Earlier studies on the distribution of geological environmental indicators in China revealed drastic changes from a zonal climate pattern (planetary-wave-dominant pattern) in the Paleogene to a monsoon-dominant one in the Neogene, which suggested an inception of the initial East-Asian summer monsoon. However, there are different views about the time and causes of the changes.Here, we attempt to compile a series of paleoenvironmental maps based on newly collected climate indicators from the literatures and chronologically constrained evidence of geological maps in order to re-examine the temporal and spatial evolution of climate belts in China during the Cenozoic with special emphasis on the changes of the arid belt. These indicators include mammalian fauna, coal, carbonate concretions, jarosite, salt, gypsum deposits and pollen assemblages etc, with chronological controls that we believe reliable. Pollen assemblages and mammalian fauna have been classified into three categories (arid, semi-arid/sub-humid, humid) to reflect the intensity of aridity/humidity. Salt, jarosite and gypsum deposits are classified as the arid indicators. Carbonate concretions and coal are classified into the semi-arid/sub-humid and humid one respectively. Paleoenvironmental maps at 8 time slices have been reconstructed. They are the Paleocene, Eocene, Oligocene, Miocene, Early Miocene, Middle Miocene, Late Miocene and Pliocene.And furthermore, we attempt to use IAP^AGCM to simulate the evolution of climate belts in emphasizing on the changes of the rain band, and compare the results with the paleoenvironmental maps in order to examine the causes of the drastic paleoenvironmental changes near the Oligocene/Miocene boundary. 36 sensitive numerical experiments are carried out using the IAP__AGCM to analyze the impacts of the uplift of the Himalayan-Tibetan complex, shrinkage of the Paratethys Sea, expansion of the South China Sea and the development of the polar ice sheets on rain band in China.The main conclusions are as follows:The obtained results essentially confirm the earlier conclusions about a zonal climate pattern in the Paleogene and a different pattern in the Neogene, and illustrate that a monsoon-dominant environmental pattern with inland aridity formed by the Early Miocene, which is temporally consist with the onset of eolian deposits in China.Cenozoic cooling and the formation of polar ice sheets are unlikely the main causes to the changes of environmental patterns mentioned above in China. But northern hemispheric cooling and the ice-sheets can intensify the Siberian High Pressure, and strengthen the winter monsoon circulations and enhance the aridity in the west part of China. These results support the earlier studies.Shrinkage of the Paratethys Sea and uplift of the Himalayan-Tibetan complex played important roles in strengthening the East Asian monsoon and induceing the above changes of environmental pattern, which is consistent with the earlier studies. Furthermore, "the monsoon-dominant pattern" appears when the Himalayan-Tibetan complex reaches to about 1000-2000 meters high and the Paratethys Sea retreats to the Turan Plate.4) Expansion of the South China Sea is another significant factor that drives the evolution of environmental patterns. We believe that the above three factors co-act and drive the change of the environmental patterns from a planetary-wave-dominant one to a monsoon-dominant one. However, the impacts of each factor vary by regions. The uplift mainly increases the humidity in Southwestern China and the aridity in northwestern country. The shrinkage mainly increases the humidity in Northern China and also enhances the aridity in the northwestern country. The expansion greatly increases the humidity in the south part of China.

In 2003, the remains of an Early Iron Age bog body, known as ‘Oldcroghan Man’, were recovered during the cutting of a drainage ditch in a bog in the Irish Midlands. Only some fingernails and a withe fragment remained undisturbed in situ in the drain face, providing the sole evidence for the original position of the body. A detailed reconstruction of the depositional context of the body has been undertaken through multi-proxy analyses of a peat monolith collected at the findspot. The palynological record shows that the surrounding area was the focus of intensive human activity during the Later Bronze Age, but was largely abandoned during the Bronze Age–Iron transition in the mid-first millennium BC. In the mid-4th century BC, a bog pool developed at the site, evidenced in the stratigraphic, plant macrofossil, testate amoebae and coleopteran records. Plant macrofossil and pollen analysis of peat samples associated with the fingernails suggests that the body was deposited in this pool most likely during the 3rd century BC. The absence of carrion beetle fauna points to complete submergence of the body within the pool. Deposition occurred shortly before or around the time that the surrounding area again became the focus of woodland clearance, as seen in the extended pollen record from the peat monolith. This period corresponds to the Early Iron Age in Ireland, during which renewed cultural connections with Britain and continental Europe can be seen in the archaeological record and widespread forest clearance is recorded in pollen records from across Ireland. The palaeoenvironmental results indicate, therefore, that the demise of Oldcroghan Man took place at a pivotal time of socio-economic and perhaps political change.

