kulmalla. Tämän jälkeen kulmaa ja kuormaa nostettiin kolmen minuutin välein aina 12 asteen kulmaan ja 6 km∙h-1 nopeuteen asti. Suoritusten aikana mitattiin ventilaatiota (VE), hapenkulutusta (VO2), sykettä (HR), subjektiivista tuntemusta (RPE) ja laktaattiarvoa (La). Tutkimustulosten perusteella taakankantotestit erosivat toisistaan submaksimaalisen suorituksen aikana VE- ja HR-arvojen perusteella (p<0,05). Taakankantotestien suoritusaika ja maksimaalinen hapenotto (ml∙min-1) olivat yhteydessä toisiinsa (29kg: r=0,80 p≤0,05, 45kg: r=0,92 p≤0,05). Suoritusaika ja suhteellinen hapenoton (ml∙kg-1∙min-1) arvo eivät olleet yhteydessä toisiinsa. Maksimaalinen isometrinen penkkipunnerrus oli yhteydessä kummankin taakankantotestin suoritusajan kanssa (29kg: r=0,75 p≤0,05, 45kg r=0,81 p≤0,05). Maksimaalinen hapenotto ja penkkipunnerrus olivat ainoat alkumittausten tulokset, jotka olivat yhteydessä molempien taakankantotestien loppuaikojen kanssa. Kehonpaino ja rasvaton massa olivat yhteydessä raskaamman taakan kanssa suoritetun loppuajan kanssa (r=0,72 p≤0,05), (r=0,74 p≤0,05). Vauhditon pituushyppy oli yhteydessä kevyemmän varustuksen loppuajan kanssa (r=0,80, p≤0,05). Regressioanalyysi selitti menestymistä raskaamman taakan kanssa pituuden ja penkkipunnerrustestien tuloksilla selitysasteen ollessa 79 % (p<0,01). Taakankantotestien ja alkumittausten tulosten perusteella riittävän suuret ylävartalon voimaominaisuudet ovat merkittävä tekijä taakankantotehtävissä menestymiseen. Maksimaalistahapenottokykyä vaaditaan kannettaessa taakkaa lähellä maksimaalista suorituskykyä. Kehonpainolla ei ole suoritusta heikentävää vaikutusta, painvastoin kehonpaino ja riittävä rasvattoman massan määrä näyttäisi tämän tutkimuksen mukaan parantavan taakankantokykyä.

Bromelia serra Griseb. (Bromeliaceae) es una hierba terrestre que habita en el sotobosque de bosques abiertos del Chaco y el Cerrado. En este trabajo se analizó el efecto de la temperatura, el clima lumínico y la escarificación sobre la germinación de semillas. Se evaluaron dos regímenes de temperatura (15/20 ºC y 20/30 ºC), tres ambientes lumínicos (Luz, Filtro y Oscuridad; 100% luz y 0.65 R:FR, 100% luz y 0.09 R:FR y sin luz respectivamente) y dos tratamientos de escarificación (Escarificación durante 1 min. con H2SO4 al 30% y Control sin escarificar). La temperatura fue un factor clave ya que solo se registró germinación (una reducida fracción de las semillas y en forma lenta) en el tratamiento a 20/30 ºC. Las semillas fueron indiferentes a la intensidad de luz, germinando tanto en condiciones de luz como de oscuridad. La calidad de la luz afectó la germinación, registrándose mayores porcentajes de germinación a altas relaciones R:FR. No se detectaron efectos de la escarificación en la germinación de las semillas. Los resultados de este trabajo aportan información básica sobre la germinación de semillas, que resultan útiles para el manejo de la especie.

This article evaluates the effects that monetary policy actions exert on Brazilian stock market. By the measures defined to estimate the surprise caused by Comitê de Política Monetária do Banco Central do Brasil (COPOM) decisions, it was verified that to a hypothetic unexpected 1% increase in the target rate is associated an 1.3% average fall of Bovespa Index. Additional tests did not show distinct reactions caused by direction decisions, neither evidences from relevant recent economic events or decision contexts having influences on the surprise responses.

