This work describes the evaluation of several parameters for the preparation of a tuna fish candidate as a reference material (RM) in order to measure the total As mass fraction by slurry sampling graphite furnace atomic absorption spectrometry (SLS-GF AAS) and slurry sampling hydride generation atomic absorption spectrometry (SLS-HG AAS). The main parameters investigated were the homogeneity, analyte segregation and composition during material production. For candidate RM preparation, tuna fish was collected at a local market, cleaned, freeze-dried and treated using different procedures as follows: (1) ground in a cutting mill and separated in different particle sizes (2) ground in cryogenic mill. The mass fraction of As in the cryogenically ground sample was (4.77 +/- A 0.19) mu g g(-1) for SLS-GF AAS and (4.61 +/- A 0.34) mu g g(-1) for SLS-HG AAS. The accuracy of the procedures was checked with tuna fish certified reference material (BCR 627) with recoveries of 102 and 94% for SLS-GF AAS and SLS-HG AAS, respectively. The homogeneity factor was calculated for different pretreatment procedures and for particle sizes in the range of 500-150 mu g, indicating good homogeneity, except for raw fish. There was no observed analyte segregation and no losses, no contamination and no changes in the microdistribution of material during preparation.

Syftet med denna uppsats har varit att studera hur barnavårdsnämnden i Stora Tuna verkställde lagstiftningen om uppfostran åt vanartade och i sedligt avseende försummade barn. För att uppnå syftet med studien har barnavårdsnämndens ledamöter undersökts, lika så de anmälda barnen. Vad anmäldes de för, vilka blev konsekvenserna och vilka var det som stod för anmälningarna? Resultaten jämförs med tidigare forskning från andra platser. Huvudsakligen har protokoll från barnavårdsnämndens sammanträden mellan 1903 och 1917 använts för att besvara frågeställningarna.Resultatet visar att en klar majoritet av nämndens ledamöter representerade medelklassen eller överklassen, endast ett fåtal kom från arbetarklassen. Men de barn som anmäldes kom nästan uteslutande från arbetarklassen. Detta är något som den tidigare forskningen också framhåller. Pojkar anmäldes i betydligt högre utsträckning än flickor, men de flickor som anmäldes dömdes betydligt hårdare än pojkarna. Flickorna anmäldes främst på grund av de bristande hemförhållanden och påföljderna blev oftast att de skiljdes från hemmet till fosterhem eller till barnhem. Pojkarna anmäldes främst för stöld och den vanligaste påföljden blev en allvarlig varning. Polisen var den grupp som anmälde flest barn till nämnden.

[EN] The information provided by the International Commission for the Conservation of Atlantic Tunas (ICCAT) on captures of skipjack tuna (Katsuwonus pelamis) in the central-east Atlantic has a number of limitations, such as gaps in the statistics for certain fleets and the level of spatiotemporal detail at which catches are reported. As a result, the quality of these data and their effectiveness for providing management advice is limited. In order to reconstruct missing spatiotemporal data of catches, the present study uses Data INterpolating Empirical Orthogonal Functions (DINEOF), a technique for missing data reconstruction, applied here for the first time to fisheries data. DINEOF is based on an Empirical Orthogonal Functions decomposition performed with a Lanczos method. DINEOF was tested with different amounts of missing data, intentionally removing values from 3.4% to 95.2% of data loss, and then compared with the same data set with no missing data. These validation analyses show that DINEOF is a reliable methodological approach of data reconstruction for the purposes of fishery management advice, even when the amount of missing data is very high.

