The properties of a previously unknown enzyme, denominated cyclodextrin glycosyltransferase, produced from Bacillus lehensis, were evaluated using affinity chromatography for protein purification. Enzyme characteristics (optimum pH and temperature; pH and temperature stability), the influence of substances on the enzyme activity, enzyme kinetics, and cyclodextrin production were analysed. Cyclodextrin glycosyltransferase was purified up to 320.74-fold by affinity chromatography using beta-cyclodextrin as the binder and it exhibited 8.71% activity recovery. This enzyme is a monomer with a molecular weight of 81.27 kDa, as estimated by SDS-PAGE. Optimum temperature and pH for cydodextrin glycosyltransferase were 55 degrees C and 8.0, respectively. The Michaelis-Menten constant was 8.62 g/l during maximum velocity of 0.858 g/l.h.

The biodegradability properties of poly(epsilon-caprolactone) (PCL) and modified adipate-starch (AS) blends, using Edenol-3203 (E) as a starch plasticizer, were investigated in laboratory by burial tests of the samples in previously analyzed agricultural soil. The biodegradation process was carried out using the respirometric test according to ASTM D 5988-96, and the mineralization was followed by both variables such as carbon dioxide evolution and mass loss. The results indicated that the presence of AS-E accelerated the biodegradation rate as expected.

Starch isolated from non-edible Aesculus hippocastanum seeds was characterized and used for preparing starch-based materials. The apparent amylose content of the isolated starch was 33.1%. The size of starch granules ranged from 0.7 to 35 pm, and correlated with the shape of granules (spherical, oval and irregular). The chain length distribution profile of amylopectin showed two peaks, at polymerization degree (DP) of 12 and 41-43. Around 53% of branch unit chains had DP in the range of 11-20. A. hippocastanum starch displayed a typical C-type pattern and the maximum decomposition temperature was 317 degrees C.Thermoplastic starch (TPS) prepared from A. hippocastanum with glycerol and processed by melt blending exhibited adequate mechanical and thermal properties. In contrast, plasticized TPS with glycerol:malic acid (1:1) showed lower thermal stability and a pasty and sticky behavior, indicating that malic acid accelerates degradation of starch during processing. The aim of this study is to analyze the impact of food shortage on growth performance, by means of energetic reserves (proteins, glycogen and lipids) mobilization and hepatopancreas cells analysis in C. quadricarinatus juveniles maintained in groups, as well as the effect on culture water quality. Two experiments were performed, each of them with two feeding regimes during 45 days. The Control feeding regime, in which crayfish were fed daily (once a day) throughout the experimental period (DF), and the Cyclic feeding regime, in which juveniles were fed for 2 or 4 days (once a day) followed by 2 or 4 days of food deprivation (2F/2D and 4F/4D, respectively) in repeated cycles. Cyclic feeding influenced growth, biochemical composition from hepatopancreas and muscle, and water quality. Juveniles cyclically fed were unable to maintain a normal growth trajectory during 45 days. Apparent feed conversion ratio, apparent protein efficiency ratio, hepatosomatic index and relative pleon mass were similar in cyclic and daily fed animals and no structural damage was found in the hepatopancreas of juveniles subjected to cyclic feeding. The novelty of this study was the significant accumulation of proteins in pleonal muscle in both cyclic feeding regimes (approx. 18%) suggesting that the storage of this constitutive material during food shortage may be an adaptation for a compensatory growth when food becomes abundant again. The cyclic feeding regimes had a positive effect on water quality decreasing inorganic nitrogen concentration. This was due to the reduction in the amount of animal excretes and feces in the group that received approx. 50% less feed. Additionally, water pH was higher in cyclic feeding tanks, as a result of lower organic matter decomposition and consequent release of CO2. Accordingly, total ammonia in the water was significantly lower for the cyclic feeding regimes compared to their respective controls. This study suggests that the protocol of cyclic feeding could be applied at least 45 days in 1 g juveniles maintained in group conditions, without affecting the energetic reserves and hepatopancreas structure, emphasizing the high tolerance of this species to food restriction.

The aims of this study were to assess the turmeric oleoresin microencapsulation by freeze-drying with modified starch/gelatin and to evaluate its stability during storage at different temperatures and light. Encapsulated turmeric oleoresin w stored at −20, 25 and 60C, in the absence of light, and at 25C in the presence of light, and analyzed over a period of 6 weeks for curcumin and total phenolic contents and color. The different concentrations of wall material showed no significant effect on the curcumin retention. The best conditions for microencapsulation of turmeric oleoresin were: wall material composed of 30 g/100 g of modified starch + 1 g/100 g gelatin and mechanical homogenization. Encapsulated material was more stable during storage at −20C and less stable at 25C in the presence of light.
