Objectives We have investigated the effects of a multi–species probiotic preparation containing a combination of probiotic bacterial genera that included Bifidobacteria, Lactobacilli and a Streptococcus in a mouse model of high fat diet/obesity induced liver steatosis. Methods Three groups of C57B1/6J mice were fed either a standard chow or a high fat diet for 20 weeks, while a third group was fed a high fat diet for 10 weeks and then concomitantly administered probiotics for a further 10 weeks. Serum, liver and large bowel samples were collected for analysis. Results The expression of the tight junction proteins ZO-1 and ZO-2 was reduced (p < 0.05) in high fat diet fed mice compared to chow fed mice. Probiotic supplementation helped to maintain tight ZO-1 and ZO-2 expression compared with the high fat diet group (p < 0.05), but did not restore ZO-1 or ZO-2 expression compared with chow fed mice. Mice fed a high fat diet ± probiotics had significant steatosis development compared to chow fed mice (p < 0.05); steatosis was less severe in the probiotics group compared to the high fat diet group. Hepatic triglycerides concentration was higher in mice fed a high fat diet ± probiotics compared to the chow group (p < 0.05), and was lower in the probiotics group compared to the high fat diet group (p < 0.05). Compared to chow fed mice, serum glucose and cholesterol concentrations, and the activity of alanine transaminase were higher (p < 0.05), whereas serum triglyceride concentration was lower (p < 0.05) in mice fed a high fat diet ± probiotics. Conclusions Supplementation with a multi-species probiotic formulation helped to maintain tight junction proteins ZO-1 and ZO-2, and reduced hepatic triglyceride concentrations compared with a HFD alone.

Background The incidence of obesity amongst patients presenting for elective Total Hip Arthroplasty (THA) has increased in the last decade and the relationship between obesity and the need for joint replacement has been demonstrated. This study evaluates the effects of morbid obesity on outcomes following primary THA by comparing short-term outcomes in THA between a morbidly obese (BMI ≥40) and a normal weight (BMI 18.5 - <25) cohort at our institution between January 2003 and December 2010. Methods Thirty-nine patients included in the morbidly obese group were compared with 186 in the normal weight group. Operative time, length of stay, complications, readmission and length of readmission were compared. Results Operative time was increased in the morbidly obese group at 122 minutes compared with 100 minutes (p=0.002). Post-operatively there was an increased 30-day readmission rate related to surgery of 12.8% associated with BMI ≥40 compared with 2.7% (p= 0.005) as well as a 5.1 fold increase in surgery related readmitted bed days - 0.32 bed days per patient for normal weight compared with 1.64 per patient for the morbidly obese (p=0.026). Conclusion Morbidly obese patients present a technical challenge and likely this and the resultant complications are underestimated. More work needs to be performed in order to enable suitable allocation of resources.

