Nous avons utilisé une approche ethnobotanique pour identifier des espèces de plantes utilisées par les Cris afin de traiter les symptômes du diabète de type 2. Larix laricina du Roi (L. laricina) a récemment été identifiée comme une des meilleures plantes qui a stimulé le transport de glucose dans les cellules C2C12 et fortement potentialisé la différenciation des 3T3-L1 en indiquant une sensibilité potentiellement accrue à l’insuline. Ensuite, ces études de criblage ont été effectuées sur des extraits éthanolique (EE) en utilisant une série de bioessais in vitro. Cependant, les préparations traditionnelles des plantes sont souvent faites avec l’eau chaude. Le but de cette thèse de doctorat était d’isoler les principes actifs de L. laricina par un fractionnement guidé par l’adipogenèse; d’évaluer et de comparer l’activité et les mécanismes antidiabétiques des EE et des extraits aqueux (HWE) de ces 17 plantes. Pour le fractionnement de L. laricina, on a isolé plusieurs composés connus et identifié un nouveau composé actif cycloartane triterpene, qui a amélioré fortement l’adipogenèse et a été responsable en partie de l’activité adipogénique (potentiellement similaire à l’effet sensibilisateur à l’insuline des glitazone) de l’extrait éthanolique issu de l’écorce de L. laricina. Pour le métabolisme lipidique, nos résultats ont confirmé que 10 parmi les 17 EE ont augmenté la différenciation des adipocytes alors que 2 extraits seulement l’ont inhibée. Les HWE ont montré une faible activité adipogénique ou antiadipogénique. Les EE de R. groenlandicum et K. angustifolia ont le PPAR γ (peroxisome proliferator-activated receptor γ), le SREBP-1 (sterol regulatory element binding protein-1) et le C/EBP (CCAAT-enhancer binding proteins) α, alors que ceux de P. balsamifera et A. incana les ont inhibés. L’effet inhibiteur de P. balsamifera a également été prouvé d’avoir impliqué l’activation de la protéine kinase activée par l’AMP (AMPK). Les EE et HWE de R. groenlandicum ont stimulé les mêmes facteurs de transcription alors que les extraits aqueux d’autres plantes sélectionnées ont perdu ces effets en comparaison avec leurs extraits éthanoliques respectifs. L’analyse phytochimique a également identifié le groupe des espèces actives et inactives, notamment lorsque les espèces ont été séparées par famille de plante. Finalement concernant l’homéostasie de glucose, nos résultats ont confirmé que plusieurs EE ont stimulé le transport de glucose musculaire et inhibé l’activité de la glucose-6-phosphatase (G6Pase) hépatique. Certains des HWE ont partiellement ou complètement perdu ces activités antidiabétiques par rapport aux EE, tandis qu’une seule plante (R.groenlandicum) a juste conservé un potentiel similaire entre les EE et HWE dans les deux essais. Dans les cellules musculaires, les EE de R.groenlandicum, A. incana et S. purpurea ont stimulé le transport de glucose en activant la voie de signalisation de l’AMPK et en augmentant le niveau d’expression des GLUT4. En comparaison avec les EE, les HWE de R.groenlandicum ont montré des activités similaires; les HWE de A. incana ont complètement perdu leur effet sur tous les paramètres étudiés; les HWE de S. purpurea ont activé la voie de l’insuline au lieu de celle de l’AMPK pour augmenter le transport de glucose. Dans les cellules H4IIE, les EE et HWE des 5 plantes ont activé la voie de l’AMPK, et en plus les EE et HWE de 2 plantes ont activé la voie de l’insuline. La quercétine-3-O-galactoside et la quercétine 3-O-α-L-arabinopyranoside ont été identifiées comme des composés ayant un fort potentiel antidiabétique et donc responsables de l'activité biologique des plantes HWE actifs avec le transport du glucose. En conclusion, on a isolé plusieurs composés connus et identifié un nouveau triterpène actif à partir du fractionnement de L. laricina. Nous avons fourni également une preuve directe pour l'évaluation et la comparaison d'une action analogue à l'insuline ou insulino-sensibilisateur des EE et HWE de plantes médicinales Cris au niveau de muscle, de foie et de tissus adipeux. Une partie de leur action peut être liée à la stimulation des voies de signalisation intracellulaire insulino-dépendante et non-insulino-dépendante, ainsi que l’activation de PPARγ. Nos résultats indiquent que les espèces de plantes, les tissus ou les cellules cibles, ainsi que les méthodes d'extraction sont tous des déterminants significatifs de l'activité biologique de plantes médicinales Cris sur le métabolisme glucidique et lipidique.

