Small mammals can impede tree regeneration by injuring seedlings and saplings in several ways. One fatal way is by severing their stems, but apparently this type of predation is not well-studied in tropical rain forest. Here, we report on the incidence of 'stem-cutting' to new, wild seedlings of two locally dominant, canopy tree species monitored in 40 paired forest understorey and gap-habitat areas in Korup, Cameroon following a 2007 masting event. In gap areas, which are required for the upward growth and sapling recruitment of both species, 137 seedlings of the long-lived, light-demanding, fast-growing large tropical tree (Microberlinia bisulcata) were highly susceptible to stem-cutting (83% of deaths) - it killed 39% of all seedlings over a c. 2-y period. In stark contrast, seedlings of the more shade-tolerant, slower-growing tree species (Tetraberlinia bifoliolata) were hardly attacked (4.3%). In the understorey, however, stem-cutting was virtually absent. Across the gap areas, the incidence of stem-cutting of M. bisulcata seedlings showed significant spatial variation that could not be explained significantly by either canopy openness or Janzen-Connell type effects (proximity and basal area of conspecific adult trees). To examine physical and chemical traits that might explain the species difference to being cut, bark and wood tissues were collected from a separate sample of seedlings in gaps (i.e. not monitored for stem-cutting). These analyses suggested that, compared with T. bifoliolata, the lower stem density, higher Mg and K and fatty acid concentrations in bark, and fewer phenolic and terpene compounds in M. bisulcata seedlings made them more palatable and attractive to small-mammal predators, likely rodents. We conclude that selective stem-cutting is a potent countervailing force to the current local canopy dominance of the grove-forming M. bisulcata by limiting the recruitment and abundance of its saplings. Given the ubiquity of gaps and ground-dwelling rodents in pantropical forests, it would be surprising if this form of lethal browsing was restricted to Korup.

The purpose of this dissertation research was to investigate potential mechanisms through which mutations in two ubiquitously expressed genes, inosine monophosphate dehydrogenase 1 (IMPDH1) and pre-mRNA processing factor 31 (PRPF31), cause autosomal dominant retinitis pigmentosa (adRP) but have no other apparent clinical consequences. Basic properties of the gene and gene product, such as expression and protein levels, were examined. The purpose of our research is to understand the genetic basis of inherited retinopathies such as retinitis pigmentosa (RP). RP is a heterogeneous retinal dystrophy that affects approximately one in 3,700 individuals, making it the most common heritable retinal degenerative disease worldwide. Currently, mutations in 35 genes are known to cause RP and additional loci have been mapped but the underlying gene is not yet known. Often the genes associated with RP are integral to the biological processes underlying vision, making their role in retinal disease easy to explain. However, the mechanisms by which other genes cause RP are not apparent, especially widely-expressed genes. For IMPDH1, this research characterized the enzymatic properties of retinal isoforms. Results show that the retinal isoforms have enzymatic functions similar to the previously known canonical IMPDH1 whether or not an adRP pigmentosa mutation is included in the protein. For PRPF31, this research tested the hypothesis that functional haploinsufficiency is the cause of disease and relates to nonpenetrance in some individuals. Studies in patients with known mutations show that haploinsufficiency is the likely cause of disease, however, we did not confirm that non-penetrant individuals are protected from disease via increased expression of the wild type allele. Information gleaned from these functional studies, and the testing methods developed in tandem, will contribute to future research on disease mechanism related to adRP. ^

