Introduction: La circulation extracorporelle (CEC) peut entraîner une dysfonction endothéliale pulmonaire et l’hypertension pulmonaire. Le SN50 agit au niveau de la signalisation cellulaire pour prévenir ces réactions à la CEC et pourrait renverser la dysfonction endothéliale pulmonaire post-CEC sans effets néfastes sur l’hémodynamie. Méthodes: Quatre groups de porcs ont reçu un parmi quatre traîtements avant de subir 90 minutes de CEC et 60 minutes de reperfusion: (1) milrinone nébulisé; (2) sildenafil nébulisé; (3) placebo nébulisé; et (4) SN-50 intraveineux. Un monitoring hémodynamique invasif a été utilisé. La réactivité vasculaire des artères pulmonaires de deuxième ordre a été évaluée face à l’acétylcholine et la bradykinine. Résultats: Le sildénafil produit une augmentation significative de la pression de l’artère pulmonaire (PAP) moyenne à 60 minutes de reperfusion par rapport au début de la chirurgie. Les relaxations dépendantes de l’endothélium face à la bradykinine étaient meilleurs dans les groupes milrinone et SN-50 et surtout dans le groupe sildénafil par rapport au groupe placébo. Le SN-50 produisait de moins bonnes relaxations dépendantes de l’endothélium face à l’acétylcholine que les autres traitements incluant placébo. Conclusion: Le sildénafil prévient mieux la dysfonction endothéliale pulmonaire que les autres traitements. Les bénéfices du SN-50 sont possiblement sous-estimés vu que la dose n’a pas pu être ajustée à la durée de CEC. Le sildenafil inhalé mérite une étude plus importante chez l’humain et le SN-50 dans un model de CEC animal.

Les cellules dendritiques (DC) sont parmi les premières cellules à rencontrer le virus d’immunodéficience humaine (VIH) au niveau des muqueuses. De plus, le fait que les DC sont, de manière directe ou indirecte par le virus et ses composantes, altérées tant par leur nombre, leur phénotype et leur fonction suggère leur implication dans les dérégulations des cellules B. Selon cette hypothèse, des études longitudinales impliquant des individus infectés au VIH-1 présentant différents profils de progression clinique menées dans notre laboratoire ont démontré que les altérations des cellules B sont concomitantes à une augmentation de l’expression de BLyS/BAFF dans le sang ainsi que par les DC myéloïdes (mDC) sanguines. De plus, lors de travaux antérieurs utilisant le modèle murin VIH-transgénique, les altérations des cellules B ont démontré une implication des DC et d’un excès de BLyS/BAFF, et ce, dépendamment du facteur négatif du VIH (Nef). Dans cette optique, nous investiguons dans cette présente étude l’implication de Nef dans la modulation du phénotype des DC ainsi que dans les dérégulations des cellules B. Chez tous les patients virémiques infectés au VIH-1, nous avons détecté la présence de Nef dans le plasma ainsi qu’au niveau des mDC et de leurs précurseurs d’origine monocytaire, tout au long du suivi de la progression clinique et au-delà de la thérapie antirétrovirale (ART). La surexpression de BLyS/BAFF est associée à la présence de Nef au niveau des mDC et de leur précurseur.. Des essais in vitro ont permis de démontrer l’induction d’un phénotype proinflammatoire par des mDC dérivés de monocytes lorsqu’en présence de Nef soluble, via l’augmentation de l’expression de BLyS/BAFF et de TNF-α, et où cet effet est bloqué par l’ajout de l’acide rétinoïque. Nos résultats suggèrent donc que Nef est impliquée dans le déclenchement et la persistance des dérégulations des cellules B retrouvées chez les individus infectés au VIH-1. Basé sur nos observations, une thérapie adjointe impliquant le blocage de BLyS/BAFF et/ou Nef pourrait contribuer au contrôle de l’inflammation et des altérations des cellules B. De plus, la quantification de Nef post-ART pourrait s’avérer utile dans l’évaluation du statut des réservoirs. Précédemment, nous avons démontré que les dérégulations des cellules B sanguines de ces mêmes individus présentant un profil de progression rapide et classique sont accompagnées par l’augmentation de la fréquence d’une population partageant des caractéristiques des cellules B transitionnelles immatures (TI) et des cellules B de la zone marginale (ZM), que nous avons nommé les cellules B précurseur de la ZM. Toutefois, cette population est préservée chez les contrôleurs élites, chez qui nous avons trouvé une diminution significative de la fréquence des cellules B de la ZM présentant des marqueurs phénotypiques plus matures. Récemment, ces cellules ont été associées à un potentiel de fonction régulatrice (Breg), motivant ainsi notre poursuite, dans cette étude, de la caractérisation de ces cellules B. Comme pour les individus non infectés au VIH-1, nous avons démontré que les cellules B matures de la ZM contrôlent leur capacité de production d’IL-10 chez les contrôleurs élites, contrairement à une augmentation chez les progresseurs rapides et classiques. Aussi, les cellules B précurseur de la ZM des contrôleurs élites fournissent une expression importante de LT-α lorsque comparés aux individus non infectés au VIH-1, alors que cet apport de LT-α est attribué aux cellules B TI chez les progresseurs. Le contrôle de la progression clinique semble associé à un ratio en faveur de LT-α vs IL-10 au niveau des cellules B précurseur de la ZM. Nos résultats suggèrent qu’un maintien de l’intégrité du potentiel régulateur ainsi qu’une expression augmentée de LT-α par les cellules B de première ligne, telles les populations de la ZM, sont impliqués dans le contrôle de la progression clinique du VIH-1, possiblement par leur contribution à la modulation et l’homéostasie immunitaire. De telles populations doivent être considérées lors de l’élaboration de vaccins, ces derniers cherchant à générer une réponse protectrice de première ligne et adaptative.

