Peats are an important reserve of humified carbon in terrestrial ecosystems. The interest in the use of humic substances as plant growth promoters is continuously increasing. The objective of this study was to evaluate the bioactivity of alkaline soluble humic substances (HS), humic (HA) and fulvic acids (FA) isolated from peats with different decomposition stages of organic matter (sapric, fibric and hemic) in the Serra do Espinhaço Meridional, state of Minas Gerais. Dose-response curves were established for the number of lateral roots growing from the main plant axis of tomato seedlings. The bioactivity of HA was greatest (highest response in lateral roots at lowest concentration) while FA did not intensify root growth. Both HS and HA stimulated root hair formation. At low concentrations, HS and HA induced root hair formation near the root cap, a typical hormonal imbalance effect in plants. Transgenic tomato with reporter gene DR5::GUS allowed the observation that the auxin-related signalling pathway was involved in root growth promotion by HA.

Cette étude envisage le film de Gus Van Sant à partir des théories de l'énonciation narrative, rediscutées à travers cet exemple singulier où le spectateur est convié - du moins en apparence - à adopter successivement, grâce à une « caméra subjective » dont on sonde toute l'ambiguïté, le point de vue de différents personnages. Ce contexte théorique permet de dévoiler certains mécanismes de l'organisation du film et les raisons de l'enraiement de l'identification aux figures de l'écran, fantômes qui semblent s'effacer devant une présence appuyée de la caméra rappelant qu'il s'agit avant tout d'un regard porté sur un fait divers, non d'une explication basée sur des phénomènes d'intériorisation psychologique - Elephant trouve à cet égard, comme l'auteur le montre ici, une position intermédiaire entre le film homonyme réalisé par Alan Clarke en 1989 pour la BBC et Bowling for Columbine de Michael Moore (2002). On y envisage la démarche du cinéaste de façon concrète, comme une manière d'arpenter un espace en prenant des personnages en « filature ».

Huntington's disease is an inherited neurodegenerative disease that causes motor, cognitive and psychiatric impairment, including an early decline in ability to recognize emotional states in others. The pathophysiology underlying the earliest manifestations of the disease is not fully understood; the objective of our study was to clarify this. We used functional magnetic resonance imaging to investigate changes in brain mechanisms of emotion recognition in pre-manifest carriers of the abnormal Huntington's disease gene (subjects with pre-manifest Huntington's disease): 16 subjects with pre-manifest Huntington's disease and 14 control subjects underwent 1.5 tesla magnetic resonance scanning while viewing pictures of facial expressions from the Ekman and Friesen series. Disgust, anger and happiness were chosen as emotions of interest. Disgust is the emotion in which recognition deficits have most commonly been detected in Huntington's disease; anger is the emotion in which impaired recognition was detected in the largest behavioural study of emotion recognition in pre-manifest Huntington's disease to date; and happiness is a positive emotion to contrast with disgust and anger. Ekman facial expressions were also used to quantify emotion recognition accuracy outside the scanner and structural magnetic resonance imaging with voxel-based morphometry was used to assess the relationship between emotion recognition accuracy and regional grey matter volume. Emotion processing in pre-manifest Huntington's disease was associated with reduced neural activity for all three emotions in partially separable functional networks. Furthermore, the Huntington's disease-associated modulation of disgust and happiness processing was negatively correlated with genetic markers of pre-manifest disease progression in distributed, largely extrastriatal networks. The modulated disgust network included insulae, cingulate cortices, pre- and postcentral gyri, precunei, cunei, bilateral putamena, right pallidum, right thalamus, cerebellum, middle frontal, middle occipital, right superior and left inferior temporal gyri, and left superior parietal lobule. The modulated happiness network included postcentral gyri, left caudate, right cingulate cortex, right superior and inferior parietal lobules, and right superior frontal, middle temporal, middle occipital and precentral gyri. These effects were not driven merely by striatal dysfunction. We did not find equivalent associations between brain structure and emotion recognition, and the pre-manifest Huntington's disease cohort did not have a behavioural deficit in out-of-scanner emotion recognition relative to controls. In addition, we found increased neural activity in the pre-manifest subjects in response to all three emotions in frontal regions, predominantly in the middle frontal gyri. Overall, these findings suggest that pathophysiological effects of Huntington's disease may precede the development of overt clinical symptoms and detectable cerebral atrophy.

March 21, 2012 THIS WEEK: Quick Work at Carlisle(From the Indianola Tribune, December 20, 1877) BACKGROUND: Lewis Todhunter was born in Fayette County, Ohio, April 6, 1817. He was admitted to the bar in Highland County, Ohio, in 1848. Todhunter came to Iowa in 1850, settling along the Des Moines River in Polk County, where he was engaged in farming and selling goods. In 1854, he moved to Indianola and began the practice of law, continuing in that profession for more than a quarter of a century. He served as prosecuting attorney, county auditor, treasurer, and mayor of the city. He was a member of the third constitutional convention, which met at Iowa City in 1857, representing Warren, Madison, Adair, and Cass counties in that body and had the honor of assisting in making the first laws of the state. Todhunter served in the army during the Civil War from 1863-1865 as assistant quartermaster with rank of Captain. He is perhaps widest known, however, as a temperance leader and earnest worker. He joined in the Washingtonian movement in 1840 and was active in temperance reformation for the rest of his life.