Tree-ring analysis of subfossil Pinus sylvestris L., from nine new peatland sites located beyond the species’ current northern limit in Scotland, established a regional chronology called WRATH-9. The chronology has been provisionally dated against Irish pine chronologies and provides the first annual resolution picture of Scots pine expansion from c. 3200 bc and subsequent demise from c. 3000 bc. Pine germination and growth is suggested to be associated with a widespread fall in bog water-tables that indicates a regional climatic control. Bog pines progressively declined in number, rather than died out in a single event, reflecting their growth in a marginal habitat, close to a critical ecological threshold. The use of tree-ring sequences from in situ bog pine macrofossils provides a higher resolution insight into past conditions than possible with existing radiocarbon and pollen-based chronologies.

It is commonly believed that trees were absent in Scandinavia during the last glaciation and first recolonized the Scandinavian Peninsula with the retreat of its ice sheet some 9000 years ago. Here, we show the presence of a rare mitochondrial DNA haplotype of spruce that appears unique to Scandinavia and with its highest frequency to the west—an area believed to sustain ice-free refugia during most of the last ice age. We further show the survival of DNA from this haplotype in lake sediments and pollen of Trøndelag in central Norway dating back ~10,300 years and chloroplast DNA of pine and spruce in lake sediments adjacent to the ice-free Andøya refugium in northwestern Norway as early as ~22,000 and 17,700 years ago, respectively. Our findings imply that conifer trees survived in ice-free refugia of Scandinavia during the last glaciation, challenging current views on survival and spread of trees as a response to climate changes.

The late-glacial vegetation development in northern Norway in response to climate changes during the Allerod, Younger Dryas (YD), and the transition to the Holocene is poorly known. Here we present a high-resolution record of floral and vegetation changes at lake Lusvatnet, south-west Andoya, between 13500 and 8000 cal b.p. Plant macrofossil and pollen analyses were done on the same sediment core and the proxy records follow each other very closely. The core has also been analyzed using an ITRAX XRF scanner in order to check the sediment sequence for disturbances or hiatuses. The core has a good radiocarbon-based chronology. The Saksunarvatn tephra fits very well chronostratigraphically. During both the Allerod and the Younger Dryas time-periods arctic vegetation prevailed, dominated by Salix polaris associated with many typically arctic herbs such as Saxifraga cespitosa, Saxifraga rivularis and Oxyria digyna. Both periods were cold and dry. Between 12450 and 12250 cal b.p. during the Younger Dryas chronozone, the assemblage changed, particularly in the increased abundance of Papaver sect. Scapiflora and other high-Arctic herbs, suggesting the development of polar desert vegetation mainly as a response to increased aridity. After 11520 cal b.p. a gradually warmer and more oceanic climate initiated a succession to dwarf-shrub vegetation and the establishment of Betula woodland after 1,000 years at c. 10520 cal b.p. The overall late-glacial aridity contrasts with oceanic conditions in southern Norway and is probably related to sea-ice extent.

Complex animals use a wide variety of adaptor proteins to produce specialized sites of interaction between actin and membranes. Plants do not have these protein families, yet actin-membrane interactions within plant cells are critical for the positioning of subcellular compartments, for coordinating intercellular communication, and for membrane deformation [1]. Novel factors are therefore likely to provide interfaces at actin-membrane contacts in plants, but their identity has remained obscure. Here we identify the plantspecific Networked (NET) superfamily of actin-binding proteins, members of which localize to the actin cytoskeleton and specify different membrane compartments. The founding member of the NET superfamily, NET1A, is anchored at the plasma membrane and predominates at cell junctions, the plasmodesmata. NET1A binds directly to actin filaments via a novel actin-binding domain that defines a superfamily of thirteen Arabidopsis proteins divided into four distinct phylogenetic clades. Members of other clades identify interactions at the tonoplast, nuclear membrane, and pollen tube plasma membrane, emphasizing the role of this superfamily in mediating actin-membrane interactions.