Le motif imidazole, un hétérocycle à 5 atomes contenant 2 atomes d’azote et trois atomes de carbone, présente des propriétés physico-chimiques intéressantes qui en font un composé de choix pour plusieurs applications. Parmi ces propriétés, la fonctionnalisation simple des deux atomes d’azote pour former un sel d’imidazolium est très intéressante. Ces sels sont d’excellents précurseurs de carbènes N-hétérocycliques (NHC) et sont couramment utilisés pour synthétiser des ligands en vue d’une utilisation en catalyse organométallique. D’autre part, cette famille de composés possède des propriétés anionophores permettant une utilisation en transport anionique. Le présent travail contient les résultats de travaux concernant ces deux domaines, soit la catalyse et le transport anionique. Dans un premier temps, les propriétés de dérivés de l’imidazole sont exploitées pour former un catalyseur de type palladium-NHC qui est utilisé pour catalyser la réaction de Suzuki-Miyaura en milieu aqueux. L’efficacité de ce catalyseur a été démontrée en utilisant aussi peu que 0,001 mol% pour un rendement quantitatif. Il s’agit de la première occurrence d’un processus hétérogène et recyclable dans l’eau, utilisant un catalyseur de type Pd-NHC et qui ne nécessite aucun additif ou co-solvant. Le recyclage a été prouvé jusqu’à 10 cycles sans diminution apparente de l’activité du catalyseur. Dans un second temps, plusieurs sels d’imidazolium ont été testés en tant que transporteurs transmembranaires d’anions chlorures. Les propriétés intrinsèques des sels utilisés qui en font des transporteurs efficaces ont été élucidées. Ainsi, les paramètres qui semblent affecter le plus le transport anionique sont le changement du contre-anion du sel d’imidazolium de même que la propension de ce dernier à s’auto-assembler via une succession d’empilements-π. De plus, les propriétés du transport ont été élucidées, montrant la formation de canaux transmembranaires qui permettent non-seulement la diffusion d’ions Cl-, mais aussi le transport de protons et d’ions Ca2+. L’intérêt de cette recherche repose d’abord dans le traitement de diverses pathologies voyant leur origine dans le dysfonctionnement du transport anionique. Cependant, les propriétés bactéricides des sels d’imidazolium utilisés ont été identifiées lors des dernières expériences.

The research work which was carried out to Synergic Reactions in the Estuarine Environment leading to Modulation of Aluminium metal during Transport Processes (in Cochin Estuary)Estuaries are considered as sink or source for terrestrial and various anthropogenically generated materials. These include naturally occurring elements Al, Si, Fe or trace inorganics or industrial pollutants of different types. There have been reports on both positive and negative impacts by the introduction of above materials into the ecosystem.This thesis deals with the trace metal Aluminium (Al) whose average concentration (about 8%) in the earths crust is surpassed only by that of Oxygen and Silicon. There can be no doubt that most of the land derived materials reaches the ocean through rivers via estuaries. An important aspect noticed here is that the concentration of dissolved Al is much lower in sea water than in river water.On critically analysing Cochin estuary, for the entire cycles, covering monsoon, postmonsoon and premonsoon, the following salient features are documented as hereunder. Dissolved Al exhibits high and variable trends in Cochin estuary, the influencing parameters being salinity, SPM, pH and dissolved Si. A general profile showed removal in upper/mid estuary followed by regeneration in the mid/lower estuary and further decrease seawards in the southern/northem arms.Distribution appears to be a function of freshwater input, the monsoon season exhibiting very high concentrations throughout the estuary. As the river discharge decreased with the progress of seasons, dissolved Al concentration also decreased, the metal limiting itself to the upper and mid estuary.

