Este trabalho teve como objetivo avaliar a ocorrência, isolar e identificar fungos micorrízicos arbusculares associados à cultura da mandioca (Manihot esculenta). Amostras de solo rizosférico e de várias partes da planta (raízes, tubérculos, manivas e folhas) de locais nos Estados do Rio de Janeiro, São Paulo e Paraná, foram inoculadas nos meios LGI-P, NFb-malato e NFb-GOC, avaliando-se o número mais provável de células e a atividade de redução de acetileno. Bactérias diazotróficas foram isoladas de todas as partes da planta, com exceção das folhas, sendo identificadas como Klebsiella sp., Azospirillum lipoferum e uma bactéria denominada "E", provavelmente pertencente ao gênero Burkholderia. A Bactéria E acumulou de 7,63 mg a 14,84 mg de N/g de C em meio semi-sólido, isento de N, e conseguiu manter a capacidade de fixação biológica de N, mesmo após uma dezena de repicagens consecutivas. A colonização micorrízica variou de 31% a 69%, e a densidade de esporos de 10 a 384 esporos/100 mL de solo, predominando as espécies Entrophospora colombiana e Acaulospora scrobiculata no Rio de Janeiro, A. scrobiculata e Scutellospora heterogama no Paraná e em Piracicaba (São Paulo) e A. appendicula e S. pellucida em Campinas (São Paulo).

Este trabalho objetivou avaliar os efeitos da calagem e de fontes e doses de N sobre a disponibilidade de nitrato, em amostras (020 cm) de solos brasileiros. Foram conduzidos dois experimentos: um com cinco solos da Bahia e o outro com cinco solos de Minas Gerais. Os materiais de solo foram previamente umedecidos e incubados por sete dias, a 26°C, visando ativar a flora microbiana. Nas amostras de solo da Bahia, aplicou-se a uréia ou o sulfato de amônio nas concentrações de 50, 100, 150 e 200 mg de N por kg de solo e nas de Minas Gerais, foi avaliado o efeito da calagem sobre a nitrificação. Os materiais de solo foram incubados a 26°C por 15 dias. Independentemente da dose de N aplicada, auréia nitrificou mais rapidamente do que o sulfato de amônio. Na Areia Quartzosa não foi observada nitrificação líquida do Nsulfato de amônio, sendo constatada, porém, a oxidação do Nuréia. A calagem afetou a nitrificação, notando-se nos solos que receberam calcário maior disponibilidade de nitrato. A saturação por bases e o teor de matéria orgânica apresentaram, respectivamente, a melhor correlação com o nitrato formado e os teores de uréia presentes no solo ao término da incubação.

Pyogenic liver abscess is a severe condition and a therapeutic challenge. Treatment failure may be due to an unrecognized ingested foreign body that migrated from the gastrointestinal tract. There has recently been a marked increase in the number of reported cases of this condition, but initial misdiagnosis as cryptogenic liver abscess still occurs in the majority of cases. We conducted the current study to characterize this entity and provide a diagnostic strategy applicable worldwide. To this end, data were collected from our case and from a systematic review that identified 59 well-described cases. Another systematic review identified series of cryptogenic-and Asian Klebsiella-liver abscess; these data were pooled and compared with the data from the cases of migrated foreign body liver abscess. The review points out the low diagnostic accuracy of history taking, modern imaging, and even surgical exploration. A fistula found through imaging procedures or endoscopy warrants surgical exploration. Findings suggestive of foreign body migration are symptoms of gastrointestinal perforation, computed tomography demonstration of a thickened gastrointestinal wall in continuity with the abscess, and adhesions seen during surgery. Treatment failure, left lobe location, unique location (that is, only 1 abscess location within the liver), and absence of underlying conditions also point to the diagnosis, as shown by comparison with the cryptogenic liver abscess series. This study demonstrates that migrated foreign body liver abscess is a specific entity, increasingly reported. It usually is not cured when unrecognized, and diagnosis is mainly delayed. This study provides what we consider the best available evidence for timely diagnosis with worldwide applicability. Increased awareness is required to treat this underestimated condition effectively, and further studies are needed.