La multiplication dans le corps de Galois à 2^m éléments (i.e. GF(2^m)) est une opérations très importante pour les applications de la théorie des correcteurs et de la cryptographie. Dans ce mémoire, nous nous intéressons aux réalisations parallèles de multiplicateurs dans GF(2^m) lorsque ce dernier est généré par des trinômes irréductibles. Notre point de départ est le multiplicateur de Montgomery qui calcule A(x)B(x)x^(-u) efficacement, étant donné A(x), B(x) in GF(2^m) pour u choisi judicieusement. Nous étudions ensuite l'algorithme diviser pour régner PCHS qui permet de partitionner les multiplicandes d'un produit dans GF(2^m) lorsque m est impair. Nous l'appliquons pour la partitionnement de A(x) et de B(x) dans la multiplication de Montgomery A(x)B(x)x^(-u) pour GF(2^m) même si m est pair. Basé sur cette nouvelle approche, nous construisons un multiplicateur dans GF(2^m) généré par des trinôme irréductibles. Une nouvelle astuce de réutilisation des résultats intermédiaires nous permet d'éliminer plusieurs portes XOR redondantes. Les complexités de temps (i.e. le délais) et d'espace (i.e. le nombre de portes logiques) du nouveau multiplicateur sont ensuite analysées: 1. Le nouveau multiplicateur demande environ 25% moins de portes logiques que les multiplicateurs de Montgomery et de Mastrovito lorsque GF(2^m) est généré par des trinômes irréductible et m est suffisamment grand. Le nombre de portes du nouveau multiplicateur est presque identique à celui du multiplicateur de Karatsuba proposé par Elia. 2. Le délai de calcul du nouveau multiplicateur excède celui des meilleurs multiplicateurs d'au plus deux évaluations de portes XOR. 3. Nous determinons le délai et le nombre de portes logiques du nouveau multiplicateur sur les deux corps de Galois recommandés par le National Institute of Standards and Technology (NIST). Nous montrons que notre multiplicateurs contient 15% moins de portes logiques que les multiplicateurs de Montgomery et de Mastrovito au coût d'un délai d'au plus une porte XOR supplémentaire. De plus, notre multiplicateur a un délai d'une porte XOR moindre que celui du multiplicateur d'Elia au coût d'une augmentation de moins de 1% du nombre total de portes logiques.