Uveal melanoma (UM) is the second most common primary intraocular cancer worldwide. It is a relatively rare cancer, but still the second most common type of primary malignant melanoma in humans. UM is a slowly growing tumor, and gives rise to distant metastasis mainly to the liver via the bloodstream. About 40% of patients with UM die of metastatic disease within 10 years of diagnosis, irrespective of the type of treatment. During the last decade, two main lines of research have aimed to achieve enhanced understanding of the metastasis process and accurate prognosis of patients with UM. One emphasizes the characteristics of tumor cells, particularly their nucleoli, and markers of proliferation, and the other the characteristics of tumor blood vessels. Of several morphometric measurements, the mean diameter of the ten largest nucleoli (MLN) has become the most widely applied. A large MLN has consistently been associated with high likelihood of dying from UM. Blood vessels are of paramount importance in metastasis of UM. Different extravascular matrix patterns can be seen in UM, like loops and networks. This presence is associated with death from metastatic melanoma. However, the density of microvessels is also of prognostic importance. This study was undertaken to help understanding some histopathological factors which might contribute to developing metastasis in UM patients. Factors which could be related to tumor progression to metastasis disease, namely nucleolar size, MLN, microvascular density (MVD), cell proliferation, and The Insulin-like Growth Factor 1 Receptor(IGF-1R), were investigated. The primary aim of this thesis was to study the relationship between prognostic factors such as tumor cell nucleolar size, proliferation, extravascular matrix patterns, and dissemination of UM, and to assess to what extent there is a relationship to metastasis. The secondary goal was to develop a multivariate model which includes MLN and cell proliferation in addition to MVD, and which would fit better with population-based, melanoma-related survival data than previous models. I studied 167 patients with UM, who developed metastasis even after a very long time following removal of the eye, metastatic disease was the main cause of death, as documented in the Finnish Cancer Registry and on death certificates. Using an independent population-based data set, it was confirmed that MLN and extravascular matrix loops and networks were unrelated, independent predictors of survival in UM. Also, it has been found that multivariate models including MVD in addition to MLN fitted significantly better with survival data than models which excluded MVD. This supports the idea that both the characteristics of the blood vessels and the cells are important, and the future direction would be to look for the gene expression profile, whether it is associated more with MVD or MLN. The former relates to the host response to the tumor and may not be as tightly associated with the gene expression profile, yet most likely involved in the process of hematogenous metastasis. Because fresh tumor material is needed for reliable genetic analysis, such analysis could not be performed Although noninvasive detection of certain extravascular matrix patterns is now technically possible,in managing patients with UM, this study and tumor genetics suggest that such noninvasive methods will not fully capture the process of clinical metastasis. Progress in resection and biopsy techniques is likely in the near future to result in fresh material for the ophthalmic pathologist to correlate angiographic data, histopathological characteristics such as MLN, and genetic data. This study supported the theory that tumors containing epithelioid cells grow faster and have poorer prognosis when studied by cell proliferation in UM based on Ki-67 immunoreactivity. Cell proliferation index fitted best with the survival data when combined with MVD, MLN, and presence of epithelioid cells. Analogous with the finding that high MVD in primary UM is associated with shorter time to metastasis than low MVD, high MVD in hepatic metastasis tends to be associated with shorter survival after diagnosis of metastasis. Because the liver is the main organ for metastasis from UM, growth factors largely produced in the liver hepatocyte growth factor, epidermal growth factor and insulin-like growth factor-1 (IGF-1) together with their receptors may have a role in the homing and survival of metastatic cells. Therefore the association between immunoreactivity for IGF-1R in primary UM and metastatic death was studied. It was found that immunoreactivity for IGF-IR did not independently predict metastasis from primary UM in my series.

A protease inhibitor from the seeds of Butea monosperma (BmPI) was purified, characterized and studied for its influence on developmental physiology of Helicover-pa armigera. BmPI on two-dimensional separations indicated the presence of a 14 kDa protein with an isoelectric point in the acidic region (pl 5.6). Multiple Sequence Analysis data suggested that the BmPI contains a sequence motif which is conserved in various trypsin and chymotrypsin inhibitors of Kunitz-type. The inhibitor exhibited trypsin inhibitory activity in a broad range of pH (4-10) and temperature (10-80 degrees C). The enzyme kinetic studies revealed BmPI as a competitive inhibitor with a K-i value of 1.2 x 10(-9) M. In vitro studies with BmPI indicated measurable inhibitory activity on total gut proteolytic enzymes of H. armigera (IC(50)2.0 mu g/ml) and bovine trypsin. BmPI supplemented artificial diet caused dose dependent mortality and reduction in growth and weight. The fertility and fecundity of H. armigera, declined whereas the larval-pupal duration of the insect life cycle extended. These detrimental effects on H. armigera suggest the usefulness of BmPl in insect pest management of food crops. (C) 2014 Elsevier Ltd. All rights reserved.