O objetivo deste trabalho foi avaliar a composição químico-bromatológica e a digestibilidade in vitro do bagaço de cana-de-açúcar, submetido à amonização com uréia. Os tratamentos constaram de quatro doses de uréia, adicionadas ao bagaço de cana-de-açúcar, e adição de 1,2% de soja como fonte de urease. O delineamento experimental utilizado foi o inteiramente casualizado. O bagaço foi armazenado em silos de PVC com capacidade para 5,3 L, que foram abertos depois de 110 dias de amonização. Com a adição das doses de uréia ao bagaço, houve aumento linear nos teores de proteína bruta. Os teores de fibra em detergente neutro, fibra em detergente ácido, hemicelulose, celulose, lignina e carboidratos totais diminuíram, enquanto os teores de nitrogênio insolúvel em detergente ácido, nitrogênio insolúvel em detergente neutro, carboidratos não fibrosos, nitrogênio amoniacal e pH aumentaram, significativamente, com a adição de uréia. Nas doses avaliadas, a uréia adicionada ao bagaço promove aumento no conteúdo dos compostos nitrogenados, diminuição dos componentes fibrosos e aumenta a digestibilidade in vitro da matéria seca. Estimada pela equação de regressão, a adição de 2,62% de uréia ao bagaço de cana fornece o teor mínimo de proteína bruta para o bom funcionamento do rúmen.

O objetivo deste trabalho foi caracterizar a comunidade bacteriana endofítica de plantas assintomáticas (escapes) e afetadas pela clorose variegada dos citros (CVC) por meio de isolamento em meio de cultura, técnica de gradiente desnaturante em gel de eletroforese (DGGE) e detecção de Methylobacterium mesophilicum e Xyllela fastidiosa por meio de PCR específico, para estudar esta comunidade e sua relação com a ocorrência da CVC. A análise da comunidade bacteriana via DGGE permitiu a detecção de X. fastidiosa, bem como Klebsiella sp. e Acinetobacter sp. como endófitos de citros. Foram observados também Curtobacterium sp., Pseudomonas sp., Enterobacter sp. e Bacillus spp. Utilizando primers específicos, Methylobacterium mesophilicum e X. fastidiosa também foram observadas, reforçando hipóteses de que estas bactérias podem estar interagindo no interior da planta hospedeira.

OBJECTIVES: To test the activity of tigecycline combined with 16 antimicrobials in vitro against 22 gram-positive and 55 gram-negative clinical isolates. METHODS: Antibiotic interactions were determined by chequerboard and time-kill methods. RESULTS: By chequerboard, of 891 organism-drug interactions tested, 97 (11%) were synergistic, 793 (89%) were indifferent and 1 (0.1%) was antagonistic. Among gram-positive pathogens, most synergisms occurred against Enterococcus spp. (7/11 isolates) with the tigecycline/rifampicin combination. No antagonism was detected. Among gram-negative organisms, synergism was observed mainly with trimethoprim/sulfamethoxazole against Serratia marcescens (5/5 isolates), Proteus spp. (2/5) and Stenotrophomonas maltophilia (2/5), with aztreonam against S. maltophilia (3/5), with cefepime and imipenem against Enterobacter cloacae (3/5), with ceftazidime against Morganella morganii (3/5), and with ceftriaxone against Klebsiella pneumoniae (3/5). The only case of antagonism occurred against one S. marcescens with the tigecycline/imipenem combination. Selected time-kill assays confirmed the bacteriostatic interactions observed by the chequerboard method. Moreover, they revealed a bactericidal synergism of tigecycline with piperacillin/tazobactam against one penicillin-resistant Streptococcus pneumoniae and with amikacin against Proteus vulgaris. CONCLUSIONS: Combinations of tigecycline with other antimicrobials produce primarily an indifferent response. Specific synergisms, especially against enterococci and problematic gram-negative isolates, might be worth investigating in in vitro models and/or in animal models simulating the human environment.