Canned tuna is one of the most widespread and recognizable fish commodities in the world. Over all oceans 80% of the total tuna catches are caught by purse seine fishery and in tropical waters their target species are: yellowfin (Thunnus albacares), bigeye (Thunnus obesus) and skipjack (Katsuwonus pelamis). Even if this fishing gear is claimed to be very selective, there are high levels of by-catch especially when operating under Fish Aggregating Devices (FADs). The main problem is underestimation of by-catch data. In order to solve this problem the scientific community has developed many specific programs (e.g. Observe Program) to collect data about both target species and by-catch with observers onboard. The purposes of this study are to estimate the quantity and composition of target species and by-catch by tuna purse seiner fishery operating in tropical waters and to underline a possible seasonal variability in the by-catch ratio (tunas versus by-catch). Data were collected with the French scientific program ”Observe” on board of the French tuna purse seiner “Via Avenir” during a fishing trip in the Gulf of Guinea (C-E Atlantic) from August to September 2012. Furthermore some by-catch specimens have been sampled to obtain more information about size class composition. In order to achieve those purposes we have shared our data with the French Institute of Research for the Development (IRD), which has data collected by observers onboard in the same study area. Yellowfin tuna results to be the main specie caught in all trips considered (around 71% of the total catches) especially on free swimming schools (FSC) sets. Instead skipjack tuna is the main specie caught under FADs. Different percentages of by-catch with the two fishing modes are observed: the by-catch incidence is higher on FADs sets (96.5% of total by-catch) than on FSC sets (3.5%) and the main category of by-catch is little-tuna (73%). When pooling data for both fishing sets used in purse seine fishery the overall by-catch/catch ratio is 5%, a lower level than in other fishing gears like long-lining and trawling.

Yellowfin tuna (Thunnus albacares, YFT, Bonnaterre 1788) is one of the most important market tuna species in the world. The high mortality of juveniles is in part caused by their bycatch. Indeed, if unregulated, it could permanently destabilize stocks health. For this reason investigating and better knowing the stock boundaries represent a crucial concern. Aim of this thesis was to preliminary investigate the YFT population structure within and between Atlantic and Pacific Oceans through the analysis of genetic variation at eight microsatellite loci and assess the occurrence of barriers to the gene flow between Oceans. For this propouse we collected 4 geographical samples coming from Atlantic and Pacific Ocean and selected a panel of 8 microsatellites loci developped by Antoni et al., (2014). Samples 71-2-Y and 77-2-Y, came from rispectively west central pacific ocean (WCPO) and east central pacific ocean (ECPO), instead samples 41-1-Y and 34-2-Y derive from west central atlantic ocean (WCAO) and east central atlantic ocean (ECAO). Total 160 specimens were analyzed (40 per sample) and were carried out several genetic information as allele frequencies, allele number, allelic richness, HWE (using He and Ho) and pairwise Fst genetic distance. Results obtained, may support the panmictic theory of this species, only one of pairwise Fst obtained is statistically significant (Fst= 0.00927; pV= 0.00218) between 41-1-Y and 71-2-Y samples. Results suggest low genetic differentiation and consequent high level of gene flow between Atlantic and Pacific populations. Furthermore, we performed an analysis of molecular taxonomy through the use of ATCO (the flaking region between ATPse6 and cytochrome oxidase subunit III genes mt DNA, to discriminate within the gener Thunnus two of the related species (Yellofin and bigeye tuna) according with their difficult recognition at certain size (<40 cm). ATCO analysis in this thesis, has provided strong discriminate evidence between the target species proving to be one of the most reliable genetic tools capable to indagate within the genus Thunnus. Thus, our study has provided useful information for possible use of this protocol for conservation plans and management of this fish stocks.