La presente investigación se llevó a efecto en el Centro Nacional de Investigación Agropecuaria (CNIA- INTA), ubicado en el Km. 14 de la carretera norte, 2 Km. al sur en el municipio de Managua, departamento de Managua. El ensayo se estableció en la época de postrera del 2001, en suelos de origen volcánico y textura franco arenosa. El propósito del experimento fue contribuir a la obtención de líneas de sorgo [Sorghum bicolor (L.) Moench) resistentes y/o tolerantes a plagas y enfermedades. Los materiales utilizados (provenientes de la Universidad de Texas A & M) son parte de un ADIN (AII Disease and lnsect Nursery). La parcela experimental tuvo un tamaño de 3.75 m2 que constó de 1 surco de 5 metros lineales (1 línea por surco). El ensayo se estableció utilizando la metodología de la Universidad de Texas A & M con 20 tratamientos (líneas) y 2 repeticiones. Las variables evaluadas fueron: a) Daño foliar causado por el gusano cogollero (Spodoptera frugiperda J. E. Smith), b) Días a floración, e) Número de mosquitas (Stenodipfosis sorghicola Coquillett) por panoja, d) Número de parasitoides de mosquita (Aprostocetus diplosidis Crawford) por panoja, y e) Severidad de las enfermedades foliares. Para la primera y la última variable se utilizó la escala de daño propuesta por la Universidad de Texas A & M, a estos datos se les efectuó un análisis estadístico descriptivo. Para las variables b, e y d se realizó un análisis de varianza y prueba de rangos múltiples de Duncan (P:s:0.05), además se les efectuó un análisis de correlación parcial. En cuanto al daño foliar causado por el gusano cogollero (Spodoptera frugiperda J. E. Smith), no existen diferencias en el comportamiento de las líneas. Este aspecto no representó una incidencia relevante. Con respecto al período de floración, la línea MB1088 presentó el mayor número de dias a floración, en tanto que la línea Tx2911/9281941 presentó el menor número de días a floración. Los resultados obtenidos indican que el comportamiento de las 20 líneas de sorgo [Sorghum bicolor (L.) Moench] ante el ataque de mosquita (Stenodiplosis sorghico/a Coquillett) y la incidencia de su parasitoide (Aprostocetus diplosidis Crawford) es similar para cada línea; la línea R.9618 fue la que presentó el mayor valor con respecto a esta variable, mientras que la línea que presentó el menor número de mosquitas por panoja fue Tx7078; Existe relación entre la variable número de mosquitas (Stenodip/osis sorghico/a Coquillett) por panoja y días a floración y esta relación es afectada por el parasitoide de la mosquita (Aprostocetus diplosidis Crawford). Puede identificarse que la línea Tx2783 presentó el menor porcentaje de severidad por enfermedades foliares (12.5%) mientras que el mayor porcentaje de severidad por enfermedades foliares lo obtuvo la línea R.9618 (65.12%). De las 6 enfermedades que se presentaron en el ensayo, la más predominante fue antracnosis (Collectotrichum graminicola G. W. Wilson).