O objetivo deste trabalho foi avaliar os efeitos da amonização com diferentes níveis de ureia e do uso de grãos de soja como fonte de urease, na melhoria da composição químico-bromatológica do resíduo agroindustrial da carnaúba (Copernicia prunifera), conhecido como bagana (BC). Os níveis de ureia utilizados foram 0, 2,5, 5, 7,5 e 10%, em relação à percentagem de matéria seca total da BC; e os níveis de grãos de soja tostados foram de 0 e 20%. Foram também avaliados o consumo de ração e o desempenho de cordeiros confinados, submetidos a dietas com níveis crescentes de substituição de feno de capim Tifton 85 (Cynodon spp.) (FT) pela BC: 100 FT; 25% BC e 75% FT; 50% BC e 50% FT; 75% BC e 25% FT; 100% BC. Foram utilizados 30 cordeiros mestiços inteiros, recém-desmamados, com peso corporal médio de 17 kg e idade média em torno de 80 dias. Os teores de proteína bruta na forragem aumentaram com o aumento das dosagens de ureia. Foram observados, ainda, aumentos nos teores de fibra em detergente neutro e celulose e redução da digestibilidade da BC. Os consumos diários mais elevados de matéria seca, matéria orgânica, proteína bruta, extrato etéreo e fibra em detergente neutro foram observados nos animais que receberam apenas FT como volumoso. O desempenho animal foi reduzido com o aumento da BC na dieta.

The purpose of this study was to evaluate the postmortem distributions of procalcitonin (PCT), C-reactive protein (CRP), soluble triggering receptor expressed on myeloid cells-1 (sTREM-1) and soluble interleukin-2 receptor (sIL-2R) levels in postmortem serum from femoral blood, pericardial fluid and pleural fluid in a series of sepsis-related fatalities (12 subjects) and control cases (20 subjects) that underwent medico-legal investigations. Our aim was to assess the diagnostic potential of the results obtained from pericardial and pleural fluid analysis in identifying sepsis-related deaths. All sepsis-related cases had a documented, clinical diagnosis that was established in vivo during hospitalization. Pneumonia was the main infectious focus identified during autopsy and histology. Pseudomonas aeruginosa, Klebsiella pnemoniae and Escherichia coli were the most commonly identified bacteria in blood and lung tissue cultures. The preliminary results corroborate the usefulness of PCT, CRP, sTREM-1 and sIL-2R determination in postmortem serum for the identification of sepsis-related deaths. Moreover, the data suggest that, as far as PCT, CRP, sTREM-1 and sIL-2R measurements are concerned, pericardial and pleural fluids can be considered suitable alternatives to postmortem serum should femoral blood prove unavailable at autopsy.

BACKGROUND: The thiomethyl group of S-adenosylmethionine is often recycled as methionine from methylthioadenosine. The corresponding pathway has been unravelled in Bacillus subtilis. However methylthioadenosine is subjected to alternative degradative pathways depending on the organism. RESULTS: This work uses genome in silico analysis to propose methionine salvage pathways for Klebsiella pneumoniae, Leptospira interrogans, Thermoanaerobacter tengcongensis and Xylella fastidiosa. Experiments performed with mutants of B. subtilis and Pseudomonas aeruginosa substantiate the hypotheses proposed. The enzymes that catalyze the reactions are recruited from a variety of origins. The first, ubiquitous, enzyme of the pathway, MtnA (methylthioribose-1-phosphate isomerase), belongs to a family of proteins related to eukaryotic intiation factor 2B alpha. mtnB codes for a methylthioribulose-1-phosphate dehydratase. Two reactions follow, that of an enolase and that of a phosphatase. While in B. subtilis this is performed by two distinct polypeptides, in the other organisms analyzed here an enolase-phosphatase yields 1,2-dihydroxy-3-keto-5-methylthiopentene. In the presence of dioxygen an aci-reductone dioxygenase yields the immediate precursor of methionine, ketomethylthiobutyrate. Under some conditions this enzyme produces carbon monoxide in B. subtilis, suggesting a route for a new gaseous mediator in bacteria. Ketomethylthiobutyrate is finally transaminated by an aminotransferase that exists usually as a broad specificity enzyme (often able to transaminate aromatic aminoacid keto-acid precursors or histidinol-phosphate). CONCLUSION: A functional methionine salvage pathway was experimentally demonstrated, for the first time, in P. aeruginosa. Apparently, methionine salvage pathways are frequent in Bacteria (and in Eukarya), with recruitment of different polypeptides to perform the needed reactions (an ancestor of a translation initiation factor and RuBisCO, as an enolase, in some Firmicutes). Many are highly dependent on the presence of oxygen, suggesting that the ecological niche may play an important role for the existence and/or metabolic steps of the pathway, even in phylogenetically related bacteria. Further work is needed to uncover the corresponding steps when dioxygen is scarce or absent (this is important to explore the presence of the pathway in Archaea). The thermophile T. tengcongensis, that thrives in the absence of oxygen, appears to possess the pathway. It will be an interesting link to uncover the missing reactions in anaerobic environments.

