<p>After artificial activation or fertilization of non-nucleate fragments or eggs of the sea urchin, the mitochondria actively synthesize RNA. The RNA made in non-nucleate fragments is shown to be mostly single stranded and to be associated primarily with the low speed pellet of centrifuged cellular homogenates.</p> <p>Protein synthesis is observed in non-nucleate fragments in the presence or absence of the mitochondrial RNA synthesis: it is found to be qualitatively similar but quantitatively less in the absence of the RNA synthesis. The continued syntheses of proteins in the non-nucleate fragments in the absence of mitochondrial RNA synthesis provides additional evidence for the presence of a stable messenger RNA component in the unfertilized sea urchin egg.</p> <p>Since the uptake or actinomycin D was found to be inhibited by the presence of a fertilization membrane, ethidium bromide, at 10 μgs/ml, is used as an effective inhibitor of RNA synthesis in non-nucleate fragments and in early cleavage stage embryos. However, this same concentration of ethidium bromide is found to be only partially effective in blocking RNA synthesis at the mesenchyme blastula stage of development.</p> <p>Low concentrations of ethidium bromide (2 and 5 μgs/ml) are found not to be lethal but to be capable of producing moderate developmental defects. In the presence of concentrations of ethidium bromide adequate to inhibit all the mitochondrial RNA synthesis (10 μgs/ml of ethidium bromide), from fertilization on, the embryos do not cleave beyond the 4-8 cell stages. When similar concentrations of ethidium bromide are added at an early mesenchyme blastula stage, the embryos do not gastrulate but continue to swim for more than 24 additional hours (adequate for control embryos to develop to a late prism stage). These results lead to the conclusion that mitochondrial RNA synthesis may be very essential for normal development to occur. </p> <p>DNA is synthesized in the non-nucleate fragments of sea urchin eggs. None of the newly synthesized DNA is found in the closed circular form. When phenol extracted directly from the fragments, the DNA is found to sediment at approximately 38 and 27s in sucrose gradients but neither of these size classes could be found associated with the isolated mitochondria. The template for the synthesis of DNA in non-nucleate fragments remains unknown.</p>

In recent years, the performance of semi-supervised learning has been theoretically investigated. However, most of this theoretical development has focussed on binary classification problems. In this paper, we take it a step further by extending the work of Castelli and Cover [1] [2] to the multi-class paradigm. Particularly, we consider the key problem in semi-supervised learning of classifying an unseen instance x into one of K different classes, using a training dataset sampled from a mixture density distribution and composed of l labelled records and u unlabelled examples. Even under the assumption of identifiability of the mixture and having infinite unlabelled examples, labelled records are needed to determine the K decision regions. Therefore, in this paper, we first investigate the minimum number of labelled examples needed to accomplish that task. Then, we propose an optimal multi-class learning algorithm which is a generalisation of the optimal procedure proposed in the literature for binary problems. Finally, we make use of this generalisation to study the probability of error when the binary class constraint is relaxed.

We used 25 years of conventional tagging data (n= 6173 recoveries) and 3 years of ultrasonic telemetry data (n=105 transmitters deployed) to examine movement rates and directional preferences of four age classes of red drum (Sciaenops ocellatus) in estuarine and coastal waters of North Carolina. Movement rates of conventionally tagged red drum were dependent on the age, region, and season of tagging. Age-1 and age-2 red drum tagged along the coast generally moved along the coast, whereas fish tagged in oligohaline waters far from the coast were primarily recovered in coastal regions in fall months. Adult (age-4+) red drum moved from overwintering grounds on the continental shelf through inlets into Pamlico Sound in spring and summer months and departed in fall. Few tagged red drum were recovered in adjacent states (0.6% of all recoveries); however, some adult red drum migrated seasonally from overwintering grounds in coastal North Carolina northward to Virginia in spring, returning in fall. Age-2 transmitter-tracked red drum displayed seasonal emigration from a small tributary, but upstream and downstream movements within the tributary were correlated with fluctuating salinity regimes and not season. Large-scale conventional tagging and ultrasonic telemetry programs can provide valuable insights into the complex movement patterns of estuarine fish.

