对生长在青藏高原不同海拔地区的多年生高山植物珠芽蓼(Polygonum viviparum L.)的抗氧化系统进行了测试,以探讨高山植物对于高寒环境的适应机理.结果表明:随着海拔的升高,叶绿素a(chla)、叶绿素b(chlb)含量明显下降,chla/chlb增大,珠芽蓼叶和根细胞的膜脂过氧化均加剧,丙二醛(MDA)含量明显增加.3种抗氧化酶的活性受到明显影响,其中叶片中过氧化物酶(POD)和超氧化物歧化酶(SOD)活性,随海拔升高而活性降低,而均与根中的变化趋势相反.过氧化氢酶(CAT)活性随海拔升高呈增强的趋势,且叶片中活性较根中变化明显.抗坏血酸(ASA)随海拔的升高,含量呈明显增加.膜脂过氧化与抗氧化物酶的变化具有不一致性,这可能表明两种抗氧化系统之间存在一定的协同作用,特别是高含量的抗坏血酸,可能在高山植物适应高寒环境的胁迫方面具有更加重要的作用.图5表1参22

对生长在青藏高原不同海拔自然生境下的多年生典型抗寒植物-矮蒿草(Kobresia humilis)的抗氧化系统进行了比较研究。结果表明,矮蒿草的叶组织中,非酶抗氧化系统物质脯氨酸(Pro)和抗坏血酸(AsA),随着海拔升高具有明显的增加趋势。在抗氧化酶系统中,过氧化物酶(POD)和过氧化氢酶(CAT)活性均随海拔的升高,而明显增强。但叶中的超氧化物歧化酶(SOD),随着海拔的升高,其活性有下降趋势,三者变化趋势并不一致。高海拔矮蒿草的植株与低海拔的植株相比,叶细胞内的膜脂过氧化加剧,丙二醛(MDA)含量明显增。细胞可溶性蛋白也随海拔升高显著增加。根中的抗氧化系统变化与叶中的有所不同。根中AsA含量随海拔而显著升高,且较叶中的增加明显,但Pro含量则有所减少。根中的CAT和POD活性变化与叶中的变化趋势基本一致,且随海拔高度的增加,根中的CAT活性较叶中的变化更为明显。而根中的SOD活性变化不如叶中明显,MDA含量随海拔增高,其变化趋势比叶中的小。可见,青藏高原典型抗寒植物矮蒿草体内的两类抗氧化系统,在不同海拔条件下可能存在互补协同的调节作用,这可能是矮蒿草适应或抵抗高原极端高寒低温和强UV-B辐射等环境胁迫的重要生理机制之一。

目的 观察藏成药安神丸饱和脂及不饱和脂提取物对大鼠脂质代谢及抗氧化功能的影响。方法 测定连续ig给药4周、8周及给药8周又停药4周后大鼠血清中丙二醛(MDA)、还原型谷胱甘肽(GSH)及高密谋脂蛋白(HDL)的含量。结果 安神丸的脂溶性提取物可明显增加较大龄鼠血清中的GSH和HDL含量。结论 安神丸脂溶性提取物可改善较大龄鼠脂质代谢功能,提高其抗氧化能力。

为了确定高在鼢鼠(Myospalax baileyi)体内抗缺氧水成分,通过超临界萃取(SCEF)方法从高原鼢鼠肌肉提取脂溶性部分,我们对其成分通过气相色谱-质谱(GC-MS)联用分析。为了证实高原鼢鼠肌肉脂溶性成分的抗缺氧效果,探讨其抗缺氧机制,我们以小鼠为实验动物,分为5个组,即高剂量实验组(HEG),中剂量实验组(MEG),低剂量实验组(LEG),阳性药物对照组(PEG),空白对照组(CEG),分别以浓度为20%萃取物,10%萃取物,5%萃取物,10%的红景天和蒸馏水连续饲喂10天,然后进行小鼠常压抗缺氧实验,测定小鼠血清超氧化物歧化酶(SOD),谷光甘肽过氧化物酶(GSH-PX),乳酸脱氢酶(LDH)活性及血清丙二(MAD)含量。结果发现,高原鼢鼠肌肉脂溶性物质提取率为11.5%,共确定了17种成分,主要成分是n-十六烷酸(32.293%),n-十廿烷酸(1.109%),n-十八烷酸(6.03%),二-羟基-环十五酮(1.198%),顺十六烯酸(6.13%),反十八烯酸(27.3%),和亚油酸(81.4%),小鼠抗缺氧实验和生化指标测试结果表明,高原鼢鼠肌从脂溶性成分在20%,10%,5%时,与空白对照组相比,都不同程度地延长小鼠在缺氧条件下的存活时间,提高小鼠血清SOD和GSH-PX活性,降低血清LDH活性,减少血清MDA含量。