Alkylation of phenol with methanol has been carried out over Sn-La and Sn-Sm mixed oxides of varying compositions at 623 K in a vapour phase flow reactor. It is found that the product selectivity is greatly influenced by the acid-base properties of the catalysts. Ortho-cresol formation is favoured over catalysts with weak acid sites whereas formation of 2,6-xylenol occurs in the presence of stronger acid sites. The cyclohexanol decomposition reaction and titrimetric method using Hammett indicators have been employed to elucidate the acid-base properties of the catalysts.

The catalyst compositions of the Zn1−xCOxFe2O4 (x= 0, 0.2, 0.5, 0.8 and 1.0) spiel series possessing ‘x’ values, x less than or equal to 0.5, are unique for selective N-monomethylation of aniline using methanol as the alkylating agent. Since dimethyl carbonate (DMC) is another potential non-toxic alkylating agent, alkylation of aniline was investigated over various Zn–Co ferrites using DMC as the alkylating agent. The merits and demerits of the two alkylating agents are compared. Catalytic activity followed a similar trend with respect to the composition of the ferrospinel systems. DMC is active at comparatively low temperature, where methanol shows only mild activity. However, on the selectivity basis, DMC as an alkylating agent could not compete with methanol, since the former gave appreciable amounts of N,N-dimethylaniline (NNDMA) even at low temperature where methanol gave nearly 99% N-methylaniline (NMA) selectivity. As in the case of methanol, DMC also did not give any C-alkylated products.

The reaction of aniline with methanol was carried out over Zn1-xNixFe2O4 (x= 0, 0.2, 0.5, 0.8 and 1) type systems in a fixed-bed down-flow reactor. It was observed that systems possessing low ``x'' values are highly selective and active for mono N-alkylation of aniline leading to N-methyl aniline. Selectivity for N-methyl aniline over ZnFe2O4 was more than 99% under the optimized reaction conditions. Even at methanol to aniline molar ratio of 2, the yield of N-methyl aniline was nearly 55.5%, whereas its yield exceeded 67% at the molar ratio of 7. The Lewis acid sites of the catalysts are mainly responsible for the good catalytic performance. Cation distribution in the spinel lattice influences their acido-basic properties, and hence, these factors have been considered as helpful to evaluate the activity and stability of the systems.

Lanthana modified sol-gel titania is prepared through particulate sol-gel route and the physico-chemical characterizations of the prepared systems were done using X-Ray diffration, EDX, BET surface area-pore volume measurements and TG-DTG analysis. Benzophenone was observed to be the sole product in the TiO2 photocatalyzed oxidation of benzhydrol in oxygen purged acetonitrile. The influence of various parameters, like irradiation time, amount of catalyst, concentration of the catalyst and other factors on the photocatalytic oxidation has been investigated. The proposed mechanism envisages the involvement of a superoxide radical anion.