The Retinoblastoma tumor suppressor gene (RB) plays a role in a variety of human cancers. Experimental analyses have indicated that the protein product of the RB gene (pRb) plays a role in cell cycle regulation, and that this protein is required in cellular differentiation, senescence, and cell survival. pRb function is dependent on its ability to bind to cellular factors. There are multiple protein binding domains within pRb. Mutations within these domains which eliminate the ability of pRb to bind its targets result in loss of function. Loss of pRb function leads to tumorigenesis, although uncontrolled cellular proliferation is not a universal response to pRb inactivation. The ultimate response to the loss of pRb is influenced by both the genetic and epigenetic environments. Targeted disruption of RB in mice results in embryonic lethality, demonstrating the requirement for functional pRb in development. Close examination of various tissues from the embryos which lack wildtype RB shows problems in differentiation as well as showing induction of apoptosis. Although disruption of RB has provided useful information, complete inactivation of a gene precludes the possibility of discovering the functions that separate domains may have within the system. Creation of a dominant negative mutant by domain deletion whose phenotype is expressed in the presence of the wildtype may provide information about the intermediate functions of the protein. In addition, tissue specific targeting of a dominant negative mutant of pRb allows for comprehensive analysis of pRb function in organogenesis. In this thesis, a series of RB deletion mutants were created and tested for dominant negative activity as well as cellular localization. A tissue culture assay for dominant negative activity was developed which screens for the phenotype of apoptosis due to loss of pRb function. Two mutants from this series scored positive for dominant negative activity in this assay. The effect of these mutants within the assay environment can be explained by a model in which pRb acts as a facilitator of cell fate pathway decisions. ^

La obra crítica y la poesía de Edelweis Serra poseen un particular sentido en el contexto cultural argentino en el que se desarrollan. En una época de manifiesta secularización, su tentativa se orienta hacia el rescate de la dimensión sagrada de la realidad. El estudio ofrece un examen panorámico sobre la labor de Edelweis Serra. Dicho examen atiende especialmente a los principales temas y los rasgos estilísticos dominantes en su lírica. En lo que se refiere a este último punto, el interés se centra en los aspectos que resaltan la singularidad de su poesía: el arraigo de símbolos y figuras retóricas propias del lenguaje bíblico. Tales características guardan relación con su concepción de la poesía como don divino. Se propone, de este modo, la difusión de una labor importante, tanto crítica como de creación literaria, y el señalamiento de las claves interpretativas que favorecen su lectura.

El presente trabajo tiene por finalidad redescubrir y analizar una obra que recrea la lucha entre unitarios y federales, pero con una visión totalmente superadora del conflicto, a través de la mirada de sus personajes principales: Agustina Palacio de Libarona y Juan Felipe Ibarra. Además, la temática abordada se relaciona íntimamente con rasgos que son definitorios de la narrativa de Abelardo Arias. Entre ellos pueden mencionarse: 1) la calidad y fuerza de expresión de su prosa, cualidades que confieren un espesor particular a la caracterización de los personajes tanto principales como secundarios y configura un ambiente plásticamente muy logrado; 2) el trabajo de fondo que evidencia una dedicación exhaustiva al estudio historiográfico y a la precisión geográfica; 3) la simbología que se desprende de la esencia misma de los personajes. Basándonos en estos tres aspectos, analizamos tanto la base histórica como la construcción ficcional de dichos personajes, con particular referencia a su simbología, e intentaremos revelar aspectos que definen la obra de Arias como exponente de la literatura regional, pero a la vez, inserta en el marco de las corrientes hispanoamericanas reinantes.

Data on chlorophyll a concentration and species composition and biomass of phytoplankton have been collected for two years continues monitoring in the central part and the slope zone of the Black Sea. Seasonal dynamics of chlorophyll a concentration correspond to U-type. It is characterized by low values in summer, high values in autumn, which are going to winter-spring bloom of phytoplankton. Oscillations of mean values of chlorophyll concentrations are equal to 3-4 in the central part and 2 - in the slope zone. In the autumn-winter-spring period high values of phytoplankton biomass are related to typical blooming diatom algae in the first year and other taxons - in the second year. Relationship between relative content of phaeopigment and chlorophyll concentration has been determined for the central western part of the sea. Seasonal dynamics of carbon/chlorophyll a ratio has been shown. For both regions mean values of this ratio were equal to ~20 in autumn-winter-spring time and 40-50 - in summer time.
