The reaction of 2-chloro-3-methyl-1,4-naphthoquinone (3) with the anion of ethyl cyanoacetate led to a mixture of two epimeric fused-ring cyclopropane compounds, characterised as exo- and endo-1-cyano-1 -ethoxycarbonyl-1a-methyl-1a,7a-dihydro-1H-cyclopropa[b]naphthalene-2,7-dione (8) and (9). Various hydrolysis products of these were prepared and an X-ray crystallographic analysis was carried out on one of them, 1-carbamoyl-1 -carboxy-1a-methyl-1a,7a-dihydro-1H-cyclopropa[b]-naphthalene-2,7-dione (17). The reaction of 2-methyl-1,4-naphthoquinone (1) with ethyl diazoacetate gave a fused pyrazoline derivative, 3-ethoxycarbonyl-4-hydroxy-9a-methyl-1,9a-dihydro-benz[f]indazol-9-one (22), while reaction of 2-methyl-3-nitro-1,4-naphthoquinone (5) with diazomethane led to a fused Δ2-isoxazoline N-oxide, 3a-methyl-3,3a-dihydroisoxazolo[3,4-b]naphthalene-4,9-dione 1-oxide (26).

The cycloaddition of acetylenes with azides to give the corresponding 1,4-disubstituted 1,2,3-triazoles is reported using immobilised reagents and scavengers in pre-packed glass tubes in a modular flow reactor.

By performing at) initio density functional theory (DFT) calculations and electronic transport simulations based on the OFT nonequilibrium Green`s functions method we investigate how the conformational changes of a benzene-1,4-dithiol molecule bonded to gold affect the molecular transport as the electrodes are separated from each other. In particular we consider the full evolution of the stretching process until the Junction breaking point and compare results obtained with a standard semilocal exchange and correlation functional to those computed with a self-interaction corrected method. We conclude that the inclusion of self-interaction corrections is fundamental for describing both the molecule conductance and its stability against conformational fluctuations.

alpha-diamines, such as ethylendiamine and o-phenylendiamine, add to 3,4-aryl-disubstituted 1,2,5-thiadiazole 1,1-dioxides to give dihydropyrazines or quinoxalines, respectively and sulfamide. The new compound acenaphtho [5,6-b]-2,3-dihydropyrazine was synthesized and characterized. The addition of ethylendiamine to 3,4-diphenyl-1,2,5-thiadiazoline 1,1-dioxide gives 3,4-disubstituted thiadiazoildine 1,1-dioxide, dihydropyrazines, or pyrazines, depending on the reaction condition used. The reactions were followed by cyclic voltammetry and NMR spectroscopy which, in some cases, allowed the detection of the thiadiazolidine intermediate. Copyright (c) 2008 John Wiley & Sons, Ltd.