Adubos verdes (AVs) são condicionadores do solo utilizados para a melhoria da estrutura e fertilidade do solo. Este estudo avaliou o efeito de Crotalaria spectabilis Roth (crotalária), Cajanus cajan (L) Millsp (guandu) e Brachiaria decumbens Stapf (braquiária) nas propriedades microbiológicas e bioquímicas do solo sob laranjal. As leguminosas foram plantadas nas entrelinhas do laranjal e, após 3 meses, cortadas e lançadas nas entrelinhas. A braquiária, já estabelecida, foi roçada e lançada nas entrelinhas. Após 5 meses, as amostras de solo foram coletadas na entrelinha e na linha, na profundidade de 0-20 cm. O delineamento experimental foi em parcela subdividida. Efeito significativo (p< 0,05%) da aplicação dos AV sobre as contagens de bactérias, as atividades da desidrogenase e nitrificante foram obtidas e aumentaram, em média, de 20, 39 e 190%, respectivamente. O número de fungos diminuiu de 57%. A atividade da urease e a solubilizadora, e os conteúdos de matéria orgânica e umidade não foram influenciados pela aplicação dos AVs. Dentre as plantas, respostas significativas foram obtidas decorrentes do estímulo da atividade da desidrogenase em 57% pelo guandu, da urease em 58% pela braquiária, solubilizadora em 346% pela crotalária e nitrificante em 236% pelo guandu ou braquiária em relação aos demais AVs. Estes resultados sugerem que a aplicação de AVs durante anos consecutivos pode estimular a ação dos microrganismos e melhorar a qualidade do solo.

This study was performed in order to evaluate the efficacy of different mouthrinses whose use is extended in Spain. Six different antiseptic mouthrinses were studied by means of determination of Minimal Inhibitory Concentration (MIC) values against Klebsiella pneumoniae, Serratia marcescens, Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, Salmonella typhimurium, Bacillus subtilis, Streptococcus mutans, Prevotella intermedia, Porphyromonas gingivalis and Actinobacillus actinomycetemcomitans. Also in vivo experiments were carried out in volunteers by the use of mouthrinses and evaluation of bacterial populations before and after the treatment. Finally, the kinetics of bacterial death was determined. Results suggested that the determination of MIC values is not a reliable method to evaluate the antibacterial effect of such products. On the other hand those rinsing solutions based on the effect of oxygen, such as those containing carbamide peroxide have a greater efficacy against anaerobic bacteria compared with rinses whose active molecule is a disinfectant. Finally, the kinetics of bacterial death demonstrates that the essential oil rinse kills bacteria much faster. All tested mouthrinses were active as antibacterial although those based on oxygen production or essential oils were more active than solutions based on chlorhexidine and Triclosan

O objetivo deste trabalho foi quantificar os teores de fenóis e flavonoides totais, bem como avaliar as atividades antioxidante e antimicrobiana de extratos obtidos dos talos e folhas de atemoia (A. cherimola Mill. x A. squamosa L.), que pertence à família Annonaceae. A atividade antioxidante foi avaliada pelos métodos de sequestro dos radicais 2,2-difenil-1-picrilhidrazil (DPPH) e 2,2'-azinobis-3-etilbenzotiazolina-6-ácido sulfônico (ABTS), bem como pelo método da cooxidação do β-caroteno/ácido linoleico. A avaliação da atividade antimicrobiana dos extratos foi analisada contra 10 cepas de bactérias. Os resultados da atividade antioxidante dos extratos mostraram que o extrato etanólico dos talos (EEt) foi o antioxidante mais efetivo (IC50 = 10,44 ± 1,25 µg/mL) no método do sequestro do DPPH, bem como no sequestro do radical ABTS (24,81 ± 0,49%). O extrato hexânico das folhas apresentou o melhor percentual de atividade antioxidante no ensaio do β-caroteno/ácido linoleico (41,12 ± 4,35%). Os extratos etanólico dos talos e metanólico das folhas mostraram-se ativos contra cepas de Bacillus cereus, Klebsiella pneumoniae, Staphylococcus aureus resistente à meticilina (MRSA), Staphylococcus aureus e Staphylococcus epidermidis.