El ensayo se estableció utilizando la metodología de la Universidad de Texas A & M con19 tratamientos (líneas) y 2 repeticiones. Las variables evaluadas fueron: a) daño foliar causado por el gusano cogollero ( Spodoptera frugiperda J. E. Smith ), b) días a floración, c) número de mosquitas d el sorgo ( Stenodiplosis sorghicola Coquillett ) por panoja, d) número de parasitoides de mosquita del sorgo ( Aprostocetus diplosidis Crawford) por panoja, e) severidad de las enfermedades foliares y vasculares. Para las variables b, c y d se realizó un análisis de varianza y prueba de rangos múltiples de Duncan (P ≤ 0.05), además se les efectuó un análisis de correlación de Pearson. En cuanto al daño foliar causado por el gusano cogollero ( Spodoptera frugiperda J. E. Smith ), no existieron diferencias en el comportamiento de las líneas. Este aspecto no representó una incidencia relevante. Con respecto al período de floración, la línea 96CA5986 presentó el mayor número de días a floración, en tanto que la línea Tx2880 presentó el menor número de días a floración. Los resultados obtenidos indican que el comportamiento de las 19 líneas de sorgo [ Sorghum bicolor ( L.) Moench ] ante el ataque de mosquita del sorgo ( Stenodiplosis sorghicola Coquillett ) y la incidencia de su parasitoide ( Aprostocetus diplosidis Crawford ) es similar para cada línea; La línea 98BRON122 fue la que presentó el mayor valor con respecto a esta variable, mientras que la línea que presentó el menor número de mosquitas por panoja fue B9104; Existe relación entre la variable número de mosquitas ( Stenodiplosis sorghicola Coquillett ) por panoja y días a floración y esta relación es afectada por el parasitoide de la mosquita ( Aprostocetus diplosidis Crawford ). Puede identificarse que la Linea 87EO366*9EO328 fue la que presentó la mayor severidad de enfermedades foliares tales como antracnosis ( Colletotrichum graminicola G. W. Wilson ), mancha zonada ( Gloeocercospora sorghi D.Bain.& Edgerton ex Deighton) , mancha gris de la hoja (Cercospora sorghi Ellis y Everth) tizón de la hoja ( Exserohilum turcicum (pass.). La línea que presentó la menor severidad fue 94B1055. La enfermedad que presentó mayor severidad en el ensayo fue Antracnosis.

Carfentrazone-ethyl (CE) is a reduced risk herbicide that is currently being evaluated for the control of aquatic weeds. Greenhouse trials were conducted to determine efficacy of CE on water hyacinth ( Eichhornia crassipes (Mart.) Solms- Laub.), water lettuce ( Pistia stratiotes L.), salvinia ( Salvinia minima Baker) and landoltia (Landoltia punctata (G. Mey.) Les & D. J. Crawford ) . CE controlled water lettuce, water hyacinth and salvinia at rates less than the maximum proposed use rate of 224 g ha -1 . Water lettuce was the most susceptible to CE with an EC 90 of 26.9 and 33.0 g ha -1 in two separate trials. Water hyacinth EC 90 values were calculated to be 86.2 to 116.3 g ha -1 , and salvinia had a similar susceptibility to water hyacinth with an EC 90 of 79.1 g ha -1 . Landoltia was not adequately controlled at the rates evaluated. In addition, CE was applied to one-half of a 0.08 ha pond located in North Central, Florida to determine dissipation rates in water and hydrosoil when applied at an equivalent rate of 224 g ha -1 . The half-life of CE plus the primary metabolite, CE-chloropropionic acid, was calculated to be 83.0 h from the whole pond, and no residues were detected in water above the limit of quantification (5 μg L -1 ) 168 h after treatment. CE dissipated rapidly from the water column, did not occur in the sediment above the levels of quantification, and in greenhouse studies effectively controlled three species of aquatic weeds at relatively low rates.(PDF contains 6 pages.)

The Tanzania Coastal Management Partnership (TCMP) works to implement the National Integrated Coastal Environmental Management Strategy (ICEMS) in Tanzania’s coastal landscapes and seascapes, funded in large measure by the U.S. Agency for International Development. The overarching goal of the Sustainable Coastal Communities and Ecosystems in Tanzania (SUCCESS Tanzania) initiative is to conserve coastal and marine biodiversity while improving the well being of coastal residents through implementation of the Tanzania ICEMS and related ICM policies and strategies. It does this by focusing on three key results: -Policies and Laws that Integrate Conservation and Development Applied -Participatory Landscape Scale Conservation Practiced -Conservation Enterprises Generate Increased and Equitable Benefits from Sustainable Use An additional result sought in the program is gender equity and HIV/AIDS preventive behaviors promoted through communicating HIV/AIDS, environment, and equity messages. Northern Ireland has approximately 1670 lakes, which cover 4.4% of the land surface. However, most of the water area is accounted for by the large lakes such as Lough Neagh (385 km2) and Lower Lough Erne (109.5 km ). The majority of lakes are less than 100 hectares in area. They tend to be distributed towards the south and west of the Province, where extensive drumlin swarms are rich in small waterbodies. In 1988-1991, 610 of the 708 lakes between one and 100 hectares were sampled by the Northern Ireland Lake Survey. The objective was to assess their conservation status based on their aquatic macrophyte flora, but in addition to extensive plant surveys, the water of each lake was analysed for a range of chemical variables. This article reports on a full-scale survey carried out in early March 2002. The survey was taken with help of two helicopters. The authorise summarise the results of the chemical analysis of the survey.

