In his work on human knowledge, Vygotsky reveals the second human nature, the one which is historical and cultural, due to people´s learning throughout life, through the mediation of others and the concrete conditions of life and education. In this eminently social process, the child grows into the intellectual life having the adult as a peer and learns human skills from this adult-child interaction. This means that, for working with abstract formulation, it is necessary understanding it as a complex, dynamic and functional act that is built by the insertion of individual performance into culture that is mediated by interaction with others. In this setting, each individual reaches knowledge through formal and non-formal learning that help on the formulation of scientific and everyday concepts. To make studies on the process of concept formation, Vygotsky adopted an experimental methodology based on the philosophical assumptions of Marxist theory of how mental processes occur, once he perceived these processes in a constantly changing and moving. Thus, the method called “Instrumental, Cultural and Historical” differed from conventional experimental studies focused on the performance of the task itself. The method adopted by Vygotsky was concerned with the process of concept formation and not only with fragmentary cutouts of cognitive processes. According to our study, the formation of the social nature of man develops from processes of appropriation and objectification of knowledge, which makes individual the historically constructed achievements by mankind, as, for example, types of sophisticated thinking, which requires the discussion of concept formation.

Para a recuperação da estética facial,há necessidade de reposição do volume orbitário perdido em decorrência de eviscerações ou enucleações. Este estudo foi realizado com os objetivos de avaliar comparativamente a biocompatibilidade e a manutenção do volume orbitário com o uso de esferas de hidroxiapatita sintética e de polietileno poroso, na reconstrução de cavidades evisceradas de coelhos. Métodos: Para isso foram utilizados 56 coelhos albinos, submetidos à evisceração do olho direito, com colocação de esferas de hidroxiapatita sintética (G1 – 28 animais) ou polietileno poroso (G2 – 28 animais).Quatro coelhos de cada grupo foram sacrificados em 7 momentos experimentais: 7, 15, 30, 60, 90, 120 e 180 dias após a evisceração. Após o sacrifício, o conteúdo da cavidade orbitária direita foi removido e seccionado na porção central em duas hemi-metades,uma das quais foi preparada para exame histopatológico; exame ultra-estrutural em microscópio eletrônico de varredura foi feito em dois animais de 7, 60 e 180 dias, de ambos os grupos experimentais. Resumo 118 Resultados: Observou-se aos 7 dias, tecido conjuntivo frouxo, constituído de células inflamatórias e hemáceas, em meio à rede de fibrina, principalmente na periferia da esfera de hidroxiapatita e chegando até ao centro da esfera de polietileno; a reação tecidual tornou-se mais densa com o passar do tempo. Decorridos 60 dias, observou-se em G1, início de metaplasia óssea que se acentuou ao longo do experimento. Em todos os momentos experimentais, a inflamação,predominantemente macrofágica, foi muito mais acentuada em G1.O volume da cavidade esclero-corneana foi melhor mantido em G2. Conclusão: a esfera de hidroxiapatita sintética provoca mais inflamação que a de polietileno poroso e é menos eficiente na manutenção do volume orbitário... This experimental study aimed the evaluation of the use of anthropomorphic phantoms in the analysis of the influence of the acquisition parameters in the contrast of radiographic images. The analyzed factors were the variation of the peak kilovoltage (kVp) and the product of the filament circuit by the time of exposition (mAs). The influence of these factors was verified for different anthropomorphic phantoms: foot, knee and chest. The image contrast behavior with the simulators was compared to values obtained with the use of an aluminum ladder being the behavior of this reference for analysis and discussion. To assure the reproducibility of images, quality control tests were made and evaluation of procedure conditions before the experiments. The results obtained are shown in a scale of images where it was possible to evaluate the impact in the darkness of images. Regions with different compositions were analyzed which were different in image, this way the values of optical density and contrast are represented as charts and graphics. We conclude that the use of anthropomorphic phantoms in the evaluation of the influence of tension of the tube and time of exposition in the contrast of the radiographic images is not indicated for a quantitative analysis through optical density, once they present incompatible results with the data as reference as the aluminum ladder even so these simulators present a great property in qualitative analysis regarding the differentiation of structures and subjective evaluations

