In un quadro internazionale di forte interesse verso uno sviluppo sostenibile e sfide energetiche per il futuro, il DIEM, in collaborazione con altri istituti di ricerca ed imprese private, sta progettando l’integrazione di componentistica avanzata su di una caldaia alimentata a biomasse. Lo scopo finale è quello di realizzare una caldaia a biomasse che produca energia in maniera più efficiente e con un impatto ambientale ridotto. L’applicazione è indirizzata inizialmente verso caldaie di piccola-media taglia (fino a 350 kW termici) vista la larga diffusione di questa tipologia di impianto. La componentistica in oggetto è: - filtro sperimentale ad alta efficienza per la rimozione del particolato; - celle a effetto Seebeck per la produzione di energia elettrica direttamente da energia termica senza parti meccaniche in movimento; - pompa Ogden per la produzione di energia meccanica direttamente da energia termica; La finalità dell’attività di ricerca è la progettazione dell’integrazione dei suddetti dispositivi con una caldaia a biomassa da 290 kW termici per la realizzazione di un prototipo di caldaia stand-alone ad impatto ambientale ridotto: in particolare, la caldaia è in grado, una volta raggiunte le condizioni di regime, di autoalimentare le proprie utenze elettriche, garantendo il funzionamento in sicurezza in caso di black-out o consentendo l’installazione della caldaia medesima in zone remote e prive di allaccio alla rete elettrica. Inoltre, la caldaia può fornire, tramite l'utilizzo di una pompa a vapore o pompa Ogden, energia meccanica per il pompaggio di fluidi: tale opportunità si ritiene particolarmente interessante per l'integrazione della caldaia nel caso di installazione in ambito agricolo. Infine, l'abbinamento di un filtro ad alta efficienza e basso costo consente l'abbattimento delle emissioni inquinanti, favorendo una maggiore diffusione della tecnologia senza ulteriori impatti sull'ambiente.

La semplicità, affidabilità, robustezza ed economicità del motore asincrono trifase ne fanno un motore largamente utilizzato sia in ambiente industriale che civile (pompe, ascensori, montacarichi, nastri trasportatori, ventilatori, locomotive ferroviarie, …). È ipotizzabile che in futuro vi sarà sempre più largo impiego di motori elettrici e i recenti sviluppi nella tecnologia dei convertitori elettronici per il controllo dei motori asincroni ne consentiranno l’uso in sempre più estesi ambiti applicativi. La messa sotto tensione di motori a media/alta potenza presenta però, in fase di avviamento, correnti di spunto e coppie elevate che devono essere controllate. In questa tesi saranno pertanto studiate le tecniche di protezione, avviamento e controllo dei motori asincroni trifase Nel primo capitolo esamineremo le funzioni di sezionamento, comando e protezione da cortocircuiti e sovraccarichi e i dispositivi adatti a svolgere tale compito in modo coordinato. Nel secondo capitolo esamineremo le principali tecnologie usate per l’avviamento dei motori, da quelle più semplici a quelle che impiegano dispositivi elettronici allo stato solido: Soft-Starters a tiristori (solo regolazione AC), Inverters a transistori (controllo sia in frequenza che ampiezza della forma d’onda AC generata). Fra tutti i possibili convertitori DC/AC analizzeremo dettagliatamente esclusivamente quelli a controllo PWM analogico che costituiranno l’oggetto della trattazione dei capitoli seguenti.