The plant actin cytoskeleton is a highly dynamic, fibrous structure essential in many cellular processes including cell division and cytoplasmic streaming. This structure is stimulus responsive, being affected by internal stimuli, by biotic and abiotic stresses mediated in signal transduction pathways by actin-binding proteins. The completion of the Arabidopsis genome sequence has allowed a comparative identification of many actin-binding proteins. However, not all are conserved in plants, which possibly reflects the differences in the processes involved in morphogenesis between plant and other cells. Here we have searched for the Arabidopsis equivalents of 67 animal/fungal actin-binding proteins and show that 36 are not conserved in plants. One protein that is conserved across phylogeny is actin-depolymerizing factor or cofilin and we describe our work on the activity of vegetative tissue and pollen-specific isoforms of this protein in plant cells, concluding that they are functionally distinct.

The acquisition of high quality, well-dated local site records is essential for progressing regional environmental reconstructions. As part of a wider study designed to examine intra- and extra- site ecosystem responses to environmental change, this paper presents new palaeoecological data from the floodplain of the River Torne in the Humberhead Levels, South Yorkshire. The sampling site lies adjacent to the lowland raised mire of Hatfield Moors, a location with a long history of palaeoecological investigations. The potential of using floodplain records to reconstruct local variations in ecosystem response to environmental change is also considered. Coleoptera and pollen are used to reconstruct floodplain ecosystem dynamics, whilst chronologies are established using Bayesian age–depth modelling. Between 10,200 cal BP and 2300 cal BP, the floodplain experienced multiple phases of ecological change. At 10,200–9910 cal BP, a cut-off channel began to infill with peat, while the surrounding floodplain remained relatively dry with Pinus forest growing nearby. Between 9630–9500 cal BP and 7270–7020 cal BP, a depositional hiatus occurred in the sedimentary record. By the end of this period, the local woodland had diversified and expanded to mixed deciduous tree cover. A wet shift identified at 6870–6160 cal BP was shortly followed by a rise in Alnus and Tilia from 6410–6160 cal BP. At this time, widespread floodplain paludification had occurred in the Humberhead Levels, which was largely controlled by relative sea-level (RSL) rise and the associated rise in regional water tables. Floodplain expansion also resulted in the widespread occurrence of Alnus dominated fen woodland. The local Torne floodplain experienced varying levels of wetness that influenced the decline and subsequent regeneration of the woodland from 5870–5640 cal BP. At this time, the Ulmus decline is identified in the pollen stratigraphic record. Floodplain hydrology appears to have been controlled by a combination of water table fluctuations and changes in channel pattern/flow, both of which can be linked to RSL variations recorded in the Humber Estuary. Floodplain alluviation, also linked to rising water tables, is dated to 4360–4160 cal BP. Anthropogenic woodland clearance further upstream may have further compounded this event.

Subsistence farmers are exposed to a range of mycotoxins. This study applied novel urinary multi-mycotoxin LC-MS/MS methods to determine multiple exposure biomarkers in the high oesophageal cancer region, Transkei, South Africa. Fifty-three female participants donated part of their maize-based evening meal and first void morning urine, which was analysed both with sample clean-up (single and multi-biomarker) and by a 'dilute-and-shoot' multi-biomarker method. Results were corrected for recovery with LOD for not detected. A single biomarker method detected fumonisin B1 (FB1) (87% incidence; mean±standard deviation 0.342±0.466 ng/mg creatinine) and deoxynivalenol (100%; mean 20.4±49.4 ng/mg creatinine) after hydrolysis with β-glucuronidase. The multi-biomarker 'dilute-and-shoot' method indicated deoxynivalenol-15-glucuronide was predominantly present. A multi-biomarker method with β-glucuronidase and immunoaffinity clean-up determined zearalenone (100%; 0.529±1.60 ng/mg creatinine), FB1 (96%; 1.52±2.17 ng/mg creatinine), α-zearalenol (92%; 0.614±1.91 ng/mg creatinine), deoxynivalenol (87%; 11.3±27.1 ng/mg creatinine), β-zearalenol (75%; 0.702±2.95 ng/mg creatinine) and ochratoxin A (98%; 0.041±0.086 ng/mg creatinine). These demonstrate the value of multi-biomarker methods in measuring exposures in populations exposed to multiple mycotoxins. This is the first finding of urinary deoxynivalenol, zearalenone, their conjugates, ochratoxin A and zearalenols in Transkei.