The alpha-aminoketone 1,4-diamino-2-butanone (DAB), a putrescine analogue, is highly toxic to various microorganisms, including Trypanosoma cruzi. However, little is known about the molecular mechanisms underlying DAB`s cytotoxic properties. We report here that DAB (pK(a) 7.5 and 9.5) undergoes aerobic oxidation in phosphate buffer, pH 7.4, at 37 degrees C, catalyzed by Fe(II) and Cu(II) ions yielding NH(4)(+) ion, H(2)O(2), and 4-amino-2-oxobutanal (oxoDAB). OxoDAB, like methylglyoxal and other alpha-oxoaldehydes, is expected to cause protein aggregation and nucleobase lesions. Propagation of DAB oxidation by superoxide radical was confirmed by the inhibitory effect of added SOD (50 U ml(-1)) and stimulatory effect of xanthine/xanthine oxidase, a source of superoxide radical. EPR spin trapping studies with 5,5-dimethyl-1-pyrroline-1-oxide (DMPO) revealed an adduct attributable to DMPO-HO(center dot), and those with alpha-(4-pyridyl-1-oxide)-N-tert-butylnitrone or 3,5-dibromo-4-nitrosobenzenesulfonic acid, a six-line adduct assignable to a DAB(center dot) resonant enoyl radical adduct. Added horse spleen ferritin (HoSF) and bovine apo-transferrin underwent oxidative changes in tryptophan residues in the presence of 1.0-10 mM DAB. Iron release from HoSF was observed as well. Assays performed with fluorescein-encapsulated liposomes of cardiolipin and phosphatidylcholine (20:80) incubated with DAB resulted in extensive lipid peroxidation and consequent vesicle permeabilization. DAB (0-10 mM) administration to cultured LLC-MK2 epithelial cells caused a decline in cell viability, which was inhibited by preaddition of either catalase (4.5 mu M) or aminoguanidine (25 mM). Our findings support the hypothesis that DAB toxicity to several pathogenic microorganisms previously described may involve not only reported inhibition of polyamine metabolism but also DAB pro-oxidant activity. Nicotinic acetylcholine receptors (nAChRs) were studied in detail in the past regarding their interaction with therapeutic and drug addiction related compounds. Using fast kinetic whole-cell recording, we have now studied effects of tacrine, an agent used clinically to treat Alzheimer`s disease, on currents elicited by activation of rat alpha(3)beta(4) nAChR heterologously expressed in KX alpha(3)beta(4)R2 cells. Characterization of receptor activation by nicotine used as agonist revealed a K(d) of 23 +/- 0.2 mu M and 4.3 +/- 1.3 for the channel opening equilibrium constant, Phi(-1). Experiments were performed to investigate whether tacrine is able to activate the alpha(3)beta(4) nAChR. Tacrine did not activate whole-cell currents in KX alpha(3)beta(4)R2 cells but inhibited receptor activity at submicromolar concentration. Dose response curves obtained with increasing agonist or inhibitor concentration revealed competitive inhibition of nAChRs by tacrine, with an apparent inhibition constant, K(I), of 0.8 mu M. The increase of Phi(-1) in the presence of tacrine suggests that the drug stabilizes a nonconducting open channel form of the receptor. Binding studies with TCP and MK-801 ruled out tacrine binding to common allosteric sites of the receptor. Our study suggests a novel mechanism for action of tacrine on nAChRs besides inhibition of acetylcholine esterase.

The presence of paramagnetic species in the aqueous ring opening metathesis polymerizations of the exo,exo-7-oxabicyclo[2.2.1]hept-5-ene-2,3-dicarboxylic acid monomer with RuCl(3) and K(2)[RuCl(5)H(2)O] compounds was studied using ESR techniques. It was observed that the intensities of the Ru(III) signals in the ESR spectra decrease on the time scale of the induction period so that the ROMP can take place. The intensity of the Ru(III) signal almost disappeared 50 min after reacting with K(2)[RuCl(5)H(2)O] and after 100 mm in the case of RuCl(3). Reactions of the cis-[Ru(NH(3))(4)(H(2)O)(2)](tfms)(3) and [Ru(NH(3))(5)H(2)O](tfms)(3) complexes with the monomer and different organic compounds representing the organic functions in the monomer (furan, norbornene, but-2-ene-1,4-diol and formic, acetic, oxalic and maleic acids) were also monitored by ESR and UV/vis spectra. It was deduced that the organic acids provide the disappearance of the Ru(III) signal. The proton NMR relaxation times of the residual water in D(2)O for reactions with oxalic acid suggested that the presence of paramagnetic ions in the solution decreases along with

Syftet med denna studie är att synliggöra pedagogernas arbete med matematik i förskolans inomhusmiljö med barn i åldern 4 - 5 år. Utgångspunkt för synliggörande är Lpfö 98/10 mål och uttrycksformer i matematik. Förhoppningen är att resultatet från undersökningen kan ligga till grund för att utveckla verksamheten. För att nå syftet har följande frågeställningar använts:1. Hur arbetar pedagogerna med barns matematiklärande? Vilket matematikinnehåll? Vilka uttrycksformer?2. I vilka situationer sker pedagogers arbete med matematik? I vilka av de sex vardagssituationerna: barns lek med närvarande pedagog, samling, utgång, ingång, lunch och läsvila?3. Hur ser de verksamma pedagogerna på behov, möjligheter och hinder för att utveckla arbetet med barns matematiklärande?För att få svar på frågeställning 1 och 2 genomfördes strukturerade observationer med hjälp av observationsscheman. Sammanlagt observerades fem avdelningar och på varje avdelning observerades samtliga sex situationer, det vill säga vid 30 tillfällen. Barnen var i åldern 4-5 år och med pedagog menas i undersökningen all personal som arbetade på avdelningarna. För att få svar på den tredje och sista frågeställningen genomfördes kvalitativa intervjuer genom fokusgruppssamtal vilket innebär att samtalet utfördes avdelningsvis med samma fem avdelningar vilka ingick i observationerna.Resultatet visar att pedagoger arbetar med barns matematiklärande i större utsträckning än de själva tror och upplever. Observationerna åskådliggör att de matematiska aktiviteterna räkna och förklara förekommer oftast, följt av lokalisera, mäta och konstruera. Den matematiska aktiviteten leka förekommer sällan. Av de sex undersökta uttrycksformerna dominerar samtal, följt av rörelse, lek/drama, sång/musik, bild/form och dans. Pedagogerna har en önskan om fortbildning i matematik, så att de kan synliggöra matematiklärandet för sig själva, barnen och hela verksamheten. Av resultatet framgår att de matematiska strävansmålen i läroplanen för förskolan inte är helt tydliga för personalen, och att uttrycksformerna inte används fullt ut av pedagogerna i det dagliga arbetet.En slutsats av undersökningens resultat är att pedagogerna behöver ökad kunskap i arbetet med barns matematiklärande samt stöd för att förtydliga och omsätta de olika matematiska strävansmålen i läroplanen för förskolan i verksamheten.