Space-based (satellite, scientific probe, space station, etc.) and millimeter – to – microscale (such as are used in high power electronics cooling, weapons cooling in aircraft, etc.) condensers and boilers are shear/pressure driven. They are of increasing interest to system engineers for thermal management because flow boilers and flow condensers offer both high fluid flow-rate-specific heat transfer capacity and very low thermal resistance between the fluid and the heat exchange surface, so large amounts of heat may be removed using reasonably-sized devices without the need for excessive temperature differences. However, flow stability issues and degradation of performance of shear/pressure driven condensers and boilers due to non-desirable flow morphology over large portions of their lengths have mostly prevented their use in these applications. This research is part of an ongoing investigation seeking to close the gap between science and engineering by analyzing two key innovations which could help address these problems. First, it is recommended that the condenser and boiler be operated in an innovative flow configuration which provides a non-participating core vapor stream to stabilize the annular flow regime throughout the device length, accomplished in an energy-efficient manner by means of ducted vapor re-circulation. This is demonstrated experimentally. Second, suitable pulsations applied to the vapor entering the condenser or boiler (from the re-circulating vapor stream) greatly reduce the thermal resistance of the already effective annular flow regime. For experiments reported here, application of pulsations increased time-averaged heat-flux up to 900 % at a location within the flow condenser and up to 200 % at a location within the flow boiler, measured at the heat-exchange surface. Traditional fully condensing flows, reported here for comparison purposes, show similar heat-flux enhancements due to imposed pulsations over a range of frequencies. Shear/pressure driven condensing and boiling flow experiments are carried out in horizontal mm-scale channels with heat exchange through the bottom surface. The sides and top of the flow channel are insulated. The fluid is FC-72 from 3M Corporation.

Whole-body vibration exposure of locomotive engineers and the vibration attenuation of seats in 22 U.S. locomotives (built between 1959 and 2000) was studied during normal revenue service and following international measurement guidelines. Triaxial vibration measurements (duration mean 155 min, range 84-383 min) on the seat and on the floor were compared. In addition to the basic vibration evaluation (aw rms), the vector sum (av), the maximum transient vibration value (MTVV/aw), the vibration dose value (VDV/(aw T1/4)), and the vibration seat effective transmissibility factor (SEAT) were calculated. The power spectral densities are also reported. The mean basic vibration level (aw rms) was for the fore-aft axis x = 0.18 m/sec2, the lateral axis y = 0.28 m/sec2, and the vertical axis z = 0.32 m/sec2. The mean vector sum was 0.59 m/sec2 (range 0.27 to 1.44). The crest factors were generally at or above 9 in the horizontal and vertical axis. The mean MTVV/aw was 5.3 (x), 5.1 (y), and 4.8 (z), and the VDV/(aw T1/4) values ranged from 1.32 to 2.3 (x-axis), 1.33 to 1.7 (y-axis), and 1.38 to 1.86 (z-axis), generally indicating high levels of shocks. The mean seat transmissibility factor (SEAT) was 1.4 (x) and 1.2 (y) and 1 (z), demonstrating a general ineffectiveness of any of the seat suspension systems. In conclusion, these data indicate that locomotive rides are characterized by relatively high shock content (acceleration peaks) of the vibration signal in all directions. Locomotive vertical and lateral vibrations are similar, which appears to be characteristic for rail vehicles compared with many road/off-road vehicles. Tested locomotive cab seats currently in use (new or old) appear inadequate to reduce potentially harmful vibration and shocks transmitted to the seated operator, and older seats particularly lack basic ergonomic features regarding adjustability and postural support.

The influence of the nest location and the placement of nipple drinkers on nest use by laying hens in a commercial aviary was assessed. Twenty pens in a laying hen house were equipped with the same commercial aviary system, but the pens differed in the nest location and the placement of nipple drinkers. Nests were placed along the walls in 10 pens, and nipple drinkers were installed in front of the nests in 5 of these pens. The other 10 pens were equipped with nests placed on a tier within the aviary (integrated nests). Nipple drinkers were installed in front of the nests in 5 of these pens. A total of 225 Lohmann Selected Leghorns were housed per pen. The hens were offered 4 nests per pen: 2 facing the service corridor of the laying hen house and 2 facing the outdoor area. The numbers of nest eggs and mislaid eggs were counted daily per pen. At 25, 36, and 43 wk of age, the nest platforms were videotaped and the behavior of laying hens in front of the nests was analyzed. The nest location affected the stationary and locomotive behaviors in front of the nests. Hens in front of the integrated nests and the nests with drinkers displayed more stationary behaviors than hens in front of wall-placed nests or nests without drinkers. No difference in the number of nest eggs could be detected, but the integration of the nests inside the aviary led to a more even distribution of hens while nest searching. In the pens with wall-placed nests, significantly more hens laid eggs in the nests at the wall near the service corridor than at the wall near the outdoor area. Due to this imbalance, crowding in front of the preferred nests occurred and pushing and agonistic interactions on the nest platforms were significantly more frequent. Placement of nipple drinkers in front of nests had no effect on the number of eggs laid in those nests.