Sediment transport in the nearshore areas is an important process in deciding the coastline stability. The design and effective maintenance of navigable waterways, harbours and marine structures depend on the stability of the sediment substrate and the nature of sedimentation in the nearshore zone. The nearshore zone is a complex environment and the exact relationships existing between water motions and the resulting sediment transports are not well understood. During the rough weather season, when the sediment movement is considerable, processes occurring in the nearshore area are much less understood. Moreover, there is a general lack of field measurements, especially during the time of severe storm conditions. The increasing pressures and the concern on the preservation of the valuable coastal environment have led to the development of shore protection programmes. Conservation not only demands knowledge of what needs to be done, but also requires the basic processes to be fully understood. Considering the fragile nature of barrier beaches and intense occupancy of these areas by man, these coastal features have long been a subject of study by coastal oceanographers, geomorphologists and engineers. The present study is an attempt to understand the sediment movement in relation to beach dynamics, especially in the surf zone, along some part of Kerala coast and the response of the beaches to various forcing functions over different seasons

A case of long-range transport of a biomass burning plume from Alaska to Europe is analyzed using a Lagrangian approach. This plume was sampled several times in the free troposphere over North America, the North Atlantic and Europe by three different aircraft during the IGAC Lagrangian 2K4 experiment which was part of the ICARTT/ITOP measurement intensive in summer 2004. Measurements in the plume showed enhanced values of CO, VOCs and NOy, mainly in form of PAN. Observed O3 levels increased by 17 ppbv over 5 days. A photochemical trajectory model, CiTTyCAT, was used to examine processes responsible for the chemical evolution of the plume. The model was initialized with upwind data and compared with downwind measurements. The influence of high aerosol loading on photolysis rates in the plume was investigated using in situ aerosol measurements in the plume and lidar retrievals of optical depth as input into a photolysis code (Fast-J), run in the model. Significant impacts on photochemistry are found with a decrease of 18% in O3 production and 24% in O3 destruction over 5 days when including aerosols. The plume is found to be chemically active with large O3 increases attributed primarily to PAN decomposition during descent of the plume toward Europe. The predicted O3 changes are very dependent on temperature changes during transport and also on water vapor levels in the lower troposphere which can lead to O3 destruction. Simulation of mixing/dilution was necessary to reproduce observed pollutant levels in the plume. Mixing was simulated using background concentrations from measurements in air masses in close proximity to the plume, and mixing timescales (averaging 6.25 days) were derived from CO changes. Observed and simulated O3/CO correlations in the plume were also compared in order to evaluate the photochemistry in the model. Observed slopes change from negative to positive over 5 days. This change, which can be attributed largely to photochemistry, is well reproduced by multiple model runs even if slope values are slightly underestimated suggesting a small underestimation in modeled photochemical O3 production. The possible impact of this biomass burning plume on O3 levels in the European boundary layer was also examined by running the model for a further 5 days and comparing with data collected at surface sites, such as Jungfraujoch, which showed small O3 increases and elevated CO levels. The model predicts significant changes in O3 over the entire 10 day period due to photochemistry but the signal is largely lost because of the effects of dilution. However, measurements in several other BB plumes over Europe show that O3 impact of Alaskan fires can be potentially significant over Europe.

