The use of stable isotopes in Brazil is being improved, mainly through research conducted at main universities in the country. Some applications in health allow to studying, for example, processes involving synthesis and protein degradation, energy expenditure, body composition, kinetics of vitamins, mineral absorption and diagnose diseases related to Helicobacter pylori. The big motivation is to encourage the growth of investments in health in the few centers that have mass spectrometers in Brazil, as the technique is harmless to humans, in other words, no has problems to use it like when you use radioactive isotopes

Baccharis trimera (Less.) DC. Asteraceae e Casearia sylvestris Swartz Salicaceae são plantas nativas do Brasil e de uso tradicional em doenças do trato digestório. Seus extratos e substâncias purificadas apresentaram diversas atividades farmacológicas (ex.antiulcerogênica, antimicrobiana) e baixa toxicidade. O Helicobacter pylori (bacilo gram-negativo) coloniza o estômago humano, associada ao maior risco de incidência de várias doenças (úlceras pépticas, linfomas e adenocarcinomas gástricos). Com o objetivo de avaliar a atividade antibacteriana de diferentes extratos, frações e substâncias de ambas as plantas medicinais, extratos de partes aéreas de B. trimera e de folhas de C. sylvestris foram preparados utilizando-se técnicas de maceração, obtendo-se o extrato acetato de etila e etanólico de C. sylvestris e acetato de etila, etanólico, etanol 70% e hexânico de B. trimera. Estes foram fracionados e analisados por técnicas cromatográficas. Os extratos e suas frações e substâncias foram submetidos a ensaios para avaliação da atividade antibacteriana para a determinação da concentração inibitória mínima (CIM). O método utilizado para os ensaios foi a diluição em microplacas usando as bactérias Helicobacter pylori ATCC 43504, Staphylococcus aureus ATCC 25923 e Escherichia coli ATCC 25922. A partir do fracionamento do extrato acetato de etila de B. trimera e do extrato etanólico de C. sylvestris foram obtidas oito frações distintas do EAcBt e a fração de diterpenos do EEtCs. A análise em CCD demonstrou a presença de casearina X, B e caseargrewiina F na fração de diterpenos (EFSCs2). Os extratos da B. trimera EAcBt, EEtBt e EEt70Bt e da C. sylvestris EAcCs e EEtCs apresentaram atividade anti-H. pylori, com valores de CIM de 250, 500, 500, 125 e 750 g/mL, respectivamente... This report describes a protocol for managing the accidental swallowing of dental instruments in implant dentistry, illustrated by a patient who accidentally swallowed a hexagonal wrench. The first step was to refer the patient to the medical emergency hospital service for radiographic and clinical evaluation. The hexagonal wrench was located in the stomach and was immediately removed with an endoscopic procedure. The gastric mucosa was sampled via biopsy and the sample submitted to the urease test, which was positive for Helicobacter pylori. Triple treatment was instituted for gastritis caused by H pylori to avoid exposing the patient to unnecessary risk. Removal of a foreign body by means of an endoscopic procedure constitutes a safe and effective treatment.

Introduction: much studies regarding risk factors associated with oral cancer. Factors such as smoking and alcohol and solar radiation are well established, but others, such as bacterial influence in the development or progression of a tumor still remain unexplained. Interest in the possible relationship between bacteria and different stages of cancer development has increased since the classification of H. pylori by the WHO as a definite carcinogen. Subsequently, links between infection and the onset of cancer in various sites in the body were discovered. Review of literature: this literature review attempts to show the influence of poor oral hygiene, with consequent bacterial accumulation as a possible important risk factor for the initiation and development of mouth cancer, and correlate the possible mechanisms by which bacteria can initiate or promote carcinogenesis. Conclusion: there is much evidence that bacteria in the oral cavity and periodontopathic are present in tumor tissue, however, still can not affirm that these bacteria initiate or promote carcinogenesis.

BACKGROUND: Toll-like receptors (TLR) are membrane proteins that recognize conserved molecules derived from bacterial, viral, fungal or host tissues. They are responsible for promoting the production of cytokines and chemokines, increasing the expression of costimulatory molecules and influencing the T Helper response (Th) toward either a Th1 or Th2 profile, thereby modulating the regulatory T cell response and controlling the integrity of the epithelial barrier. The key factors responsible for increased susceptibility to recurrent aphthous ulceration (RAU) are unclear, and because TLRs are involved in both immune regulation and control of the epithelial barrier, a deficiency in TLR activity is likely to cause increased susceptibility. METHODS: We investigated the gene expression of TLRs one through 10 in tissue samples and peripheral blood mononuclear cells (PBMC) of RAU patients in comparison to healthy controls using real-time quantitative reverse transcription PCR. RESULTS: The analysis of mRNA expression levels in oral lesion showed significant (P < 0.01) overexpression of the TLR2(similar to 6-fold) gene and decreased expression of the TLR3 (similar to 5-fold) and TLR5 (similar to 6-fold) genes in comparison with healthy oral mucosa. The analysis of mRNA expression in PBMC indicated a down-regulation of TLR5 gene expression in the cells from RAU patients (P < 0.05; similar to 2-fold). CONCLUSION: Our results support the hypothesis that a subset of RAU patients has fewer TLR expression that have been tentatively implicated in antiinflammatory effects. This derangement of TLR gene expression may cause an overlay exuberant inflammation reaction in situations where normal individuals are resistant. J Oral Pathol Med (2012) 41: 8085