The nematode Caenorhabditis elegans is a well-known model organism used to investigate fundamental questions in biology. Motility assays of this small roundworm are designed to study the relationships between genes and behavior. Commonly, motility analysis is used to classify nematode movements and characterize them quantitatively. Over the past years, C. elegans' motility has been studied across a wide range of environments, including crawling on substrates, swimming in fluids, and locomoting through microfluidic substrates. However, each environment often requires customized image processing tools relying on heuristic parameter tuning. In the present study, we propose a novel Multi-Environment Model Estimation (MEME) framework for automated image segmentation that is versatile across various environments. The MEME platform is constructed around the concept of Mixture of Gaussian (MOG) models, where statistical models for both the background environment and the nematode appearance are explicitly learned and used to accurately segment a target nematode. Our method is designed to simplify the burden often imposed on users; here, only a single image which includes a nematode in its environment must be provided for model learning. In addition, our platform enables the extraction of nematode ‘skeletons’ for straightforward motility quantification. We test our algorithm on various locomotive environments and compare performances with an intensity-based thresholding method. Overall, MEME outperforms the threshold-based approach for the overwhelming majority of cases examined. Ultimately, MEME provides researchers with an attractive platform for C. elegans' segmentation and ‘skeletonizing’ across a wide range of motility assays.

Scientific background: Marine mammals use sound for communication, navigation and prey detection. Acoustic sensors therefore allow the detection of marine mammals, even during polar winter months, when restricted visibility prohibits visual sightings. The animals are surrounded by a permanent natural soundscape, which, in polar waters, is mainly dominated by the movement of ice. In addition to the detection of marine mammals, acoustic long-term recordings provide information on intensity and temporal variability of characteristic natural and anthropogenic background sounds, as well as their influence on the vocalization of marine mammals Scientific objectives: The PerenniAL Acoustic Observatory in the Antarctic Ocean (PALAOA, Hawaiian "whale") near Neumayer Station is intended to record the underwater soundscape in the vicinity of the shelf ice edge over the duration of several years. These long-term recordings will allow studying the acoustic repertoire of whales and seals continuously in an environment almost undisturbed by humans. The data will be analyzed to (1) register species specific vocalizations, (2) infer the approximate number of animals inside the measuring range, (3) calculate their movements relative to the observatory, and (4) examine possible effects of the sporadic shipping traffic on the acoustic and locomotive behaviour of marine mammals. The data, which are largely free of anthropogenic noise, provide also a base to set up passive acoustic mitigation systems used on research vessels. Noise-free bioacoustic data thereby represent the foundation for the development of automatic pattern recognition procedures in the presence of interfering sounds, e.g. propeller noise.

El alcance del proyecto es describir las directrices técnicas, la definición de criterios, y la estrategia de suministro de energía (Electricidad. Calefacción y Agua Caliente Sanitaria (ACS)) a un barrio modelo situado en un entorno urbano. De inicio se estudia los diversos modelos energéticos atendiendo a la normativa y tecnología, que se pueden aplicar en un conjunto residencial, dando como resultado el modelo propuesto de abastecimiento energético, mediante calefacción de distrito, que incorporara el diseño de una planta de producción de energía termo-eléctrica o Central de Energías basada en la tecnología de condensación de baja temperatura para calefacción y A.C.S. incluyendo una cogeneración con pila de combustible. Al mismo tiempo se han calculado y diseñado una serie de chimeneas externas para dar cumplida necesidad técnica y legal al proyecto. Estos estudios nos sirven de punto de partida para analizar la amortización de la inversión y por tanto la rentabilidad y viabilidad del proyecto, comparándose con los costes económicos derivados de la generación por sistemas convencionales. Para finalizar se hace mención a las ventajas medioambientales y a los grados de seguridad en la planta de producción ABSTRACT The scope of this work is the description of an energy supply project ( Electricity, heat and hot water ) to a housing development in a urban neibourhood , including technical criteria in their different options. Initially, several solutions are studied based on available technologies and legal restrictions. The final proposal is based on the district hearing model including electricity production in cogeneration via fuel cell technology as well as heating and hot water produced by low temperature condensation boilers It includes calculations and design criteria of the exhaust gases system and chimeneys in compliance with legal requirement in urban areas. This work also includes an economical model including payback, IRR and VAN analysis and an economical comparaison with the standard solutions. Finally, environmental advantages of the preferred solution over other standards as well as safety issues are also presented.