The Integrated Catchment Model of Nitrogen (INCA-N) was applied to the River Lambourn, a Chalk river-system in southern England. The model's abilities to simulate the long-term trend and seasonal patterns in observed stream water nitrate concentrations from 1920 to 2003 were tested. This is the first time a semi-distributed, daily time-step model has been applied to simulate such a long time period and then used to calculate detailed catchment nutrient budgets which span the conversion of pasture to arable during the late 1930s and 1940s. Thus, this work goes beyond source apportionment and looks to demonstrate how such simulations can be used to assess the state of the catchment and develop an understanding of system behaviour. The mass-balance results from 1921, 1922, 1991, 2001 and 2002 are presented and those for 1991 are compared to other modelled and literature values of loads associated with nitrogen soil processes and export. The variations highlighted the problem of comparing modelled fluxes with point measurements but proved useful for identifying the most poorly understood inputs and processes thereby providing an assessment of input data and model structural uncertainty. The modelled terrestrial and instream mass-balances also highlight the importance of the hydrological conditions in pollutant transport. Between 1922 and 2002, increased inputs of nitrogen from fertiliser, livestock and deposition have altered the nitrogen balance with a shift from possible reduction in soil fertility but little environmental impact in 1922, to a situation of nitrogen accumulation in the soil, groundwater and instream biota in 2002. In 1922 and 2002 it was estimated that approximately 2 and 18 kg N ha(-1) yr(-1) respectively were exported from the land to the stream. Aquatic sediments often remove hydrophobic contaminants from fresh waters. The subsequent distribution and concentration of contaminants in bed sediments determines their effect on benthic organisms and the risk of re-entry into the water and/or leaching to groundwater. This study examines the transport of simazine and lindane in aquatic bed sediments with the aim of understanding the processes that determine their depth distribution. Experiments in flume channels (water flow of 10 cm s(-1)) determined the persistence of the compounds in the absence of sediment with (a) de-ionised water and (b) a solution that had been in contact with river sediment. In further experiments with river bed sediments in light and dark conditions, measurements were made of the concentration of the compounds in the overlying water and the development of bacterial/algal biofilms and bioturbation activity. At the end of the experiments, concentrations in sediments and associated pore waters were determined in sections of the sediment at 1 mm resolution down to 5 mm and then at 10 mm resolution to 50 mm depth and these distributions analysed using a sorption-diffusion-degradation model. The fine resolution in the depth profile permitted the detection of a maximum in the concentration of the compounds in the pore water near the surface, whereas concentrations in the sediment increased to a maximum at the surface itself. Experimental distribution coefficients determined from the pore water and sediment concentrations indicated a gradient with depth that was partly explained by an increase in organic matter content and specific surface area of the solids near the interface. The modelling showed that degradation of lindane within the sediment was necessary to explain the concentration profiles, with the optimum agreement between the measured and theoretical profiles obtained with differential degradation in the oxic and anoxic zones. This paper investigates phosphorus (P) transport and transformation dynamics in two contrasting sub-catchments of the River Kennel, England. Samples were collected daily under baseflow and hourly under stormflow conditions using autosamplers for 2 years and analysed for a range of determinands (full P fractionation, suspended sediment (SS), cations, pH, alkalinity, temperature and oxygen). Concentrations of SRP, SUP, PP and SS were higher in the flashy River Enborne (means of 0.186, 0.071, 0.101 and 34 mg l(-1), respectively) than the groundwater-fed River Lambourn (0.079, 0.057, 0.028 and 9 mg l(-1), respectively). A seasonal trend in the daily P dataset was evident, with lower concentrations during intermediate flows and the spring (caused by a dilution effect and macrophyte uptake) than during baseflow conditions. However, in the hourly P dataset, highest concentrations were observed during storm events in the autumn and winter (reflecting higher scour with increased capacity to entrain particles). Storm events were more significant in contributing to the total P load in the River Enborne than the River Lambourn, especially during August to October, when dry antecedent conditions were observed in the catchment. Re-suspension of P-rich sediment that accumulated within the channel during summer low flows might account for these observations. It is suggested that a P-calcite co-precipitation mechanism was operating during summer in the River Lambourn, while adsorption by metal oxyhydroxide groups was an important mechanism controlling P fractionation in the River Enborne. The influence of flow conditions and channel storage/release mechanisms on P dynamics in these two lowland rivers is assessed. (C) 2004 Elsevier B.V. All rights reserved.

