The present work consists of the development and validation of analytical method for evaluation of glycyrrhizic acid, salicylic acid, and caffeine in chitosan-alginate nanoparticles by high performance liquid chromatography. Method validation investigated parameters such as linearity, precision, accuracy, robustness and specificity, which gave results within the acceptable range. The methods were applied to nanoparticles suspensions containing the drugs and were able to determine the entrapment efficiency successfully. The best entrapment efficiency was achieved with the glycyrrhizic acid (95.4%).

A simple analytical method for extraction and quantification of lutein colorant added to yogurt was developed and validated. The method allowed complete extraction of carotenoids using tetrahydrofuran in vortex, followed by centrifugation, partition to diethyl ether/petroleum ether, and drying. The carotenoids dissolved in ethanol were quantified by UV-Vis spectrophotometry. This method showed linearity in the range tested (1.41-13.42 µg g-1), limits of detection and quantification of 0.42 and 1.28 µg g-1, respectively, low relative standard deviation (3.4%) and recovery ranging from 95 to 103%. The method proved reliable for quantification of lutein added to yogurt.

Analytical methods were developed and validated to determine residues of veterinary drugs in buffalo milk and meat, using the QuEChERS method and gas chromatography-mass spectrometry. Both milk and meat, at 2 g of sample, 4 mL of acetonitrile, 0.8 g of MgSO4 and 0.2 g of NaCl, were used in the liquid-liquid partition, whereas 50 mg of C18, 50 mg of PSA and 150 mg of MgSO4 were employed in the dispersive solid-phase extraction (d-SPE). The methods showed sensitivity, precision and accuracy. The quantitation limits were in agreement with the maximum residue limit established by the Codex Alimentarius, FAO and WHO.

A multiresidue method using HPLC/DAD for the determination of fourteen pesticides in water based on SPE, using SDVB (styrene divynilbenzene copolymer) as adsorbent was validated. Recoveries from 61 to 120%, relative standard deviation between 2 and 15% and detection limits from 0.07 to 0.75 µg L-1 were obtained. It was applied to 66 surface water samples collected in a degraded area at the headwaters of São Lourenço river, Mato Grosso, Brazil. Eight pesticides were detected in concentrations ranging from 0.15 to 35.25 µg L-1. Considering ecotoxicological data, carbendazim and carbofuran may represent a risk to aquatic organisms. These results draw attention to the contamination of this vulnerable degraded area.

The need for effective and reliable quality control in products from pharmaceutical industries renders the analyses of their active ingredients and constituents of great importance. This study presents the theoretical basis of ¹H NMR for quantitative analyses and an example of the method validation according to Resolution RE Nº 899 by the Brazilian National Health Surveillance Agency (ANVISA), in which the compound paracetamol was the active ingredient. All evaluated parameters (selectivity, linearity, accuracy, repeatability and robustness) showed satisfactory results. It was concluded that a single NMR measurement provides structural and quantitative information of active components and excipients in the sample.

This paper reports the development and validation of a new analytical method using UV spectrophotometry to quantify carvedilol (CRV) in hydrophilic matrices and raw material. This method was shown to be linear, accurate, precise, robust and to have adequate limits of quantification and detection (LQ and LD, respectively), allowing its use in the dissolution test of hydrophilic matrices. The content of CRV determined through this method was compared with two previously validated methods based on the reference techniques of High Performance Liquid Chromatography (HPLC) and Potentiometric Titrations (PT). ANOVA confirmed the equivalence of these methods, showing no significant differences.

Blooms of cyanobacteria represent a public health risk due to their cyanotoxins such as microcystins. Liquid chromatography techniques to separate and quantify microcystins invariably use acetonitrile as the organic component of the mobile phase. The price and availability of acetonitrile together with its elevated toxicity encourage the validation of acetonitrile-free methods of microcystin analysis. In this work, methanol was employed as the organic solvent of the mobile phase and the validation method was performed with different environmental water samples. The method showed limits of detection between 0.17 and 0.25 µg/L and of quantification between 0.55 and 0.82 µg/L for the microcystin variants: -RR, -YR, -LR, -LA.

Difenoconazole residues in strawberry fruit cultivated in pots were estimated using the solid-liquid extraction with low temperature partition (SLE/LTP) method for sample preparation and gas chromatography with electron capture detection (GC/ECD) for analysis. The optimized method presented excellent recovery values from fortified samples and reproducibility (average recovery values ≥ 98%; CV values < 15%). Linearity of response was demonstrated (r = 0.995) with a detection limit of 9 µg kg-1. The method was successfully applied for the determination of difenoconazole residues in strawberries. Based on these results, the fungicide dissipates quickly, but the residual concentration increases after multiple applications.