Despite advancement in breast cancer treatment, 30% of patients with early breast cancers experience relapse with distant metastasis. It is a challenge to identify patients at risk for relapse; therefore, the identification of markers and therapeutic targets for metastatic breast cancers is imperative. Here, we identified DP103 as a biomarker and metastasis-driving oncogene in human breast cancers and determined that DP103 elevates matrix metallopeptidase 9 (MMP9) levels, which are associated with metastasis and invasion through activation of NF-κB. In turn, NF-κB signaling positively activated DP103 expression. Furthermore, DP103 enhanced TGF-β-activated kinase-1 (TAK1) phosphorylation of NF-κB-activating IκB kinase 2 (IKK2), leading to increased NF-κB activity. Reduction of DP103 expression in invasive breast cancer cells reduced phosphorylation of IKK2, abrogated NF-κB-mediated MMP9 expression, and impeded metastasis in a murine xenograft model. In breast cancer patient tissues, elevated levels of DP103 correlated with enhanced MMP9, reduced overall survival, and reduced survival after relapse. Together, these data indicate that a positive DP103/NF-κB feedback loop promotes constitutive NF-κB activation in invasive breast cancers and activation of this pathway is linked to cancer progression and the acquisition of chemotherapy resistance. Furthermore, our results suggest that DP103 has potential as a therapeutic target for breast cancer treatment.

Molecular characterization of genome-wide association study (GWAS) loci can uncover key genes and biological mechanisms underpinning complex traits and diseases. Here we present deep, high-throughput characterization of gene regulatory mechanisms underlying prostate cancer risk loci. Our methodology integrates data from 295 prostate cancer chromatin immunoprecipitation and sequencing experiments with genotype and gene expression data from 602 prostate tumor samples. The analysis identifies new gene regulatory mechanisms affected by risk locus SNPs, including widespread disruption of ternary androgen receptor (AR)-FOXA1 and AR-HOXB13 complexes and competitive binding mechanisms. We identify 57 expression quantitative trait loci at 35 risk loci, which we validate through analysis of allele-specific expression. We further validate predicted regulatory SNPs and target genes in prostate cancer cell line models. Finally, our integrated analysis can be accessed through an interactive visualization tool. This analysis elucidates how genome sequence variation affects disease predisposition via gene regulatory mechanisms and identifies relevant genes for downstream biomarker and drug development.

La malnutrition est identifiée comme l’un des facteurs potentiellement responsables des mortalités élevées de colonies d’abeilles des dernières années au Québec. Pour contrer cela, les apiculteurs donnent des suppléments de pollen à leurs colonies, mais les impacts d’une telle pratique à diverses périodes sont méconnus. Les effets de la disponibilité du pollen sur le développement de colonies d’abeilles ont été mesurés pendant 3 différentes périodes : au printemps, durant la pollinisation de la canneberge et à la fin de l’été. À chacune des périodes correspondait une expérience distincte utilisant 40 colonies. Pour chaque expérience, des conditions d’abondance de supplément de pollen et de restriction de pollen naturel étaient créées chez les colonies pendant un mois selon un plan d’expérience factorielle 2x2. L’élevage du couvain et la récolte de miel ont été mesurés jusqu’à la fin de l’été (début de l’été suivant pour l’expérience de fin d’été). Au printemps, les colonies restreintes en pollen naturel ont élevé 18% moins de couvain (p<0.05) pendant la période de restriction et 11% de moins à la fin de l’été alors que l’utilisation du supplément n’a eu aucun effet (p>0.05). Les colonies supplémentées durant la pollinisation des canneberges ont élevé moins de couvain (p<0.05) à la fin de l’été. Pour l’expérience de fin d’été, les colonies supplémentées ont eut une meilleure reprise printanière (p<0.05) de l’élevage du couvain (60% de plus) alors qu’une restriction en pollen naturel avait un effet négatif (p>0.05). Les récoltes de miel ont été augmentées (p<0.05) de 1,3 kg pendant la pollinisation de la canneberge alors qu’elles ont été diminuées (p<0.05)par une restriction en pollen naturel de 4,2 kg à la fin de l’été et de 15 kg au printemps.

