The hydrolysis of cell wall pectins by tomato (Lycopersicon esculentum) polygalacturonase (PG) in vitro is more extensive than the degradation affecting these polymers during ripening. We examined the hydrolysis of polygalacturonic acid and cell walls by PG isozyme 2 (PG2) under conditions widely adopted in the literature (pH 4.5 and containing Na+) and under conditions approximating the apoplastic environment of tomato fruit (pH 6.0 and K+ as the predominate cation). The pH optima for PG2 in the presence of K+ were 1.5 and 0.5 units higher for the hydrolysis of polygalacturonic acid and cell walls, respectively, compared with activity in the presence of Na+. Increasing K+ concentration stimulated pectin solubilization at pH 4.5 but had little influence at pH 6.0. Pectin depolymerization by PG2 was extensive at pH values from 4.0 to 5.0 and was further enhanced at high K+ levels. Oligomers were abundant products in in vitro reactions at pH 4.0 to 5.0, decreased sharply at pH 5.5, and were negligible at pH 6.0. EDTA stimulated PG-mediated pectin solubilization at pH 6.0 but did not promote oligomer production. Ca2+ suppressed PG-mediated pectin release at pH 4.5 yet had minimal influence on the proportional recovery of oligomers. Extensive pectin breakdown in processed tomato might be explained in part by cation- and low-pH-induced stimulation of PG and other wall-associated enzymes.

The voltage-dependent K+ channel responsible for the slowly activating delayed K+ current IKs is composed of pore-forming KCNQ1 and regulatory KCNE1 subunits, which are mutated in familial forms of cardiac long QT syndrome. Because KCNQ1 and KCNE1 genes also are expressed in epithelial tissues, such as the kidneys and the intestine, we have investigated the adaptation of KCNE1-deficient mice to different K+ and Na+ intakes. On a normal K+ diet, homozygous kcne1−/− mice exhibit signs of chronic volume depletion associated with fecal Na+ and K+ wasting and have lower plasma K+ concentration and higher levels of aldosterone than wild-type mice. Although plasma aldosterone can be suppressed by low K+ diets or stimulated by low Na+ diets, a high K+ diet provokes a tremendous increase of plasma aldosterone levels in kcne1−/− mice as compared with wild-type mice (7.1-fold vs. 1.8-fold) despite lower plasma K+ in kcne1−/− mice. This exacerbated aldosterone production in kcne1−/− mice is accompanied by an abnormally high plasma renin concentration, which could partly explain the hyperaldosteronism. In addition, we found that KCNE1 and KCNQ1 mRNAs are expressed in the zona glomerulosa of adrenal glands where IKs may directly participate in the control of aldosterone production by plasma K+. These results, which show that KCNE1 and IKs are involved in K+ homeostasis, might have important implications for patients with IKs-related long QT syndrome, because hypokalemia is a well known risk factor for the occurrence of torsades de pointes ventricular arrhythmia.

Adenoviral vectors can direct high-level expression of a transgene, but, due to a host immune response to adenoviral antigens, expression is of limited duration, and repetitive administration has generally been unsuccessful. Exposure to foreign proteins beginning in the neonatal period may alter or ablate the immune response. We injected adult and neonatal (immunocompetent) CD-1 mice intravenously with an adenoviral vector expressing human blood coagulation factor IX. In both groups of mice, expression of human factor IX persisted for 12-16 weeks. However, in mice initially injected as adults, repeat administration of the vector resulted in no detectable expression of the transgene, whereas in mice initially injected in the neonatal period, repeat administration resulted in high-level expression of human factor IX. We show that animals that fail to express the transgene on repeat administration have developed high-titer neutralizing antibodies to adenovirus, whereas those that do express factor IX have not. This experimental model suggests that newborn mice can be tolerized to adenoviral vectors and demonstrates that at least one repeat injection of the adenoviral vector is possible; the model will be useful in elucidating the immunologic mechanisms underlying successful repeat administration of adenoviral vectors.

Amperometry has been used for real-time electrochemical detection of the quantal release of catecholamines and indolamines from secretory granules in chromaffin and mast cells. Using improved-sensitivity carbon fiber electrodes, we now report the detection of quantal catecholamine release at the surface of somas of neonatal superior cervical ganglion neurons that are studded with axon varicosities containing synaptic vesicles. Local application of a bath solution containing high K+ or black widow spider venom, each of which greatly enhances spontaneous quantal release of transmitter at synapses, evoked barrages of small-amplitude (2-20 pA), short-duration (0.5-2 ms) amperometric quantal "spikes". The median spike charge was calculated as 11.3 fC. This figure corresponds to 3.5 x 10(4) catecholamine molecules per quantum of release, or approximately 1% that evoked by the discharge of the contents of a chromaffin granule.