Amostras de borracha natural foram reticuladas por meio de radiação gama (doses de 2, 4, 6 e 8 MRad) e intumescidas: a) em n-octano, ciclo-hexano e esqualeno; b) em polibutadieno líquido (Mn=1830 g/mol; alto teor 1,4- cis) a 45°C; C) em solução de polibutadieno de baixo peso molecular (M,= 1830 g/mol; alto teor 1,4-cis) em n-octano e ciclo-hexano; d) em solução de polibutadieno de alto peso molecular (Mn= 175.000 g/rnol; alto teor 1,4-cis) emn-octano e ciclo-hexano ; e) em solução de borracha natural (Mn=779.000 g/mol) em ciclo- hexano, a 25°C. Amostras de polibutadieno (alto teor 1,4-cis) foram reticuladas por meio de radiação gama (doses de 1,s; 18 e 25 MRad) e intumescidas: a) em n-octano, ciclo-hexano e esquaieno. Salvo quando mencionado em contrário, as amostras de borracha natural e de polibutadieno foram intumescidas nas temperaturas de 25, 35 e 45°C. A partir dos ensaios de intumescirnento acima mencionados, foram determinadas as solubilidades de borracha natural e de polibutadieno em n-octano, ciclo-hexano e esqualeno através dos parâmetros de Flory-Huggins, calculados com o emprego da equação de Flory-Rehner. Verificou-se a influência da temperatura e da concentração de polimero no gel sobre o valor do parâmetro de Flory-Huggins. Também através desse parâmetro procurou-se determinar a compatibilidade entre borracha natural e polibutadieno. Foi demonstrado que cadeias lineares de baixo peso molecular de polibutadieno conseguem penetrar em amostras reticuladas de borracha natural, quando o peso molecular das cadeias lineares for bem inferior ao do arco de rede Mc das amostras reticuladas. Cadeias lineares de polibutadieno e de borracha natural com peso molecular superior ao do arco de rede da borracha natural reticulada, entretanto, não conseguem penetrar no retículo. Quranto mais alta a concentração da solução externa em moléculas de alto peso molecular, tanto menor é o grau de intumescimento da amostra reticulada.

Neste trabalho, foi obtido o xerogel híbrido 3-(1,4-fenilenodiamina) propil/sílica, usando-se o método sol-gel de síntese, variando-se as condições experimentais de síntese. Foram usados como reagentes precursores o tetraetilortosilicato (TEOS) e o 3-[(1,4-fenilenodiamina)propil]trimetoxisilano (FDAPS) sintetizado em nosso laboratório. As condições experimentais de síntese variadas foram: a concentração de precursor orgânico (FDAPS), a temperatura de gelificação, o tipo de solvente e o pH do meio reacional. O trabalho foi dividido em duas etapas: na primeira, foram obtidas duas séries de materiais onde se variou a temperatura de gelificação (5, 25, 50 e 70 °C), além da quantidade de precursor orgânico (FDAPS), adicionado à síntese (1,5 e 5,0 mmol). Na segunda etapa variou-se o pH do meio reacional (4, 7 e 10) além do tipo de solvente (etanol, butanol e octanol), mantendo-se a quantidade de precursor orgânico adicionado e a temperatura de gelificação constantes em 5,0 mmol e 25 oC, respectivamente. Em ambas etapas utilizou-se HF como catalisador e manteve-se o sistema fechado, porém não vedado, durante a gelificação. Na caracterização dos xerogéis híbridos foram usadas as seguintes técnicas: a) termoanálise no infravermelho, para estimar a estabilidade térmica do componente orgânico além da fração de orgânicos dispersos na superfície; b) isotermas de adsorção e dessorção de nitrogênio, para determinação da área superficial específica, do volume e da distribuição de tamanho de poros; c) análise elementar para estimar a fração de componente orgânico presente no xerogel e d) microscopia eletrônica de varredura onde foi possível observar textura, compactação e presença de partículas primárias nos xerogéis. A partir dos resultados de caracterização foi possível avaliar a influência dos parâmetros experimentais de síntese nas características dos xerogéis híbridos obtidos. Xerogéis híbridos com maior teor de orgânicos foram mais influenciados pela variação da temperatura de gelificação. Um aumento na temperatura de gelificação produz xerogéis com menor porosidade, entretanto, com maior estabilidade térmica do componente orgânico. Considerando-se estabilidade térmica e porosidade, as amostras gelificadas a 25 oC apresentaram os melhores resultados. Em relação à variação de pH e solvente, as amostras gelificadas em pH ácido foram as que apresentaram maior porosidade, enquanto que a maior estabilidade térmica foi alcançada usando-se etanol como solvente.