Rising nitrate levels have been observed in UK Chalk catchments in recent decades, with concentrations now approaching or exceeding legislated maximum values in many areas. In response, strategies seeking to contain concentrations through appropriate land management are now in place. However, there is an increasing consensus that Chalk systems, a predominant landscape type over England and indeed northwest Europe, can retard decades of prior nitrate loading within their deep unsaturated zones. Current levels may not fully reflect the long-term impact of present-day practices, and stringent land management controls may not be enough to avert further medium-term rises. This paper discusses these issues in the context of the EU Water Framework Directive, drawing on data from recent experimental work and a new model (INCA-Chalk) that allows the impacts of different land use management practices to be explored. Results strongly imply that timelines for water quality improvement demanded by the Water Framework directive are not realistic for the Chalk, and give an indication of time-scales over which improvements might be achieved. However, important unresolved scientific issues remain, and further monitoring and targeted data collection is recommended to reduce prediction uncertainties and allow cost effective strategies for mitigation to be designed and implemented. The unsaturated zone exerts a major control on the delivery of nutrients to Chalk streams, yet flow and transport processes in this complex, dual-porosity medium have remained controversial. A major challenge arises in characterising these processes, both at the detailed mechanistic level and at an appropriate level for inclusion within catchment-scale models for nutrient management. The lowland catchment research (LOCAR) programme in the UK has provided a unique set of comprehensively instrumented groundwater-dominated catchments. Of these, the Pang and Lambourn, tributaries of the Thames near Reading, have been a particular focus for research into subsurface processes and surface water-groundwater interactions. Data from LOCAR and other sources, along with a new dual permeability numerical model of the Chalk, have been used to explore the relative roles of matrix and fracture flow within the unsaturated zone and resolve conflicting hypotheses of response. From the improved understanding gained through these explorations, a parsimonious conceptualisation of the general response of flow and transport within the Chalk unsaturated zone was formulated. This paper summarises the modelling and data findings of these explorations, and describes the integration of the new simplified unsaturated zone representation with a catchment-scale model of nutrients (INCA), resulting in a new model for catchment-scale flow and transport within Chalk systems: INCA-Chalk. This model is applied to the Lambourn, and results, including hindcast and forecast simulations, are presented. These clearly illustrate the decadal time-scales that need to be considered in the context of nutrient management and the EU Water Framework Directive. (C) 2007 Elsevier B.V. All rights reserved.

The year 2000 radiative forcing (RF) due to changes in O3 and CH4 (and the CH4-induced stratospheric water vapour) as a result of emissions of short-lived gases (oxides of nitrogen (NOx), carbon monoxide and non-methane hydrocarbons) from three transport sectors (ROAD, maritime SHIPping and AIRcraft) are calculated using results from five global atmospheric chemistry models. Using results from these models plus other published data, we quantify the uncertainties. The RF due to short-term O3 changes (i.e. as an immediate response to the emissions without allowing for the long-term CH4 changes) is positive and highest for ROAD transport (31mWm-2) compared to SHIP (24 mWm-2) and AIR (17 mWm-2) sectors in four of the models. All five models calculate negative RF from the CH4 perturbations, with a larger impact from the SHIP sector than for ROAD and AIR. The net RF of O3 and CH4 combined (i.e. including the impact of CH4 on ozone and stratospheric water vapour) is positive for ROAD (+16(±13)(one standard deviation) mWm-2) and AIR (+6(±5) mWm-2) traffic sectors and is negative for SHIP (-18(±10) mWm-2) sector in all five models. Global Warming Potentials (GWP) and Global Temperature change Potentials (GTP) are presented for AIR NOx emissions; there is a wide spread in the results from the 5 chemistry models, and it is shown that differences in the methane response relative to the O3 response drive much of the spread.

Waterbirds have been proposed as important vectors for the passive dispersal of those aquatic invertebrates and plants that lack a capacity for active dispersal between isolated water bodies. We analysed the frequency of internal transport of bryozoan propagules (statoblasts) by waterbirds in Donana, Spain, by examining their presence in the intestines and ceca of dead birds and analysing the role of different aspects of gut characteristics in explaining variation in the presence/absence and abundance of statoblasts. Of the 228 samples examined, 7.9% presented intact statoblasts of Plumatella fungosa (Pallas, 1768), Plumatella emarginata Allman, 1844, and two unidentified Plumatella species. For a given bird species, individuals with heavier gizzards and shorter ceca had a lower incidence and abundance of statoblasts in the lower gut. Grit mass and intestine length were unrelated to the presence or abundance of statoblasts. Our results suggest that waterbirds frequently transport bryozoans on a local scale, with lighter gizzards and longer ceca favouring such transport. Lighter gizzards are likely to destroy fewer propagules before they reach the lower gut. Species and individuals with longer ceca are particularly good candidates for long-distance dispersal of bryozoans, given the longer passage time of propagules that enter the ceca.

