The whale shark is the largest fish in the ocean, A tourism industry based on interacting with whale sharks has developed recently in Ningaloo Marine Park, off the coast of Western Australia, This is the only known, accessible place in the world where whale sharks congregate in significant numbers, Results from surveys of participants in the 'whale shark experience' are reported, with the experience found to be extremely satisfying for most consumers, Yet management questions relating to diver-to-shark separation distances, the appropriate number of operators, and the use of regulation, self-regulation and economic instruments, remain, The nexus between experiential aspects of the whale shark attraction and such management concerns is examined, and the usefulness of appropriate research in resolving management questions established, It is concluded that a judicious combination of management approaches is required to ensure the sustainable development of the whale shark tourism industry. The Egr proteins, Egr-1, Egr-2, Egr-3 and Egr-4, are closely related members of a subclass of immediate early gene-encoded, inducible transcription factors. They share a highly homologous DNA-binding domain which recognises an identical DNA response element. In addition, they have several less-well conserved structural features in common. As immediate early proteins, the Egr transcription factors are rapidly induced by diverse extracellular stimuli within the nervous system in a discretely controlled manner. The basal expression of the Egr proteins in the developing and adult rat brain and the induction of Egr proteins by neurotransmitter analogue stimulation, physiological mimetic and brain injury paradigms is reviewed. We review evidence indicating that Egr proteins are subject to tight differential control through diverse mechanisms at several levels of regulation. These include transcriptional, translational and posttranslational (including glycosylation, phosphorylation and redox) mechanisms and protein-protein interaction. Ultimately the differentially co-ordinated Egr response may lead to discrete effects on target gene expression. Some of the known target genes of Egr proteins and functions of the Egr proteins in different cell types are also highlighted. Future directions for research into the control and function of the different Egr proteins are also explored. (C) 1997 Elsevier Science Ltd.

This paper outlines the ethical arguments used in the Australian debate about whether or not to relax the prohibition on cannabis use by adults. Over the past two decades a rising prevalence of cannabis use in the Australian population has led to proposals for the decriminalization of the personal use of cannabis. Three states and territories have removed criminal penalties for personal use while criminal penalties are rarefy imposed in the remaining states. Libertarian arguments for legalization of cannabis use have attracted a great deal of media interest but very little public and political support. Other arguments in favour of decriminalization have attracted more support. One has been the utilitarian argument that prohibition has failed to deter cannabis use and the social costs of its continuation outweigh any benefits that it produces. Another has been the argument from hypocrisy that cannabis is less harmful than alcohol and so, on the grounds of consistency, if alcohol is legally available then so should cannabis. To date public opinion has not favoured legalization, although support for the decriminalization of personal cannabis use has increased. In the long term, the outcome of the debate may depend more upon trends in cannabis use and social attitudes among young adults than upon the persuasiveness of the arguments for a relaxation of the prohibition of cannabis.

Glioblastoma is the most frequent and malignant brain tumor, characterized by an elevated capacity for cellular proliferation and invasion. Recently, it was demonstrated that podoplanin membrane sialo-glycoprotein encoded by PDPN gene is over-expressed and related to cellular invasion in astrocytic tumors; however the mechanisms of regulation are still unknown. MicroRNAs are noncoding RNAs that regulate gene expression and several biological processes and diseases, including cancer. Nevertheless, their roles in invasion, proliferation, and apoptosis of glioblastoma are not completely understood. In this study, we focused on miR-29b and miR-125a, which were predicted to regulate PDPN, and demonstrated that these microRNAs directly target the 30 untranslated region of PDPN and inhibit invasion, apoptosis, and proliferation of glioblastomas. Furthermore, we report that miR-29b and miR-125a are downregulated in glioblastomas and also in CD133-positive cells. Taken together, these results suggest that miR-29b and miR-125a represent potential therapeutic targets in glioblastoma. (C) 2010 Wiley-Liss, Inc.