This article describes the application of a recently developed general unknown screening (GUS) strategy based on LC coupled to a hybrid linear IT-triple quadrupole mass spectrometer (LC-MS/MS-LIT) for the simultaneous detection and identification of drug metabolites following in vitro incubation with human liver microsomes. The histamine H1 receptor antagonist loratadine was chosen as a model compound to demonstrate the interest of such approach, because of its previously described complex and extensive metabolism. Detection and mass spectral characterization were based on data-dependent acquisition, switching between a survey scan acquired in the ion-trapping Q3 scan mode with dynamic subtraction of background noise, and a dependent scan in the ion-trapping product ion scan mode of automatically selected parent ions. In addition, the MS(3) mode was used in a second step to confirm the structure of a few fragment ions. The sensitivity of the ion-trapping modes combined with the selectivity of the triple quadrupole modes allowed, with only one injection, the detection and identification of 17 phase I metabolites of loratadine. The GUS procedure used in this study may be applicable as a generic technique for the characterization of drug metabolites after in vitro incubation, as well as probably in vivo experiments.

Before the Iowa Department of Transportation (DOT) was established by legislation in July 1974, there were several state agencies that handled the tasks that are now the responsibility of an integrated, multimodal Iowa DOT. Among those agencies was the Iowa State Highway Commission (IHC). You are invited to read a brief history of the Iowa DOT here:http://www.iowadot.gov/about/organizationalhistory.htm The IHC operated as an independent state agency between 1913 and 1974. In 1968, the IHC created and released This is YOUR Highway Commission, a 24 ½- minute film that showcased the responsibilities and functions of the IHC. The narrator describes the activities of various offices and employees, and explains how those activities benefited Iowa’s citizens and motorists. The film journeys through all areas of IHC responsibility to Iowa’s roadways, including administration, planning, design, bidding, right of way, materials, construction, maintenance and facilities. As part of the Iowa DOT’s effort to preserve and archive its historical resources, the original 16mm film was professionally cleaned, restored and digitized so that it could be made available via this website. The Iowa DOT is currently researching and compiling information necessary to prepare detailed biographies of the IHC employees identified in the film. Included in each biography will be still frames taken from the film, as well as other images from the Iowa DOT’s archives. This more comprehensive description of the film will be available in the future. In the meantime, below is a list of the IHC employees who have been identified. The list is arranged in the order in which each employee first appears in the film. The development and optimization of efficient transformation protocols is essential in new citrus breeding programs, not only for rootstock, but also for scion improvement. Transgenic 'Hamlin' sweet orange (Citrus sinensis (L.) Osbeck) plants were obtained by Agrobacterium tumefaciens-mediated transformation of epicotyl segments collected from seedlings germinated in vitro. Factors influencing genetic transformation efficiency were evaluated including seedling incubation conditions, time of inoculation with Agrobacterium and co-culture conditions. Epicotyl segments were adequate explants for transformation, regenerating plants by direct organogenesis. Higher percentage of transformation was obtained with explants collected from seedlings germinated in darkness, transferred to 16 hours photoperiod for 2-3 weeks, and inoculated with Agrobacterium for 15-45 min. The best co-culture condition was the incubation of the explants in darkness, for three days in culture medium supplemented with 100 muM of acetosyringone. Genetic transformation was confirmed by performing beta-glucoronidase (GUS) assays and, subsequently, by PCR amplification for the nptII and GUS genes.

O objetivo deste trabalho foi avaliar a dispersão de pólen transgênico em soja. Plantas transgênicas de soja contendo os genes ahas, para tolerância ao herbicida imazapyr, e uidA (GUS), foram cultivadas com plantas não-transgênicas. A dispersão do pólen transgênico foi avaliada pela presença de ambos os genes dominantes na progênie de plantas não-transgênicas. A maior freqüência de disseminação de pólen transgênico foi observada na primeira linha, distante 0,5 m da parcela central (0,44% a 0,45%). Esta freqüência foi reduzida drasticamente na linha 2 (0,04% a 0,14%), atingindo 0 na linha 13, a 6,5 m da parcela central.

