The paper describes the rapid and label-free detection of the white spot syndrome virus (WSSV) using a surface plasmon resonance (SPR) device based on gold films prepared by electroless plating. The plating condition for obtaining films suitable for SPR measurements was optimized. Gold nanoparticles adsorbed on glass slides were characterized by transmission electron microscopy (TEM). Detection of the WSSV was performed through the binding between WSSV in solution and the anti-WSSV single chain variable fragment (scFv antibody) preimmobilized onto the sensor surface. Morphologies of the as-prepared gold films, gold films modified with self-assembled alkanethiol monolayers, and films covered with antibody were examined using an atomic force microscope (AFM). To demonstrate the viability of the method for real sample analysis, WSSV of different concentrations present in a shrimp hemolymph matrix was determined upon optimizing the surface density of the antibody molecules. The SPR device based on the electroless-plated gold films is capable of detecting concentration of WSSV as low as 2.5 ng/mL in 2% shrimp hemolymph, which is one to two orders of magnitude lower than the level measurable by enzyme-linked immunosorbant assays. Plasmon resonance spectra of supported Ag nanoparticles are studied by depositing the particles on different substrates. It was found that the dielectric properties of the substrates have significant effects on the spectral line shape, except the resonance frequency. Beyond the plasmon resonance band, the spectral shape is mainly governed by the dielectric function, particularly its imaginary part, of the substrate. The plasmon resonance band, on the other hand, may be severely distorted if the substrate is absorbing strongly.

Gold nanowires with diameters (d) between,15 run and 200 urn and with length/diameter ratio of 700 were prepared in ion-track templates with electrode position method. The morphology and crystal structure of the gold nanowires were Studied by scanning electron microscopy (SEM) and X-ray diffraction (XRD). The 200 nm (d) gold nanowires preferred orientation along the [100] direction were formed at the deposition voltage of 1.5 V (Without reference electrode). The optical properties of gold nanowire arrays embedded in ion-track templates were studied by UV-Vis spectrophotometer. There was a strong absorption peak at 539 nm for 45 nm (d) gold nanowire arrays. With the diameter of gold nanowires increasing, the absorption peak shifted to the longer wavelength. At last, the result was discussed combined with surface plasmon resonance of gold nanoparticles.

We present a newly designed colormetric sensor sensitive to pH value based on a gold nanocomposite composed of gold nanoparticles and pH-sensitive polymer{dodecylthioether end functionalized poly[2-(diethlamino) ethyl methacrylate], poly(DEAEMA)-DDT}. We have shown that this design can produce stable GNP precipitate under weakly basic condition(pH=7.5) and this precipitate can be dispersed in acidic solution(pH=4.0), due to the 2-(diethylamino) ethyl methacrylate protonated by H+.

A novel hydrogen peroxide biosensor was fabricated that is based on horseradish peroxidase-Au nanoparticles immobilized on a viologen-modified glassy carbon electrode (GCE) by amino cation radical oxidation in basic solution. The immobilized BAPV acts as a mediator and a covalent linker between GCE and the Au nanoparticles. The biosensor exhibited fast response, good reproducibility, and long-term stability.

A simple, rapid and ultrasensitive colorimetric detection of protein using aptamer-Au nanoparticles (AuNPs) conjugates based on a dot-blot array has been developed, which was combined with the unique optical properties of AuNPs, enabling the visual detection of protein within minutes without any instrument.

The stability of the complex of cationic lipid with nucleic acid, especially when facing serum, is crucial for the efficiency of gene delivery. Here, we demonstrated that the stability of the complex of didodecyldimethylammonium bromide (DDAB, a cationic lipid) with DNA in the presence of serum dramatically increased after coating DDAB onto the surface of the gold nanoparticles. The stability of the complex was demonstrated with dye intercalation assay, and agarose gel electrophoresis.

Electrodeposition of novel Au/Pd bimetallic nanostructures with dendrimer films as matrices has been reported. The dendrimers exhibited highly open structures arising from protonation of amines and this made them have good penetrability for solvent molecules. The unique properties of dendrimers obviously affected the morphologies and compositions of deposited bimetallic nanostructures compared with those from unmodified surfaces. Field-emitted scanning electron microscopy, energy dispersive spectroscopy, X-ray photoelectron spectroscopy and UV-vis spectroscopy were used to characterize these nanostructures.

An electrochemiluminescent glucose biosensor was proposed based on gold nanoparticle-catalyzed luminol electrochemiluminescence (ECL). Gold nanoparticles were self-assembled onto silica sol-gel network, and then glucose oxidase was adsorbed on the surface of gold nanoparticles. The surface assembly process and the electrochemistry and ECL behaviors of the biosensor were investigated. The assembled gold nanoparticles could efficiently electrocatalyze luminol ECL ECL intensity of the biosensor depended on scan rate, luminol concentration, and size of gold nanoparticles.

In order to develop a novel high-throughput tool for monitoring carbohydrate-protein interactions, we prepared carbohydrate or glycoprotein microarrays by immobilizing amino modified carbohydrates on aldehyde-derivatized glass slides or glycoprotein on epoxide-derivatized glass slides and carried out lectin binding experiments by using these microarrays, respectively. The interaction events are marked by attachment of gold nanoparticles followed by silver deposition for signal enhancement. The attachment of the gold nanoparticles is achieved by standard avidin-biotin chemistry.

microarray approach based on surface-enhanced Raman spectroscopic (SERS) was developed for detection of spotted peptide, peptide-protein or protein-antibody interaction. The procedure involves the attachment of peptide-capped gold nanoparticles followed by silver deposition for signal enhancement. The attachment of the gold nanoparticles is achieved by standard avidin-biotin chemistry. The well-known biomolecular recognition pairs, IgG/protein A and biotin/avidin, were used to demonstrate proof-of-concept of the SERS assay.

In this paper, we report a novel approach using peptide CALNN and its derivative CALNNGGRRRRRRRR (CALNNR(8)) to functionalize gold nanoparticles for intracellular component targeting. The translocation is effected by the nanoparticle diameter and CALNNR8 surface coverage. The intracellular distributions of the complexes are change from the cellular nucleus to the endoplasmic reticulum by increasing the density of CALNNR8 at a constant nanoparticle diameter. Additionally, increasing the nanoparticle diameter at a constant density of CALNNR8 leads to less cellular internalization.

In the present work, a sensitive spectroscopic assay based on surface-enhanced Raman spectroscopy (SERS) using gold nanoparticles as substrates was developed for the rapid detection protein-protein interactions. Detection is achieved by specific binding biotin-modification antibodies with protein-stabilized 30 nm gold nanoparticles, followed by the attachment of avidin-modification Raman-active dyes. As a proof-of-principle experiment, a well-known biomolecular recognition system, IgG with protein A, was chosen to establish this new spectroscopic assay. Highly selective recognition of IgG down to 1 ng/ml in solution has been demonstrated.

Multiwalled carbon nanotube (MWCNT)/ionic liquid/gold nanoparticle hybrid materials have been prepared by a chemical route that involves functionalization of MWCNT with amine-terminated ionic liquids followed by deposition of Au. Transmission electron microscopy revealed well-distributed Au with a narrow size distribution centered around 3.3 nm. The identity of the hybrid material was confirmed through Raman and X-ray photoelectron spectroscopy.

Horseradish peroxidase (HRP) was incorporated into multiwalled carbon nanotube/thionine/Au (MTAu) composite film by electrostatic interactions between positively charged HRP and negatively charged MTAu composite. The results of electrochemical impedance spectroscopy (EIS) confirmed adsorption of HRP on the surface of MTAu modified GC electrode.