This doctoral thesis addresses the growing concern about the significant changes in the climatic and weather patterns due to the aerosol loading that have taken place in the Indo Gangetic Plain(IGP)which includes most of the Northern Indian region. The study region comprises of major industrial cities in India (New Delhi, Kanpur, Allahabad, Jamshedpur and Kolkata). Northern and central parts of India are one of the most thickly populated areas in the world and have the most intensely farmed areas. Rapid increase in population and urbanization has resulted in an abrupt increase in aerosol concentrations in recent years. The IGP has a major source of coal; therefore most of the industries including numerous thermal power plants that run on coal are located around this region. They inject copious amount of aerosols into the atmosphere. Moreover, the transport of dust aerosols from arid locations is prevalent during the dry months which increase the aerosol loading in theatmosphere. The topography of the place is also ideal for the congregation of aerosols. It is bounded by the Himalayas in the north, Thar Desert in the west, the Vindhyan range in the south and Brahmaputra ridge in the east. During the non‐monsoon months (October to May) the weather in the location is dry with very little rainfall. Surface winds are weak during most of the time in this dry season. The aerosols that reach the location by means of long distance transport and from regional sources get accumulated under these favourable conditions. The increase in aerosol concentration due to the complex combination of aerosol transport and anthropogenic factors mixed with the contribution from the natural sources alters the optical properties and the life time of clouds in the region. The associated perturbations in radiative balance have a significant impact on the meteorological parameters and this in turn determines the precipitation forming process. Therefore, any change in weather which disturbs the normal hydrological pattern is alarming in the socio‐economic point of view. Hence, the main focus of this work is to determine the variation in transport and distribution of aerosols in the region and to understand the interaction of these aerosols with meteorological parameters and cloud properties.

Regional climate models are becoming increasingly popular to provide high resolution climate change information for impacts assessments to inform adaptation options. Many countries and provinces requiring these assessments are as small as 200,000 km2 in size, significantly smaller than an ideal domain needed for successful applications of one-way nested regional climate models. Therefore assessments on sub-regional scales (e.g., river basins) are generally carried out using climate change simulations performed for relatively larger regions. Here we show that the seasonal mean hydrological cycle and the day-to-day precipitation variations of a sub-region within the model domain are sensitive to the domain size, even though the large scale circulation features over the region are largely insensitive. On seasonal timescales, the relatively smaller domains intensify the hydrological cycle by increasing the net transport of moisture into the study region and thereby enhancing the precipitation and local recycling of moisture. On daily timescales, the simulations run over smaller domains produce higher number of moderate precipitation days in the sub-region relative to the corresponding larger domain simulations. An assessment of daily variations of water vapor and the vertical velocity within the sub-region indicates that the smaller domains may favor more frequent moderate uplifting and subsequent precipitation in the region. The results remained largely insensitive to the horizontal resolution of the model, indicating the robustness of the domain size influence on the regional model solutions. These domain size dependent precipitation characteristics have the potential to add one more level of uncertainty to the downscaled projections.

Después de los ataques del 11 de septiembre de 2001, Pakistán se ha convertido en uno de los principales aliados de Estados Unidos para combatir las fuerzas terroristas de Al-Qaeda y los talibán en el centro y sur de Asia. La alianza bilateral no ha manifestado resultados determinantes para aliviar los problemas de seguridad en Pakistán, por el contrario, la yihad islámica se ha fortalecido en su población y los ataques terroristas que atentan contra la población civil y el aparato estatal se vuelven cada vez mas frecuentes, en razón de lo anterior, los grupos insurgentes en Pakistán han fortalecido su capacidad operativa y expandido su escenario de influencia.