Part I. Proton Magnetic Resonance of Polynucleotides and Transfer RNA.
Proton magnetic resonance was used to follow the temperature dependent intramolecular stacking of the bases in the polynucleotides of adenine and cytosine. Analysis of the results on the basis of a two state stacked-unstacked model yielded values of -4.5 kcal/mole and -9.5 kcal/mole for the enthalpies of stacking in polyadenylic and polycytidylic acid, respectively.
The interaction of purine with these molecules was also studied by pmr. Analysis of these results and the comparison of the thermal unstacking of polynucleotides and short chain nucleotides indicates that the bases contained in stacks within the long chain poly nucleotides are, on the average, closer together than the bases contained in stacks in the short chain nucleotides.
Temperature and purine studies were also carried out with an aqueous solution of formylmethionine transfer ribonucleic acid. Comparison of these results with the results of similar experiments with the homopolynucleotides of adenine, cytosine and uracil indicate that the purine is probably intercalating into loop regions of the molecule.
The solvent denaturation of phenylalanine transfer ribonucleic acid was followed by pmr. In a solvent mixture containing 83 volume per cent dimethylsulf oxide and 17 per cent deuterium oxide, the tRNA molecule is rendered quite flexible. It is possible to resolve resonances of protons on the common bases and on certain modified bases.
Part II. Electron Spin Relaxation Studies of Manganese (II) Complexes in Acetonitrile.
The electron paramagnetic resonance spectra of three Mn+2 complexes, [Mn(CH3CN)6]+2, [MnCl4]-2, and [MnBr4]-2, in acetonitrile were studied in detail. The objective of this study was to relate changes in the effective spin Hamiltonian parameters and the resonance line widths to the structure of these molecular complexes as well as to dynamical processes in solution.
Of the three systems studied, the results obtained from the [Mn(CH3CN)6]+2 system were the most straight-forward to interpret. Resonance broadening attributable to manganese spin-spin dipolar interactions was observed as the manganese concentration was increased.
In the [MnCl4]-2 system, solvent fluctuations and dynamical ion-pairing appear to be significant in determining electron spin relaxation.
In the [MnBr4]-2 system, solvent fluctuations, ion-pairing, and Br- ligand exchange provide the principal means of electron spin relaxation. It was also found that the spin relaxation in this system is dependent upon the field strength and is directly related to the manganese concentration. A relaxation theory based on a two state collisional model was developed to account for the observed behavior.

We tested the hypothesis that larger juvenile sockeye salmon (Oncorhynchus nerka) in Bristol Bay, Alaska, have higher marine-stage survival rates than smaller juvenile salmon. We used scales from returning adults (33 years of data) and trawl samples of juveniles (n= 3572) collected along the eastern Bering Sea shelf during August through September 2000−02. The size of juvenile sockeye salmon mirrored indices of their marine-stage survival rate (e.g., smaller fish had lower indices of marine-stage survival rate). However, there was no relationship between the size of sockeye salmon after their first year at sea, as estimated from archived scales, and brood-year survival size was relatively uniform over the time series, possibly indicating size-selective mortality on smaller individuals during their marine residence. Variation in size, relative abundance, and marine-stage survival rate of juvenile sockeye salmon is likely related to ocean conditions affecting their early marine migratory pathways along the eastern Bering Sea shelf.