La finalidad del proyecto consiste en realizar la sustitución de calderas de gasóleo por calderas de biomasa para suministrar calefacción a edificios urbanos. En primer lugar, se ha hecho un estudio de la demanda calorífica de los edificios, y posteriormente se ha estudiado el ciclo completo de la biomasa, incluyendo el suministro y tipo de biomasa, dimensiones de los equipos y espacios necesarios para la instalación. Después de estudiar la viabilidad técnica y económica, podemos concluir que utilizar biomasa en edificios con calderas centralizadas nos aporta un gran ahorro en comparación con el gasóleo, además de evitar emisiones de CO2 y utilizar un combustible renovable y de producción nacional. ABSTRACT The purpose of the project consists of performing the substitution of diesel boilers by biomass boilers to provide the heating to urban buildings. In first place, it has been made a study of the calorific demand of the buildings. In addition, it has been studied the completed cycle of the biomass even including the supply and the type of biomass, the dimensions of the machines and the necessary spaces to carry out the installation. After studying the technical and economical viability, we can conclude that using biomass in buildings with central boilers provides us an important saving in comparison with diesel as well as to avoid CO2 emissions and using a renewable combustible of national production.

En este escrito se pretende llevar a cabo la realización de un proyecto técnico sobre eficiencia energética en edificios. El proyecto comienza con el estudio de la demanda térmica de un edificio, es decir, de sus necesidades de calefacción, refrigeración y agua caliente sanitaria (ACS). Una vez realizado este análisis, se procede al estudio de distintas alternativas para la disminución del coste asociado al consumo energético del edificio, las cuales son: mejora del aislamiento de la envolvente térmica del edificio, instalación de colectores solares para la producción de ACS y el cambio de los quemadores de las calderas. De estas medidas se han presupuestado aquellas que poseen un bajo periodo de retorno de la inversión, y se ha desarrollado su planificación, programación temporal y análisis económico. ABSTRACT In this document is pretended to be carry out the performance of a technical project about energy efficiency in buildings. The project begins with the study of the thermal demand of the building, that is to say, the heating, cooling and domestic hot water (DHW) needs. Once the analysis has been done, is proceeded the study of different alternatives for diminishing the cost associate to the building energy consumption, which are: improvement of the insulation of the building, solar collector installation to produce DHW and change of the burner of the boilers. Among this measures, has been budgeted the ones that possess a low invest pay-back, and it is been developed their planning, their temporal programming and their economic analysis.

Concentrating Solar Power (CSP) plants typically incorporate one or various auxiliary boilers operating in parallel to the solar field to facilitate start up operations, provide system stability, avoid freezing of heat transfer fluid (HTF) and increase generation capacity. The environmental performance of these plants is highly influenced by the energy input and the type of auxiliary fuel, which in most cases is natural gas (NG). Replacing the NG with biogas or biomethane (BM) in commercial CSP installations is being considered as a means to produce electricity that is fully renewable and free from fossil inputs. Despite their renewable nature, the use of these biofuels also generates environmental impacts that need to be adequately identified and quantified. This paper investigates the environmental performance of a commercial wet-cooled parabolic trough 50 MWe CSP plant in Spain operating according to two strategies: solar-only, with minimum technically viable energy non-solar contribution; and hybrid operation, where 12 % of the electricity derives from auxiliary fuels (as permitted by Spanish legislation). The analysis was based on standard Life Cycle Assessment (LCA) methodology (ISO 14040-14040). The technical viability and the environmental profile of operating the CSP plant with different auxiliary fuels was evaluated, including: NG; biogas from an adjacent plant; and BM withdrawn from the gas network. The effect of using different substrates (biowaste, sewage sludge, grass and a mix of biowaste with animal manure) for the production of the biofuels was also investigated. The results showed that NG is responsible for most of the environmental damage associated with the operation of the plant in hybrid mode. Replacing NG with biogas resulted in a significant improvement of the environmental performance of the installation, primarily due to reduced impact in the following categories: natural land transformation, depletion of fossil resources, and climate change. However, despite the renewable nature of the biofuels, other environmental categories like human toxicity, eutrophication, acidification and marine ecotoxicity scored higher when using biogas and BM.