We previously established an 80 kb haplotype upstream of TNFSF4 as a susceptibility locus in the autoimmune disease SLE. SLE-associated alleles at this locus are associated with inflammatory disorders, including atherosclerosis and ischaemic stroke. In Europeans, the TNFSF4 causal variants have remained elusive due to strong linkage disequilibrium exhibited by alleles spanning the region. Using a trans-ancestral approach to fine-map the locus, utilising 17,900 SLE and control subjects including Amerindian/Hispanics (1348 cases, 717 controls), African-Americans (AA) (1529, 2048) and better powered cohorts of Europeans and East Asians, we find strong association of risk alleles in all ethnicities; the AA association replicates in African-American Gullah (152,122). The best evidence of association comes from two adjacent markers: rs2205960-T (P = 1.71×10-34, OR = 1.43[1.26-1.60]) and rs1234317-T (P = 1.16×10-28, OR = 1.38[1.24-1.54]). Inference of fine-scale recombination rates for all populations tested finds the 80 kb risk and non-risk haplotypes in all except African-Americans. In this population the decay of recombination equates to an 11 kb risk haplotype, anchored in the 5′ region proximal to TNFSF4 and tagged by rs2205960-T after 1000 Genomes phase 1 (v3) imputation. Conditional regression analyses delineate the 5′ risk signal to rs2205960-T and the independent non-risk signal to rs1234314-C. Our case-only and SLE-control cohorts demonstrate robust association of rs2205960-T with autoantibody production. The rs2205960-T is predicted to form part of a decameric motif which binds NF-κBp65 with increased affinity compared to rs2205960-G. ChIP-seq data also indicate NF-κB interaction with the DNA sequence at this position in LCL cells. Our research suggests association of rs2205960-T with SLE across multiple groups and an independent non-risk signal at rs1234314-C. rs2205960-T is associated with autoantibody production and lymphopenia. Our data confirm a global signal at TNFSF4 and a role for the expressed product at multiple stages of lymphocyte dysregulation during SLE pathogenesis. Siguiendo un marco teórico integrado por varios autores entorno a los sistemas de control de gestión a lo largo de varias décadas, este trabajo pretende estudiar y contrastar la relación entre el desarrollo de dichos sistemas y los recursos y capacidades. Para tal fin, se desarrolló un estudio de caso en Teleperformance Colombia (TC), una empresa dedicada a prestación de servicio de tercerización de procesos o business process outsourcing. En el estudio se establecieron dos variables para evaluar el desarrollo de sistema de control de gestión: el diseño y el uso. A su vez, para cada uno de ellos, se definieron los indicadores y preguntas que permitieran realizar la observación y posterior análisis. De igual manera, se seleccionaron los recursos y capacidades más importantes para el desarrollo del negocio: innovación, aprendizaje organizacional y capital humano. Sobre estos se validó la existencia de relación con el SCG implementado en TC. La información obtenida fue analizada y contrastada a través de pruebas estadísticas ampliamente utilizadas en este tipo de estudios en las ciencias sociales. Finalmente, se analizaron seis posibles relaciones de las cuales, solamente se ratificó el relacionamiento positivo entre uso de sistema de control gestión y el recurso y capacidad capital humano. El resto de relacionamientos, refutaron los planteamientos teóricos que establecían cierta influencia de los sistemas de control de gestión sobre recursos y capacidades de innovación y aprendizaje organizacional.

The influence on the summer flow over Asia of both the orographic and thermal forcing of the Tibetan Plateau is investigated using a sequence of idealised experiments with a global primitive equation model. The zonally averaged flow is prescribed and both realistic and idealised orography and heating are used. There is some similarity between the responses to the two forcings when applied separately. The upper tropospheric Tibetan anticyclone is predominantly forced by the heating but also weakly by the orography. Below this, both forcings tend to give air descending in an equatorward anticyclonic circulation down the isentropes to the west and rising in a similar poleward circulation to the east. However the heating-only response has a strong ascending southwesterly flow that is guided around the south and south-east of the orography when it is included. On the northern side, the westerly flow over the orography gives ascent on the upslope and descent on the downslope. It is found that heating over the Plateau leads to a potential vorticity (PV) minimum and that if it is sufficiently strong the flow is unstable, producing a quasi-biweekly oscillation. During this oscillation the Tibetan anticyclone changes between a single centre over the southwestern side of the Plateau and a split/double structure with centres over China and the Middle East. These characteristics are similar to observed variability in the region. Associated with this quasi-biweekly oscillation are significant variations in the strength of the ascent over the Plateau and the Rossby wave pattern over the North Pacific. The origin of the variability is instability associated with the zonally extended potential vorticity PV minimum on a θ-surface, as proposed by Hsu and Plumb (2000). This minimum is due to the tendency to reduce the PV above the heating over the Plateau and to advection by the consequent anticyclone of high PV around from the east and low PV to the west. The deep convection to the south and southeast of the Plateau tends to suppress the quasi-biweekly oscillation because the low PV produced above it acts to reduce the meridional PV gradient reversal. The occurrence of the oscillation depends on the relative magnitude of the heating in the two regions.