The effects of La3+ on the antioxidant enzyme activities and the relative indices of cellular damage in cucumber seedling leaves were studied. When cucumber seedlings were treated with low concentrations of LaCl3 (0.002 and 0.02 mM), peroxidase (PO) activity increased, and catalase (CAT) activity was similar to that of control leaves at 0.002 mM La3+ and increased at 0.02 mM La3+, whereas superoxide dismutase (SOD) activity did not change significantly. The increase in the contents of chlorophyll (including chlorophylls a and b), carotenoids in parallel with the decrease in the level of malondialdehyde (MDA) suggested that low concentration of La3+ promoted plant growth. However, except the increase in SOD activity at 2 mM La3+, CAT and PO activities and the contents of pigments decreased at high concentrations of La3+ (0.2 and 2 mM), leading to the increase of MDA content and the inhibition of plant growth. It is suggested that lanthanum ion is involved in the regulation of active oxygen-scavenging enzyme activities during plant growth.

Various carbonaceous deposits are formed during the course of methane dehydro-aromatization (MDA) under non-oxidative conditions on Mo/MCM-22 catalysts. These carbon species were investigated by various temperature-programmed techniques such as TPH and TPCO2, combining with TPO and TGA results in order to reveal their chemical nature and determine their amounts. The TPO profiles recorded from coked Mo/MCM-22 catalysts show two temperature peaks: one at about 756 K and the other at about 876 K. The coke amounts related to these two peaks were determined on the basis of the corresponding corrected and deconvoluted TPO profiles, combining with the TGA profiles concerned.

Carbonaceous deposits formed during the temperature-programmed surface reaction (TPSR) of methane dehydro-aromatization (MDA) over Mo/HZSM-5 catalysts have been investigated by TPH, TPCO2 and TPO, in combination with thermal gravimetric analysis (TG). The TPO profiles of the coked catalyst after TPSR of MDA show two temperature peaks: one is at about 776 K and the other at about 865 K. The succeeding TPH experiments only resulted in the diminishing of the area of the high-temperature peak, and had no effect on the area of the low-temperature peak. On the other hand, the TPO profiles of the coked catalyst after succeeding TPCO2 experiments exhibited obvious reduction in the areas of both the high-and low-temperature peaks, particularly in the area of the low-temperature peak. On the basis of TPSR, TPR and TPCO2 experiments and the corresponding TG analysis, quantitative analysis of the coke and the kinetics of its burning-off process have been studied. A single-pass process with the combination of oxidative coupling (OCM) and dehydro-aromatization (MDA) for the direct conversion of methane is carried out. With the assistance of the OCM reaction over the SrO-La2O3/CaO catalyst loaded on top of the catalyst bed, the duration of the dehydro-aromatization reaction catalyzed by a 6Mo/HMCM-49 catalyst shows a significant improvement, and. the initial deactivation rate constant of the overall process revealed about 1.5 x 10(-6) s(-1). Up to 72 h on stream, the yield of aromatics was still maintained at 5.0% with a methane conversion of 9.6%, which is obviously higher than that reported for the conventional MDA process with single catalyst. Upon the TPR results, this wonderful enhancement would be attributed to an in-situ formation of CO2 and H2O through the OCM reaction, which serves as a scavenger for actively removing the coke formed during the MDA reaction via a reverse Boudouard reaction and the water gas reaction as well.