Vinculin, a major constituent of focal adhesions and zonula adherens junctions, is thought to be involved in linking the microfilaments to areas of cell-substrate and cell-cell contacts. To test the role of vinculin in cell adhesion and motility, we used homologous recombination to generate F9 embryonal carcinoma and embryonic stem cell clones homozygous for a disrupted vinculin gene. When compared to wild-type cells, vinculin-mutant cells displayed a rounder morphology and a reduced ability to adhere and spread on plastic or fibronectin. Decreased adhesion of the mutant cells was associated with a reduction in lamellipodial extensions, as observed by time-lapse video microscopy. The locomotive activities of control F9 and the vinculin-null cells were compared in two assays. Loss of vinculin resulted in a 2.4-fold increase in cell motility. These results demonstrate an important role for vinculin in determining cell shape, adhesion, surface protrusive activity, and cell locomotion.

O principal resíduo da indústria de café solúvel é a borra de café, gerada após a extração de sólidos solúveis com água, sendo usualmente queimada em caldeiras para geração de energia para a própria indústria. Entretanto, este co-produto pode conter de 15 a 20 % de óleo, componente de grande interesse na indústria alimentícia. Em paralelo, na indústria de produção de óleos vegetais o solvente frequentemente utilizado é o hexano. Contudo, por este ser um derivado de combustíveis fósseis, alternativas para sua substituição por solventes mais amigáveis ao meio ambiente, que podem ser obtidos por via biotecnológica, estão sendo estudadas. Deste modo, o objetivo principal desta dissertação de mestrado foi a viabilização técnica do emprego de solventes alcoólicos no processo de extração de óleo de borra de café proveniente da indústria de processamento de café solúvel. Foram realizadas extrações sólido-líquido em um estágio para estudar a influência das variáveis de processo temperatura (60 a 90 °C), tipo de solvente (etanol, ET ou isopropanol, IPA) e a hidratação do solvente (absoluto ou azeotrópico) nas características das fases extrato e rafinado, em termos de rendimento de extração de óleo, de ácidos clorogênicos (ACG), de carboidratos totais, teor de proteínas e índice de solubilidade de nitrogênio (ISN) da fase rafinado. Pré-tratamento enzimático ao processo de extração também foi realizado para investigar a sua atuação sobre o rendimento de extração de óleo e ACG, além do ISN das fases rafinado obtidas na temperatura de 70 °C. De forma geral, pode-se inferir que a temperatura favorece a extração de compostos lipídicos, mas a hidratação do solvente prejudica a extração destes compostos pelo aumento da polaridade do solvente. Do mesmo modo, como o ET é mais polar que o IPA, o primeiro solvente proporcionou a obtenção de menores rendimentos de extração de óleo. A temperatura de processo também influenciou a extração de ACG, a qual foi beneficiada a temperaturas mais baixas pelo aumento da polaridade dos solventes utilizados. De tal forma, que a 60 °C, nos experimentos utilizando etanol azeotrópico obteve-se os menores rendimentos de extração de óleo, porém maior rendimento de extração de ACG. O pré-tratamento enzimático apresentou diferenças significativas nas características das fases extrato e rafinado. No entanto, somente os experimentos com etanol absoluto resultaram em rendimentos de extração de óleo economicamente viáveis. De fato, será relevante um estudo mais aprofundado das variáveis de pré-tratamento enzimático para obter resultados mais expressivos em relação à extração de compostos lipídicos. Diante dos dados experimentais obtidos, conclui-se que é tecnicamente viável o emprego de solventes alcoólicos no processo de extração de óleo de borra de café. Entretanto, nota-se que as condições de processo devem ser avaliadas minuciosamente com o objetivo de se obter altos rendimentos de extração de óleo com maior teor de ACG.