Oceanic incidence and spawning frequency of Chesapeake Bay striped bass (Morone saxatilis) were estimated by using microchemical analysis of strontium in otoliths. Otoliths from 40 males and 82 females sampled from Marylandâs portion of the Chesapeake Bay were analyzed for seasonal and age-specific patterns in strontium and calcium levels. The proportion of oceanic females increased from 50% to 75% between ages seven to 13; the proportion of oceanic males increased from 20% to ~50% between ages four to 13. Contrary to an earliermodel of Chesapeake Bay striped bass migration, results indicated that a substantial number of males undertook oceanic migrations. Further, we observed no mass emigration of females from three to four years of age from the Chesapeake Bay. Seasonal patterns of estuarine habitat use were consistent with annual spawning runs by striped bass of mature age classes, but with noteworthy exceptions for newly mature females. Evidence of an early oceanic presence indicated that Chesapeake Bay yearlings move into coastal regionsâa pattern observed also for Hudson River striped bass. Otolith microchemical analyses revealed two types of behaviors (estuarine and oceanic) that confirm migratory behaviors recently determined for other populations of striped bass and diadromous species (e.g., American eels [Anguilla rostrata] American shad [Alosa sapidissima] and white perch [Morone Americana]).

BCL-2 family proteins are key regulators of the mitochondrial apoptotic machinery, controlling the mitochondrial outer membrane (MOM) permeabilization (MOMP). BCL-2 related Ovarian Killer (BOK) is a poorly understood pro-apoptotic member of this protein family. It has been reported that BOK localizes predominantly (although not exclusively) at membranes of the endoplasmic reticulum and of the Golgi apparatus. However, it is unclear whether BOK also operates at the MOM to promote apoptosis, as other pro-apoptotic BCL-2 family members do. Basing on the fact that the other two BAX-like pro-apoptotic members have been reported to oligomerize in order to induce MOMP, site-directed mutagenesis was used to generate two point mutations that predictably eliminated BOKâs oligomerization capacity. Then, the effect of such mutations on BOKâs membrane activity was examined using fluorescence spectroscopy.

Inter and intra-annual variation in year-class strength was analyzed for San Francisco Bay Pacific herring (Clupea pallasi) by using otoliths of juveniles. Juvenile herring were collected from March through June in 1999 and 2000 and otoliths from subsamples of these collections were aged by daily otolith increment analysis. The composition of the year classes in 1999 and 2000 were determined by back-calculating the birth date distribution for surviving juvenile herring. In 2000, 729% more juveniles were captured than in 1999, even though an estimated 12% fewer eggs were spawned in 2000. Spawning-date distributions show that survival for the 2000 year class was exceptionally good for a short (approximately 1 month) period of spawning, resulting in a large abundance of juvenile recruits. Analysis of age at size shows that growth rate increased significantly as the spawning season progressed both in 1999 and 2000. However, only in 2000 were the bulk of surviving juveniles a product of the fast growth period. In the two years examined, year-class strength was not predicted by the estimated number of eggs spawned, but rather appeared to depend on survival of eggs or larvae (or both) through the juvenile stage. Fast growth through the larval stage may have little effect on year-class strength if mortality during the egg stage is high and few larvae are available.

Mortality associated with the incidental catch and release by commercial trollers of two size classes of chinook salmon, Oncorhynchus tshawytscha, was assessed. Observed cumulative mortality 4-6 days after hooking was 18.3 percent for sublegal-sizefish « 66 cm FL) and 19.0 percent for legal-sizefish. Size of fish was not significantly related to mortality; however, when the results were combined with data from a previous experiment, there was a significant inverse relationship between fish length and mortality. Hooking mortality estimates calculated from tagging experiments and observed relative mortality of legal-and sublegal-size fish held in net pens, were used to derive a range for total hooking mortality of 22.0-26.4 percent for sublegal-size chinook salmon and 18.5-26.4 percent for legal-size chinook salmon.

A benthic survey was carried out from November 1998 to December 1999 in the tidal flats of Bahía Samborombón (Río de la Plata estuary, Argentina), in order to study the population structure, reproductive aspects, growth and secondary production of Capitella capitata (Fabricius, 1780). Growth was analyzed using ELEFAN routine, and the secondary production was estimated by Hynes and Coleman's method (1968). C. capitata did not present periods of very important recruitments throughout the year; however, the abundance of smallest size classes was higher during summer and autumn. The summer cohort showed a growth rate (K) of 2.05 and a seasonal growth oscillation (C) of 0.6, pointing out that worms grew very slowly during winter months. The life span of this cohort was 13 months. The autumn cohort showed a lower growth rate (K= 1.5) and its growth was lowest during winter. The life span was 15 months for this cohort. C. capitata in Punta Rasa presented an extended reproductive period, with absence of activity during winter months. The type of eggs and larvae suggest that C. capitata has benthic larval development in the study area, destining its reproductive effort to the production of a low number of eggs, and assuring larvae survival through incubation in brooding tubes. The annual mean biomass in Punta Rasa was 0.117 g m-2 (AFDW), with a mean secondary production of 0.23 g m-2 y-1 and a P/B ratio of 1.96 y-1. The relatively low density, biomass production and P/B ratio of C. capitata in Punta Rasa can be considered as reference values for this species inhabiting undisturbed or moderately disturbed areas.