La tuna es una fruta presente principalmente en zonas áridas y semiáridas. Su aporte nutricional podría aprovecharse consumiéndola en forma directa, como así también a través de productos elaborados en forma artesanal. Dichos productos podrían incorporarse a la economía del mercado de consumo interno y eventualmente externo. Por tal motivo, se evalúa la posibilidad de desarrollar mermeladas con tres ecotipos de tuna de la misma variedad, considerando la aceptación del producto. Se trabajó con tunas (Opuntia ficus indica f. inerme), cultivadas en el Centro Regional de Investigaciones Científicas y Tecnológicas (CRICYT) ubicado en el Parque General San Martín, Capital, Mendoza. El objetivo general del trabajo estuvo dirigido a la elaboración de mermeladas de tres ecotipos de tuna: tuna roja o morada, tuna anaranjada o amarilla y tuna verde o blanca acorde a la legislación vigente. Los ítems específicos estuvieron conducidos hacia el desarrollo de una mermelada utilizando una fruta no convencional, para determinar el valor nutricional del mismo, y sus características fisicoquímicas, ampliar la producción de subproductos de tuna en la región árida y semiárida, para así finalmente evaluar el grado de aceptabilidad sensorial y comercial del producto. Se caracterizaron los tres ecotipos de tuna y sus respectivas mermeladas mediante determinaciones fisicoquímicas, se verificó el contenido nutricional y se realizaron los análisis microbiológicos de cada una de ellas. Dentro de las determinaciones fisicoquímicas de las mermeladas se incluyó K, P, Ca y vitamina C, teniendo en cuenta que se parte de una materia prima con buenas cantidades de estos minerales y vitamina C con respecto a otras frutas. Se llevaron a cabo pruebas de evaluación sensorial del producto terminado y se consultó el precio que estarían dispuestos a pagar los potenciales consumidores. Los jueces entrenados contribuyeron al diseño de un perfil sensorial para cada una de las mermeladas y evaluaron en forma global al producto obtenido. Los resultados obtenidos en cuanto a la caracterización de la materia prima arrojan un buen contenido en sólidos solubles, vitamina C y en minerales como el fósforo, calcio, potasio y un contenido bajo en sodio con respecto a otras frutas que se consumen habitualmente. Los análisis microbiológicos realizados para evaluar la conservación permiten expresar que se trata de un producto de bajo riesgo microbiológico, bajo las condiciones de conservación que se obtuvieron. En base a lo expuesto, se puede concluir que es técnicamente posible elaborar artesanalmente mermeladas de tres ecotipos de tuna, las cuales poseen un buen aporte nutricional, son aptas para el consumo humano desde el punto de vista bromatológico y aceptadas sensorialmente por los consumidores.

The development of the ecosystem approach and models for the management of ocean marine resources requires easy access to standard validated datasets of historical catch data for the main exploited species. They are used to measure the impact of biomass removal by fisheries and to evaluate the models skills, while the use of standard dataset facilitates models inter-comparison. North Atlantic albacore tuna is exploited all year round by longline and in summer and autumn by surface fisheries and fishery statistics compiled by the International Commission for the Conservation of Atlantic Tunas (ICCAT). Catch and effort with geographical coordinates at monthly spatial resolution of 1° or 5° squares were extracted for this species with a careful definition of fisheries and data screening. In total, thirteen fisheries were defined for the period 1956-2010, with fishing gears longline, troll, mid-water trawl and bait fishing. However, the spatialized catch effort data available in ICCAT database represent a fraction of the entire total catch. Length frequencies of catch were also extracted according to the definition of fisheries above for the period 1956-2010 with a quarterly temporal resolution and spatial resolutions varying from 1°x 1° to 10°x 20°. The resolution used to measure the fish also varies with size-bins of 1, 2 or 5 cm (Fork Length). The screening of data allowed detecting inconsistencies with a relatively large number of samples larger than 150 cm while all studies on the growth of albacore suggest that fish rarely grow up over 130 cm. Therefore, a threshold value of 130 cm has been arbitrarily fixed and all length frequency data above this value removed from the original data set.

North Atlantic albacore tuna is exploited all year round by longline and in summer and autumn by surface fisheries and fishery statistics compiled by the International Commission for the Conservation of Atlantic Tunas (ICCAT). Catch and effort with geographical coordinates at monthly spatial resolution of 1° or 5° squares were extracted for this species with a careful definition of fisheries and data screening. Length frequencies of catch were also extracted according to the definition of fisheries for the period 1956-2010. Using these data, an application of the spatial ecosystem and population dynamics model (SEAPODYM) was developed for the North Atlantic albacore population and fisheries and provided the first spatially explicit estimate of albacore density in the North Atlantic by life stage. These densities by life stage (larval recruits, young immature fish adult mature fish and total biomass) are provided in gridded file (Netcdf) at resolution of 2° x 2° x month.