Here, we report the culture and characterization of an alphaproteobacterium of the order Rhizobiales, isolated from the gut of the honey bee Apis mellifera. Strain PEB0122T shares >95 % 16S rRNA gene sequence similarity with species of the genus Bartonella, a group of mammalian pathogens transmitted by bloodsucking arthropods. Phylogenetic analyses showed that PEB0122T and related strains from the honey bee gut form a sister clade of the genus Bartonella. Optimal growth of strain PEB0122T was obtained on solid media supplemented with defibrinated sheep blood under microaerophilic conditions at 35-37 °C, which is consistent with the cultural characteristics of other species of the genus Bartonella. Reduced growth of strain PEB0122T also occurred under aerobic conditions. The rod-shaped cells of strain PEB0122T had a mean length of 1.2-1.8 μm and revealed hairy surface structures. Strain PEB0122T was positive for catalase, cytochrome c oxidase, urease and nitrate reductase. The fatty acid composition was comparable to those of other species of the genus Bartonella, with palmitic acid (C16 : 0) and isomers of 18- and 19-carbon chains being the most abundant. The genomic DNA G+C content of PEB0122T was determined to be about 45.5 mol%. The high 16S rRNA gene sequence similarity with species of Bartonella and its close phylogenetic position suggest that strain PEB0122T represents a novel species within the genus Bartonella, for which we propose the name Bartonella apis sp. nov. The type strain is PEB0122T ( = NCIMB 14961T = DSM 29779T).

Since the introduction of antibiotic agents, the amount and prevalence of Beta-lactam resistant enterobacteria has become an increasing problem. Many enterobacteria are opportunistic pathogens that easily acquire resistance mechanisms and genes, which make the situation menacing. These bacteria have acquired resistance and can hydrolyse extended spectrum cephalosporins and penicillins by producing enzymes called extended-spectrum Beta-lactamases (ESBLs). ESBL-producing bacteria are most commonly found in the gastro-intestinal tract of colonised patients. These resistant strains can be found in both health-care associated and community-acquired isolates. The detection and treatment of infections caused by bacteria producing ESBLs are problematic. This study investigated the genetic basis of extended-spectrum Beta-lactamases in Enterobacteriaceae, especially in Escherichia coli and Klebsiella pneumoniae isolates. A total of 994 Finnish Enterobacteriaceae strains, collected at 26 hospital laboratories, during 2000 and 2007 were analysed. For the genetic basis studies, PCR, sequencing and pyrosequencing methods were optimised. In addition, international standard methods, the agar dilution and disk diffusion methods were performed for the resistance studies, and the susceptibility of these strains was tested for antimicrobial agents that are used for treating patients. The genetic analysis showed that blaCTX-M was the most prevalent gene among the E. coli isolates, while blaSHV-12 was the most common Beta-lactamase gene in K. pneumoniae. The susceptibility testing results showed that about 60% of the strains were multidrug resistant. The prevalence of ESBL-producing isolates in Finland has been increasing since 2000. However, the situation in Finland is still much better than in many other European countries.

The data presented describe the development of an enzymatic process in vegetable oils. Six bacterial lipases were tested for their ability to hydrolyze. For each lipase assay, the p-NPP method was applied to obtain maximum enzymatic activities. The lipase from Burkholderia cepacia (lipase B-10) was the most effective in buriti oil, releasing 4840 µmol p-NP mL-1. The lipase from Klebsiella variicola (lipase B-22) was superior in passion fruit oil, releasing 4140 µmol p-NP mL-1 and also in babassu palm oil, releasing 2934 µmol p-NP mL-1. Research into the bioprocessing of oils aims to provide added value for this regional raw material.