Modal analysis of middle Miocene to Pleistocene volcaniclastic sands and sandstones recovered from Sites 1108, 1109, 1118, 1112, 1115, 1116, and 1114 within the Woodlark Basin during Leg 180 of the Ocean Drilling Program indicates a complex source history for sand-sized detritus deposited within the basin. Volcaniclastic detritus (i.e., feldspar, ferromagnesian minerals, and volcanic rock fragments) varies substantially throughout the Woodlark Basin. Miocene sandstones of the inferred Trobriand forearc succession contain mafic and subordinate silicic volcanic grains, probably derived from the contemporary Trobriand arc. During the late Miocene, the Trobriand outerarc/forearc (including Paleogene ophiolitic rocks) was subaerially exposed and eroded, yielding sandstones of dominantly mafic composition. Rift-related extension during the late Miocene-late Pliocene led to a transition from terrestrial to neritic and finally bathyal deposition. The sandstones deposited during this period are composed dominantly of silicic volcanic detritus, probably derived from the Amphlett Islands and surrounding areas where volcanic rocks of Pliocene-Pleistocene age occur. During this time terrigenous and metamorphic detritus derived from the Papua New Guinea mainland reached the single turbiditic Woodlark rift basin (or several subbasins) as fine-grained sediments. At Sites 1108, 1109, 1118, 1116, and 1114, serpentinite and metamorphic grains (schist and gneiss) appear as detritus in sandstones younger than ~3 Ma. This is thought to reflect a major pulse of rifting that resulted in the deepening of the Woodlark rift basin and the prevention of terrigenous and metamorphic detritus from reaching the northern rift margin (Site 1115). The Paleogene Papuan ophiolite belt and the Owen Stanley metamorphics were unroofed as the southern margin of the rift was exhumed (e.g., Moresby Seamount) and, in places, subaerially exposed (e.g., D'Entrecasteaux Islands and onshore Cape Vogel Basin), resulting in new and more proximal sources of metamorphic, igneous, and ophiolitic detritus. Continued emergence of the Moresby Seamount during the late Pliocene-early Pleistocene bounded by a major inclined fault scarp yielded talus deposits of similar composition to the above sandstones. Upper Pliocene-Pleistocene sandstones were deposited at bathyal depths by turbidity currents and as subordinate air-fall ash. Silicic glassy (high-K calc-alkaline) volcanic fragments, probably derived from volcanic centers located in Dawson and Moresby Straits, dominated these sandstones.

The potassium (K) nutrition and high K requirement of tropical root crops may be affected by their sodium (Na) status, as has been observed in a number of plant species. Solution culture was used to study the effects of K and Na supplies in tannia [Xanthosoma sagittifolium (L.) Schott.], sweetpotato [Ipomoea batatas (L.) Lam.] and taro [Colocasia esculenta (L.) Schott]. At low K supply, Na ameliorated symptoms of K deficiency and increased growth in tannia, and to a lesser extent in sweetpotato, but not in taro. None of the species responded to Na at adequate K supply. Differences in response to Na were attributed to differences in Na translocation to plant tops. At maximum Na supply, the Na concentration in index leaves averaged 1.82% in tannia, 0.205% in sweetpotato, and 0.0067% in taro. An increase in the supply of Na resulted in a shift in the critical K concentration for deficiency (i.e., 90% of maximum yield) in index leaves from 2.9% to 1.2% in tannia, and from 4.8% to 2.5% in sweetpotato. The critical K concentration in taro was 3.3%, irrespective of Na supply. To overcome the problem in tannia and sweetpotato of determining the critical concentration relevant to a leaf sample of unknown K status, a relationship was established for each species relating the critical K concentration to the concentration of Na in the index leaves.