It is now clear that fisheries resources are among the key assets contributing to the national development objective of poverty eradication through providing food, employment, income and export earnings. It was recently reported in the papers that monthly fish exports had increased by 23% and fetched about US$ 10 Million during the month of November 2001 alone. This value may be underestimated as it is based solely on recorded exports from fish processing factories numbering 12. Although fisheries resources are renewable they can be depleted through unsustainable exploitation. It is therefore important to ensure that there is guided development and management of this asset so that it can continue contributing to the livelihood of the people who depend on it. Therefore, FIRRI contributes to the fisheries sub-sector developmental objective of ensuring increased and sustainable fishery production and utilization by providing information to guide sustainable management of capture fisheries resources and development of aquaculture.

Cells communicate with their external environment via focal adhesions and generate activation signals that in turn trigger the activity of the intracellular contractile machinery. These signals can be triggered by mechanical loading that gives rise to a cooperative feedback loop among signaling, focal adhesion formation, and cytoskeletal contractility, which in turn equilibrates with the applied mechanical loads. We devise a signaling model that couples stress fiber contractility and mechano-sensitive focal adhesion models to complete this above mentioned feedback loop. The signaling model is based on a biochemical pathway where IP3 molecules are generated when focal adhesions grow. These IP3 molecules diffuse through the cytosol leading to the opening of ion channels that disgorge Ca2+ from the endoplasmic reticulum leading to the activation of the actin/myosin contractile machinery. A simple numerical example is presented where a one-dimensional cell adhered to a rigid substrate is pulled at one end, and the evolution of the stress fiber activation signal, stress fiber concentrations, and focal adhesion distributions are investigated. We demonstrate that while it is sufficient to approximate the activation signal as spatially uniform due to the rapid diffusion of the IP3 through the cytosol, the level of the activation signal is sensitive to the rate of application of the mechanical loads. This suggests that ad hoc signaling models may not be able to capture the mechanical response of cells to a wide range of mechanical loading events. © 2011 American Society of Mechanical Engineers.

Class Condrichthyes consist of two sub-classes: Holocehali and. Elasmobranchii, the second one are more divers and has more importance. Selachimorpha (sharkes) and Batidoidimorpha (rays and skates) are two super-order of Elasmobranchii, which they have crucial role in ecological balance in marine ecosystmes. Except few cases, most of sharks and rays (rays and skates) are not well identified, so a lot of works need to be done in this regards. The area of study is located between 49°, 35' and 52°, 33' E and between 27°, 21' and 30°, 02' N in depth of 7 to 78 in. Study were conducted diuring August 1998, September 1999. Samples were taken during 3 sea cruises from 70 bottom trawl net. All sharks and batoid fishes were identified based on biometric specifications (weight, total, length and sexuality for both group and extra biometric specifications, disc width, disc length, and tail length only for, batoid fishes). The 1140 specimens of batoid fishes identified belonged to 6 families, and 18 species. 3 new species were identified, they are Himantura sp.1 and Himantura sp.2 belonged to Dasyatidae family and Rhinobatos sp. belonged to Rhinobatidae Familiy. It needs more works and more adaquate ducuments for cl.earfing scientific names of these species. The 275 specimens of sharks identified belonged to 6 families and 10 species. Chiloscyllium sp. belonged to Hemiscylliidae, family as a new species was identified.

Forecasting the returns of assets at high frequency is the key challenge for high-frequency algorithmic trading strategies. In this paper, we propose a jump-diffusion model for asset price movements that models price and its trend and allows a momentum strategy to be developed. Conditional on jump times, we derive closed-form transition densities for this model. We show how this allows us to extract a trend from high-frequency finance data by using a Rao-Blackwellized variable rate particle filter to filter incoming price data. Our results show that even in the presence of transaction costs our algorithm can achieve a Sharpe ratio above 1 when applied across a portfolio of 75 futures contracts at high frequency. © 2011 IEEE.