Maternal undernutrition affects the foetal development, promoting renal alterations and adult hypertension. The present study investigates, in adult male rats, the effect of food restriction in utero on arterial blood pressure changes (AP), and its possible association with the number of nephrons, renal function and angiotensin II (AT1R/AT2R), glucocorticoid (GR) and mineralocorticoid (MCR) receptors expression. The daily food supply to pregnant rats was measured and one group (n=5) received normal quantity of food (NF) while the other group received 50% of that (FR50) (n=5). The AP was measured weekly. At 16 weeks of life, fractionator’s method was used to estimate glomeruli number in histological slices. The renal function was estimate by creatinine and lithium clearances. Blood and urine samples were collected to biochemical determination of creatinine, sodium, potassium and lithium. At 90th and 23rd days of life, kidneys were also processed to AT1R, AT2R, GR and MCR immunolocalization and for western blotting analysis. FR50 offspring shows a significant reduction in BW (FR50: 5.67 ± 0.16 vs. 6.84 ± 0.13g in NF, P<0.001) and increased AP from 6th to 12nd week (6thwk FR50: 149.1 ± 3.4 vs. 125.1 ± 3.2mmHg in NF, P<0.001and, 12ndwk FR50: 164.4 ± 4.9 vs. 144.0 ± 3.3 mmHg in NF, P=0.02). Expression of AT1R and AT2R were significantly decreased in FR50 (AT1, 59080 ± 2709 vs. 77000 ± 3591 in NF, P=0.05; AT2, 27500 ± 95.50 vs. 67870 ± 1509 in NF, P=0.001) while the expression of GR increased in FR50 (36090 ± 781.5 vs. 4446 ± 364.5 in NF, P=0.0007). The expression of MCR did not change significantly. We also verified a pronounced decrease in fractional urinary sodium excretion in FR50 offspring (0.03 ± 0.02 vs. 0.06 ± 0.04 in NF, p=0.03). Durante a infecção experimental por Trypanosoma cruzi, a resposta imune adaptativa mediada por células CD8+ é direcionada a poucos epítopos imunodominantes e subdominantes. Em estudos anteriores, foi demonstrado que, em diferentes linhagens de camundongos, a imunização com plasmídeos ou proteínas recombinantes, que expressam epítopos CD8 imunodominantes, antecipa a resposta imune após o desafio e proporciona resposta imune protetora contra infecções letais com T.cruzi. Em nosso estudo atual, desenvolvemos a hipótese que também é possível gerar resposta imune protetora a partir de imunizações com plasmídeos que expressam epítopos subdominantes. Neste caso, esta resposta poderia se somar a resposta imune dominante ampliando o espectro da resposta imuno-protetora. Para testar nossa hipótese, nós tivemos que construir diferentes plasmídeos contendo a ORF do gene da proteína 2 de superfície de amastigota (asp-2) de T.cruzi contendo mutações que: i) retiraram os epítopos imunodominantes CD8 (VNHRFTLV para H-2Kb ou TEWETGQI para H-2Kk) ;ii) trocaram o epítopo imunodominante por epítopos sub-dominantes (TsKb-18 ANYDFTLV ou TsKb-20 ANYKFTLV). Além disso, também foi construído um plasmídeo contendo somente a porção P4-P7 (261° a 500° aminoácidos) da asp-2. Após a construção destes plasmídeos, testamos a imunogencidade de dois deles, denominados pRE-P4P7 e o pIgSP-VNHRATLA, foram testados individualmente na vacinação de camundongos F1(C57BL/6 x B10.A) (H-2Kb e H- 2Kk) ou de CB10 (H-2Kb), respectivamente, e desafiados com tripomastigotas da cepa Y de T.cruzi. A vacinação de camundongos F1 (C57BL/6 x B10.A) com o plasmídeo pré- P4-P7 aumentou significativamente a resposta imune de células CD8+ específicas ao epítopo sub-dominante TEWETGQI, restrito ao MHC H-2kk, porém não diminuiu a resposta ...(Resumo completo, clicar acesso eletrônico abaixo)

The behavior and experience of pain are characterized by alertness and anxiety, which disappear soon after the healing process begins. Based on this area, the present study aimed to quantify the expression of c-fos in segments of the brain of rats after surgical stimulation in one rear hind paw (analgesia), using anesthesia with sodium thiopental and practices of acupuncture pre and post operative. The animals were stimulated with intraperitoneal injections of solution (NaCl) 0.2% (2ml), and divided into three groups, control, preoperative and postoperative. Each treatment was divided in manual acupuncture (AM) and electroacupuncture (EA). The animals were randomly distributed in each group. The c-fos expression was quantitated using immunohistochemistry for all situations. The results showed a great efficiency of all treatment compared to the control group (p <0.001), thus reenforcing data in the literature on the potential of acupuncture analgesia. There were no statistical differences among the different treatments, although there was a trend of EA being more efficient than AM