Gastric cancer is the second leading cause of cancer-related death worldwide. The identification of new cancer biomarkers is necessary to reduce the mortality rates through the development of new screening assays and early diagnosis, as well as new target therapies. In this study, we performed a proteomic analysis of noncardia gastric neoplasias of individuals from Northern Brazil. The proteins were analyzed by two-dimensional electrophoresis and mass spectrometry. For the identification of differentially expressed proteins, we used statistical tests with bootstrapping resampling to control the type I error in the multiple comparison analyses. We identified 111 proteins involved in gastric carcinogenesis. The computational analysis revealed several proteins involved in the energy production processes and reinforced the Warburg effect in gastric cancer. ENO1 and HSPB1 expression were further evaluated. ENO1 was selected due to its role in aerobic glycolysis that may contribute to the Warburg effect. Although we observed two up-regulated spots of ENO1 in the proteomic analysis, the mean expression of ENO1 was reduced in gastric tumors by western blot. However, mean ENO1 expression seems to increase in more invasive tumors. This lack of correlation between proteomic and western blot analyses may be due to the presence of other ENO1 spots that present a slightly reduced expression, but with a high impact in the mean protein expression. In neoplasias, HSPB1 is induced by cellular stress to protect cells against apoptosis. In the present study, HSPB1 presented an elevated protein and mRNA expression in a subset of gastric cancer samples. However, no association was observed between HSPB1 expression and clinicopathological characteristics. Here, we identified several possible biomarkers of gastric cancer in individuals from Northern Brazil. These biomarkers may be useful for the assessment of prognosis and stratification for therapy if validated in larger clinical study sets.

Metronidazole is a BCS (Biopharmaceutics Classification System) class 1 drug, traditionally considered the choice drug in the infections treatment caused by protozoa and anaerobic microorganisms. This study aimed to evaluate bioequivalence between 2 different marketed 250 mg metronidazole immediate release tablets. A randomized, open-label, 2 x 2 crossover study was performed in healthy Brazilian volunteers under fasting conditions with a 7-day washout period. The formulations were administered as single oral dose and blood was sampled over 48 h. Metronidazole plasma concentrations were determined by a liquid chromatography mass spectrometry (LC-MS/MS) method. The plasma concentration vs. time profile was generated for each volunteer and the pharmacokinetic parameters C-max, T-max, AUC(0-t), AUC(0-infinity), k(e), and t(1/2) were calculated using a noncompartmental model. Bioequivalence between pharmaceutical formulations was determined by calculating 90% CIs (Confidence Intervall) for the ratios of C-max, AUC(0-t), and AUC(0-infinity) values for test and reference using log-transformed data. 22 healthy volunteers (11 men, 11 women; mean (SD) age, 28 (6.5) years [range, 21-45 years]; mean (SD) weight, 66 (9.3) kg [range, 51-81 kg]; mean (SD) height, 169 (6.5) cm [range, 156-186 cm]) were enrolled in and completed the study. The 90% CIs for C-max (0.92-1.06), AUC(0-t) (0.97-1.02), and AUC(0-infinity) (0.97-1.03) values for the test and reference products fitted in the interval of 0.80-1.25 proposed by most regulatory agencies, including the Brazilian agency ANVISA. No clinically significant adverse effects were reported. After pharmacokinetics analysis, it concluded that test 250 mg metronidazole formulation is bioequivalent to the reference product according to the Brazilian agency requirements.

In functional dairy products, polyunsaturated fatty acids such as, conjugated linoleic acid (CLA) and alpha-linolenic acid (ALA) have been highlighted for their benefits related to prevention of some chronic diseases. In order to study the effect of type of milk (conventional vs. organic, characterized by a specific fatty acid composition), Bifidobacterium animalis subsp. lactis (BB12, B94, BL04 and HN019) counts, acidification activity and chemical composition (pH, lactose, lactic acid contents and fatty acids profile) were investigated before fermentation and after 24 h of products stored at 4 degrees C. Organic and conventional milk influenced acidification performance and bacteria counts, which was strain-dependent. Higher counts of BB12 were observed in organic milk, whereas superior counts of BL04 were found in conventional milk. Organic fermented milk showed lower levels in saturated fatty acids (FA) and higher in monounsaturated FA contents. Similarly, among bioactive FA, organic fermented milks have higher amounts of trans vaccenic acid (TVA-C18:1t), conjugated linoleic acid (CLA) and slightly higher contents of alpha-linoleic acid (ALA).