Diets with more than 30 g K/kg DM have previously been associated with hypomagnesaemia in grazing cattle, and to test whether such diets lead to mineral disorders in sheep, the absorption of Mg and other elements was investigated using experimental diets to which KC I had been added to provide 27, 29, 32 or 34 g K/kg DM. The apparent absorption, balance and apparent retention of Mg, and to a lesser extent Ca, were reduced for sheep offered the diets with 32 or 34 g K/kg DM. The absorption and retention of K, Na, P, Zn, Pb and Cd was not affected by treatment. The blood intracellular Ca concentration was reduced by the diets with 29, 32 or 34 g K/kg DM, compared to the diet with 27 g K/kg DM, but the concentration of other elements was unaffected. Blood plasma Ca concentration was increased at the highest level of K inclusion, providing evidence of mild hyperkalaemia and the involvement of Ca homeostatic mechanisms. It is concluded that Mg absorption by sheep will be impaired if the diet contains more than 30 g K/kg DM, equivalent to an intake of approximately 13 g K/d, but that a high K diet may be beneficial before parturition to accustom the sheep to Ca mobilization before lactation. Ar-40/Ar-39 incremental heating ages for twenty one grains of cryptomelane, collected at 0, 42, 45, and 60 in depths in the Cachoeira Mine weathering profile, Minas Gerais, permit calculating long-term (10 Ma time scale) weathering rate (saprolitization rate) in SE Brazil. Pure well-crystallized cryptomelane grains with high K contents (3-5 wt.%) yield reliable geochronological results. The Ar-40/Ar-39 plateau ages obtained decrease from the top to the bottom of the profile (12.7 +/- 0.1 to 7.6 +/- 0.1 Ma at surface; 7.6 +/- 0.2 to 6.1 +/- 0.2 Ma at 42 m; and 7.1 +/- 0.2 to 5.9 +/- 0.1 Ma at 45 in; 6.6 +/- 0.1 to 5.2 +/- 0.1 Ma at 60 in), yielding a weathering front propagation rate of 8.9 +/- 1.1 m/m.y. From the geochronological results and the mineral transformations implicit by the current mineralogy in the weathering profiles, it is possible to calculate the saprolitization rate for the Cachoeira Mine lithologies and for adjacent weathering profiles developed on granodiorites and scbists. The measured weathering front propagation rate yields a saprolitization rate of 24.9 +/- 3.1 t/km(2)/yr. This average long-term (> 10 Ma) saprolitization rate is consistent with mass balance calculations results for present saprolitization rates in weathering watersheds. These results are also consistent with longterm saprolitization rates estimated by combining cosmogenic isotope denudation rates with mass balance calculations. This paper examines the effects of non-metallic particles on fatigue performance and, in particular, their influence on fatigue crack propagation at high ΔK (Kmax) levels. The nature and properties of a number of common non-metallic particles found in Fe- and Al- based alloys are described, and consideration is given to the consequences of mismatch of physical and chemical properties between particle and matrix. Effects of particles on fatigue in conventional alloys are illustrated and compared with the behaviour of Al/SiCp MMC. The problems associated with developing particulate reinforced MMC with adequate fatigue crack growth resistance and toughness for structural applications are discussed. © 1991.

Surface modification by means of nanostructures is of interest to enhance boiling heat transfer in various applications including the organic Rankine cycle (ORC). With the goal of obtaining rough and dense aluminum oxide (Al2O3) nanofilms, the optimal combination of process parameters for electrophoretic deposition (EPD) based on the uniform design (UD) method is explored in this paper. The detailed procedures for the EPD process and UD method are presented. Four main influencing conditions controlling the EPD process were identified as nanofluid concentration, deposition time, applied voltage and suspension pH. A series of tests were carried out based on the UD experimental design. A regression model and statistical analysis were applied to the results. Sensitivity analyses of the effect of the four main parameters on the roughness and deposited mass of Al2O3 films were also carried out. The results showed that Al2O3 nanofilms were deposited compactly and uniformly on the substrate. Within the range of the experiments, the preferred combination of process parameters was determined to be nanofluid concentration of 2 wt.%, deposition time of 15 min, applied voltage of 23 V and suspension pH of 3, yielding roughness and deposited mass of 520.9 nm and 161.6 × 10− 4 g/cm2, respectively. A verification experiment was carried out at these conditions and gave values of roughness and deposited mass within 8% error of the expected ones as determined from the UD approach. It is concluded that uniform design is useful for the optimization of electrophoretic deposition requiring only 7 tests compared to 49 using the orthogonal design method.

The studied area is situated in the northeastern extremity of the Rio Grande do Norte State, between the municipalities of Taipu and Poço Branco, and is geologically inserted into the São José do Campestre Crystalline Terrain within the Borborema Province, where the analysis of field relationships, petrographic and geochemical data allowed the distinction of three plutons named: Gameleira, Taipu and Pitombeira. The Gamaleira Pluton is composed of granodioritic rocks characterized by zoned plagioclase phenocrysts, with amphibole and biotite as the main mafic phases. Geochemically, these are metaluminous rocks of calc-alkaline nature and magnesian character. The Pitombeira Pluton encompasses two facies: (a) a coarse-grained to porphyritic monzo- to syenogranitic facies marked by K-feldspar phenocrysts; and (b) a quartz dioritic to tonalitic facies with partially zoned plagioclase laths showing chilled rims. Geochemically, rocks of the monzo- to syenogranitic facies are transitional between metaluminous and peraluminous, display a subalkaline nature (high K calc-alkaline) and a ferroan character, whereas rocks of the quartz dioritic to tonalitic facies are metaluminous, with shoshonitic affinity and ferroan character. Lastly, the Taipu Pluton is made of monzoto syenogranitic rocks with biotite as the chief mafic mineral. They are peraluminous rocks of subalkaline nature (high-K calc-alkaline) and ferroan character. Regarding the rare-earth elements (REE), it is possible to conclude that the three studied plutons display negative Eu anomalies and a relative enrichment of LREE over HREE, with LaN/YbN ratios between 9.39 to 16.20 (Gameleira Pluton), 17.99 to 31.39 (granitic facies of the Pitombeira Pluton), 14.15 to 21.81 (dioritic facies of the Pitombeira Pluton) and 15.17 to 175.41 (Taipu Pluton). Based on the combined investigation of geochemical data and discrimination tectonic diagrams, a late- to post-collisional tectonic environment is suggested for the plutons here studied