Background: Periodontal wound healing and regeneration require that new matrix be synthesized, creating an environment into which cells can migrate. One agent which has been described as promoting periodontal regeneration is an enamel matrix protein derivative (EMD). Since no specific growth factors have been identified in EMD preparations, it is postulated that EMD acts as a matrix enhancement factor. This study was designed to investigate the effect of EMD in vitro on matrix synthesis by cultured periodontal fibroblasts. Methods: The matrix response of the cells was evaluated by determination of the total proteoglycan synthesis, glycosaminoglycan profile, and hyaluronan synthesis by the uptake of radiolabeled precursors. The response of the individual proteoglycans, versican, decorin, and biglycan were examined at the mRNA level by Northern blot analysis. Hyaluronan synthesis was probed by identifying the isotypes of hyaluronan synthase (HAS) expressed in periodontal fibroblasts as HAS-2 and HAS-3 and the effect of EMD on the levels of mRNA for each enzyme was monitored by reverse transcription polymerase chain reaction (RTPCR). Comparisons were made between gingival fibroblast (GF) cells and periodontal ligament (PDLF) cells. Results: EMD was found to significantly affect the synthesis of the mRNAs for the matrix proteoglycans versican, biglycan, and decorin, producing a response similar to, but potentially greater than, mitogenic cytokines. EMD also stimulated hyaluronan synthesis in both GF and PDLF cells. Although mRNA for HAS-2 was elevated in GF after exposure to EMD, the PDLF did not show a similar response. Therefore, the point at which the stimulation of hyaluronan becomes effective may not be at the level of stimulation of the mRNA for hyaluronan synthase, but, rather, at a later point in the pathway of regulation of hyaluronan synthesis. In all cases, GF cells appeared to be more responsive to EMD than PDLF cells in vitro. Conclusions: EMD has the potential to significantly modulate matrix synthesis in a manner consistent with early regenerative events.

Interactions between Eph receptors and their ligands the ephrin proteins are critically important in many key developmental processes. Emerging evidence also supports a role for these molecules in postembryonic tissues, particularly in pathological processes, including tissue injury and tumor metastasis. We review the signaling mechanisms that allow the 14 Eph and nine ephrin proteins to deliver intracellular signals that regulate cell shape and movement. What emerges is that the initiation of these signals is critically dependent on which Eph and ephrin proteins are expressed, the level of their expression, and, in some cases, which splice variants are expressed. Diversity at the level of initial interaction and in the downstream signaling processes regulated by Eph-ephrin signaling provides a subtle, versatile system of regulation of intercellular adhesion, cell shape, and cell motility.

A partir da década de 1970, a literatura que trata daqueles setores de infraestrutura, cujo funcionamento requer um acompanhamento específico do Estado, vale dizer através da regulação, absorveu grande parte dos conceitos fundamentais desenvolvidos pelas escolas institucionalistas, notadamente pela Nova Economia Institucional (NEI). Entretanto, apesar dos avanços alcançados, esta literatura ainda carece de alguns desenvolvimentos teóricos e estudos empíricos que contribuam de forma decisiva. Tais questões estão ligadas, por exemplo, à evolução dos cenários e às condições econômicas que são alteradas a partir de mudanças significativas nas condições tecnológicas e que acabam por alterar significativamente aquela estrutura industrial para qual o modelo regulatório foi desenhado. Nesse sentido, as contribuições da Economia da Regulação e da NEI carecem de elementos evolucionários, notadamente schumpeterianos. O principal objetivo da dissertação é investigar como alguns elementos de inspiração schumpeteriana podem em algum grau ser aplicados à economia da regulação e neste sentido contribuir com proposições naqueles temas que essa escola fincou sua abordagem característica: o progresso tecnológico e o comportamento estratégico inovativo das empresas. Nesse sentido, o problema de pesquisa desse trabalho pode ser assim colocado: como pode ser entendido o papel do progresso tecnológico e da inovação nas teorias da regulação? A hipótese assumida neste trabalho é que não se pode desconsiderar a tecnologia e suas diversas implicações para a análise dos setores regulados, constatação essa cada vez mais clara, dadas as recentes transformações dessas indústrias: uma das principais características dos setores regulados, a partir da década de 1990, é o surgimento de novos marcos regulatórios e uma alteração progressiva dos modelos organizacionais que predominavam até então e um movimento de adaptação à margem da regulação vigente. Os resultados empíricos mostram que as transformações tecnológicas e institucionais ocorridas a partir de 1995 na indústria brasileira de telecomunicações alteraram significativamente a estrutura industrial e a dinâmica concorrencial do setor, colocando ao desenho regulatório – da Anatel – novos e complexos desafios, sobretudo, o de se adaptar à margem da dinamicidade característica das telecomunicações. Neste novo contexto, as funções regulatórias são mais complexas, o que exige um duplo processo de aprendizagem: quanto às estruturas de mercado e quanto ao comportamento estratégico das empresas.