The current classification system for spinal cord injury (SCI) considers only somatic information and neglects autonomic damage after injiuy. Heart rate variability (HRV) has the potential to be a valuable measure of cardiac autonomic control after (SCI). Five individuals with tetraplegia and four able-bodied controls underwent 1 min continuous ECG recordings during rest, after Metoprolol administration (max dose=3x5mg) and after Atropine administration (0.02mg/kg) in both supine and 40° head-up tilt. After Metoprolol administration there was a 61.8% decrease in the LF:HF ratio in the SCI participants suggesting that the LF:HF ratio is a reflection of cardiac sympathetic outflow. After Atropine administration there was a 99.1% decrease in the HF power in the SCI participants suggesting that HF power is highly representative of cardiac parasympathetic outflow. There were no significant differences between the SCI and able-bodied participants. Thus, HRV measures are a valid index of cardiac autonomic control after SCI.

Pyruvate dehydrogenase (PDH) is an important regulator of carbohydrate oxidation during exercise and its activity can be down-regulated by an increase in dietary fat. The purpose of this study was to determine the acute metabolic effects of differential dietary fatty acids on the activation of PDH in its active form (PDHa) at rest and at the onset of moderate-intensity exercise. University-aged male subjects (n=7) underwent 2 fat loading trials spaced at least 2 weeks apart. Subjects consumed saturated (SFA) or polyunsaturated (PUFA) fat over the course of 5 hours. Following this, participants cycled at 65% VO2 max for 15 min. Muscle biopsies were taken prior to and following fat loading and at 1 min exercise. Plasma free fatty acids increased from 0.15 ± 0.07 to 0.54 ± 0.19 mM over 5 hours with SFA and from 0.1 1 ± 0.04 to 0.35 ±0.13 mM with PUFA. PDHa activity was unchanged following fat loading, but increased at the onset of exercise in the SFA trial, from 1 .4 ± 0.4 to 2.2 ± 0.4 /xmol/min/kg wet wt. This effect was negated in the PUFA trial (1 .2 ± 0.3 to 1 .3 ± 0.3 pimol/min/kg wet wt.). PDH kinase (PDK) was unchanged in both trials, suggesting that the attenuation of PDHa activity with PUFA was a result of changes in the concentrations of intramitochondrial effectors, more specifically intramitochondrial NADH or Ca^*. Our findings suggest that attenuated PDHa activity participates in the preferential oxidation of PUFA during moderateintensity exercise.

Abstract Background Children have been shown to have higher lactate (LaTh) and ventilatory (VeTh) thresholds than adults, which might be explained by lower levels of type-II motor-unit (MU) recruitment. However, the electromyographic threshold (EMGTh), regarded as indicating the onset of accelerated type-II MU recruitment, has been investigated only in adults. Purpose To compare the relative exercise intensity at which the EMGTh occurs in boys versus men. Methods Participants were 21 men (23.4 ± 4.1 years) and 23 boys (11.1 ± 1.1 years), with similar habitual physical activity and peak oxygen consumption (VO2pk) (49.7 ± 5.5 vs. 50.1 ± 7.4 ml kg−1 min−1, respectively). Ramped cycle ergometry was conducted to volitional exhaustion with surface EMG recorded from the right and left vastus lateralis muscles throughout the test (~10 min). The composite right–left EMG root mean square (EMGRMS) was then calculated per pedal revolution. The EMGTh was then determined as the exercise intensity at the point of least residual sum of squares for any two regression line divisions of the EMGRMS plot. Results EMGTh was detected in 20/21 of the men (95.2 %) and only in 18/23 of the boys (78.3 %). The boys’ EMGTh was significantly higher than the men’s (86.4 ± 9.6 vs. 79.7 ± 10.0 % of peak power output at exhaustion; p < 0.05). The pattern was similar when EMGTh was expressed as percentage of VO2pk. Conclusions The boys’ higher EMGTh suggests delayed and hence lesser utilization of type-II MUs in progressive exercise, compared with men. The boys–men EMGTh differences were of similar magnitude as those shown for LaTh and VeTh, further suggesting a common underlying factor.