Phenylpropanoid glycoside verbascoside was isolated and identified from the ethyl acetate fraction of the aerial parts of Buddleja stachyoides Cham. & Schltdl. by 1H-NMR. A method using high-performance liquid chromatography has been developed and validated for determination of verbascoside in alcoholic crude extract of the aerial parts of B. stachyoides. Analysis was performed on a Phenomenex® Gemini-NX C18 analytical column (250 mm × 4.6 mm; 5 µm) using a mobile phase (pump A - aqueous solution containing H2SO4 (0.01 M), H3PO4 (0.4%), and (C2H5)2NH (0.4%); pump B - methanol:aqueous (95:5) solution containing H2SO4 (0.05 M), H3PO4 (2%), and (C2H5)2NH (0.2%); pump C - acetonitrile:aqueous (90:10) solution containing H2SO4 (0.05 M) and H3PO4 (2%)) and a diode array detector at 325 nm. The method was validated in accordance with ANVISA guidelines and may be applied to quality control of herbal medicine with aerial parts of B. stachyoides.

In this study, a method of solid-liquid extraction and purification at low temperature (SLE-PLT) to determine 16 polycyclic aromatic hydrocarbons (PAHs) in sewage sludge was optimized and validated. The analyses were performed by HPLC-UV. The extraction phase, homogenization procedure, influence of pH, ionic strength and clean-up of the extracts were optimized. Recoveries were higher than 63.4% for 11 PAHs. The correlation coefficients were greater than 0.99 and limits of detection and quantitation were less than 0.060 and 0.15 µg g-1, respectively. These values were lower than the maximum residue limits of PAHs established by European legislation. SLE-PLT proved a more practical, economical method with fewer steps compared to Soxhlet extraction (reference method) for PAHs in sewage sludge.

Validation of a rapid method for the determination of ascorbic, citric, fumaric and tartaric acids in stored pulp fruit and its application as a quality parameter was performed. The validation parameters showed that for the four evaluated acids, the method presented low limits of detection (LOD) and quantification (LOQ), indicating good precision and accuracy, thus representing an important tool for quality assessment of stored fruit pulp. The results showed that the concentration of organic acids generally decreased with longer storage time in the fruit pulp under study. Amongst all the organic acids under investigation, ascorbic proved the least stable.

17α-methyltestosterone (17α-MT) is a hormone used in medicine and aquaculture which can be found in natural waters. It is considered an emerging organic pollutant and its accurate and precise analysis is required. Therefore, this study presents the in-house validation of a new voltammetric methodology for 17α-MT analysis in natural waters. A hanging mercury drop electrode and NaOH 5.0×10-3 mol L-1 solution as the electrolyte were employed in the voltammetric determinations. To confirm the presence of 17α-MT in water samples quantitative analysis was performed by ESI-MS/MS. The methodology exhibited good selectivity, linearity, accuracy (recovery of between 100.4 and 108.8%) and precision (RSDs for repeatability of 2.85% and for intermediate precision of 14.10%, with n = 5, respectively). LOD and LOQ were 3.07 µg L-1 and 10.78 µg L-1, respectively. The methodology was suitable for determination of 17α-MT in natural waters due to low-cost, good performance and no sample pre-treatment required.

Através de quatro experimentos realizados em campo, no período de setembro/98 a dezembro/99, avaliou-se a aplicabilidade de dois sistemas de previsão no controle da queima das folhas da cenoura (Daucus carotae). Seis esquemas de aplicação de fungicida (protetor ou sistêmico), baseados no acúmulo de valores diários de severidade (VDS) para Alternaria dauci ou equivalentes de infecção (EI) para Cercospora carotae, foram comparados a dois calendários fixos de tratamento e uma testemunha sem fungicida. Enquanto o número de VDS registrados por um coletor eletrônico (Colpam) variou de 25 a 94 por ciclo da cultura, nenhum EI foi detectado ao longo dos quatro experimentos. Com a adoção do sistema de VDS, o número de aplicações do fungicida protetor clorotalonil diminuiu de 7,5 (fixo, sete dias) para 3,5 (VDS 3-5), 2,7 (VDS 6-8) e dois (VDS 9-11) por cultivo. Nas mesmas condições, o fungicida sistêmico metconazole foi aplicado 3,7 (fixo, 14 dias), 2,5 (VDS 3-5), 1,7 (VDS 6-8) e 1,5 (VDS 9-11) vezes. A relação entre a área abaixo da curva de progresso de doença (AACPD) e o peso de raiz foi significativa em dois experimentos (P = 0,005 e 0,016). Quanto ao rendimento de raízes, o limiar de 3-5 VDS igualou-se ao esquema de calendário fixo em 3/4 das situações. Portanto, nas condições locais, o controle da queima das folhas pelo sistema de VDSs está condicionado ao limite de 3-5 VDS e à utilização de fungicidas sistêmicos com eficácia igual ou superior à do metconazole.