The catalytic performances of methane dehydroaromatization (MDA) under non-oxidative conditions over 6 wt.% Mo/HZSM-5 catalysts calcined for different durations of time at 773 K have been investigated in combination with ex situ H-1 MAS NMR characterization. Prolongation of the calcination time at 773 K is in favor of the diffusion of the Mo species on the external surface and the migration of Mo species into the channels, resulting in a further decrease in the number of Bronsted acid sites, while causing only a slight change in the Mo contents of the bulk and in the framework structure of the HZSM-5 zeolite. The MoQ(x) species associated and non-associated with the Bronsted acid sites can be estimated quantitatively based on the 1H MAS NMR measurements as well as on the assumption of a stoichiometry ratio of 1: 1 between the Mo species and the Bronsted acid sites. Calcining the 6 wt.% Mo/HZSM-5 catalyst at 773 K for 18 h can cause the MoOx species to associate with the Bronsted acid sites, while a 6 Wt-% MO/SiO2 sample can be taken as a catalyst in which all MoOx species are non-associated with the Bronsted acid sites. The TOF data at different times on stream on the 6 wt.% Mo/HZSM-5 catalyst calcined at 773 K for 18 h and on the 6 Wt-% MO/SiO2 catalyst reveal that the MoCx species formed from MoOx associated with the Bronsted acid sites are more active and stable than those formed from MoOx non-associated with the Bronsted acid sites. An analysis of the TPO profiles recorded on the used 6 wt.% Mo/HZSM-5 catalysts calcined for different durations of time combined with the TGA measurements also reveals that the more of the MoCx species formed from MoOx species associated with the Br6nsted acid sites, the lower the amount of coke that will be deposited on it. Exercise improves functional capacity in spinal cord injury (SCI). However, exhaustive exercise, especially when sporadic, is linked to the production of reactive oxygen species that may have a detrimental effect on SCI. We aimed to study the effect of a single bout of exhaustive exercise on systemic oxidative stress parameters and on the expression of antioxidant enzymes in individuals with paraplegia. The study was conducted in the Physical Therapy department and the Physical Education and Sports department of the University of Valencia. Sixteen paraplegic subjects were submitted to a graded exercise test (GET) until volitional exhaustion. They were divided into active or non-active groups. Blood samples were drawn immediately, 1 and 2 h after the GET. We determined plasma malondialdehyde (MDA) and protein carbonylation as markers of oxidative damage. Antioxidant gene expression (catalase and glutathione peroxidase-GPx) was determined in peripheral blood mononuclear cells. We found a significant increase in plasma MDA and protein carbonyls immediately after the GET (P<0.05). This increment correlated significantly with the lactate levels. Active paraplegics showed lower levels of exercise-induced oxidative damage (P<0.05) and higher exercise-induced catalase (P<0.01) and GPx (P<0.05) gene expression after the GET. These results suggest that exercise training may be useful in SCI patients to develop systemic antioxidant defenses that may protect them against exercise-induced oxidative damage.

The transcription factor Ets-1 is implicated in various physiological processes and invasive pathologies. We identified a novel variant of ets-1, ets-1Delta(III-VI), resulting from the alternative splicing of exons III to VI. This variant encodes a 27 kDa isoform, named Ets-1 p27. Ets-1 p27 lacks the threonine-38 residue, the Pointed domain and the transactivation domain, all of which are required for the transactivation of Ets-1 target genes. Both inhibitory domains surrounding the DNA-binding domain are conserved, suggesting that Ets-1 p27, like the full-length Ets-1 p51 isoform, is autoinhibited for DNA binding. We showed that Ets-1 p27 binds DNA in the same way as Ets-1 p51 does and that it acts both at a transcriptional and a subcellular localization level, thereby constituting a dual-acting dominant negative of Ets-1 p51. Ets-1 p27 blocks Ets-1 p51-mediated transactivation of target genes and induces the translocation of Ets-1 p51 from the nucleus to the cytoplasm. Furthermore, Ets-1 p27 overexpression represses the tumor properties of MDA-MB-231 mammary carcinoma cells in correlation with the known implication of Ets-1 in various cellular mechanisms. Thus the dual-acting dominant-negative function of Ets-1 p27 gives to the Ets-1 p27/Ets-1 p51 ratio a determining effect on cell fate.

This work proceeds from the assumption that a European environmental information and communication system (EEICS) is already established. In the context of primary users (land-use planners, conservationists, and environmental researchers) we ask what use may be made of the EEICS for building models and tools which is of use in building decision support systems for the land-use planner. The complex task facing the next generation of environmental and forest modellers is described, and a range of relevant modelling approaches are reviewed. These include visualization and GIS; statistical tabulation and database SQL, MDA and OLAP methods. The major problem of noncomparability of the definitions and measures of forest area and timber volume is introduced and the possibility of a model-based solution is considered. The possibility of using an ambitious and challenging biogeochemical modelling approach to understanding and managing European forests sustainably is discussed. It is emphasised that all modern methodological disciplines must be brought to bear, and a heuristic hybrid modelling approach should be used so as to ensure that the benefits of practical empirical modelling approaches are utilised in addition to the scientifically well-founded and holistic ecosystem and environmental modelling. The data and information system required is likely to end up as a grid-based-framework because of the heavy use of computationally intensive model-based facilities.