As infecções do trato urinário (ITU) são definidas como a invasão e posterior multiplicação de microorganismos nos tecidos das vias urinárias, sendo a bactéria gram-negativa E.coli o principal uropatógeno envolvido nessa infecção. A avaliação para a presença de ITU é feita através do quadro clínico característico desse tipo de paciente e o exame de cultura de urina. As amostras positivas para as culturas são avaliadas quanto ao perfil de sensibilidade aos antimicrobianos dos agentes etiológicos encontrados. Descrever a porcentagem de ocorrência dos agentes etiológicos e o perfil de sensibilidade aos antimicrobianos das Infecções do Trato Urinário em pacientes ambulatoriais de Botucatu e região. Foram incluídas neste estudo culturas de urinas de pacientes atendidos em UBS de Botucatu e região Pólo Cuesta, no período de Janeiro de 2008 a Maio de 2010. As culturas positivas foram avaliadas quanto a porcentagem de ocorrência dos uropatógenos segundo sexo, faixa etária e origem. Foi utilizado o teste do qui-quadrado para verificar associação entre cada antimicrobiano e as porcentagens de sensibilidade e resistência encontradas. Foram avaliadas 11.768 culturas de urina provenientes de 11 municípios da região, sendo consideradas as culturas positivas (28%). Quanto à ocorrência dos agentes etiológicos verificou-se que a E.coli é responsável 58,76% dos casos. A distribuição dos uropatógenos segundo sexo revelou maior ocorrência de E.coli nos dois sexos. O segundo agente etiológico de maior ocorrência foi, nos homens o Proteus mirabilis e nas mulheres, o S. saprophyticus. Dentre as faixas etárias, a E.coli foi o patógeno de maior ocorrência. O perfil de sensibilidade aos antimicrobianos revelou que a E.coli é mais sensível a Ertapenem, Imipenem, Meropenem, Fosfomicina, Amicacina, Ceftriaxona, Nitrofurantoína e Norfloxacina; e mais resistente... Uremic syndrome, arising from kidney malfunction, consists of a set of systemic changes caused by the accumulation of toxic substances to the body. Since, with the advance of medicine, the animals lived more reaching advanced age and entering thus on track of senility, chronic renal disease, became a common complaint in the routine of the ambulatory. This high rate of morbidity generates an increased need for clarification of pathophysiology involved in this disease. The components of the uremic syndrome include water and sodium imbalance, anemia, intolerance to carbohydrate, neurological disorders, disorders of the gastrointestinal tract, osteoarthritis, immunological incompetence and metabolic acidosis. The clinical manifestations occur in isolation or in combination. In most cases canines patients are subject to an assessment when the kidney disease has evolved to the final stage with uremic syndrome and installed already, under these conditions, the prognosis is reserved

Diuron (3-(3,4-dichlorophenyl)-1,1-dimethylurea) is a substituted urea herbicide widely used in crops of sugar cane, cotton and soybeans. In 1997, this agent has been classified by the United States Environmental Protection Agency as known/likely human carcinogen because it induced tumors in the urinary bladder and renal pelvis of rats, and breast and skin of mice exposed to 2500 ppm for feed for two years. A previous study from our group demonstrated dose-response relationship in the gene expression profile associated with severe necrosis on bladder urothelium and increased incidence of simple hyperplasia in male Wistar rats treated with different concentrations of diuron for 20 weeks. To check how early the molecular changes occurs, rats were fed for 7 days with diets containing diuron at 0, 125, 500 or 2500 ppm. The main observations recorded were urothelium ultrastructural alterations and disruptions of molecular pathways associated with cell-cell interaction and the tissue organization maintenance. Particularly, the gene Glypican 3 (Gpc3), a surface proteoglycan related to cellular adhesion and apoptosis induction, was down regulated on urothelium exposed to 2500ppm diuron for 7 days and 20 weeks. The aim of this study was validate by quantitative RT-PCR real time, the reduced Gpc3 gene expression in epithelial cells of the urinary bladder of male Wistar rats treated with different concentrations of diuron for 7 days and 20 weeks. The endogenous control of the quantitative PCR real time technique was the β-actin gene and the target was the gene Gpc3. The relative quantification (RQ) was obtained by the method of relative quantification 2-ΔΔCt . Animals exposed to diuron for 7 days or for 20 weeks presented reduction of Gpc3 gene expression compared to the control group. This reduction was statistically significant only for the 7 days study. Aquaporins (AQPs) are membrane protein channels expressed in a wide variety of cells and tissues that are involved in the transport of water. In the male excurrent duct, the movement of fluid and solutes across the epithelium is essential for establishing the proper luminal environment in which sperm mature and are stored. The initial aim of the present propose is to localize AQP 2 e 9 in epithelium of the initial segment, caput, corpus and cauda epididymidis in Mongolian gerbil during postnatal development (10, 30, 60, 120 and 300 days), observing possible changes in the pattern of expression of the AQPs along the postnatal development. The animals will be killed by inhalation of carbon dioxide. After euthanasia, the epididymis will be removed and the different regions will be isolated. The biological material obtained will be processed in accordance with the routines of histological techniques and imunnohistochemistry to study the expression and localization of the AQPs 2 and 9. The results obtained will be analyzed and photographed in conventional light microscope