The results of an experimental study into the oxidative degradation of proxies for atmospheric aerosol are presented. We demonstrate that the laser Raman tweezers method can be used successfully to obtain uptake coeffcients for gaseous oxidants on individual aqueous and organic droplets, whilst the size and composition of the droplets is simultaneously followed. A laser tweezers system was used to trap individual droplets containing an unsaturated organic compound in either an aqueous or organic ( alkane) solvent. The droplet was exposed to gas- phase ozone and the reaction kinetics and products followed using Raman spectroscopy. The reactions of three different organic compounds with ozone were studied: fumarate anions, benzoate anions and alpha pinene. The fumarate and benzoate anions in aqueous solution were used to represent components of humic- like substances, HULIS; a alpha- pinene in an alkane solvent was studied as a proxy for biogenic aerosol. The kinetic analysis shows that for these systems the diffusive transport and mass accommodation of ozone is relatively fast, and that liquid- phase di. ffusion and reaction are the rate determining steps. Uptake coe. ffcients, g, were found to be ( 1.1 +/- 0.7) x 10(-5), ( 1.5 +/- 0.7) x 10 (-5) and ( 3.0 - 7.5) x 10 (-3) for the reactions of ozone with the fumarate, benzoate and a- pinene containing droplets, respectively. Liquid- phase bimolecular rate coe. cients for reactions of dissolved ozone molecules with fumarate, benzoate and a- pinene were also obtained: k(fumarate) = ( 2.7 +/- 2) x 10 (5), k(benzoate) = ( 3.5 +/- 3) x 10 (5) and k(alpha-pinene) = ( 1-3) x 10(7) dm(3) mol (-1) s (- 1). The droplet size was found to remain stable over the course of the oxidation process for the HULIS- proxies and for the oxidation of a- pinene in pentadecane. The study of the alpha- pinene/ ozone system is the first using organic seed particles to show that the hygroscopicity of the particle does not increase dramatically over the course of the oxidation. No products were detected by Raman spectroscopy for the reaction of benzoate ions with ozone. One product peak, consistent with aqueous carbonate anions, was observed when following the oxidation of fumarate ions by ozone. Product peaks observed in the reaction of ozone with alpha- pinene suggest the formation of new species containing carbonyl groups.

Glutinous rice (or sticky rice) has to be soaked in water over an extended period of time before cooking. Soaking provides some of the water needed for starch gelatinisation to occur during cooking. The extent of water uptake during soaking is known to be influenced by temperature. This paper explores the use of very high pressures up to 600 MPa to accelerate water uptake kinetics during soaking. Changes occurring in length, diameter and moisture content were determined as a function of soaking time, pressure and temperature. The results show that length and diameter are positively correlated with all three parameters. However, the expansion ratios are not very high: the maximum length expansion ratio observed was 1.2, while the maximum diameter expansion ratio was 1. 1. Given these low values, it was possible to model water uptake kinetics by using the well-known Fickian model applied to a finite cylinder, assuming uniform average dimensions and effective diffusion coefficient. The results showed that the overall rates of water uptake and the equilibrium moisture content increased with pressure and temperature. The effective diffusion coefficient, on the other hand, did not follow the same trend. Temperature influenced the effective diffusion coefficient below 300 MPa, but had a marginal effect at higher pressures. Moreover, the effective diffusion coefficient increased with temperature between 20 and 50 degrees C, but dropped at higher temperatures. This drop can be attributed to the gelatinisation of starch, which restricts the transport of water. Regardless, it is possible to increase the quantity of water absorbed by rice and the rate at which it is absorbed, by using high pressures and temperatures. (c) 2004 Elsevier Ltd.