The atmospheric composition of the central North Atlantic region has been sampled using the FAAM BAe146 instrumented aircraft during the Intercontinental Transport of Ozone and Precursors (ITOP) campaign, part of the wider International Consortium for Atmospheric Research on Transport and Transformation (ICARTT). This paper presents an overview of the ITOP campaign. Between late July and early August 2004, twelve flights comprising 72 hours of measurement were made in a region from approximately 20 to 40°W and 33 to 47°N centered on Faial Island, Azores, ranging in altitude from 50 to 9000 m. The vertical profiles of O3 and CO are consistent with previous observations made in this region during 1997 and our knowledge of the seasonal cycles within the region. A cluster analysis technique is used to partition the data set into air mass types with distinct chemical signatures. Six clusters provide a suitable balance between cluster generality and specificity. The clusters are labeled as biomass burning, low level outflow, upper level outflow, moist lower troposphere, marine and upper troposphere. During this summer, boreal forest fire emissions from Alaska and northern Canada were found to provide a major perturbation of tropospheric composition in CO, PAN, organic compounds and aerosol. Anthropogenic influenced air from the continental boundary layer of the USA was clearly observed running above the marine boundary layer right across the mid-Atlantic, retaining high pollution levels in VOCs and sulfate aerosol. Upper level outflow events were found to have far lower sulfate aerosol, resulting from washout on ascent, but much higher PAN associated with the colder temperatures. Lagrangian links with flights of other aircraft over the USA and Europe show that such signatures are maintained many days downwind of emission regions. Some other features of the data set are highlighted, including the strong perturbations to many VOCs and OVOCs in this remote region.

A case of long-range transport of a biomass burning plume from Alaska to Europe is analyzed using a Lagrangian approach. This plume was sampled several times in the free troposphere over North America, the North Atlantic and Europe by three different aircraft during the IGAC Lagrangian 2K4 experiment which was part of the ICARTT/ITOP measurement intensive in summer 2004. Measurements in the plume showed enhanced values of CO, VOCs and NOy, mainly in form of PAN. Observed O3 levels increased by 17 ppbv over 5 days. A photochemical trajectory model, CiTTyCAT, was used to examine processes responsible for the chemical evolution of the plume. The model was initialized with upwind data and compared with downwind measurements. The influence of high aerosol loading on photolysis rates in the plume was investigated using in situ aerosol measurements in the plume and lidar retrievals of optical depth as input into a photolysis code (Fast-J), run in the model. Significant impacts on photochemistry are found with a decrease of 18% in O3 production and 24% in O3 destruction over 5 days when including aerosols. The plume is found to be chemically active with large O3 increases attributed primarily to PAN decomposition during descent of the plume toward Europe. The predicted O3 changes are very dependent on temperature changes during transport and also on water vapor levels in the lower troposphere which can lead to O3 destruction. Simulation of mixing/dilution was necessary to reproduce observed pollutant levels in the plume. Mixing was simulated using background concentrations from measurements in air masses in close proximity to the plume, and mixing timescales (averaging 6.25 days) were derived from CO changes. Observed and simulated O3/CO correlations in the plume were also compared in order to evaluate the photochemistry in the model. Observed slopes change from negative to positive over 5 days. This change, which can be attributed largely to photochemistry, is well reproduced by multiple model runs even if slope values are slightly underestimated suggesting a small underestimation in modeled photochemical O3 production. The possible impact of this biomass burning plume on O3 levels in the European boundary layer was also examined by running the model for a further 5 days and comparing with data collected at surface sites, such as Jungfraujoch, which showed small O3 increases and elevated CO levels. The model predicts significant changes in O3 over the entire 10 day period due to photochemistry but the signal is largely lost because of the effects of dilution. However, measurements in several other BB plumes over Europe show that O3 impact of Alaskan fires can be potentially significant over Europe.