Els objectius de l’estudi realitzat en aquest treball són: comprovar que l’educació artística també pot ser una eina d’inclusió a l’escola, conèixer els beneficis de l’experimentació/exploració en l’art, i observar com aquests influeixen en l’aprenentatge dels infants, i finalment veure els beneficis del treball cooperatiu com a mètode d’experimentació en l’art. L’estudi s’ha realitzat a partir d’ una recerca teòrica i duent a terme una part pràctica de vàries sessions amb la col·laboració dels infants de P4 de l’escola Joan XXIII d’Hostalets de Balenyà. La pràctica s’ha anomenat “Art amb pedres”, que com el seu nom indica, les pedres eren l’element natural que han utilitzat els infants per a convertir-se en verdaders artistes!

O objetivo deste trabalho foi apresentar uma modelagem da distribuição ambiental de vinte e nove poluentes orgânicos encontrados em amostras de lodos de esgoto das estações de tratamento de esgoto de Barueri e Suzano, SP. Lodos de esgoto são misturas de produtos orgânicos e inorgânicos gerados nos processos primários e secundários de tratamento que, mediante adequado controle de qualidade, podem ser reutilizados como fertilizantes agrícolas. O modelo de fugacidade nível I foi aplicado a um sistema compartimental hipotético constituído de ar, água, solo, sedimento, biota aquática e plantas. Foi utilizada a massa molecular, a pressão de vapor, a solubilidade em água, a constante de Henry, o coeficiente de partição octanol-água e a meia-vida no solo de cada um dos poluentes. O coeficiente de sorção no solo, os fatores de bioconcentração em organismos aquáticos, raízes e na seiva do xilema de plantas foram calculados por meio de expressões que correlacionam cada um desses parâmetros com o coeficiente de partição octanol-água do poluente. Foram estimados e usados os coeficientes de partição folha-ar e ar-água e o índice GUS de cada poluente. A modelagem e os cálculos dos fatores de bioconcentração e dos coeficientes de partição revelam os compartimentos preferenciais dos poluentes.

A plant regeneration method with cell suspension cultures of banana, and the effect of biobalistic on regeneration potential are described in this report. Somatic embryos of banana were obtained from indirect embryogenesis of male inflorescence of banana cultivar Maçã (AAB group). Part of the calluses formed (40%) showed embryogenic characteristics (nonfriable, compact and yellow color). The cell suspension, originated from embryogenic calluses, contained clusters of small tightly packed cells with dense cytoplasms, relatively large nuclei and very dense nucleoli. After four months of culture, somatic embryos started to regenerate. The maximum number of regenerated plants was observed between 45 and 60 days after embryo formation.In the first experiment, 401 plants were regenerated from approximately 10 mL of packed cells. In the second experiment, 399 plants were regenerated from a cell suspension six months older than that of the first experiment. Cell transformation using particle bombardment with three different plasmid constructions, containing the uid-A gene, resulted in a strong GUS expression five days after bombardment; however, plant regeneration from bombarded cells was much lower than nonbombarded ones.

O objetivo deste trabalho foi estabelecer sistemas de multiplicação de plantas de cana-de-açúcar in vitro e avaliar sua utilização, como material inicial, para a indução de regeneração a partir de ápices caulinares. Três métodos de cultivo foram avaliados: cultura em meio semi-sólido, cultura líquida estacionária e cultura líquida sob agitação. A taxa de multiplicação mais elevada foi alcançada por meio da cultura líquida sob agitação. Ápices caulinares, excisados dessas plantas, apresentaram taxas de regeneração in vitro compatíveis com sua utilização em protocolos de transformação. Calos resistentes a PPT e GUS-positivos foram obtidos de explantes da variedade Chunnee com inoculação de Agrobacterium tumefaciens C58C1 (pMP90) (pDUBarA9). O protocolo estabelecido a partir de cultivo in vitro pode ser utilizado para a produção de plantas transgênicas de cana-de-açúcar, visando à realização de estudos de regulação da expressão gênica, assim como à introdução de características de interesse agronômico.

Sabkha and deep burial set tings are the most com mon sites where diagenetic anhydrite forms. In a sabkha setting, displacive facies (iso lated nodules, bed ded nodules, enterolithic levels) of early diagenetic or primary anhydrite are generated (Shearman, 1966; Hardie, 1967). These anhydrite facies are commonly foundat the top of shoal ing cycles representing the evolution from subaqueous depositional conditions at the base (carbonates, lutites) to exposure conditions at the top where in ter stitially-grown gypsum/anhydrite de velops ( sabkha cy cles). In a deep burial setting, gypsum transforms to tally to anhydrite with in creas ing temperature and lithostatic pressure (Murray, 1964). Al though this mineral transformation usually preserves the depositional gypsum facies, a significant textural change is in volved in other cases, resulting in replacive anhydrite with a nodular-mosaic or"chicken-wire" fabric (Warren, 2006). In the two settings, how ever, the size of the individual anhydrite nodules is relatively small, rarely reaching some tens of centimetres across. More over, bedding is preserved or little disturbed, al though minor de formation is caused by the displacive sabkha nodules.