É reconhecida a importância da avicultura para o agronegócio e para o desenvolvimento do Brasil. Mas o País tem enfrentado grande concorrência internacional, que se traduz em barreiras sanitárias e exigências cada vez maiores de controle de seu rebanho por parte dos importadores. Neste sentido, o Ministério da Agricultura, Pecuária e Abastecimento (MAPA) publicou uma série de atos legais para viabilizar a organização dos programas de saúde animal, entre eles as Instruções Normativas n° 56/2007, n° 59/2009, n°36/2012 e n°10/2013, para estabelecer os procedimentos para registro e fiscalização de estabelecimentos avícolas comerciais e de reprodução, voltados à biosseguridade do sistema avícola. Como todos os setores produtivos, a avicultura é fortemente influenciada pela sua estrutura de custos, de modo que há um certo sentimento de que a adequação às medidas de biosseguridade, preconizadas pelas normativas, pode impactar a atividade de produção de ovos comerciais de modo a levar, eventualmente, alguns avicultores ao abandono da mesma. O objetivo deste trabalho foi analisar o impacto socioeconômico das políticas sanitárias para estabelecimentos avícolas comerciais de postura da regional agropecuária de Limeira, Estado de São Paulo, analisando o perfil socioeconômico da propriedade e do produtor e, estimando o custo de produção e seu impacto em atendimento às medidas de biosseguridade preconizadas pelas normativas. Para a análise do perfil das propriedades foram utilizados os dados dos 28 estabelecimentos avícolas, levantados por meio de documentação para registro. Para a análise do perfil do produtor e para estimar os custos de produção foram considerados dez estabelecimentos, voluntários à pesquisa, visitados entre os meses de junho e julho de 2013. O resultado do estudo sugere que as adequações à biosseguridade podem ser factíveis de serem realizadas economicamente, sendo o custo relativamente pequeno frente aos possíveis riscos de enfermidades, representando entre 1,61% e 2,09% do custo total de produção. No entanto, as sucessivas alterações nas legislações podem fazer com que o programa de sanidade avícola perca a credibilidade diante da sociedade e dos produtores que demonstram resistência às mudanças nos paradigmas zoosanitários

To improve the understanding of the development of locomotor capacity in layer hens, we measured how female laying hen chicks (n=120) of four different strains (LSL-lite, Hyline Brown, Dekalb White, Lohmann Brown; 3 groups of 10 chicks per line) utilized the ground, the air, elevated horizontal (platforms and perches) and inclined surfaces (ramps and ladders) in an aviary until 9 weeks of age. We used infra-red video recordings to perform all-occurrences sampling of locomotive behavioural and perching events that occurred on the ground—where bedding material, food and water were provided, in the air, and on elevated horizontal and inclined surfaces within weekly 30-min sampling periods. Chicks preferred level ground during the first week of life compared to weeks 5–9 (P<0.0001) and performed 52% of all behavioural events in this section. Elevated surface use began at 2 weeks of age and increased over time (P=0.003), where most behaviour was performed in S2 (45% of all events). Chicks preferred horizontal to inclined surfaces, which were used from weeks 2–5 with maximum use occurring during weeks 2 and 3. Lohmann LSL chicks used the space above the ground most frequently (P=0.05) and performed more aerial ascent/descent behaviour than other lines (P<0.0001). Overall activity levels declined with age (P<0.0001). In summary layer chicks almost exclusively locomoted on the ground but utilized elevated horizontal surfaces (perch, first platform) as early as 2 weeks. These results provide information for improving space use in rearing aviaries by introducing lower perches, platforms and ramps/ladders to accommodate age-dependent locomotor abilities.