O ruído é, por definição, um som desagradável ou indesejável que perturba o ambiente, contribuindo para o mal-estar físico e psíquico, podendo pôr em causa a saúde do ser humano. As situações de exposição nos espaços de repouso, lazer e trabalho a valores sonoros elevados, especialmente nos meios urbanos e suburbanos, têm-se multiplicado. Por outro lado, o desenvolvimento cultural dos cidadãos, cada vez mais cientes do direito à qualidade de vida, tem originado um aumento das exigências de conforto, influenciado directamente pela qualidade dos edifícios que habitam. Através da publicação do Regulamento Geral Sobre o Ruído – RGR – (aprovado pelo Decreto Lei nº251/87 de 24 de Junho) bem como o Regulamento dos Requisitos Acústicos dos Edifícios – RRAE - (aprovado pelo Decreto Lei nº129/02 de 11 de Maio), com as devidas alterações no Decreto-lei nº96/08, de 9 de Junho, foi permitido relacionar um conjunto de disposições normativas e legais, permitindo o estabelecimento de condições para a verificação das exigências fundamentais associadas ao conforto acústico dos edifícios (integração urbanística, isolamento sonoro a sons aéreos, isolamento a sons de percussão, exposição ao ruído durante o trabalho), justificando assim a necessidade da existência de regulamentação pelo facto da saúde dos indivíduos também se encontrar relacionada ao conforto acústico. Na presente dissertação, pretende-se dar um contributo para a elaboração da avaliação e certificação do comportamento acústico de edifícios de habitação através da definição dos principais aspectos a avaliar neste processo. Para tal, foi retratado o actual estado da arte e foi feito um levantamento das metodologias utilizadas pelas entidades acreditadas para os ensaios de acústica, tendo em conta a normalização e regulamentação existente, de maneira a obter o panorama destes diagnósticos de conforto acústico para a verificação da sua conformidade com as exigências regulamentares.

In order to develop a flexible simulator, a variety of models for Ancillary Services (AS) negotiation has been implemented in MASCEM – a multi-agent system competitive electricity markets simulator. In some of these models, the energy and the AS are addressed simultaneously while in other models they are addressed separately. This paper presents an energy and ancillary services joint market simulation. This paper proposes a deterministic approach for solving the energy and ancillary services joint market. A case study based on the dispatch of Regulation Down, Regulation Up, Spinning Reserve, and Non-Spinning Reserve services is used to demonstrate that the use of the developed methodology is suitable for solving this kind of optimization problem. The presented case study is based on CAISO real AS market data considers fifteen bids.

This paper proposes two meta-heuristics (Genetic Algorithm and Evolutionary Particle Swarm Optimization) for solving a 15 bid-based case of Ancillary Services Dispatch in an Electricity Market. A Linear Programming approach is also included for comparison purposes. A test case based on the dispatch of Regulation Down, Regulation Up, Spinning Reserve and Non-Spinning Reserve services is used to demonstrate that the use of meta-heuristics is suitable for solving this kind of optimization problem. Faster execution times and lower computational resources requirements are the most relevant advantages of the used meta-heuristics when compared with the Linear Programming approach.

Electricity market players operating in a liberalized environment requires access to an adequate decision support tool, allowing them to consider all the business opportunities and take strategic decisions. Ancillary services represent a good negotiation opportunity that must be considered by market players. For this, decision support tool must include ancillary market simulation. This paper proposes two different methods (Linear Programming and Genetic Algorithm approaches) for ancillary services dispatch. The methodologies are implemented in MASCEM, a multi-agent based electricity market simulator. A test case based on California Independent System Operator (CAISO) data concerning the dispatch of Regulation Down, Regulation Up, Spinning Reserve and Non-Spinning Reserve services is included in this paper.