Ce projet porte sur la reconstitution paléo-écologique d'un environnement forestier fossile retrouvé sur un plateau au sud-ouest de l'île Bylot. Il a comme objectifs 1) de préciser la chrono-stratigraphie du site; 2) d’établir une liste et une succession des différents taxons polliniques retrouvés dans les différentes unités stratigraphiques du site; 3) d’estimer leur âge et 4) d’en inférer des conditions climatiques (température et précipitations). Plusieurs coupes stratigraphiques ont été excavées puis échantillonnées afin de réaliser des analyses stratigraphiques, paléomagnétiques et polliniques. Un GPS différentiel fut également utilisé afin de caractériser à petite et grande échelle les unités stratigraphiques associées aux unités organiques fossiles. Les résultats des analyses granulométriques indiquent la séquence de dépôt suivante au sein d’une dépression dans la roche en place (schiste tertiaire) : 1) un diamicton glaciaire local ; 2) un sédiment limoneux d’origine glacio-lacustre ; 3) une unité organique tourbeuse; 4) une unité de type alluvial ; 5) un sédiment fluvio-glaciaire et 6) un diamicton glaciaire d’origine allochtone. Les analyses polliniques suggèrent une végétation similaire à celle présente près de la limite des arbres actuelle, environ 2000 km plus au sud. Les conditions climatiques plus humides et plus chaudes permettaient notamment la croissance du pin (Pinus type strobus et banksiana), de l’épinette (Picea cf. mariana), de l’aulne (type crispa et incana) et du mélèze (Larix, indifférencié). Enfin, les études paléomagnétiques et la présence d’espèces éteintes suggèrent un âge pour les dépôts organiques fossiles situé entre 2,14 et 2,15 Ma ou entre 2,581 et 3,040 Ma.

Perennial plants are the main pollen and nectar sources for bees in the tropical areas where most of the annual flora are burned in dry seasons. Therefore perennial plants constitute the most reliable bio materials for determining and evaluating the beekeeping regions of the Republic of Benin. A silvo-melliferous region (S-MR) is a geographical area characterised by a particular set of homogenous melliferous plants that can produce timber. Using both the prevailing climatic and the agro-ecological conditions six S-MRs could be identified, i.e. the South region, the Common Central region, the Central West region, the Central North region, the Middle North region and the Extreme North region. At the country level, the melliferous plants were dominated by Vitellaria paradoxa which is common to all regions. The most diversified family was the Caesalpiniaceae (12 species) followed by the Combretaceae (10 species) and Combretum being the richest genus. The effect of dominance is particularly high in the South region where Elaeis guineensis alone represented 72.6% of the tree density and 140% of the total plant importance. The total melliferous plant density varied from 99.3 plants ha^(−1) in the Common Central region to 178.0 plants ha^(−1) in the Central West region. On the basis of nectar and pollen source, the best region for beekeeping is the CentralWest region with 46.7% of nectar producing trees, 9.4% of pollen producing trees and 40.6% of plants that issue both, this in opposition to the South region which was characterised by an unbalanced distribution of melliferous trees.

Este estudio fue conducido para evaluar la correlación entre lactato arterial y venoso central en niños con sepsis y choque séptico de una unidad de cuidado intensivo pediátrico. Se incluyeron 42 pacientes con edades comprendidas entre 1 mes y 17 años 364 días con diagnóstico de sepsis y choque séptico que ingresaron a la Unidad de Cuidado Intensivo en un hospital universitario de referencia. Se registró el valor del lactato obtenido de una muestra de sangre arterial y de sangre venosa central tomadas simultáneamente y dentro de las primeras 24 horas del ingreso a la unidad. Por medio de la prueba de Rho de Spearman se encontró una correlación de 0,872 (p<0,001) y se ajustó al uso de medicamentos, vasoactivos, edad y peso (modelo de regresión no paramétrico quantílico), manteniéndose una correlación fuerte y significativa.