The maturation of Madeira wines usually involves exposure to relatively high temperatures and humidity levels >70%, which affect the aroma and flavor composition and lead to the formation of the typical and characteristic bouquet of these wines. To estimate the levels of sotolon [3-hydroxy4,5-dimethyl-2(5 H )-furanone] and their behavior over time, 86 aged Madeira wines samples (1-25 years old), with different sugar concentrations, respectively, 90 g L-1 for Boal, 110 g L-1 for Malvazia, 25 g L -1 for Sercial, and 65 g L-1 for Verdelho varieties, were analyzed. Isolation was performed by liquid-liquid extraction with dichloromethane followed by chromatographic analysis by GC-MS. The reproducibility of the method was found to be 4.9%. The detection and quantification limits were 1.2 and 2.0 µgL-1, respectively. The levels of sotolon found ranged from not detected to 2000 µgL-1 for wines between 1 and 25 years old. It was observed that during aging, the concentration of sotolon increased with time in a linear fashion ( r ) 0.917). The highest concentration of sotolon was found in wines with the highest residual sugar contents, considering the same time of storage. The results show that there is a strong correlation between sotolon and sugar derivatives: furfural, 5-methylfurfural, 5-hydroxymethylfurfural, and 5-ethoxymethylfurfural. These compounds are also well correlated with wine aging. These findings indicate that the kinetics of sotolon formation is closely related with residual sugar contents, suggesting that this molecule may come from a component like sugar.

It is widely acknowledged that the indoleamine neurotransmitter serotonin (5-HT) plays a dual role in the regulation of anxiety, a role that in part depends upon neuroanatomical locus of action. Thus, whereas stimulation of 5-HT1A or 5-HT2 receptors in the limbic forebrain (amygdala, hippocampus) enhances anxiety-like responding in rodents, activation of corresponding receptor populations in the midbrain periaqueductal grey (PAG) more often than not reduce anxiety-like behaviour. The present study specifically concerns the anxiety-modulating influence of 5-HT2 receptors within the mouse PAG. Experiment 1 assessed the effects of intra-PAG infusions of the 5-HT2B/2C receptor agonist mCPP (0, 0.03, 0.1 or 0.3 nmol/0.1 mu l) on the behaviour of mice exposed to the elevated plus-maze. As mCPP acts preferentially at 5-HT2B and 5-HT2C receptors, Experiment 2 investigated its effects in animals pretreated with ketanserin, a preferential 5-HT2A/2C receptor antagonist. In both cases, test sessions were videotaped and subsequently, scored for anxiety-like behaviour (e.g., percentage of open arm entries and percentage of open arm time) as well as general locomotor activity (closed arm entries). The results of Experiment I showed that mCPP microinfusions (0.03 and 0.1 nmol) into the PAG of mice decreased behavioural indices of anxiety without significantly altering general activity measures. In Experiment 2, the anxiolytic-like profile of intra-PAG mCPP (0.03 nmol) was substantially attenuated by intra-PAG pretreatment with an intrinsically inactive dose of the preferential 5-HT2A/2C receptor antagonist, ketanserin (10 nmol/0.1 mu l). Together, these data suggest that 5HT(2C) receptor populations within the midbrain PAG play an inhibitory role in plus-maze anxiety in mice. (C) 2007 Elsevier B.V. All rights reserved.