This paper discusses the correlation of thermal conductivity, density and magnetic susceptibility with composition of major and trace elements of Neoproterozoic igneous bodies from Borborema Province, Northeastern Brazil. These properties were used as potential markers among the studied magmatic suites. For the correlation between petrophysical and geochemical properties it was considered a set of 195 chemical analyzes of granitoid rocks, separated by the degree of acidity in basic, intermediate and acidic. Major (SiO2, Al2O3, Fe2O3, MgO, CaO, Na2O, K2O and TiO2) and some trace elements (Rb, Sr, Ba, Zr, Th and U) that are usually linked to the formation of the most common minerals of igneous rocks were used. The results show that SiO2 has the best positive correlation with the thermal conductivity, while Al2O3, CaO, Fe2O3, MgO and TiO2 exhibit negative correlation for the same property. The correlation with density is opposite to that one for these oxides with the thermal conductivity. The magnetic susceptibility did not correlate with the elements studied. The results for thermal conductivity and density indicate a tendency of SiO2 and oxides with higher affinity with mafic minerals (Al2O3, CaO, Fe2O3, TiO2 and MgO) in controlling these petrophysical parameters. The set of samples was divided into five different magmatic suites based on their lithogeochemical aspects into: i) peralkaline / alkaline; ii) alkaline; iii) calc-alkaline; iv) high potassium calcium alkaline; and v) shoshonitic. Data analysis showed that the thermal conductivity and density presented good results in the individualization of these suites, notably between peralkaline / alkaline, alkaline suites, calc-alkaline and shoshonitic. However, the high-K calc-alkaline suite overlapped with the other. In contrast, the magnetic susceptibility did not show effective results for separating the five chemical suites.

O objetivo deste estudo foi comparar a fusibilidade de ligas de Co-Cr-Mo-W (Remanium 2000), Ni-Cr (Durabond) e Co-Cr-Mo (Vera-PDI), incluídas em revestimentos à base de fosfato, sílica ou utilizando uma técnica mista. Uma rede de nylon quadrada (10 X 10 mm) com 100 espaços abertos serviu de modelo para construção de padrões de cera, que foram incluídos com revestimento à base de sílica, revestimento fosfatado e técnica mista (camada de revestimento fosfatado com 2 mm de espessura + revestimento à base de sílica). Quarenta e cinco espécimes (5 para cada condição experimental) foram fundidos sob chama de gás-oxigênio e a seguir jateados com óxido de alumínio. O número de segmentos fundidos completos foi contado para obter uma percentagem designada como "valor de fusibilidade", representando a precisão da liga em reproduzir os detalhes do molde. A análise estatística por meio de ANOVA a dois critérios e teste Tukey mostrou que, comparando-se as ligas, a Remanium 2000 teve fusibilidade estaticamente semelhante (p>0,05) à da Vera PDI e inferior à da liga Durabond (p<0,05). Considerando os resultados da técnica mista, a liga Remanium 2000 teve menor valor de fusibilidade (p<0,05) que as ligas Durabond e Vera PDI, que apresentaram valores estatisticamente semelhantes entre si (p>0,05). Concluindo, a fusibilidade da liga de Co-Cr-Mo-W (Remanium 2000) foi comparável à da liga de Co-Cr (Vera PDI) e inferior à da liga de Ni-Cr alloy (Durabond). À exceção da liga Remanium 2000, a técnica de inclusão mista aumentou consideravelmente a capacidade das ligas testadas de reproduzir os detalhes do molde, quando comparada à técnica de inclusão em revestimento fosfatado. A técnica de inclusão mista representa uma alternativa para melhorar a fusibilidade de ligas de metais básicos sem afetar a qualidade superficial das peças metálicas.
